1.Antiproliferative effect of curcumin on human breast cancer of MCF-7 cells

Objective To investigate the effect of curcumin on the proliferation,cell cycle distribution and gene expression of human breast cancer MCF-7 cells.Methods MCF-7 cells were treated with 10,20,40,80 ?mol/L curcumin for 24,48 or 72 h.MTT was used to assay the biologic activities of curcumin and its antiproliferative effect.FCM was used to detect cell cycle distribution and apoptosis.The expression levels of bcl-2,bax,PCNA mRNA were analyzed by RT-PCR.Results The curcumin IC_(50)value for MCF-7 cells was(18.5 ?mol/L.) The inhibitory effect of curcumin on cell growth was in a time-and dose-dependent manner.MCF-7 cells treated with 20 ?mol/L or(40 ?mol/L) curcumin for 24 h were arrested in G_0/G_1 phase and induced apoptosis.The expression levels of bcl-2,PCNA mRNA were decreased in MCF-7 cells by 10,20 and 40 ?mol/L curcumin treatment for 24 h,and bax mRNA was increased.Conclusion Curcumin is of inhibitory effect on MCF-7 cell growth,and it can arrest MCF-7 cells in G_0/G_1 phase,inhibit the proliferation and induce apoptosis.Curcumin down-regulates bcl-2,PCNA mRNA expression in MCF-7 cells,but upregulates bax mRNA expression.
2.Minimally invasive surgery for inflammatory bowel disease

Inflammatory bowel disease (IBD) could lead to the complexity of intra-abdominal pathology and patients' malnutrition,which might greatly increase the risk of operation.Laparoscopic minimally invasive surgery is playing an important role in the treatment of IBD,with the advantages of faster postoperative recovery,shorter duration of hospital stay and lower incidence of postoperative complications.And some new techniques for IBD have been reported in the western countries,such as single-incision laparoscopic surgery,natural orifice transluminal endoscopic surgery,robot-assisted laparoscopic surgery and so on.However,the new techniques are not widely used in China,and it is necessary to carry out a series of clinical practices,which should be carefully selected for IBD patients undergoing minimally invasive surgery.
3.STUDY OF CD-INDUCED DAMAGE OF ERYTHROCYTE MEMBRANES IN RATS

The blood levels of hemoglobin(Hb), cerrulo-plasmin (Cp), superoxide dismutase (SOD), glu-tathion peroxidase (GSH-Px), malonaldehyde(MAD) and the concentrition of Cd, Fe, Cu andSe in liver were measured 4 weeks after weanedSprague-Dawley rats fed on Cd-supplemented diet(Cd 100mg/kg). The results indicated that. theconcentrition of Fe, Cu and Se in liver and theblood levels of Hb, SOD and GSH-Px activity weredecreased, but the blood level of MAD was in-creased by dietary Cd. The abnormal changes andanemia induced by Cd were able to be partially al-leviated by oral supplementing Fe, Cu and Se. Itsuggested. that Cd was able to produce peroxidationof erythrocyte membrane lipids. It is a possiblemechanism of the decreased survival of erythrocyteand hemolysis by Cd.
4.Immunotherapy of Colorectal Cancer

Objective To generally analyze the current situations and advancement of the study on immunotherapy for colorectal cancer. Methods The pertinent published papers about the current situation and research advancement of the immunotherapy of colorectal cancer were retrospectively investigated. And also the immunogenicity and the varying principles of immunoresistance, the functional targets, the practicality, and some other characteristics of different immunotherapy for colorectal cancer were reviewed. Results The main treatments and the research focuses in the immunotherapy of colorectal cancer are initiative nonspecific immunotherapy, adoptive immunotherapy, monoclonal antibody immunotherapy, initiative specific immunotherapy, and targeted therapy. They work by fighting against the cancer itself, cutting off the tumor’s nutrition supply, activating the immune system specifically or breaking the immune tolerance and so on. Though there are still many problems unsolved, immunotherapy has a promising clinical prospect. Conclusion As a beneficial complement for surgery, chemotherapy and radiotherapy, immunotherapy plays an important auxiliary role in the combined therapy for colorectal cancer.
5.Relationship of 25 (OH) D levels in cord and maternal peripheral blood

Object To explore the relationship of 25 (OH) D levels in cord and maternal peripheral blood to understand the maternal and neonatal vitamin D status in Songjiang District of Shanghai. Methods One hundred newborns delivered at the First People’s Hospital afifliated to Shanghai Jiaotong University and 20 pregnant women during November 2010 to January 2011 in Songjiang district were enrolled in the study. The levels of 25 (OH) D in cord and maternal blood were measured by ELISA and compared. Results Mean materna1 serum levels of 25 (OH) D was (23.42±5.23) nmol/L, and cord blood 25 (OH) D was (29.77±12.51) nmol/L. Maternal serum 25 (OH) D was positively correlated with cord blood 25 (OH) D (r=0.84, P<0.001). Conclusions This study shows that the prevalence of vitamin D deifciency in pregnant women is high in Shanghai Songjiang District, which could have adverse effects on newbowns.
7.Clinical treatment of the intermittent claudication caused by lumbar spine disease combined lower extremity arteriosclerosis obliterans

Objective To explore the principle of diagnosis and treatment of intermittent claudication caused by lumbar spine disease combined with lower extremity arteriosclerosis obliterans. Method Eighteen cases of patients with intermittent claudication caused by lumbar spine disease combined with lower extremity arteriosclerosis obliterans, using MRI and MRA, to determine the main reason due to of disease, to take the treatment for the cause. Results The patients were followed up for 7 to 24 months, average (16.0 ± 4.7) months. Assessed according to the modified MacNab criteria of clinical efficacy, excellent in 7 cases, good in 8 cases, general in 3 cases,satisfactory rate was 83.3%(15/18). Intermittent claudication were improved with all patients, walking distance of more than 1000 meters, relief rate was 100%. Postoperative ankle brachial score(0.90±0.54 ) was obviously increased compared with preoperative average(0.58±0.36), there was significant statistical difference(P<0.01). All cases were not amputee due to circulation disturbance. Conclusion The diagnosis and treatment of intermittent claudication caused by lumbar spine disease combined lower extremity arteriosclerosis obliterans,it is necessary to collect a history of serious and careful investigation,combined with imaging, can accurately determine the responsibility of lesions, target to give the appropriate treatment, can get a good effect
8.Combined evisceration for the treatment of advanced gastric cancer

ObjectiveTo evaluate the rationale of combined evisceration for the treatment of advanced gastric cancer.MethodsThe clinical data of 137 cases with advanced gastric cancer treated with combined evisceration from 1994 to 2001 were analyzed retrospectively.ResultsEleven cases underwent combined hepatectomy, 25 cases with splenectomy, 13 with transverse colectomy, 15 with cholecystectomy, 4 with auxiliary adrenalectomy, 38 with splenectomy plus distal pancreectomy, 13 with pancreatoduodenectomy, 18 with other adjecent evisceration. The operative mortality rate was nil. The 1-,3-,5-year survival rate were 60.2%,26.3% and 16.6% respectively.Conclusions Combined evisceration for treating advanced gastric cancer was feasible and yielded a longer survival.
9.Practice and thought about laboratory security education in postgraduate in medicine under new situation

New characteristics and changes of laboratory security were summarized,and then current status and problems of awareness of laboratory security of medical postgraduate under the new situation were analyzed in detail.Based on laboratory security education in Center of Experimental Teaching for Postgraduates in Medicine at Xi'an Jiaotong University,strategies to enhance their awareness and capacity of laboratory security were proposed.Compilation of textbooks on laboratory security,development of curriculum content,training of laboratory security practice,and establishment of evaluation system and laboratory access regulation,will be helpful to maximize safety of postgraduates and to guarantee security of laboratory.
10.Clinicopathological factors on the prognosis of patients with stage lⅡ colorectal cancer

Objective To analyze the clinicopathological factors on the prognosis and investigate the necessity of adjuvant chemotherapy for patients with stage Ⅱ colorectal cancer.Methods The clinical data of 255 patients with stage Ⅱ colorectal cancer who were admitted to the First Affiliated Hospital of Sun Yat-Sen University from January 2000 to December 2005 were collected.The survival curve was drawn by Kaplan-Meier method,and the survival rate of the patients were analyzed by Log-rank test.Factors influencing the survival were analyzed by Cox regression model.Results All patients were followed up till April 23,2010,and the mean time of follow-up was (63 ± 22)months.The median survival time was 63 months.The 5-year and tumor-free survival rates were 85.3% and 83.7%,respectively.The 5-year overall and tumor-free survival rates of patients without preoperative bowel obstruction or perforation were 86.9% and 85.6%,which were sigaificantly higher than 72.7%and 68.4% of patients with preoperative bowel obstruction or perforation(x2 =4.546,4.573,P < 0.05 ).The 5-year overall and tumor-free survival rates of patients with negative resection margin were 85.5% and 83.9%,which were significantly higher than 75.0% and 75.0% of patients with positive resection margin(x2 =7.020,6.009,P < 0.05 ).The result of multivariate analysis revealed that preoperative bowel obstruction or perforation were the independent risk factors for patients with stage Ⅱ colorectal cancer(Wald =4.477,relative risk =2.371,95 % confidence interval:1.066-5.275,P < 0.05 ).The 5-year overall and tumor-free survival rates were 87.3% and 86.0% for patients who received adjuvant chemotherapy,and were 82.2% and 80.3% for patients who did not receive adjuvant chemotherapy (P > 0.05 ).Conclusions Preoperative bowel obstruction or perforation are independent risk factors for the survival of patients with stage Ⅱ colorectal cancer.Adjuvant chemotherapy could not improve the prognosis of patients with stage Ⅱ colorectal cancer.
