2.Effect of glutamine on p38MAPK and IL-8 expression in PBMC of patients with chronic obstructive pulmonary disease

Objective To investigate the effects of glutamine (Gln) on the p38MAPK activity and IL‐8 expression in periph‐eral blood mononuclear cells(PBMC) of the patients with chronic obstructive pulmonary disease(COPD) .Methods Thirty‐two pa‐tients with acute exacerbation of COPD (AECOPD ) were chosen as the research subjects(AECOPD group) .Then the patients con‐verting to the stable stage of COPD after treatment were set as the SCOPD group .PBMC of each group were extracted and conduc‐ted the Gln intervention .At the same time 16 healthy people were selected as the healthy control group .The real‐time PCR(RT‐PCR) was used to detect the levels of p38MAPK and IL‐8 expression in PBMC among the five groups .Results (1)The expressions of p38MAPK and IL‐8 in the AECOPD blank control group and the SCOPD blank control group were both higher than those in the healthy control group (P<0 .01 or P<0 .05) ,while the expression levels of p38MAPK and IL‐8 in patients with AECOPD was higher than those of patients with SCOPD ,the difference was statistically significant (P< 0 .05) .(2)The expression levels of p38MAPK and IL‐8 in the AECOPD Gln group were lower than those in the AECOPD blank control group (P<0 .01);the expres‐sion levels of p38MAPK and IL‐8 in the SCOPD Gln group were lower than those in the SCOPD blank control group (P<0 .05) . Conclusion Gln could inhibit the p38MAPK pathway activation in inflammatory cells of COPD patients and reduce the expression level of IL‐8 .
3.Expression and clinical significance of HSPA2 in pancreatic cancer

Objective To detect the expression of HSPA2 mRNA and protein in pancreatic cancer and corresponding adjacent nontumorous tissues, and investigate the expression of HSPA2 protein in pancreatic cancer and its association with clinicopathological characteristics.Methods Western blot and qPCR (real-time quantitative PCR) was used to evaluate HSPA2 expression in pancreatic cancer and corresponding adjacent nontumorous tissues.Immunohistochemistry was used to investigate the HSPA2 expression in pancreatic cancer and its association with clinicopathological characteristics.Results Both the mRNA and protein levels of HSPA2 were significant higher in pancreatic cancer tissues than those in their paired adjacent nontumorous tissues.High HSPA2 expression was detected in 68.8% (55/80) of pancreatic cancer tissues.HSPA2 protein expression was significantly associated with tumor differentiation, vascular invasion, and status of metastasis (P=0.011,0.005,0.012).Conclusion HSPA2 expression is elevated in pancreatic cancer, and positively correlated the process of invasion, metastasis and progression of pancreatic cancer.
4.Evaluation of Multiple-slice CT Pulmonary Angiography on Right Ventricular Parameters in Acute Pulmonary Embolism

Purpose To evaluate the correlation between acute pulmonary embolism (APE) and right ventricular parameters by multi-slice spiral computed tomography pulmonary angiogmphy (MSCTPA),and assess the severity and mortality of patients with APE.Materials and Methods The clinical and MSCTPA image data of APE patients from January 2012 to November 2015 was analyzed retrospectively.All the patients were divided into three groups according to the pulmonary embolism type and clinical prognosis:66 cases in mixed embolism group (25 cases of death,41 cases of survival);36 cases in peripheral embolism group,2 cases in centralembolism group.The CT post-processing software was used to measure thoracic aorta and main pulmonary artery inner diameter ratio (PA/AO),right ventricular diameter to left ventricular diameter ratio (RVD/LVD);right ventricular area to left ventricular area ratio (RVA/LVA) on axial four-chamber (4-CH) sections and the largest of maximum minor axis diameters on axial sections (RV/LV-LD).The difference of CT related right ventricular parameters among the mixed embolism groups,peripheral embolism and normal groups was compared.Results Statistical difference was found in right ventricular parameters (RVD/LVD,RV-LD/LV-LD,RVA/LVA) in mixed embolism death patients compared with the patients in other three groups (P<0.05).There was significant difference of right ventricular parameters RVD/LVD,RVA/LVA in mixed pulmonary embolism survival patients compared with patients in peripheral pulmonary embolism and normal groups (P<0.05),but there was no significant difference between peripheral APE patients and the control group in all RV and PA/AO parameters (P>0.05).According to the result of ROC curve in mixed type embolism death patients,RVA/LVA was the best indicator of predicting early death (AUC=0.881),with 100.0% sensitivity and 73.2% specificity.Conclusion The right ventricular parameters by multislice CT pulmonary angiography can assess APE patient's condition and forecast their early mortality;and the clinical diagnosis accuracy of RVA/LVA is higher.
5.Effect of bilirubin on the anti-oxidative stress ability of red cell membrane in rats undergoing high intensity training

AIM:To observe the effect of supplementing bilirubin on the anti-oxidative stress ability of red cell membrane in training rats through establishing animal models of high-intensity training. METHODS:The experiment was performed in the physiological laboratory of Physical Institute of Jiangxi Normal University from December 2004 to January 2005.①After one week of adaptive feeding,24 male SD rats were randomly divided into quiet control group,high intensity sports training and physiological saline group(training group),high intensity sports training and bilirubin group(training and supplementing group)with 8 animals in each group.②The training group and the training and supplementing group did the running on the platform with the slope of 0?at the first three weeks and 5?at the fourth week,6 days every week expect Sunday.The training and supplementing group was intragastrically infused with bilirubin by 40mg/kg body mass at 17:00-18:00 after training every day,and the other groups were given physiological saline,six times every week.Anticoagulatory blood about 2mL of heparin was harvested to prepare red cell membrane sample and measure the relevant indexes on the 4~(th)Sunday.③The serum total bilirubin level was determined using automatic biochemical analysis instrument;the malondialdehyde(MDA)and glutathione peroxidase(GSH-Px)of red cell membrane by TBA;the activity of superoxide dismutase(SOD)of red cell membrane using the method of Xanthine oxidase.All the treatment for animals was based on the ethical standards. 8.Effects of repetitive transcranial magnetic stimulation on locomotor outcomes after experimental hemisectional spinal cord injury in rats

Objective To study the effects of repetitive transcranial magnetic stimulation (rTMS) on rats'locomotor function after hemisectional spinal cord injury (SCI). Methods A model of hemisectional SCI was established at the T10 segment in 12 rats,which were then randomly divided into an SCI-rTMS group and an SCI-control group.Another 6 rats served as normal controls.The SCI-rTMS group received threshold rTMS daily for 4 weeks,while the SCI-control group was given sham rTMS.The normal control group was left without any treatment.BassoBeatti-Bresnahan (BBB) locomotor scores were recorded weekly.Motor evoked potential (MEP) was detected at the 28th day after the operation.Neurofilament-200 (NF-200) was detected with immunofluorescent staining of the spinal cord lesions. Results The BBB scores in the SCI-rTMS group improved more than those in the SCI-control group.MEP of the fight hindlimb was detected in the rTMS group,but not in the control group.Expression of NF-200 marker increased significantly more than in the rTMS group. Conclusions rTMS can improve locomotor function of rats after spinal cord hemisectioning,which may result from increased expression of NF-200 and the regeneration of axons.

The analysis of 2013 circulating foreign and Chinese books in Library of Capital Medical University showed that the number of foreign and Chinese books classified into R Medicine and Healthand I Literaturewas the largest ,which thus provided the evidence for book acquisition ,book circulation and library holding develop-ment.Certain suggestions were put forward for improving the utilization of books, such as enforcing the acquisition and catalogue , and recommending books that should be bought .
10.Overtraining induces renal cell apoptosis partly through inflammatory signal pathway in exhaustive swimming rats

Objective To observe the changes in the expression of renal tissue TNF-α , NF-κB and the interrelation to renal cell apoptosis, and their influences of Inula Britannica(an inhibitor of inflammatory signal pathway) in exhausted swimming rats, and to investigate the role of inflammatory signal pathway. Methods Forty-eight male Wistar rats were randomly divided into three groups: control group (CN, n=8), exhaustive swimming group (ES, n=24) and Inula Britannica group (IB, n=16). The rats of CN were quiet without swimming. The rats of ES swam to exhaustion and were sacrificed at immediately (ESI, n=8), 6 hour (ES 6 h, n=8) and 24 hour (ES 24 h, n=8) after exhanstiing swimming. The rats of IB group took orally Inula Britannica at the dose of 25 ml/kg body weight at 24 h before swimming and then swam to exhaustive state. The rats of IB group were sacrificed at 6 hour (IB 6 h, n=8) and 24 hour (IB 24 h, n=8) after exhaustiing swimming. The renal cell apoptosis was measured by the method of terminal-deoxynucleotidyl transferase mediated d-UTP nick end labeling (TUNEL). The expression of TNF-α in renal tissue was examined by immunohistochemistry. The changes of NF-κB in renal tissue were measured by flow cytometry and immunnhistochemistry. The interrelation between TNF-α and NF-κB was analyzed by Pearson method, and the interrelation between TNF-α, NF-κB and renal tissue cell apoptosis was analyzed by Spearman method. Resulls The number of renal tissue apoptotic cells was increased progressively from ESI to ES 24 h rats (P <0.05). Immunohistochemistry staining showed that the positive expressions of renal tissue TNF-α and NF-κB were increased progressively at 0 h (0.136±0.009, 0.129±0.011), 6 h (0.171±0.011, 0.166± 0.009) and 24 h (0.229±0.008, 0.218±0.019) after exhaustiing swimming in ES compared with control group (0.109±0.010, 0.095±0.010) ( all P<0.05). The similar changes of renal tissue NF-κB was also revcalved by flow cytometry. The expression of TNF-α was positively correldted with NF-κB (r=0.955, P<0.01 ), and renal cell apoptosis was also positively correlated with TNF-α and NF-κB (r =0.953, r=0.939, P<0.01) in ES rats. Pretreatment with Inula Britannica, inhibited the up-regulation of expressions of renal tissue TNF-α (6 h:0.142±0.012, 24 h:0.130±0.010) and NF-κB (6 h:0.138±0.010, 24 h:0.136±0.011 ) induced by exhausting swimming. Conclusion Overtraining can induce the up-regulating expressions of renal tissue TNF-α and NF-κB, and Inula Britannica can partly counter the above changes in exhaustied swimming rats, which may be one important mechanisms of overtraining-induced renal tissue cell apoptosis and the anti- apoptosis effect of Inula Britannica.
