1.Determination of the Related Substances in Nimesulide Granules by HPLC

Objective:To establish an HPLC method for the determination of the related substances in nimesulide granules. Meth-ods:An Agilent HC-C18(250 mm ×4.6 mm,5 μm)column was used, the mobile phase was composed of acetonitrile-0.01 mol ·L-1 ammonium dihydrogen phosphate (adjusting pH to 7. 0 with ammonia) (40:60) and the detection wavelength was 230 nm, the flow rate was 1. 0 ml·min-1 ,the column temperature was 30℃, and the injection volume was 20μl. Results:The linear range of nimesu-lide was 0. 10-0. 30 mg·ml-1(r=0. 999 9). Nimesulide and its related substances could be well separated with the peak separation degree above 2. 0. The limit of detection and the limit of quantification was 0. 3 ng and 1. 0 ng, respectively. Conclusion:The method is simple, accurate and reproducible, which can be used for the quality control of nimesulide granules.
2.Research advances in molecular mechanism of pharmacological actions of oxymatrine

Oxymatrine is the main effective monomer of Radix Sophorae flavescentis( Kushen) , which has a variety of pharma-cological actions and valuable clinical applications. Recently, there are many reports about molecular mechanisms of oxymatrine pharmacological actions, giving highly attention to the anti-in-flammatory, anti-fibrosis and antineoplastic effects. These effects are achieved through resisting of oxidation and free radical, anti- <br> virus, and affecting the secretion of inflammatory factor and ap-optosis and so on. This article summarizes the reports on the mo-lecular mechanism of the protection on liver, cardiovascular sys-tem, endocrine system and nervous system, hoping to provide theoretical basis for the practical application.
3.The study on expression and activity of human mutant CD59 gene

Objective: To amplify two human mutant CD59 eukaryotic expressing systems and investigate mutant CD59 functional activity. Methods: Mammalian expression vector PLATER of mutant CD59 cDNAs was transfected into CHO together with the pcDNA by lipofectamine,which confered resistance to G418(400 ?g/ml). The positive clones were tested by FIH. Activity of both mutants CD59 was determined by BCECF release assay. Results: Mutant CD59 cDNAs subcloned into the mammalian expression vector PLATER and transfected CHO together with the pcDNA,which confered resistance to G418. The positive clones were tested by FIH.Activity of both mutants CD59 before and after glycation was determined by BCECF release assay,both of them could restrict MAC formation ,and glycation could inhibit CD59. Conclusion: A eukaryotic system that expressing mutant CD59 cDNA was successfully set up.It was found that mutant CD59 could restrict MAC formation,and glycation could inhibit mutant CD59. These would be helpful for the furthur study of link mutant CD59 and the vascular proliferative of diabetes.
4.The effects and mechanisms of dimethylamiloride on cardiac function in cardiomegaly rat isolated hearts

Aim To investigate the effects and mechanisms of DMA on cardiac function in cardiomegaly rat isolated hearts.Methods With Langendorff perfusion,the changes in the indicators reflecting cardiac function were observed in cardiomegaly rat isolated hearts.Its probable mechanism was explored with calcium channel blocker and sodium calcium exchange(NCX) blockers.Results DMA(0.5~2 ?mol?L~(-1))enchanced cardiac function and exerted positive effects on LVSP-LVDP,+dp/dt_(max)and-dp/dt_(max) in Langendorff-perfused cardiomegaly rat isolated hearts.The effects of DMA on cardiac function in cardiomegaly rat isolated hearts weren′t blocked by nicardipine-one of calcium channel blockers and were blocked by NiCl_2——one of sodium calcium exchange(NCX) blockers.Conclusion DMA can enhance systolic and diastolic function in cardiomegaly rat isolated hearts.By agitating NCX,DMA enhances cardiac function in cardiomegaly rat isolated hearts,independent of L-calcium channel.
5.Metal implant fixation and repair for fresh unstable distal radial fractures with scaphoid fracture:half-year follow-up

BACKGROUND:Healing time and plaster used in fixed position and fixed time are different between distal radius fractures and scaphoid fracture. When both fractures occur simultaneously, you cannot choose a fixed position and determine a fixed time. One-stage internal fixation for the distal radial fractures and scaphoid fractures simultaneously can obtain a good function. OBJECTIVE:To retrospectively analyze the effects of titanium locking compression plate and Herbert screw fixation for fresh unstable distal radial fractures with scaphoid fracture. METHODS:A total of 12 patients with fresh unstable distal radial fractures combined with scaphoid fracture were treated in the Shanxi Dayi Hospital from November 2011 to June 2014. Al cases received open reduction and locking compression plate fixation of the distal radius, open reduction of scaphoid fracture or percutaneous Herbert screw fixation. Fracture healing was observed during folow-up. At 6 months after treatment, wrist joint function was evaluated with modified McBride score. Range of motion of palmar flexion, dorsiflexion, ulnar deviation, radial deviation, pronation and supination and grip strength were measured. The distal radial shortening (difference in height of styloid process of radius and ulna), and angles of ulnar deviation and palmar tilt were measured with X-ray films. The data of healthy side and affected side were compared. RESULTS AND CONCLUSION:(1) 12 cases were folowed up for 6-24 months. (2) The distal radius and scaphoid fractures healed. The healing time of distal radius was 6-12 weeks, with an average of seven weeks. Healing time of scaphoid fracture was 3-6 months, with an average of 4.2 months. (3) Wrist score was evaluated using modified McBride scoring criteria at 6 months after treatment. There were excelent in 5 cases, good in 6 cases, and average in 1 case, with the excelent and good rate of 92%. (4) No significant difference in range of motion of palmar flexion, dorsiflexion, ulnar deviation, radial deviation, pronation and supination and grip strength, angles of ulnar deviation and palmar tilt, difference in height of styloid process of radius and ulna and modified McBride score was detected between healthy side and affected side (P > 0.05). (5) The median nerve injury in two patients was recovered within 6 months after postoperative exploration decompression. No complications appeared, such as infection, dorsal muscle tendon irritation symptoms, carpal tunnel syndrome, and internal fixation failure. (6) These findings confirm that fresh unstable distal radial fractures with scaphoid fractures can be treated with open reduction of the distal radius and locking plate fixation, open reduction of scaphoid bone or percutaneous Herbert screw fixation, with reliable fixation, high fracture healing rate, and good wrist function recovery rate; the repair effect is satisfactory.

7.Effects of Different Anesthesia Techniques on the Balance of Th1/Th2 in Patients Undergoing Radical Resection of Gastric Cancer

Objective To explore the optimal method of anesthesia in radical resection of gastric cancer by comparing the effects of two anesthesia techniques on the balance of Th1/Th2 in order to provide evidence for clinical anesthesia manage-ment.Methods Forty patients who underwent elective radical resection of gastric cancer were randomized into two groups:in-halation anesthesia group(group S)and total intravenous anesthesia group(group P),in which sevoflurane and propofol were ad-ministered for the induction and maintenance of anesthesia,respectively.The general data of the patients were collected,the tis-sue type of tumors obtained and the fluid infusion and blood loss volumes recoded.The VAS score was obtained 1 h,6 h and 5 d after the surgery.Before the induction of anesthesia,6 h and 5 d after surgery,the blood samples were drawn to determine the serum levels of IL-1β,IFN-γ,TNF-α,IL-2,IL-6 and IL-10.Flow cytometry was used to detect the proportion of CD3+CD8-IFN-γ+ and CD3+CD8-IL-10+ cells and the Th1/Th2 ratio was calculated.Results The level of IL-6 was increased post-sur-gically in groups S and P(P<0.05).There were no significant changes in the levels of IL-1β,IL-2 and IL-10 in groups S and P and in the levels of TNF-αand IFN-γ,and the Th1/Th2 ratio in group S between before the induction of anesthesia and different time points after the surgery(P>0.05).The levels of TNF-αand IFN-γ,and the Th1/Th2 ratio were significantly increased in group P 5 d after the surgery(P<0.05).Conclusion Total intravenous anesthesia with propofol can enhance Th1 response and it is an ideal anesthetic method for radical resection of gastric cancer.
8.Markers of hepatitis B virus infection:current application and prospect

The natural history of chronic HBV infection is diverse and variable, ranging from inactive carriers to progressive chronic hepatitis B ( CHB), cirrhosis and hepatocelluar carcinoma.It is estimated that 93 million people are chronically infected with HBV and 20 million cases suffering from chronic hepatitis B in China.Hepatocelluar carcinoma has been the second leading cause of death for male in China.Liver cirrhosis and HCC which have high mortality and morbidity have become the heavy burden for the limited medication resource of China.Here the current clinical applications and consensus progression based on antigen and nuclear acid detection were acknowledged.The reasonable application as well as appropriate clinical interpretation are emphasized indicating that laboratory medicine practitioners should be more actively involved in clinical diagnosis and treatment.More efforts and contributions should be made by the laboratory medicine practitioner for optimizing clinical management of HBV-related diseases in future.
9.The association between serum leptin level and periodontal parameters in patients with type 2 diabetes mel-litus and periodontitis

Objective:To explore the association of serum leptin(LEP)level in patients of type 2 diabetes mellitus and periodontitis with clinical periodontal indexes.Methods:30 healthy subjects(group A),30 chronic periodontitis patients(group B)and 30 type 2 diabetic patients with chronic periodontitis(DMCP)(group C)were included.Probing depth(PD),attachment loss(AL),plaque index (PLI)and bleeding on probe(BOP)were examined.High performance liquid chromatography detection was used to determine the ex-pression of the fasting plasma glucose (FPG),glycated hemoglobin(HbA1c)in plasma samples.Serum level of LEP was measured by ELISA.Results:Levels of serum HbA1c,FPG and LEP in group C were significantly higher than those in group B and A(P <0.05);serum LEP in group B were significantly higher than those in group A(P <0.05).However,no virtual disparity was found in serum HbA1c and FPG between group A and B(P >0.05);The r values of plasma LEP level to PD,AL,PLI and BOP were 0.506,0.551, 0.519 and 0.582 respectively(P <0.05).Conclusion:The LEP level is elevated in patients with DMCP.Circulating LEP level is positively related to PD,AL,PLI and BOP.
10.Construction of the teaching case library on pharmacology

Case teaching is one of the important means to improve teaching effect in the process of pharmacology teaching. According to the teaching object, teaching objectives and requirements to build one useful teaching case library about pharmacology, which can be used and shared for a long time, is the premise of case teaching in pharmacology. According to the actual situation of case teach-ing in our university, this study researched and explored the background of case library building, the basic principles of case compilation, the establishment of team, the teacher-led and the student-oriented processes of case building, and the process of sorting and summarizing cases in the case library build-ing, which promotes the reform in the pharmacology case teaching, improves teaching quality, and also provides a reference for other universities or schools and disciplines on teaching case library building.
