5.Effect of losartan on transforming growth factor β1 in alcoholic nephropathy in rats
Journal of Chinese Physician 2009;11(1):21-24
Objective To investigate the effect of losartan,an angiotensin 1I type 1 receptor antagonist(ATlRA),on transforming growth factor-β1 expression in alcoholic nephropathy in rats. Methods Male adult Wistar rats were random divided into three subgroups and treated daily for 12 weeks as follows, rats were treated with normal saline[20ml/(kg·d)gavage twicely]in vehicle group, rats were treatedwith alcohol[400g/L,8g/(kg·d)gavage twieely]in alcoholic nephropathy, and rats were concomitantly treated with alcohol and Iosartan [200mg/L,ling/(kg·d),gavage]in alcohol+losartan.HE staining,Sirius red staining, electronic microscope, and immunohistochemistry were used to examine histologic changes, collagen fibers deposition, and the expression of TGF-BI.Results Compared with vehicle treated rats, alcoholic nephropathy rats showed tubular vacuolation, granular degeneration, swelling, tubulointerstitial inflammatory cell infil tration, and rarely mesangial proliferation and matrix deposition.Sirius red staining displayed a significant increase in collagen fibers in alias of glomemli, tubulointerstitium and vessels.Concomitant administration of losartan reversed all of above parameters(P<0.05).The upregulated expression of TGF-β1 proteins was significantly suppressed by losartan treatment(P<0.05).Conclusions These findings suggest that alcoholic nephropathy is characterized by the tubular granular degeneration, vacuolation, swelling, and tubulointemtitial inflammato-ry cell infiltration, and this is associated with upregulation of TGF-β1.Losartan treatment offers protection effect by downregulating TGF-β1 expression.
6.Tumor necrosis factor-α and cerebral ischemia-reperfusion injury
International Journal of Cerebrovascular Diseases 2009;17(1):60-62
As an important inflammatory cytokine, tumor necrosis factor-α (TNT-α) plays an important role in the process of cerebral ischemia-reperfusion. The discussion of the dynamic changes, the mechanism of neurotoxicity and antagonizing the therapeutic effect of TNF-α in the process of cerebral ischemia-reperfusion injury will provide theoretical basis for the treatment of cerebrovascular diseases.
7.The establishment and identification of human umbilical artery endothelial cells-smooth muscle cells co-culture model
Fudan University Journal of Medical Sciences 2009;36(6):768-775
Objective To establish a human umbilical artery EC-SMC co-culture model, and mimic the morphological and functional characteristics of human arterial wall, for further reseach of the pathological mechanism and therapy of atherosclerosis and imflammatory damage. Methods We secceeded in the primary culture of human umbilical artery endothelial cells (HUAEC) and human umbilical artery smooth muscle cells (HUASMC) by collagenase perfusion digestion and tissue planting, respectively. HUASMCs were incubated in a medium with ascorbic acid at the concentration greater than 50 μg/mL to produce collagen, which was considered as the extracellular matrix for ECs. Then HUAECs were seeded directly upon HUASMCs in a saturate density for sufficient direct physical interaction between ECs and SMCs. The morphological characteristic of EC-SMC co-culture was identified by immunofluorescence staining, and the function of EC-SMC co-culture was identified by Dil-Ac-LDL uptake test. Results The morphological identification showed that the entire surface of HUASMCs was covered by a confluent monolayer confluent monolayer, which indicated that the model had simulated the morphological characteristic of human arterial wall. The results of Dil-Ac-LDL uptake test showed that there was a fluorescent signal in HUAECs. Compared with EC monoculture, the Dil-Ac-LDL uptake of HUAECs was increased significantly in the co-culture system. All the reseach results indicated that there was an interaction between HUAECs and HUASMCs in the co-culture system. Conclusions In the present study, human umbilical artery EC-SMC co-culture model was constructed successfully, which could mimic the morphological characteristic and basic functions of human arterial wall.
8.Study on the correlation between hs-CRP and coronary heart disease as well as acute myocardial infarction
International Journal of Laboratory Medicine 2009;30(1):36-38
Objective To explore the relationship between high-sensitivity C-reactive protein (hs-CRP) and the occurrence of certain diseases such as coronary heart disease (CHD) and acute myocardial infarction (AMI).Methods Sixty-one CHD patients, 45 AMI patients (patients group, excluding high hs-CRP level diseases caused by other etiological factors) and 35 healthy controls (control group) were enrolled in the study. The serum level of hs-CRP and myocardial zymogram was measured in the Hitachi 7060 automatic analyzer by latex-enhance immunoturbidimetry for all subjects, and cTnI of AMI group was detected with immunofluorescence method at the same time. The difference of hs-CRP level was compared and analyzed between control group and patients group; control group before and after treatment. Moreover, the relationship between hs-CRP and myocardial zymogram as well as cTnI was analyzed. Results The level of hs-CRP was significantly higher in patients group than that in control group (P<0.01). There was statistical difference of hs-CRP level before and after treatment in control group (P<0.01). hs-CRP was positively correlative with myocardial zymogram and cTnI.Conclusion The serum level of hs-CRP is significantly elevated in CHD and AMI patients, indicating the measurement of hs-CRP contributes to auxiliary diagnosis, therapeutic effect observation, and prognosis evaluation.
9.Effect of curcumin on acute lung injury induced by one-lung ventilation in mice
The Journal of Clinical Anesthesiology 2016;32(4):387-392
Objective To investigate the effect of curcumin (Cur)on acute lung injury (ALI) induced by one-lung ventilation (OLV)in mice. Methods Sixty C57BL/6J mice were randomly alloca-ted into six groups (n =10):two-lung ventilation group (TLV group),OLV group,OLV+Cur pre-treated with 50 mg/kg group (Cur50 group),100 mg/kg group (Cur100 group),1 50 mg/kggroup (Cur1 50 group)and200 mg/kg group (Cur200 group).Peak and averaged airway pressure (Ppeak and Paw)of mice in each group were recorded.Mice were euthanized and the end of experiment, and left lung tissue was excised.Wet lung weight to dry lung weight (W/D)and total lung water content (TLW)were tested.Pathological changes of lung tissue were observed under light microscope, and changes of ultrastructure of lung tissue were observed by transmission electron microscope.Index of quantitative evaluation for alveolar damage (IQA)and lung injury scores were tested under light mi-croscope.The expression of c-Jun N-terminal kinase (JNK)mRNA and phosphorylated JNK (p-JNK)protein were detected respectively by reverse transcription-polymerase chain reaction (RT-PCR)and Western Blot.Apoptosis index (AI)of lung tissue was determined by terminal dexynucle-otidyl transferase(TdT)-mediated dUTP nick end labeling (TUNEL)method. Results There was no statistical significance at Ppeak and Paw of rats in each group.Compared to TLV group, the expres-sion of JNK mRNA and p-JNK protein were all significantly increased (P <0.05)in OLV group, and W/D,TLW,IQA,lung injury scores and AI were all notably higher (P <0.05);morphological and ultrastructural injuries in lung tissue were notably occured in OLV group.Compared to OLV group, the expression levels of JNK mRNA and p-JNK protein were decreasingly lower (P <0.05 )in the groups of Cur100,Cur1 50 and Cur200,W/D,TLW,IQA,lung injury scores and AI were also de-creased (P <0.05);morphological and ultrastructural injuries in lung tissue were gradually alleviated in the groups of Cur100,Cur1 50 and Cur200.However,there was no statistical significance in the in-dexes mentioned above between Cur50 group and OLV group. Conclusion Cur at doses of 100 to 200 mg/kg has protective effects on lung against OLV injury in mice, which may be related to inhibition of pneumocyte apoptosis induced by JNK.
10.Cases of Clinical Application of Abdominal Acupuncture Combined with Body Acupuncture for Treatment of Allergic Skin Diseases
Chinese Journal of Information on Traditional Chinese Medicine 2017;24(3):114-116
In recent years, the incidence of the allergic skin diseases is increasing as the growing of the pressure, the changes of the environment and other factors. The clinical features of the allergic skin diseases are easily repeated and in long course. It is found that the incidence of clinical allergic skin diseases, in addition to the season, diet and emotional relevant, is also closely linked to the physical deterioration of patients. Therefore, in addition to the emphasis on the treatment of the current symptoms of patients, adjustment and treatment of phsical constitution of patients and improvement of the adaptability of patients to environmental changes should attract attention, thereby reducing the frequency of incidence and alleviating the symptoms. The application of the abdominal acupuncture combined with body acupuncture for the improvement of the symptoms and physical constitution of allergic skin disease patients has obvious efficacy, which should be applied and promoted in clinic.
