2.The Effect of MPTP on Movement and TH-immunoreactive Neuronal Changes of C57BL/6 Mice in Relation to Aging.

The effects of 1-methyl-4-phenyl-1, 2, 3, 6-tetrahydropyridine (MPTP) on movement and tyrosine hydroxylase (TH)-immunoreactive (ir) neuronal changes in young (5~6 weeks) and aged (10~12 months) C57BL/6 mice were studied. Locomotor activity was measured during 180 minutes after a single injection of 30 mg/kg of MPTP. For immunohistochemistry both young and aged mice were injected four repeated dosages of 10 mg/kg of MPTP 12 hours apart. We counted the numbers of TH-ir cell bodies using immunohistochemical technique in substantia nigra (SN), ventral tegmental area (VTA) and locus ceruleus (LC) 7 days after the last injection of MPTP. There was a marked decrease of locomotor activity in MPTP-treated young and aged mice, and a delay in recovery of locomotor activity in MPTP-treated aged mice. In young mice, there was a decrease in the number of TH-ir cell bodies in the SN of young mice, but not in VTA or LC. In aged mice, there was a significant decrease in the number of TH-ir cell bodies in VTA as well as SN. It was concluded that aged mice were more sensitive to MPTP than young mice, and MPTP-treated aged mice a more useful animal model for studing the characteristics of Parkinson's disease.

3.Effect of Pinealectomy on the Renin-Angiotensin System in Sprague-Dawley Rats.

Experiments were performed to investigate the effects of pinealectomy on the renin-angiotensin system in Sprague-Dawley rats. The results obtained were as follows. 1) There was no differences of systolic blood pressure, heart rate, water intake, and plasma electrolytes between pinealectomized and sham-operated rats. 2) Renin activity in plasma and pituitary gland did not differ in two experimental groups. 3) The pituitary gland contained the renin activity of about 50pg AI/hr/mg of wet weight. The results suggest that the pineal gland has no correlation with cardiovascular and renin-angiotensin system.

5.Comparison of the Effects of NOS Inhibitor or HS-1580 on Nigrostriatal Dopaminergic System in MPTP-induced Parkinson's Disease Animal Model.

The antioxidant effects of HS-1580 derived from Brown seaweeds in comparison with NOS inhibitor, L-NAME, were tested in the Parkinson's disease animal model. C57BL/6 mice were implanted with osmotic pump containing vehicle, HS-1580 or L-NAME intraperitoneally 2 days before MPTP injection. The Parkinson's animal model were prepared by intraperitonal injection of MPTP (20 mg/kg, 4 times with 2 hours intervals in a ay).The mice were perfused with Zamboni fixative 2 days or 7days after MPTP injection. The 30 micrometer cryostat section were immunostained with free-floating method. Rabbit anti-tyrosine hydroxylase (TH)and rat anti-mouse MAC-1 were used as the primary antibodies. The following results were obtained. The number of TH-immunoreactive (ir)neurons in substantia nigra (SN)and the relative density of TH-ir axon terminals in striatum were decreased by MPTP injection. But the ecrease was significantly attenuated with 10%HS-1580 or L-NAME (50 mg/kg)pretreatment before MPTP injection. MAC-1-ir activated microglia were observed in substantia nigra 2 days after MPTP injection. Activated microglia were showed as thickened processes with round cell bodies. The morphological changes of MAC-1-ir activated microglia were inhibited by HS-15980 or L-NAME pretreatment before MPTP injction. Above results mean that the damage of nigrostriatal dopaminergic system was rescued by pretreatment of HS-1580 or L-NAME in MPTP-induced Parkinson's disease animal model.

6.Report for Development of Korean Portable Cardiopulmonary Bypass II. Experimental Study of Portable Cardiopulmonary Bypass for Emergency Cardiopulmonary Resuscitation after Cardiac Arrest in Normal Dogs.

BACKGROUND: Portable cardiopulmonary bypass (CPB) technique has been used increasingly as a potent and effective option for emergency cardiopulmonary resuscitation (CPR) because it can maintain more stable hemodynamics and provide better survival than conventional CPR techniques. This study was designed to develop a prototype of Korean portable CPB system and, by applying it to CPR, to discriminate whether it would be superior to standard open-chest CPR. MATERIAL AND METHOD: By using adult mongrel dogs, open-chest CPR (OCPR group, n=4) and portable-CPB CPR (CPB group, n=4) were compared with respects to restoration of spontaneous circulation (ROSC), hemodynamics, effects on blood cells, blood gas patterns, biochemical markers, and survivals. Ventricular fibrillation-cardiac arrest (VF-CA) of arrest (VF-CA) of 4 minutes followed by basic life support (BLS) of 15 minutes was applied in either group, which was standardized by the protocol of American Heart Association. Then, advanced life support (ALS) was applied to either group under the support of internal cardiac massage or CPB. ALS was maintained until ROSC was achieved but not longer than 30 minutes regardless of the presence of ROSC. All of the measured values were expressed as means+/-SD percent change from baseline. RESULT: During the early ALS, higher mean arterial pressure was maintained in CPB group than in OCPR group (90+/-19 vs. 71+/-32 %; p<.05) and lower mean pulmonary arterial pressure was also maintained in CPB group than in OCPR group (105+/-24 vs. 146+/-6%; p<.05). ROSC was achieved in all dogs. Post-ROSC levels of hematocrit, RBC, and platelet were decreased and plasma free hemoglobin was increased significantly in CPB group compared to OCPR group (p<.05). Changes in blood gas patterns, lactate, and CK-MB levels were not different between groups. Early mortality was seen in 3 dogs in OCPR group (survival time 31+/-36 hours) and 2 in CPB group (228+/-153 hours, p=ns). The remainders in both groups showed prolonged survival. CONCLUSION: These findings indicate that portable CPB can be effective to maintain stable hemodynamics during cardiac arrest, to achieve ROSC and to prolong survival. Further study is needed to refine the portable CPB system and to meet clinical challenges.

Over 26 kinds of tablets and capsules, such as Tetracycline, Quinidine and Potassium preparations were reported to cause esopbageal ulcerations, eharacterized in various size, shape and number, ind sually in mid-esophagus, Recently authors experienced 10 cases of drug-induced esophageal ulcerations due to antibiotics and antiinflammatory agents such as Tetracycline, Aspirin, etc. Four cases were men and six were women. Four cases were in third dades, three in fourth cleeades, two in fifth decades and one in sixth decades. Presenting symptoras were odynophagia(4/10), dysphagia(3/10), substernal pain(7/10) and epigastric pain(3/10). Endoscopic examination of the esophgus showed single or multiple, small and shallow ulcers on the mid-esophgeal mucosa at the level of 30cm from the ineisor in eight cases, one Iarge and deep ulcer at the level of 40cm from incisor in one patient and one amall, shallow and one large, deep ulcers at the same time. in one patient The shape of alcers were various from a round to a large horseshoe shaped one. The clinical course was mild without complications. It was suggested that drug-induced esophsgeal ulcers with antibiotics and antiinflammatory agents could be found frequently and they had benign and mild clinicalc ourse,

9.Pseudoaneurysm of the popliteal artery mimicking tumorous condition.

Diagnosing pseudoaneurysms of the popliteal artery is usually straightforward in physical examinations and imaging findings. However, when a pseudoaneurysm shows a soft tissue mass with adjacent osseous change, it can mimic a bone tumor or a soft tissue sarcoma. We present a case of a 65-year-old man who had a pseudoaneurysm of the popliteal artery showing soft tissue mass and insinuating into the intramedullary cavity of the tibia. This presented case emphasizes the importance of considering pseudoaneurysms in the differential diagnosis of an apparent soft tissue mass with pressure erosion in adjacent bone.

10.A Micropncture Study on Renal Function and Hemodynamic Factors in Obese Rats.

It has been commonly noted that male patients have worse prognosis in adult polycystic kidney disease, IgA nephropathy, and membranous nephropathy. Several animal experiments were performed to clarify the mechanisms by which male gender confers a more adverse renal prognosis. Obese Zucker rats are characterized by severe obesity, insulin resistance, extremely high serum insulin level, high cholesterol level and spontaneous glomerular sclerosis. The aim of current study was to investigate whether there was a gender difference in 1) mean arterial pressure and renal function 2) glomerular volume by renal perfusion 3) glomerular hemodynamic factors including glomerular blood flow, glomerular pressure, renal vascular resistance, ultrafiltration coefficient by micropuncture techniques in obese male and female Zucker rats. There was no difference in blood glucose, serum insulin level, mean arterial pressure and glomerular sclerosis rate between male and female Zucker rats. Male Zucker rats had slightly higher glomerular filtration rate, kidney weight, single nephron glomerular filtration rate, glomerular capillary hydraulic pressure which was statistically insignificant. These findings suggest that gender difference in hemodynamic factors does not play a role in the progression of renal disease in obese Zucker rats.
