1.Macrophages in the Corpus Luteum of the Rat : Immunohistochemical and Transmission Electron Microscopic Study.

Macrophages in the corpus luteum have many important roles during the periods of functional development and luteal regression. Not only phagocyte the apoptotic luteal cells, but also they secrete many cytokines and exert their effects via autocrine/paracrine actions.In this study, we investigated the changes of number and immunoreactivity of macrophages at various developmental periods of the corpus luteum in the rat ovary. The rats (Sprague-Dawley strain, female)at age of 8 weeks (ovulatory period), GD 6 (pregnant period), and postpartum 5 days (postpartum period)were sacrificed under ether anesthesia and obtained both ovaries, one used for macrophages immunohistochemistry and the other used for TEM. The results were as follows; 1.In the corpora lutea of the rat, macrophages were observed all the developmental periods including ovulatory, pregnant and postpartum periods. 2.In the corpora lutea of the rat, number of macrophages was highest in the ovulatory period, and decreased at postpartum period and pregnant period in order. The immunoreactivity of macrophages was high at ovulatory period, moderate at postpartum period, and low at pregnant period. 3.In TEM observations, two types of macrophages were observed: One type was non-phagocytic macrophage and the other type was phagocytic macrophage. Phagocytic macrophages were observed in the corpora lutea at ovulatory and postpartum periods and contained apoptotic bodies, phagocytic vacuoles and many lipid droplets. Non-phagocytic macrophages were observed in the corpora lutea at pregnancy period and showed slender cell body with long cytoplasmic processes and contained no apoptotic bodies. In the rat, the number and the degree of immunoreactivity of macrophages in the corpus luteum varied with the changes of functional state of the corpus luteum. It was suggested that the main function of the macrophages at the ovulatory and postpartum periods was elimination of apoptotic luteal cells and that at pregnancy period was autocrine/paracrine action.Ultrastructurally, two types, phagocytic and nonphagocytic types, of macrophages confirmed. These results will provide valuable informations on the study of the role macrophages during development and regression of corpus luteum.

2.Neodothiora pruni sp. nov., a Biosurfactant-Producing Ascomycetous Yeast Species Isolated from Flower of Prunus mume

A yeast strain, designated as JAF-11 T , was isolated from flower ofPrunus mume Sieb. et Zucc. in Gwangyang, Republic of Korea. Phylogenetic analysis showed that strain JAF-11 T was closely related to Neodothiora populina CPC 39399 T with 2.07 % sequence divergence (12 nucleotide substitutions and three gaps in 581 nucleotides) in the D1/D2 domain of the large subunit (LSU) rRNA gene, and Rhizosphaera macrospora CBS 208.79 T with 4.66 % sequence divergence (25 nucleotide substitutions and five gaps in 535 nucleotides) in the internal transcribed spacer (ITS) region. Further analysis based on the concatenated sequen ces of the D1/D2 domain of the LSU rRNA gene and the ITS region confirmed that strain JAF-11 T was well-separated from Neodothiora populina CPC 39399 T . In addition to the phylo genetic differences, strain JAF-11 T was distinguished from its closest species, Neodothiora populina CPC 39399 T and Rhizosphaera macrospora CBS 208.79 T belonging to the family Dothioraceae by its phenotypic characteristics, such as assimilation of carbon sources. Hence, the name Neodothiora pruni sp. nov. is proposed with type strain JAF-11 T (KACC 48808 T ; MB 850034).
3.Efficacy and Safety of Biodegradable Microparticles in the Regeneration of Injured Rabbit Corpus Cavernosum: Primary Report.

PURPOSE: This study analyzed the effectiveness of poly (lactic-co-glycolic acid) (PLGA) as a tissue recovery agent and determines the in vivo safety and efficacy of microparticle-based PLGA. MATERIALS AND METHODS: Fifteen 3-month-old male white rabbits were used. Allogenic adipose tissue derived stromal vascular fraction (SVF) was cultured and labeled with the fluorescent dye PKH26. The rabbits were divided into 4 groups: the SVF group, the PLGA group, the normal control group, and the disease control group. The right corpus cavernosal tissue of the rabbits was surgically removed in the selected portion, except in the normal control group. The defect space of each rabbit was replaced with 10(6) SVF cells in the SVF group and 0.1 g of biodegradable polymer solution in the PLGA group. Microscopic confirmation and analysis of tissue regeneration were performed after 8 weeks. Using confocal microscopy, the nuclei of the smooth muscle cells and SVF migration were examined. The composition of smooth muscle and fibrosis of the injured corpus cavernosum were compared and analyzed by Masson's trichrome stain. RESULTS: There were no signs of migration or rejection of the injected materials in any of the experimental groups. The mean amount of smooth muscle in the normal control group was 15.25+/-1.34 microm2 (right) and 13.90+/-0.703 microm2 (left); in the disease control group it was 11.10+/-0.87 microm2 (right) and 12.80+/-1.01 microm2 (left); in the SVF group it was 13.82+/-4.10 microm2 (right) and 13.96+/-3.94 microm2 (left); and in the PLGA group it was 12.89+/-1.39 microm2 (right) and 13.24+/-1.43 microm2 (left). Only the disease control group showed significant decreased smooth muscle in the left cavernosum (p<0.05). No significant difference was found between the left and right side of each rabbit's cavernosal smooth muscle in the SVF or PLGA group (p>0.05). Furthermore, no difference was found between any two groups (normal control versus SVF (p=0.705), normal control versus PLGA (p=0.88), SVF versus PLGA (p=0.23). CONCLUSIONS: PLGA microparticles had the same tissue restoring effect when compared with SVF and no adverse effect or migration of particles was found through the injection of PLGA or SVF. PLGA is safe and has the proper tissue recovery effect, saving additional tissue harvesting.

Closed chest trauma occasionally results in the development of traumatic lung cyst or pulmonary hematocele.Radiologically, this latter rarely mimicks posterior mediastinal mass, which can cause unnecessary surgicalresection, We encountered two cases of pulmonary hematocele simulating posterior mediastinal mass. Multiplicity ofthe lesion, fracture of surrounding bony structure, decrease of mass size at follow-up examination, an acute anglebetween the mass and chest wall, peripheral rim enhancement of the mass, as seen on CT scans, or characteristicsignal intensity suggesting hematoma, as seen on MR images, helped differentiate pulmonary hematocele fromposterior mediastinal mass.

5.Prevalence of Hepatitis C Virus Genotype in Pusan, Korea.

The authors investigated the distribution of Hepatitis C virus (HCV) genotype in blood donors with positive for anti-HCV (n=34), health check-up examiness with positive for anti-HCV (n=29), and in patients with various chronic liver disases positive for anti-HCV (n=63) in Pusan, Korea. HCV genotype was determined by using the molecular typing method through the reverse transcription - polymerase chain reaction (RT - PCR) with four type specific primers. Among 116 anti-HCV positive study subjects, 66.4% were positive HCV RNA by RT-PCR. The major HCV genotype was type II (31.9%) and it was followed by type III (27.6%). Two cases were type IV (1.7%). Double infection with two different HCV genotypes (mixed type) was found in three cases (2.6%). Three cases (2.6%) were not determined by the four type specific primers, it may have different subtype. Type II was more prevalent than type III in the blood donors and health check-up examinees, but the reverse was true in the chronic liver disease patients including hepatocellular carcinoma patients. Type II was more prevalent than type III among the anti-HCV positive subjects with risk factors such as acupuncture history, surgical operation history, and trans-fusion history. In contrast type III was more prevalent than type II among the subjects without the above risk factors. It is supposed that the pathogenicity of different kind of HCV genotype might be different. The results of this study suggest that the type II and type III may be the major CV genotype in Korea. The differences of HCV genotype distribution between the study groups support that the clinical significance according to the HCV genotype may be different.

A benign metastasizing leiomyoma is a rare condition that affects women with a history of uterine leiomyoma, usually after a myomectomy or hysterectomy. Typical radiographic findings include well-circumscribed solitary or multiple pulmonary nodules ranging from a few millimeters to several centimeters in diameter and scattered among the normal interstitium. We report a case of a benign metastasizing leiomyoma that manifested with multiple cavitary nodules in a 46-year-old woman with no previous history of a myomectomy or hysterectomy.
