In order to develop a more efficient method for the cultivation of human epidermal cells suitable for use in the treatment of burns, a culture model using fibronectin coated substrate was established,and the effects of concentration of FN and density of epidermal cells on growth of the cells were evaluated. These results show thatthe cultured model is an effective, and simple method for cultivation of human keratinocytes and it is worthy of clinical trial. The cultured autologous epidermal cell sheets were grafted onto burn wound in 4 burn patients. In two of them the gafts grew into multi-layered keratinocyte epithelium.

Numerical simulation is one of the most significant methods to predict the temperature distribution in high-intensity focused ultrasound (HIFU) therapy. In this study, the adopted numerical simulation was used based on a transcranial ultrasound therapy model taking a human skull as a reference. The approximation of the Westervelt formula and the Pennes bio-heat conduction equation were applied to the simulation of the transcranial temperature distribution. According to the temperature distribution and the Time Reversal theory, the position of the treatable focal region was corrected and the hot spot existing in the skull was eliminated. Furthermore, the influence of the exposure time, input power and the distance between transducer and skull on the temperature distribution was analyzed. The results showed that the position of the focal region could be corrected and the hot spot was eliminated using the Time Reversal theory without affecting the focus. The focal region above 60 degrees C could be formed at the superficial tis sue located from the skull of 20 mm using the hot spot elimination method and the volume of the focal region increases with the exposure time and the input power in a nonlinear form. When the same volume of the focal region was obtained, the more power was inputted, the less the exposure time was needed. Moreover, the volume of the focal region was influenced by the distance between the transducer and the skull.

Abstract A rapid gas chromatographic ( GC ) method was established for the determination of short-chain fatty acids ( SCFAs ) in human feces. Feces samples were directly extracted by 1% HCl-75% ethanol solution, and then centrifuged at high speed for GC analysis. The chromatographic separation was performed on a DB-FFAP capillary column ( 30 m í 0 . 25 mm í 0 . 25 μm ) with a temperature program ( initial temperature at 50℃ held for 1 min, ramped to 190℃ at 10℃/min ) . The injection port temperature was 250℃ with split ratio at 50:1 . The carrier gas was high purity nitrogen with a constant linear velocity at 1. 0 mL/min. A flame ionization detector was employed to quantify SCFAs. The proposed method had been certified by systematic method validation, and used to determine feces samples. Subsequently, the health volunteer and colorectal cancer patient groups could be distinguished successfully by multivariate statistical analysis. Compared with health volunteers, the acetic acid and butyric acid of feces from colorectal cancer patients were reduced obviously, indicating that SCFAs particularly butyric acid could be considered as candidate markers for colorectal cancer diagnosis. In summary, this study provides a rapid method for the determination of SCFAs in feces form health volunteers and colorectal cancer patients. The method had a prospect for screening and diagnosing colorectal cancer rapidly.

A low calcium medium developed for epidermal keratinocytes were prepared according to the MCDB 153 modified formula and used in human epidermal keratinocyte culture compared with DMEM culture system. The observation by contrast microscopy and electron microscopy showed that in the low calcium medium keratinocytes grew as a monolayer of high proliferation and had many characteristics of basal cells, with a more rounded shape and large intercellular spaces. Increasing the calcium ion concentration in the medium or changing the other culture conditions the cells in these cultures could be induced stratification and terminal differentiation. The results suggest that the growth, proliferation and differentiation of cultured human epidermal keratinocytes can be controlled and regulated someway.

OBJECTIVE: To investigate the effects of Shenshuning Recipe (SSNR) on gene expression of hepatocyte growth factor (HGF) in renal tissues in rats with glomerulosclerosis. METHODS: Glomerulosclerosis was induced in 42 rats by unilateral nephrectomy and intravenous injection of doxorubicin. Then these 42 rats were randomly divided into three groups: untreated group, SSNR-treated group and benazepril-treated group. Another eight rats were included into sham-operation group. The rats in the SSNR-treated group and the benazepril-treated group were fed SSNR or benazepril respectively for 8 weeks. The levels of 24 h urine protein (Upr), serum creatinine (Cr) and blood urea nitrogen (BUN) of rats in each group were examined. The renal morphological changes were observed under microscope, and the diameter of glomerular capillary, mesangial matrix and glomerulosclerosis index were analyzed by image analysis software. Reverse transcription-polymerase chain reaction (RT-PCR) method was used to detect the gene expression of HGF in the renal tissues. RESULTS: The levels of 24 h Upr, serum Cr and BUN in the untreated group were remarkably increased than those in the sham-operation group (P<0.01). The pathological morphological changes in the untreated group showed that the glomerulosclerosis was diffused around the renal tissue and the capillaries were shrunk. The expression level of mesangial matrix was up-regulated and the glomerulosclerosis index was 3.32+/-0.35. The expression level of HGF mRNA in the untreated group was obviously lower than that in the sham-operation group (P<0.05). The levels of 24 h Upr, serum Cr and BUN in the SSNR-treated group and the benazepril-treated group were remarkably decreased as compared with those in the untreated group, while the expression levels of HGF mRNA were both obviously higher than that in the untreated group (P<0.01). The pathological morphological changes in the SSNR-treated group and the benazepril-treated group were both alleviated. There was no significant difference in therapeutic effect between the SSNR-treated group and the benazepril-treated group. CONCLUSION: Shenshuning Recipe can up-regulate the expression of HGF mRNA, decrease the mesangial matrix, and improve the renal function, so that it may retard the development of glomerulosclerosis.

Objective:To establish a preliminary diagnostic criteria of pathological internet use for field test in China.Methods:Through searching common "pathological internet use" diagnostic criteria or screen criteria in literature,a diagnostic criteria items pool and a preliminary diagnostic criteria of "pathological internet use" were established .According to the diagnostic criteria,each 2 senior professional psychiatrists in turn interviewed 79 patients and their parents,and then made diagnosis respectively. In addiction,44 high school students were each evaluated by one psychiatrist.The definite diagnosis was made when 2 evaluators make the same diagnosis for one patient.Result:In 123 patients,54 were pathological internet use.For diagnosis,the value of kappa for inter-rater reliability was 0.812( P<0.001).In the 12 criteria items,2 for poor inter-rater reliability and 4 for little contribution to diagnosis were eliminated.In the reserved 6 items,if 4 items(or more)were positive,the diagnostic sensitivity was 78.9% and specificity was 95.3%.Functional impairment criteria were made strictly.In the patients who were made the definite diagnosis,90.7% had duration of illness above 3 months,and 77.7% above 6 months.Conclusion:The preliminary diagnostic criteria of pathological internet use after revised includes 6 symptom criteria,3 functional impairment criteria,duration of illness and exclusive criteria.The criteria proposed is with high consistency on evaluations made by psychiatric raters,and with operational convenience.The criteria,after further revised,may fit the clinical application.

Objective To investigate the teaching effect of‘index’teaching method used in practical teaching of the medical imaging.Methods A total of 178 medical undergraduates were selected this study,113 clinical medical students using‘index’teaching method were taken as experimental group while others 65 anesthesia medical students using traditional teaching method as control group.The teaching effect of‘index’teaching method was evaluated quantitatively and qualitatively by practical test and questionnaire survey.Results The test scors of experimental group were(80.3±16.5),obviously higher than those of(71.3±25.8)in control group(P＜0.05),especially instudents with low scores.Conclusion The new teaching method had obvious advantages in culturing the thinking of imaging diagnosis and in improving the ability to solve clinical practical problems,the‘index’teaching method was more effective than traditional teaching method.

Objective To investigate the effect of stress-induced hyperglycemia (SHG) on myocardial perfusion and clinical prognosis in elderly patients with acute myocardial infarction (AMI) who underwent primary percutaneous coronary intervention (PCI). Methods In this study, 459 elderly patients with first-time occurrence of acute ST-elevation myocardial infarction (STEMI) who underwent primary PCI within 12 h from January 2012 to January 2015 were enrolled and followed up. All patients were divided into three groups according to serum glucose (SG) on admission: normal group (SG<7.0 mmol/L, 148 patients), SG elevation group (SG 7.0-11.1 mmol/L, 169 patients) and SHG group (SG>11.1 mmol/L, 142 patients). Myocardial perfusion indexes, including ST segment resolution (STR), TIMI myocardial perfusion grade (TMPG), peak value of creatine kinase (CK)-MB, left ventricular ejection (LVEF), and major adverse cardiac events (MACE) of patients in three groups were measured and compared after emergency PCI. Results The blood glucose levels were increased, ST-elevation 2 h after PCI were well declined, the percentages of patients with TMPG 2-3 were decreased and peak values of CK-MB were increased in the three groups (P<0.05). After 12 months of follow-up, Kaplan-Meier survival analysis showed that cumulative non-events survival rates in three groups had significantly different: 89.2% (132/148) vs. 85.8% (145/169) and 76.1% (108/142), P<0.05. Multivariate Cox regression analysis showed that SHG was the independent predictor for the occurrence of MACE in patients undergoing PCI after adjusting for age and gender (P<0.05). Conclusions SHG in elderly patients with STEMI can decrease myocardial perfusion level after primary PCI, which will lead to high incidence of MACE.

Objective To retrospectively analyze the distribution percentage points of molecular subtypes of breast cancer and investigate its guiding significance in the prognosis and personalized therapy in patients with breast cancer aged over 60 years.Methods A retrospective analysis of all patients with invasive breast cancer aged over 60 years from January 2009 to June 2014 in Breast Center of Beijing Hospital was performed.Based on the immunohistochemical methods recommended in the St.Gallen International Expert Consensus Report 2013,the molecular subtypes of breast cancer in these patients were identified.All patients were followed up every 6 months after operation until December 2014.Clinical pathological characteristics and prognosis indexes in different molecular subtypes of breast cancer were evaluated.Results Totally 770 patients aged over 60 years with invasive breast cancer met the criterion.The luminal A,luminal B,human epidermal growth factor receptor 2 (HER2) over-expression,triple-negative subtypes accounted for 29.1％ (224 cases),47.5 ％ (366 cases),6.5 ％ (50 cases) and 16.9 ％ (130 cases)of patients,respectively.The luminal A subtype of breast cancer had larger tumor size,more lymph node metastasis and more late TNM stage as compared with other subtypes(all P＜0.01).Univariate and multivariate analysis showed that aged ≥ 80 years and the triple-negative molecular subtype were the independent poor prognostic factors for breast cancer(both P＜0.05).The luminal A subtype of breast cancer had relatively good prognosis,while the triple-negative subtype had relatively poor prognosis.Conclusions Molecular subtypes have a significant prognostic effect on breast cancer in patients aged over 60 years,which suggests molecular subtype can be considered as a reference indicator in personalized therapy.Patients aged over 80 years has poor prognosis due to less rational treatment.The reasonable and target treatment based on the molecular subtype of breast cancer should be provided for patients aged over 80 years in clinical medicine.
