1.Diagnostic value of procalcitionin in infectious diseases

Objective To evaluate the diagnostic value of procalcitionin (PCT ) in infectious diseases .Methods Levels of PCT , C reaction protein (CRP) and white blood cells (WBC) were detected and compared among 103 cases of bacterial infection ,77 ca‐ses of viral infections and 60 cases of non‐infected patients .Results PCT level of most bacterial infection patients was higher than 0 .5 ng/mL ,and that of viral infection patients was less or equal to 0 .5 ng/mL .Proportion of bacterial infection patients with differ‐ent PCT level was different with that of viral infection patients (P<0 .05) .PCT ,CRP and WBC levels in bacterial infection patients were higher than viral infection patients and non‐infected patients (P<0 .05) .Positive rates of PCT ,CRP and WBC in bacterial in‐fection patients were higher than viral infection patients and non‐infected patients (P<0 .05) .Conclusion PCT might be with high diagnostic sensitivity and specificity to infectious diseases ,with important diagnostic value .
2.Screening of mimicry peptide of Japanese encephalitis virus E protein from phage 15-mer peptide library

Aim To study the mimicry peptide of JEV E protein by screening a phage 15-mer peptide library with anti-JEV E protein mAb 2H4. Methods After three rounds biopanning, the enriched positive phage clones were identified by ELISA. 10 positive phage clones were sequenced and compared homologously with JEV E protein. Results The short peptide displayed on screened positive phage could bind specifically to mAb 2H4, and the binding could be inhibited by natural JEV Ag. Amino acid sequences of the 10 positive phage clones were consensus, that is, RQDPQWPYANSTIAR. By homology analysis, two higher homologous sequences STXAR and WXXAXST were found in different regions of JEV E protein. The peptide displayed on positive phage could react specifically with the mouse antiserum against natural JEV Ag . Conclusion This peptide displayed on positive phage may mimic partial antigenicity of JEV E protein.
3.Immunophenotyping of eighty six children with acute lymphoblastic leukemia by three-color flow cytometry.

OBJECTIVESTo evaluate the specificity of three-color flow cytometry in childhood acute lymphoblastic leukemia (ALL) immunophenotyping.
METHODSImmunophenotyping was performed by three-color flow cytometry analysis using CD(45)/SSC gating.
RESULTSThe percentage of blasts was correlated better with leukemic cell count compared with that of FSC/SSC, and the false positive results were low. Among eighty six cases of ALL, 95.3% was B-ALL, in which common-ALL and Pro-B-ALL were 76.8% and 6.1%, respectively, and 2.3% was T-ALL. CD(34)(+) and myeloid-associated antigen expression were observed in 57.0% and 34.9% of the cases, respectively, among which Pro-B-ALL was the commonest. CD(33) was more commonly expressed than CD(13) in Pro-B-ALL cases, but no difference in the expression between these two antigens in other subtypes.
CONCLUSIONGating of CD(45)/SSC eliminated effection of normal cells to blasts in bone marrow, with which the immunophenotyping results were more reliable.

4.Effects of hydrogen-rich saline on intestinal mucosal barrier in intestinal ischemia/reperfusion injury rat

Objective:To investigate the effects of hydrogen rich-saline (HRS) on intestinal mucosal barrier in rat with intestinal ischemia/reperfusion injury (IIRI).Methods:Twenty-four healthy male Sprague-Dawley rats, aged 8 weeks, were randomly divided into 3 groups (8 in each group) by random number table method: sham group, model group and HRS group.Rats in HRS group were intraperitoneally injected with HRS (10 mL/kg) at 30 min of ischemia, and the same amount of normal saline was intraperitoneally injected in model group.After 45 min of ischemia and 6 h of reperfusion, rats were sacrificed.Serum and ileum were collected for further detection.Tumor necrosis factor alpha (TNF-α), interleukin (IL)- 1β and IL-17A expression levels in serum were detected by conducting enzyme-linked immunosorbent assay (ELISA). The localization expressions of tight junction protein Occludin was detected by immunohistochemical staining (IHC), while the localization expression of tight junction protein zonula occluden-1 (ZO-1) were detected by immunofluorescence staining (IF). The protein expression of Occludin, ZO-1, and Lysozyme were detected by performing Western blot.The mRNA expression of Lysozyme and α-defensin were detected by real-time PCR (qPCR).Results:ELISA results proved that the levels of serum TNF-α and IL-1β in HRS group rats were significantly lower than those in model group [(62.02±29.97) ng/L vs.(113.40±44.58) ng/L, (21.68±0.35) ng/L vs.(28.29±3.49) ng/L], while the level of IL-17A increased [(28.18±5.28) ng/L vs. (15.10±3.60) ng/L] (all P<0.05). IHC staining: compared with model group, the expression of Occludin in HRS group was uniform and continuous, and the staining was darker.IF results: compared with model group, the fluorescence signal intensity of ZO-1 in HRS group rats significantly increased, and the distribution was clear and continuous.Wes-tern blot results: compared with model group, the expression levels of Occludin and ZO-1 proteins in HRS group rats remarkably increased (0.79±0.06 vs. 0.54±0.04, 0.91±0.11 vs. 0.51±0.13), while Lysozyme protein decreased (1.50±0.40 vs. 2.99±0.80) (all P<0.05). qPCR results revealed that the expression level of Lysozyme mRNA in HRS group rats was lower than that in model group (1.64±0.33 vs. 2.20±0.40), while α-defensin mRNA obviously increased (0.82±0.19 vs. 0.47±0.13) (all P<0.01). Conclusions:HRS protects intestinal mucosal barrier by inhibiting the expression of tight junctions and the secretion of antimicrobial peptides in rat suffering from IIRI.
5.Transcriptome Analysis of Monozygotic Twin Brothers with Childhood Primary Myelofibrosis

Primary myelofibrosis (PMF) is a chronic myeloproliferative disorder in human bonemarrow. Over 50% of patients with myelofibrosis have mutations in JAK2, MPL, or CALR. However,these mutations are rarely detected in children, suggesting a difference in the pathogenesis ofchildhood PMF. In this study, we investigated the response to drug treatment of a monozygotic twinpair with typical childhood PMF. The twin exhibited different clinical outcomes despite following the same treatment regimen. The transcriptomic profiles of patient samples after drug treatment (E2and Y2) were significantly different between the twin pair, which is consistent with the observationthat the drug treatment was effective only in the younger brother, despite the twin being geneticallyidentical. Bioinformatics analysis of the drug-responsive genes showed that the JAK-STAT pathwaywas activated in the cured younger brother, which is opposite to the pathway inhibitionobserved in adult PMF cases following treatment. Moreover, apoptosis and cell cycle processes wereboth significantly influenced by drug treatment in the sample of younger brother (Y2), implyingtheir potential association with the pathogenesis of childhood PMF. Gene mutations in JAK2,MPL, or CALR were not observed; however, mutations in genes including SRSF2 and SF3B1occurred in this twin pair with childhood PMF. Gene fusion events were extensively screened inthe twin pair samples and the occurrence of IGLV2-14-IGLL5 gene fusion was confirmed. The currentstudy reported at transcriptomic level the different responses of monozygotic twin brothers withchildhood PMF to the same androgen/prednisone treatment regimen providing new insights into thepotential pathogenesis of childhood PMF for further research and clinical applications.
6.Therapeutic observation and pharmacoeconomic analysis of two first-line chemo-therapy regimens in advanced NSCLC

Objective To evaluate the cost-effectiveness of two chemotherapy regimens in NSCLC for the purpose of lower cost,higher quality medical care.Methods 56 patients with advanced NSCLC were divided into two groups from our hospital,group PP(pemtrexal+cisplatin,n=30)and group GP(gemcitabine+cisplatin,n=26),and analyzed by pharmaco-economic cost-effectiveness method,and retrospective analysis was also applied.Results Effective rates of two regimens were 46.67%,42.31%,no significant difference.But there was significant difference in the incidence rate of ADR between the two groups.The cost of chemotherapy regiments were 31 985.48 and 27 683.15 yuan.The average length of stay was 10.94 and 13·91 days.Compared to group GP,for each unit of efficacy,the cost in group PP were increased 986.77 yuan.Conclusion From the analysis of cost-effectiveness,group PP had higher chemotherapy costs,but better safety than group GP.
7.Progress of non-coding RNA in angiogenesis of osteosarcoma

Osteosarcoma needs to continuously induce angiogenesis to satisfy its own nutritional supply in the process of development. Therefore, the inhibition of osteosarcoma cell-induced angiogenesis as a target has become a research hot in recent years. Currently, vascular inhibitors targeting mRNA-encoded protein have been applied in clinic, but the efficacy is poor. Non-coding RNA (ncRNA) is a type of RNA molecules that do not participate in protein coding. ncRNA can regulate the angiogenesis of osteosarcoma by regulating the secretion of vascular factors such as vascular endothelial growth factor, angiogenin-2 and hypoxia-inducible factor 1 or the interaction between ncRNAs. This article reviews the role of ncRNA in the angiogenesis of osteosarcoma to provide references for clinical targeted therapy strategies for osteosarcoma.
8.Clinical characteristics and treatment of perianal abscess in neonates

Objective To evaluate the clinical characteristics and the clinical outcomes of perianal abscess (PA) in neonates.Methods A retrospective review was performed on the collected data of 185 patients of PA in neonates prospectively admitted to Binzhou Medical University Hospital from January 2008 to December 2015.Patients were divided into 2 groups on the parents' intention:nonsurgical treatment and surgical treatment,the standard surgical treatment for PA was incision and drainage with the use of packing.The standard surgical treatment for PA was surgical incision drainage of lower abscess under local anesthesia by the use of filling tamponade iodoform gauze,while the patients receiving conservative treatment took hip bath perianally with topical 1 ∶ 5 000 potassium permanganate,besmearing erythromycin eye ointment outside locally.Incision-thread-drawing procedure was recommended in fistula-in-ano (FIA) after 6 months.Antibiotics were administered in all patients in the early days.The clinical data of age,gender,accompanying diseases,abscess amount and location,treatment approach,healing time and recurrence rates were analyzed with statistical method.Results All patients were boys,time of visiting hospital was 1-25 day,the average time 7.5 days;60 cases (32.4%)had neonatal diarrhea,45 cases (24.3%)had neonatal jaundice,but no patients had severe fever.A single skin lesion was present in 145 patients (78.4%),2 lesions in 30 patients (16.2%),and 10 patients had 3 lesions (5.4%).The most commonly affected sites were at 9 o'clock clockwise direction with 115 (62.2%)lesions on lithotomy position,followed by 3 o'clock clockwise direction with 65(35.1%) lesions by 1 o'clock clockwise direction with 3 (1.6%) lesions and 6 o'clock clockwise direction with 2 (1.1%) lesions.Bacteria cultures were obtained from 123 patients (90.4%,123/136 cases) of surgical treatment and 35 patients (71.4%,35/49 cases) of nonsurgical treatment obtained the results of bacteria culture.The average healing time was (21 ±2) days (10-60 days) in the surgical treatment group,and (36 ± 3) days (9-90 days) in the nonsurgical treatment group,7 out of 136(5.1%) patients had a recurrence with surgical treatment,incision drainage was performed again with the use of packing,and FIA was not found,10 out of 49 (20.4%) patients had a recurrence with nonsurgical treatment group,and 6 out of 49 (12.2%) were spontaneously resolves within the first year of life,4 out of 49 (8.1%) developed into FIA,incision-thread-drawing procedure was performed after 6 months.The significant difference was observed between and nonsurgical treatment and surgical treatment in healing time (t =-6.707,P =0.000),recurrence (x2 =11.347,P =0.001) and FIA formation rate (x2 =10.054,P=0.002).Conclusions PA is an entity in neonates.Incision and drainage of PA is an effective and safe therapy in the early days.Surgery for PA may result in low recurrence rates,a low rate of evolution toward FIA,and a short healing time,which should be considered as the primary treatment.The key procedure is to keep the drainage unobstructed by the use of filling gauze drainage to prevent crissum abscess recurrence.Postoperative care with antibiotics is effective to shorten hospital stays.
9.Research progress in in vitro detection of urinary volatile organic compounds for early cancer diagnosis

With the further increase of the global cancer burden, various cancers are increasingly challenging human health status and quality of life. Thus, early screening of cancer is crucial. Urinary volatile organic compounds (VOCs) detection techniques have the advantages of easy access to samples, high acceptance of patients, non-invasive, and so on, which have been favored and concerned by researchers. In this paper, existing techniques and methods for cancer diagnosis based on urine VOCs were described, relevant studies on the use of urine VOCs for cancer diagnosis were reviewed, and the barriers and future perspectives of the technique were discussed. This paper can be a reference for researchers working in the direction of urinary VOCs detection, a multidisciplinary field that spans medicine and materials science.

COVID-19 is a public health emergency currently. In this study, a scale-free network model is established based on the Spring Migration data in 2020.The cities is clustered into three different modules. The epidemic of the cities in the black module was the most serious, followed by the red and the cyan. The black module contains 9 cities in Zhejiang province and 8 cities in Guangdong province, most of them located in the southeast coastal economic belt. These cities should be the key cities for epidemic prevention and control.
