1.Optimizing of ELISA assay for HBV serum markers

Objective To analyze and approach the optimizing of ELISA assay for HBV serum markers.Methods To compare the critical value rate of flushing with distilled water,flushing with cleaning tap water and flushing without anything.Results The HBsAg critical value rates of flushing with distilled water,flushing with cleaning tap water and flushing without anything were 0.52%,2.60% and 5.56%.The HBsAb critical value rates of flushing with distilled water,flushing with cleaning tap water and flushing without anything were 5.52%,8.02% and 14.86%.The HBeAg critical value rate of flushing with distilled water,flush-ing with cleaning tap water and flushing without anything were 2.82%,2.92%and 3.55%.Conclusion Flushing with distilled wa-ter or flushing with cleaning tap water can reduce the high critical value rate because of enzyme or developer pollution.Flushing with distilled water exhibits more efficacy than flushing with cleaning tap water.

The determinations of total extracting flavone content and total extracting solid yield of roots or stems of Radix Ilieis Pubescentis and their contrast studies in pharmacodynamics and toxicology have been carried out.As a result,no significant difference between roots and stems,which provide the basis for mixed use of roots and stems of Radix Ilicis Pubescentis.
3.Chemical Elements Burden in Soil of High Prevalence Areas of Birth Defects in Shanxi Province

Objective To find the chemical elements whose contents are abnormal in the soil of the high prevalence areas of birth defects in Shanxi Province. Methods 131 soil samples were collected from the high and low prevalence areas of birth defects respectively in 2005, the contents of 16 elements were determined with inductively coupled plasma (ICP)machine. The data were analyzed with regression and nonparametric tests. Results The total content of elements in the soil of the high prevalence areas was significantly higher than that of the low prevalence area. Compared with the low prevalence area, in a high prevalence area, the content of Mo, As, Pb, Ni, V and Se in soil was abnormal, Mo, As, Ni and Pb showed a significant relationship to the prevalence rate of birth defects, in another high prevalence area, the content of Sn, Se, Mo, Zn, Sr, Pb, Ni, Fe, V, Ca, Cu, Al, Mg and Na was abnormal, Pb, Mg, Ca, Al and Zn showed a significant relationship to the prevalence rate of birth defects. Conclusion Maybe the abnormal content of Mo, As, Ni, Pb, Mg, Ca, Al and Zn in the soil is one of the risk factors for birth defects.
4.Effect of LncRNA-PVT1 on Proliferation and Apoptosis of Human Pancreatic Cancer Cell Line HPAF-Ⅱ

Background:Recent studies have shown that long non-coding RNAs(lncRNAs)play important roles in carcinogenesis and cancer biology and the related context has attracted more and more attentions. PVT1,which encodes a lncRNA,is reported to be up-regulated and exhibit pro-oncogenic activity in a wide variety of human cancers. Aims:To investigate the expression of PVT1 in human pancreatic cancer cells and its effect on proliferation and apoptosis of HPAF-Ⅱ cells. Methods:One target siRNA against PVT1 was synthesized and transfected into HPAF-Ⅱ cells by using lipofactamine technique. PVT1 mRNA expression was detected by real-time PCR;capability of cell proliferation was examined by MTS and colony formation assays;cell cycle progression and apoptosis were measured by flow cytometry;and Western blotting was performed to determine the expressions of apoptosis-related proteins and proto-oncogene protein c-Myc. Results:The mRNA expression of PVT1 in several human pancreatic cancer cell lines,especially HPAF-Ⅱ cells was significantly higher than that in H6c7,a human immortalization normal pancreatic ductal epithelial cell line. Compared with HPAF-Ⅱ cells transfected with negative control siRNA or without transfection,silencing of PVT1 by siRNA-PVT1 resulted in remarkable reduction in cell proliferation,cell cycle G1 phase arrest,and notable apoptosis;meanwhile,the expressions of apoptosis-related proteins(cleaved-caspase-3 and cleaved-PARP)were up-regulated,the ratio for Bcl-2 / Bax was decreased,and the expression of c-Myc protein was down-regulated. Conclusions:LncRNA-PVT1 is highly expressed in human pancreatic cancer cell line HPAF-Ⅱ. It may affect the proliferation and apoptosis of HPAF-Ⅱ cells partially through regulating c-Myc expression.
5.Apoptosis gene expression profiling of placental trophoblast cells in patients with pregnancy induced hypertension

Objective To explore the function of placental trophoblast cell apoptosis on the pathogenetic mechanism of pregnancy induced hypertension (PIH). Methods Apoptosis of trophoblast cells in 20 cases of PIH(PIH group) and in 10 cases of normal pregnancy (control group) were directly observed using the terminal-deoxynucleotidyl transferase mediated d-UTP nick end labeling (TUNEL) method. Apoptosis gene expression patterns were screened with gene chip provided by Poxing Company, Shanghai. Standards for differently expressed genes were: (1) An absolute value of the natural logarithm of cy5(PIH group)/cy3(control group) greater than 0.69 with a difference of signal of cy5 2 times over that of cy3. (2) The signal value either cy3 or cy5 must be greater than 800. 6.Effect of rehabilitation training on deglutition disorders of children with cerebral palsy

Objective To explore effect of rehabilitation training on deglutition disorders of children with cerebral palsy . Methods Twenty-seven children patients from January to June in 2013 were set as control group and thirty-one patients from July 2013 to January 2014 as experiment group. The children in the control group were treated with tube-feeding combined with spoon feeding and bottle-feeding by professional nurse. Children in the experiment group were treated with oral rehabilitation training by professional therapists and nurses apart from the same feeding as in the control group. The two groups were compared in terms of effect of deglutition within 4 weeks, time and rate of removing the stomach tube. Results The recovery of deglutition function in the experiment group was much better than that in the control group , the total effective rate and rate of removing the stomach tube within 3 months were higher and the intubation duration was significantly lower as compared with those of the control group (P < 0.01). Conclusion Rehabilitation training can improve the recovery of deglutition disorders, improve active feeding ability of children with cerebral palsy, shorten time of tube feeding and improve their life quality.
7.The exploration of the application of the chronic patients as the standardized patients in the respiratory medicine clinical teaching

ObjectiveTo discuss the feasibility and importance of the application of the chronic respiratory patients as the standardized patients ( short for SP ) in the respiratory medicine clinical internship. MethodsThe experimental group ( 27 ) and control group ( 24 ) of the 2004 undergraduate interns were formed. The chronic respiratory patients were trained as the SP, which are used in clinical internship education to evaluate the teaching effectiveness.ResultsThe examination scores of SP group were compared with the control group with the result of SP group ( 9.02 ± 0.13 ) and the control group ( 5.34 ± 2.35 )in humanistic care and communication skills, SP group ( 18.62 ± 1.06 )and the control group ( 13.16 ±5.36 ) in history taking, SP group ( 19.66 ± 1.22 ) and the control group ( 14.35 ± 3.57 ) in physical examination; SP group ( 37.88 ± 2.11 ) and the control group ( 31.68 ± 6.42 )in medical records analysis capacity, all of them having significant difference P<0.01. ConclusionThe chronic respiratory patients as the SP which are applied to clinical internship will improve the effectiveness of clinical internship and bring broad application foreground of medical education.
8.Surgical management of liver carcinoma accompanied by portal hypertension:report of 26 cases

Obiective:To evaluate the effects of various surgical procedures on liver carcinoma accompanied by portal hypertension.Methods:Combined surgical procedures which were performed in 26 patients with liver carcinoma accompanied by portal hypertension in our department between Aug,2002 and Aug,2008 were analysed retrospectively.Results:There was no operative mortality.The postoperative complications developed in 50%(13/26) patients.The postoperative survival rates of 1,2 and 3 years were 84.6%(22/26),57.7%(15/26),34.6%(9/26),respectively.Postoperative upper digestive tract hemorrhage developed in 10 cases.Fifteen patients died during follow-up period,of whom 7 cases died of recurrence of liver carcinoma,2 cases died of liver failure,6 cases died of upper digestive tract hemorrhage.Conclusion:The survival time can be prolonged and the postoperative complications can be reduced through perioperative cares and prudent selection of surgical procedures in patients suffering from concurrent liver cancer and portal hypertension.Combined operation is safe and feasible.
9.BRCA1 inhibits progesterone-induced proliferation and migration of breast cancer cells.

OBJECTIVETo study the effect of BRCA1 in regulating the proliferation and migration of breast cancer cells stimulated by progesterone.
METHODSBreast cancer MCF-7 and T-47D cell were transfected with a vector containing the coding sequence of BRCA1 (pFlag-CMV2-BRCA1 wt) to induce BRCA1 overexpression or with the empty vector (control). The cells were then stimulated with progesterone, and the cell proliferation and migration were observed using MTT assay and wound healing assay, respectively. The proliferation and migration of MCF-7 cells were also observed following transfection with a small interfering RNA (siRNA) for BRCA1 knockdown or with a scrambled siRNA prior to progesterone stimulation.
RESULTSTransfection with the empty vector and with pFlag-CMV2-BRCA1 wt prior to progesterone stimulation caused significantly different proliferation rates in MCF-7 cells [(114.4∓6.0)% vs (82.1∓3.2)%, P<0.05] and in T-47D cells [(111.3∓4.3)% vs (84.2∓3.5)%, P<0.05], resulting also in significantly different cell migration rates (55.9% vs 15.8% in MCF-7 cells and 44.83% vs 10.43% in T-47D cells). Compared to the scrambled siRNA, BRCA1 siRNA transfection prior to progesterone stimulation significantly increased the proliferation rates [(114.4∓3.05)% vs (125.3∓4.0)%, P<0.05] and migration rate (39.2% vs 69.08%) of MCF-7 cells. The progesterone antagonist RU468 could antagonize the effects of BRCA1 knockdown in enhancing progesterone-stimulated MCF-7 cell proliferation and migration.
CONCLUSIONA decreased BRCA1 expression can enhance progesterone-stimulated tumor cell proliferation and migration in sporadic breast cancer.

10.Correlation of tissue elasticity modulus and pathological grades in a chronic pancreatitis model

Objective To investigate the correlativity between elasticity modulus and pathological severity in chronic pancreatitis (CP).Methods Twenty-one pigs were divided randomly into experimental group (n=18) and control group (n=3) using random number method.The main pancreatic duct (MPD) was incompletely ligated to establish the CP model.In control group, MPD was not ligated.The animals were killed in batches at 4th, 8th and 12th week after surgery.The pancreatic tissue was taken for elasticity modulus test and pathological examination, and the pigs were classified into control, mild, moderate and severe groups based on the severity of fibrosis.Cell density, fat infiltration and extracellular edema were observed and classified into mild and severe.The difference of elasticity modulus among different groups were compared by Variance analysis, the correlation between pancreatic fibrosis and elastic modulus was analyzed with Spearman correlation analysis, and ROC curve was used to evaluate its efficacy of diagnosing CP.Results Sixteen CP models were established successfully expected for 2 deaths (mild, n=7;moderate, n=2 and severe, n=7).All of the control group (n=3) showed normal pancreas.The elasticity modulus of control, mild and moderate to severe group were 0.4268±0.0566, 0.3203±0.0518 and 0.2235±0.0685, respectively.The difference between the groups was statistically significant (F=13.658,P<0.01), and the elastic modulus and pathological grade had a negative correlation (r=0.969, P<0.01).AUC of elasticity modulus for differentiating normal and mild CP was 1.000, the best critical value was 0.3807, and both the sensitivity and specificity of the diagnosis were 100%.AUC for differentiating mild and moderate to severe CP was 0.8730, the best critical value was 0.2646, and the sensitivity and specificity of the diagnosis were 85.7% and 77.8%, respectively.The pancreatic elasticity modulus of low parenchymal cell density group and high parenchymal cell density group were 0.1931±0.0373 and 0.3485±0.0655, respectively, which in the high cell density group was significantly higher than that in the low cell density group (t=-5.719, P<0.01).The elasticity modulus of negative infiltration or slight fatty infiltration group and severe fatty infiltration group were 0.3401±0.0697 and 0.1855±0.0344, respectively, which in the negative infiltration or slight infiltration group was significantly higher than that in severe infiltration group (t=5.102, P<0.01).The elasticity modulus of negative or mild cell edema group and moderate to severe cell edema group were 0.2760±0.0825 and 0.3024±0.1056, respectively;there was no statistically significant(t=-0.586, P >0.05).Conclusions The elasticity modulus can be used to detect the pathological changes of CP, and evaluate the CP pathologic grades.
