1.Effect of monosialo gangliosides-1 and Radix salviae miltiorrhizae on hyposemic ischemic encephalopathy

ObjectiveTo observe the effect of monosialo gangliosides-1 (GM-1) and Radix salviae miltiorrhizae (RSM) on neonatal hyposemic ischemic encephalopathy (HIE).Methods62 cases of HIE were randomly divided into treatment group and control group, and both groups received conventional therapy. Injection of RSM and GM-1 were added into the treatment group in 7~10 days therapy. ResultsHigh curative effect, average curative effect and total curative effect in the treatment group of 31 cases were 80.6%, 16.1% and 96.8%, while that in control group of 31 cases were 32.3%, 51.6% and 83.9% respectively. There was significant difference in the high curative effect and total effect between the two groups (P<0.05). ConclusionThe treatment combined with RSM and GM-1 can enhance the curative effect on HIE without any side effect.
2.Expression of chemokine receptor CCR7 and CXCR4 in human colorectal carcinoma and its significance

Objective:To observe the expression of chemokine receptor CCR7 and CXCR4 in human colorectal carcinoma and its significance,so as to assess their expression with the metastasis and prognosis of colorectal carcinoma. Methods:Immunohistochemistry was used to detect the expression of chemokine receptor CCR7 and CXCR4 in 110 patients with colon cancer.The relationship between the CCR7 and CXCR4 expression and the clinic pathological characters was statistically analyzed. Results:CCR7 and CXCR4 expression were positively expressed in 59.1% and 45.5%of the patients. The high expression rate of CCR7 and CXCR4 in lymph node metastasis positive cases was 81. 5% and 60.0% respectively,but the without lymph node metastasis positive cases were 26.7% and 24.4% respectively. The effect of cognition need on information seeking behaviors of information users and the difference in behaviors of information users were investigated in 184 undergraduates from the same grade with different back-grounds of subjects using the cognition need scale and undergraduates information-seeking behaviors scale ,respec-tively.The data were analyzed by descriptive statistical analysis, correlation analysis, rank test, and multivariate linear regression analysis , respectively , showing that the cognition need is significantly correlated with the informa-tion seeking behaviors of undergraduates in terms of information evaluation, information searching, information management and information use .
4.The effect of folic acid on DNA methylation of tumor-related genes in healthy human peripheral blood mononuclear cells

Objective To investigate the effect of folic acid on the DNA methylation of tumorrelated genes promoters in healthy human peripheral blood mononuclear cells(PBMC). Methods Ten healthy volunteers were divided into two groups, and were randomized to receive either 5 mg folic acid (n=5)or placebo(n = 5) , one time per day for 3 months. The serum folic acid concentration was detected with chemiluminescence enzyme immunoassay kit before and after the intervention. The methylation statuses of five tumor-related genes promoter, including oncogenes c-myc, c-Ha-ras,tumor suppressor genes p16INK4A, E-cadherin and mismatch repair gene hMLH1 in PBMC were detected by bisufite sequencing. Results After folic acid intervention, the level of serum folic acid increased significantly in intervention group (t= -4. 739,P<0. 05) , however no significant difference in control group. After three-month folic acid intervention, the level of methylation of oncogene c-myc promoter increased from 4%, 3. 3%, 4. 1% before intervention, one week after intervention, one month after intervention respectively to 8%(t= -4. 079,P<0. 05), while no significant change in placebo taken group. Before and after the folic acid intervention, there was no significant difference of DNA methylation of other tumor-related genes promoter, including c-Ha-ras、E-cadherin、p16INK4Aand hMLH1. Conclusion Folic acid intervention can up-regulate DNA methylation of oncogene c-myc promoter, but can not affect the promoter methylation status of tumor suppressor genes E-cadherin,p16INK4Aand hMLH1.
5.Surgical management of liver carcinoma accompanied by portal hypertension:report of 26 cases

Obiective:To evaluate the effects of various surgical procedures on liver carcinoma accompanied by portal hypertension.Methods:Combined surgical procedures which were performed in 26 patients with liver carcinoma accompanied by portal hypertension in our department between Aug,2002 and Aug,2008 were analysed retrospectively.Results:There was no operative mortality.The postoperative complications developed in 50%(13/26) patients.The postoperative survival rates of 1,2 and 3 years were 84.6%(22/26),57.7%(15/26),34.6%(9/26),respectively.Postoperative upper digestive tract hemorrhage developed in 10 cases.Fifteen patients died during follow-up period,of whom 7 cases died of recurrence of liver carcinoma,2 cases died of liver failure,6 cases died of upper digestive tract hemorrhage.Conclusion:The survival time can be prolonged and the postoperative complications can be reduced through perioperative cares and prudent selection of surgical procedures in patients suffering from concurrent liver cancer and portal hypertension.Combined operation is safe and feasible.
6.Detection of hepatic metastases by contrast-enhanced ultrasound compared with conventional ultrasound and contrast-enhanced computed tomography

Objective To evaluate the role of contrast-enhanced ultrasound (CEUS) on the the detection of hepatic metastases in comparison with conventional ultrasonography (US) and contrastenhanced computed tomography (CECT).MethodsNinety-seven patients with hepatic metastases underwent US,CEUS and CECT images.Their detection of hepatic metastases were compared.Results Hepatic metastases showed five enhancement patterns with CEUS,including bolus hyper-enhancement,peripheralrimenhancement,inhomogeneousenhancementwithnecrosis,hypo-enhancement,isoenhancement with liver parenchyma,all hepatic metastases showed dark defects in portal and delayed phase.The mean number of metastases at CEUS was greater than that of US (2.6±1.9 vs 1.6 ±1.2,P<0.05).The detection of hepatic metastases was 53.4% and 87.1% respectively(P<0.05).CEUS and CECT have no statistically significant difference in the detection of hepatic metastases (P>0.05).ConclusionsCEUS can improve detection of hepatic metastases.
7.The exploration of the application of the chronic patients as the standardized patients in the respiratory medicine clinical teaching

ObjectiveTo discuss the feasibility and importance of the application of the chronic respiratory patients as the standardized patients ( short for SP ) in the respiratory medicine clinical internship. MethodsThe experimental group ( 27 ) and control group ( 24 ) of the 2004 undergraduate interns were formed. The chronic respiratory patients were trained as the SP, which are used in clinical internship education to evaluate the teaching effectiveness.ResultsThe examination scores of SP group were compared with the control group with the result of SP group ( 9.02 ± 0.13 ) and the control group ( 5.34 ± 2.35 )in humanistic care and communication skills, SP group ( 18.62 ± 1.06 )and the control group ( 13.16 ±5.36 ) in history taking, SP group ( 19.66 ± 1.22 ) and the control group ( 14.35 ± 3.57 ) in physical examination; SP group ( 37.88 ± 2.11 ) and the control group ( 31.68 ± 6.42 )in medical records analysis capacity, all of them having significant difference P<0.01. ConclusionThe chronic respiratory patients as the SP which are applied to clinical internship will improve the effectiveness of clinical internship and bring broad application foreground of medical education.
8.Fungal infection induced by Cryptococcus neoformans aerosol inhalation in immunosuppressed Balb/c mice.

OBJECTIVETo explore the feasibility of inducing fungal infection by Cryptococcus neoformans aerosol inhalation in immunosuppressed Balb/c mice.
METHODSTwenty-four Balb/c mice were randomized into cyclophosphamide (CTX) group and control group (n=12). The mice in CTX group were subject to inhalation of Cryptococcus neoformans aerosol prepared using a ultrasonic nebulizer 4 days after intraperitoneal CTX injection, and the control mice received the inhalation only. The leukocyte count and changes in appetite, body weight, and behaviors of the mice were observed after the treatments. On days 3 and 7 after the inoculation, the mice were sacrificed to analyze the fungal counts in brain and lung tissue homogenates and examine the pulmonary pathologies.
RESULTSCTX injection caused lowered appetite and body weight loss in the mice, and significantly reduced the leukocyte counts (2.77∓0.45 vs 8.26∓0.56, P<0.05). At days 3 and 7 after inoculation, the Cryptococci load in the lungs increased obviously in CTX group with a colony-forming unit (CFU) of the lungs of 271.67∓122.22 and 41.67∓0.28, respectively, significantly higher than that in the control group (60.00∓43.36 and 3.00∓5.30, respectively, P<0.05). In CTX group, the CFU was 10.17∓5.42 and 9.17∓6.34 in the peripheral blood and 6.83∓4.92 and 11.00∓5.44 in the brain tissue at days 3 and 7, respectively, whereas no Cryptococci was detected in the peripheral blood or in the brain tissue in the control group. Pathological examination of the lungs revealed destruction of normal alveolar structure in CTX group, with numerous infiltrating inflammatory cells and visible yeast.
CONCLUSIONInhalation of Cryptococcus neoformans aerosol can cause fungal infection in the lungs of immunosuppressed Balb/c mice.

OBJECTIVETo explore the effect and mechanism of sodium butyrate on expression of human clotting factor VIII in vitro.
METHODSMouse NIH/3T3 cell line was transfected with recombinant plasmid vector pRC/RSV-BDD-hFVIII, which enclosed B-domain deleted (760aa approximately 1 639aa) human factor VIII cDNA (BDD-hFVIII cDNA). Then cells were incubated in Dulbecco's modification of Eagle's medium (DMEM) containing sodium butyrate for 24 hours, hFVIII: C and hFVIII: Ag in the cell culture medium were measured by ELISA assay and one-stage method, respectively. In addition, the effect of sodium butyrate on transcription of cDNA encoding the whole hFVIII, heavy and light chain of hFVIII was also investigated by means of run-on assay.
RESULTSAfter stimulation of sodium butyrate, the levels of hFVIII: C and hFVIII: Ag increased 70% than those of control. Run-on assay showed that sodium butyrate enhanced the transcription of cDNA which encoded heavy chain of hFVIII.
CONCLUSIONSodium butyrate can improve the expression of hFVIII through enhancing the transcription of hFVIII heavy chain encoding cDNA. It demonstrated that sodium butyrate had potential utility in inducing the expression of hFVIII in vitro.
