1.Medical equipment maintenance management based on PDCA

The problems of some hospital in medical equipment maintenance management were introduced from the aspects of maintenance time, maintenance record, maintenance service and self maintenance. Some measures were proposed including simplifying maintenance process through PDCA, informatizing maintenance record, cooperating with third-party maintainer and establishing self maintenance team, so that precision and informatized medical equipment management can be realized to maximize the benefit of medical equipment management.
2.Lunate and Perilunate Dislocation:X-ray Features and Analysis of Missing Diagnosis

Objective To improve the diagnosing level of lunate and perilunate dislocation through analysing X-ray finding and causes of missing diagnosis.Methods 16 cases of lunate and perilunate dislocation missed diagnosis were analysed.Results In 16 cases,there were 3 cases of ventral lunate dislocation,2 cases of trans-scaphoid ventral lunate dislocation,6 cases of dorsal perilunate lunate dislocation,3 cases of trans-scaphoid dorsal perilunate lunate dislocation and 2 cases of atypical dorsal perilunate lunate dislocation.6 cases were missed in diagnosis(37.5%).Conclusion Knowing well the normal X-ray sign of wrist,mastering the X-ray characteristic of each types of dislocation and fracture,is the linchpin of reducing missing diagnosis.
3.Determination of tetrahydropalmatine in Juyanbaokang Granules by SPE-HPLC

Objective: To determine tetrahydropalmatine in Juyanbaokang Granules (Rhizoma Corydalis, Radix Aconcti Lateralis Preparata, Radix Rauwolfiae, etc.) by SPE-HPLC. Methods : Samples were purified by C 18 SPE column. HPLC column was Dikma Diamonsil C 18 (250mm?4.6mm,5?m), mobile phase was 0.05M potassium dihydrogen phosphate-0.005M sodium heptanesulfonate-acetonitrile (1.2∶1.2∶1), flow rate was 1.2 mL?min -1 and detection wavelength was 230nm. Results : The calibration curve was linear in the range of 0.067 ~0.402?g for tetrahydropalmatine ( r =0.9997). The average recovery was 98.66%. Conclusion : The method with good separation is convenient, rapid, accurate and reliable.
4.Study on determining method of dl-tetrahydropalmatine by TLCS

AIM: To study the determination of dl-tetrahydropalmatine (THP) in Chi nese traditional patent medicines by TLC-Scanning from the viewpoint of the pro cess control of manufacture. METHODS: To select the 5 kinds of representative drugs and its prep arations contained, THP was extracted with solvent method. The mobile phase contains diethylamine. THP w as separated with high-efficiency silica gel plate or handcrafted silica gel p late. Iodine was used for chronogenic agent and fluorescence defectiion for dete rmination. TLC-Scanning results were compared with HPLC. RESULTS: The method is practicable, the result is correlative with the HPLC. CONCLUSION: TLC-Scanning is fit for the quality control of the Chi nese traditional patent medicine in the process of manufacture.
5.Reprogramme-induced genomic stability

BACKGROUND:Some studies have shown that more copy number variations are present in early passage human induced pluripotent stem cells than later passage human human induced pluripotent stem cells, their parental somatic fibroblasts or human embryonic stem cells. OBJECTIVE:To investigate whether the reprogramming process itself compromises genomic stability and further explore the efficiency of induced pluripotent stem cellestablishment. METHODS:Using high-resolution Affymetrix CytoScan HD array, we compared copy number variations and loss of heterozygosity in early passage induced pluripotent stem cells with their fibroblast cellorigins from genetic epilepsy patients. RESULTS AND CONCLUSION:Compared with somatic fibroblasts from genetic epilepsy patient, there was no difference in the loss of heterozygosity between the two types of cells, but more copy number variations were present in early passage human induced pluripotent stem cells which were characterized as microduplication and involved oncogenic genes. Results demonstrate the dynamic nature of genomic abnormalities during reprogramming process and the necessity of frequent monitoring human induced pluripotent stem cells to assure their genomic stability and clinical safety.
6.A quantitative investigation of E2F1-regulated cell cycle compensation mechanism.

OBJECTIVETo explore the core mechanism of cell cycle compensation using a mathematical model.
METHODSA set of ordinary differential equations were used to describe the interactions between the core cell cycle molecules. Continuous and cyclic changes of the concentrations of these molecules were computed to capture the discrete events of molecular interactions.
RESULTSThe calculated molecule concentrations and captured signaling events agreed with the experimental results.
CONCLUSIONE2F transcription factor 1 is the pivotal element linking the positive and negative feedbacks and regulating G1/S and G2/M phase compensation.

7.Diagnostic value of triangle shadow change on lateral radiograph of the wrist joint in intractable dislocation

Objective To investigate the diagnostic value of triangular shadow changes in carpus dislocation.Methods 68 patients with carpal dislocation underwent frontal and lateral X-ray films.The changes of triangular shadow on lateral films were evaluated based on imaging findings of normal wrist joints.Results Changes of triangular shadow were found in 65 (95.59%)patients,while 3 (4.41%)patients had no changes.There were 17 patients with simple carpal dislocation,in which 15 cases showed triangular shadow changes, while 2 patients showed no changes.Thirty-two patients got pericarpal dislocation,in which 31 cases with triangular shadow changes and 1 case without any changes.The triangular shadow changes were observed in all 19 patients with trans-carpal carpal dislocation or trans-carpal pericarpal dislocation.Conclusion The changes of triangular shadow are helpful in the diagnosis of carpal dislocation, it is difficult to diagnose carpal lateral dislocation.
8.Detection for chromosomal aberrations in 43 fetuses with spontaneous abortion and stillbirth by array-based comparative genomic hybridization.

OBJECTIVETo assess the value of array-based comparative genomic hybridization (array-CGH) for analyzing tissues derived from spontaneous abortion and stillbirth.
METHODSAgilent Human Genome CGH Microarray 4×44 K chip and Affymetrix Cytoscan 750 K Array were utilized to detect genome-wide copy number variations (CNV) in 43 fetuses with spontaneous abortion and stillbirth. All identified CNV were analyzed with references from Database of Genomic variants (DGV), database of DECIPHER, ISCA and OMIM, as well as comprehensive literature review to determine whether the identified CNVs were pathogenic. Parental DNA of two cases was also analyzed with the same arrays for pathogenic or unknown significant CNVs.
RESULTSAll of the 43 specimens were successfully analyzed. Clinically significant chromosomal aberrations were identified in 32 (74.4%) of the samples, which included 26 aneuploidies and 10 pathogenic CNV.
CONCLUSIONArray-CGH is a fast and effective method for analyzing tissues derived from spontaneous abortions and stillbirths which may be difficult to culture for karyotype analysis.

9.Study on Hypoglycemic Effect and Mechanism of Zicui Yin on Type Ⅱ Diabetic Rats

OBJECTIVE: To study the hypoglycemic effect and mechanism of Zicui yin on type Ⅱ diabetic rats, and to provide theoretic basis for clinical application. METHODS: Eighty male rats were fed with high fat/sugar diet for 4 weeks, and then given intraperitoneal injected with streptozotocin (35 mg/kg) to induce type 2 diabetic model. Other 10 rats were included in normal group. Model rats were randomly divided into metformin group (positive control, 0.2 g/kg), model group (constant volume of distilled water), Zicui yin high-dose, medium-dose, low-dose groups (14.0, 7.0, 3.5 g/kg), with 10 rats in each group. After 2 weeks of continuous intragastic administration, the fasting blood glucose of rats in each group was measured by automatic blood glucose meter. The serum insulin level was determined by ELISA. mRNA expression of JNK, Akt and IRS-1 were detected by RT-PCR; The phosphorylation of JNK, Akt and IRS-1 proteins in the pancreatic tissue of rats in each group was detected by Western blot method. RESULTS: Compared with normal group, the fasting blood glucose, mRNA expression of JNK, The phosphorylation of JNK and IRS-1 proteins in the pancreatic tissue of the model group were significantly increased (P<0.05 or P<0.01), while the serum insulin content, mRNA expression of Akt and IRS-1, the phosphorylation of Akt protein were significantly decreased (P<0.01). Compared with model group, the fasting blood glucose of rats in metformin group and Zicui yin high-dose and middle-dose groups decreased significantly (P<0.05), the serum insulin content of rats in both metformin group and Zicui yin high-dose group increased significantly (P<0.01), and mRNA expression of JNK in pancreatic tissue of rats in metformin group and Zicui yin groups decreased significantly (P<0.01), while mRNA expression of Akt and IRS-1 increased significantly (P<0.01). The phosphorylation of JNK and IRS-1 proteins in metformin group and Zicui yin high-dose group decreased significantly (P<0.01), while the phosphorylation of Akt protein were increased significantly (P<0.01). CONCLUSIONS: Zicui yin can significantly reduce blood glucose level, the mechanism of which may be related to decreasing the phosphorylation of JNK and IRS-1 proteins and increasing the phosphorylation of Akt proteins in pancreatic islets.
