1.Gemcitabine plus oxaliplatin with or without rituximab: treatment for elderly patients with B-cell lymphoma

Objective To study the efficacy, safety and tolerance of the therapeutic schedule of GemOx±R regimen suitable for elderly patients with B-cell lymphoma. Methods 11 elderly patients with B-cell lymphoma were enrolled in this study, which were diagnosed by biopsy. All the patients were treated according the GemOx ± R therapeutic schedule as described: rituximab (375 mg/m2 on day 0),gemcitabine (1000 mg/m2 on day 1),oxaliplatin (100 mg/m2 on day 1).Treatment response,therapy related toxic and sideeffect were assessed after inductive and consolidated treatment. Results The median age of 11 patients were 72.18 years(range 55-83 years).The overall response rate(ORR) after inductive treatment was 90 %(9/10).The ORR of 4 DLBCL patients using GemOx+R as initial treatment was 100 %. GemOx regimen were used for 4 refractory/relapse patients,and 3/4 patients got PR after inductive treatment.No patient got dose adjustment.The hematological toxicity was controllable and nobody got renal function impairment. Conclusions The GemOx ± R regimen is feasible,tolerable,effective for elderly patients with B-cell lymphoma.
2.Studies on the Volatile Component from the Flower of Tamarix chinensis

OBJECTIVE:To conduct a quantitative analysis on the volatile constituents from the flower of Tamarix chinensis. METHODS:The volatile constituents were extracted from the flower of T. chinensis by solid-phase microextraction technique and component identification was carried out by GC-MS combined with retention index. RESULTS:36 kinds of compounds were identified,among which pentadecane took up a greatest proportion (9.80%),followed by 6,10,14-trimethyl-2-pentadecanone (7.61%),5,6-dihydro-6-pentyl-2H-pyran-2-one(6.83%),hexadecane(6.25%)and 5,6,7,7a-tetrahydro-4,4,7a-trimethyl-2(4H)-benzofuranone(5.13%).CONCLUSION:This study serves as a scientific basis for the further development and utilization of T. chinensis.
3.Clinical significance and hematologic features of CD56~+ acute myeloid leukemia

Purpose:To study the clinical significance and hematologic features of CD56 + Acute Myeloid Leukemia(AML). Methods:The expression of differentiation antigens on the acute myeloid leukemia cells were detected by flow cytometry(FCM);the Epstein-Barr Virus(EBV) mRNA in CD56 + AML leukemic cells by Rt-PCR assay;the ultrastructure of the CD56 + AML leukemia cells were showed by electron microscope and immuno-electron-microscope. The hematologic features and clinical characteristics at the first visit retrospectively analysed as well as the effect of chemotherapy.Results:CD56 expression rate in AML was 30.62%(79/258).EBV-mRNA were negative in all CD56 + AML cells detected.The ultrastructural characteristics showed 1-2 masses floccose objects in nucleus of CD56 + AML leukemic cells,CD56 expression had no correlation with age,sex,WBC,Hb,BPC,Leukemic cell count in BM,CR rate and CR duration(P value is 0.128,0.877,0.181,0.866,0.629,0.407,0.998 and 0.096,respectively),but there was correlation with higher extramedullary involvement(77.78% v 61.11%,P=0.019)?higher expression of CD34 (66.67% v 46.48%,P=0.03),higher expression of P170(51.79% v 34.94%,P=0.048) and shorter survival(Median,11.5 months v 18 months,P=0.0478). Conclusions:AML was a specific subtype of AML,with a poor prognosis.It may be useful to have more detailed classification with appropriate therapy for this subtype.
4.EFFECTS OF PROTEIN FROM TRICHOLOMA MATSUTAKE MYCELIUM ON APOPTOSIS OF HeLa CELLS

Objective: To study the antitumor effect of the protein from Tricholoma matsutake against HeLa cells in vitro. Methods: Active protein was separated from the mycelium after sub-merged fermentation, and the antitumor effect against HeLa cells was studied in vitro by cell growth inhibition, scanning electron microscope, flow cytometry and DNA electrophoresis. Results: The mycelium protein-TMP-B had strong antitumor effect and induced apoptosis in vitro. TMP-B inhibited cell proliferation and induced cell apoptosis by arresting cell conversion from S to G 2/M state. HeLa cells exposed to TMP-B showed apoptotic peaks. Agarose gel electrophoresis of DNA from cells treated with TMP-B revealed "DNA ladder". Conclusion: The mycelium protein from Tricholoma matsutake can inhibit cervix cancer cells--HeLa cell proliferation and induce apoptosis.
5.The MRI findings of meningeal peripheral-type primitive neuroectodermal tumors

Objective To analyze the MRI features of Ewing sarcoma/peripheral primitive neuroectodermal tumors(pPNETs) arising from the meninges.Methods The MRI imaging of 9 patients with Ewing sarcoma/pPNET were reviewed retrospectively,and imaging features and pathological characteristics were analyzed.Results The age of most patients ranged from 10 to 20 years.Magnetic resonance revealed a spindle-like lesion with a wide base in 8 cases.The lesions showed heterogenous iso-or hypo-intense signal on T1 WI in 7 cases,heterogenous hypo-iso-intense signal on T1 WI in 2 case,and iso-or mildly hypderisointense on T2WI in all cases.The solid part of the tumor was heterogeneously enhanced after injection of gadolinium with cyst degeneration or necrosis.The dural tail sign could be seen in 3 cases.The adjacent skull erosion could be seen in 6 cases.The breakthrough of the plate of cranium and soft-tissue invasion was present in 2 cases.The right eye proptosis was present in 1 case.The distant metastasis was found in 3 cases.Pathology showed that the lesions had high cell density.Hemorrhage and necrosis could be observed.The cells were like lymphocytes and spindle cells with transparent cytoplasm.CD99 and Vimentin were expressed in all tumor cells.Conclusion The imaging findings of the meningeal pPNET are different from meningiomas,which could be useful for the diagnosis and differential diagnosis.
6.Simultaneous Determination of Contents of 4 Heavy Metal Elements in Cultured Cordyceps militaris by ICP-MS

OBJECTIVE:To establish the method for the simultaneous determination of contents of 4 heavy metal elements in cultured Cordyceps militaris. METHODS:The samples were digested by nitric acid heating method and then determined by ICP-MS including radio-frequency power of 1.15 kW,sampling depth of 65 nm,auxiliary gas flow rate of 1.0 L/min,cooling gas flow of 13.0 L/min,temperature of 25 ℃,automatic plasma. RESULTS:The linear ranges of arsenic(As),cadmium(Cd),mer-cury (Hg) and lead (Pb) were 0-20 μg/mL(r=0.9970),0-10 μg/mL(r=0.9995),0-5 μg/mL(r=0.9955),0-20 μg/mL(r=0.9960),respectively. Detectio limit were 0.128,0.003,0.002,0.004 mg. RSDs of precision,stability and reproducibility tests were all lower than 6.5%. Recoveries of them were 90.4%-100.6%(RSD=3.45%,n=9),94.3%-101.3%(RSD=2.93%,n=9), 90.0%-102.3%(RSD=4.03%,n=9),92.3%-103.0%(RSD=3.53%,n=9),respectively. CONCLUSIONS:The method is sim-ple,precise,stable,reproducible,and can be used for simultaneous determination of contents of 4 heavy metal elements in cul-tured C. militaris. The contents of As,Cd,Hg and Pb in 10 batches of sample are all in line with related national standards.
7.Simultaneous Determination of 5 Nucleosides from Different Parts of Cultured Cordyceps militaris by HPLC

OBJECTIVE:To establish a method for simultaneous determination of 5 nucleoside from different parts of cultured Cordyceps militaris. METHODS:HPLC method was adopted. The determination was performed on InertSustain C18 column with mobile phase consisting of methanol-0.02 mol/L monobasic potassium phosphate solution (gradient elution) at the flow rate of 0.6 mL/min. The detection wavelength was set at 254 nm,the column temperature was 30 ℃. RESULTS:The linear ranges of uridine, inosine,guanosine,adenosine and cordycepin were 0.568-3.408 μg(r=0.9999),0.284-1.704 μg(r=0.9999),0.264-1.584 μg(r=0.9999),0.232-1.392 μg(r=0.9999)and 1.672-10.032 μg(r=0.9998),respectively. RSDs of precision,stability and repeatabili-ty tests were all lower than 3.0%. The recoveries were 98.2%-103.9%(RSD=1.97%,n=9),96.2%-101.6%(RSD=1.76%,n=9),96.7%-102.0%(RSD=1.94%,n=9),95.1%-99.4%(RSD=1.43%,n=9) and 95.6%-101.3%(RSD=1.82%,n=9). CON-CLUSIONS:The method is simple,precise,stable and repeatable,and can be used for simultaneous determination of 5 nucleoside from cultured C. militaris,the content of nucleosides are different in different parts of C. militaris.
8.Influence of CO2 pneumoperitoneum on gastric cancer cells

Objective To investigate the influence of CO2 pneumoperitoneum on the adhesive and invasive ability of gastric cancer cells based on the expression of adhesive and invasive molecules. Methods With an artificial CO2 pneumoperitoneum model in vitro, human gastric cancer cells MKN-45, SGC-7901 and MKN-28 were exposed to 3 different CO2 gradients: 9 mm Hg, 15 mm Hg and control group (0 mm Hg). The expression of E-cadherin, ICAM-1, MMP-2 and VEGF-A were measured at 2 and 4 hours exposure by using RT-PCR, CytoMatrixTM kit and ECMatrixTM kit. The pretreated gastric cancer cells were injected into abdominal cavity of nude mice(2×106 cells per mouse). Five mice in each group were sacrificed 4 weeks later to record the number of tumor nodules in abdominal cavity. The remaining mice were kept for observation of survival time. Results The expression of E-cadherin (MKN-45: from 2.26 to 2.19, SGC-7901 :from 2.16 to 2.09、MKN-28 :from 2.06 to 1.99), ICAM-1 (MKN-45 : from 2.20 to 2.28、SGC-7901: from 2.10 to 2.18、MKN-28: from 2.00 to 2.08), MMP-2 (MKN-45:from 2.05 to 2.13、SGC-7901: from 1.95 to 2.03、MKN-28: from 1.85 to 1.93) and VEGF-A(MKN-45 : from 2.10 to 2.16、SGC-7901 :from 2.00 to 2.06、MKN-28: from 1.90 to 1.96) didn't change significantly with increasing pressure and time (P>0.05). The expression of adhesive and invasive molecules didn't change significantly between the experimental groups and the control group. There was no statistical significance of tumor metastasis in abdominal cavity of nude mice(MKN-45:from 22 to 23、SGC-7901 :from 20 to 22、MKN-28:from 21 to 22) and survival time(MKN-45 :from 23 to 21、SGC-7901 :from 22 to 21、MKN-28 :from 22 to 21) among all the groups. Conclusion Under low pressure and short time of CO2 exposure, the adhesive and invasive capacity of gastric cancer cells did not change significantly hence did not increase the possibility of neoplasm metastasis.
9.Liposoluble constituents from two Varieties of Chrysanthemum morifolium Ramat in Kaifeng City

AIM: To analyze the liposoluble constituents from two Varieties of Chrysanthemum morifolium Ramat(Changhong and Lengyan) cultivated in Kaifeng. METHODS: Changhong and Lengyan were extracted by Soxhlet extraction,and GC-MS was used to analyze their components. RESULTS: Forty compounds were identified from Changhong,amounting to 90.09% of the total constituents.Thirty-five compounds were identified from Lengyan,amounting to 91.57% of the total constituents. CONCLUSION: There appears to be considerable difference in triterpenoids and sterols between the samples of Changong and Lengyan.
10.Research projects achievements appraisal system's establishment and application

To establish a scientific and feasible evaluation index system and to assess medical research programs subsidized by the Shanghai Municipal Health Bureau in 2002-2006, so as to provide reference to aid the management department in decision-making and better guidance of research projects. Delphi expert consultation was employed to construct an evaluation index system, and used the system to comprehensively assess the funded projects. In the end, we found that the funding of the Shanghai Health Bureau had certain promoting effect in optimization of research team, startup of scientific research, training of research personnel, and the development of disciplines.
