1.The assessment of tumor hypoxia variation after radiotherapy with 99Tcm-MNLS in mice tumor model

Objective To investigate the feasibility of monitoring the tumor's hypoxic changes by 99Tcm-2-(2-methyl-5-nitro-1H-imidazol-1-yl) ethyl eihydrogen phosphate (MNLS) imaging after radiotherapy.Methods (1) H22 cells were cultured and mice model with liver cancer xenografte was made.The mice were imaged at 0.5,1,2,3,4,6 and 8 h (six mice in each group) after injected with 7.4 MBq 99TcmMNLS when the tumor size reach about 1 cm.Then the mice were sacrificed.The T/NT and %ID/g of each time point was calculated.(2) The liver cancer bearing mice of radiotherapy group (25 Gy) and control group were imaged at 0,24,48 h,and then the technique of ROI was adopted to calculate the T/NT at each time point in the two groups.Immunohistochemical stain method was used to evaluate the expression level of HIF-1α in liver cancer.(3) One-way analysis of variance,the least significant difference t test,two-sample t test and Spearman correlation analysis were performed.Results (1) The uptake of 99TcmMNLS in the liver cancer bearing mice was significant at 2 h after injection and the %ID/g was the highest.99Tcm-MNLS was excreted mainly through kidneys.(2) The T/NT and HIF-1α expression level in radiotherapy group at 24 h (2.65±0.27,(50.62±3.78)%) were lower than those at the instant (3.35±0.19,(85.32±0.94)%,t=5.640,6.701,both P<0.05),but higher than those at 48 h (2.23±0.52,(21.69±0.75)%,t=7.674,4.911,both P<0.05).The T/NT and HIF-1α expression in the radiotherapy group were significantly higher than those in the control group at the instant (2.74 ± 0.29,(28.26 ± 1.70) %,t =4.235,3.473,both P<0.05) but lower at 48 h (3.15±0.88,(67.64±3.55) %,t =7.902,3.258,both P<0.05).However,no significant difference was observed at 24 h between radiotherapy group and the control group (2.98±0.16,(58.45±0.98) %,t =0.525,2.043,both P>0.05).The change of T/NT closely correlated with the expression of HIF-1α in both the radiotherapy group and control group(r,=0.793,0.756,both P<0.05).Conclusion 99Tcm-MNLS hypoxia imaging has potential to monitor changes of hypoxia in tumor after radiotherapy.
2.99Tcm-AnnexinⅤScintigraphy of Inferior Vena Cava Thrombus in Rabbits

Purpose To explore the features of 99Tcm-AnnexinⅤscintigraphy of venous thrombus and its feasibility of discriminating fresh venous thrombus from old one. Materials and Methods The rabbits (n=15) were randomly divided into three groups (fresh thrombus group, old thrombus group and control group). The inferior vena cava thrombus models were developed in the rabbits of thrombus groups by inserting screw cooper wire into inferior vena cava. The rabbits of control group received sham operation. 99Tcm-AnnexinⅤwas injected in the rabbits of fresh thrombus group and control group one day after operation;the same was done in the rabbits of old thrombus group 14 days after operation. Planar anterior abdominal images were obtained at 30, 60, 90 and 120 min after 99Tcm-AnnexinⅤinjection in all groups respectively. The ratios of thrombus to background of the two thrombus groups and the ratios of the area correspondent to the thrombus groups to background of the control group were calculated by ROIs counts. Then rabbits were executed, and thrombus was used for pathology examination. Results 99Tcm-AnnexinⅤuptake in thrombi was clearly visualized in all rabbits of the fresh thrombus group;whilst negative images showed in all rabbits of the old thrombus group and control group. The thrombus to background ratios of the fresh thrombus group (4.06±0.49) were higher than that of the old thrombus group (2.46±0.38), and also higher than the inferior vena cava below inferior pole of right kidney level to background ratios (2.27±0.24) of the control group (t=5.746, 7.318;P<0.05). All the thrombi of the fresh thrombus group were confirmed as fresh mixed thrombi by HE staining, and those of the old thrombus group were confirmed as old mixed organized thrombi. Conclusion 99Tcm-AnnexinⅤmay become a new acute venous thrombus imaging tracer used to discriminate fresh venous thrombus from old one.
3.Study on biflavonoids from stem bark of Daphne giraldii

Object To isolate and identify the biflavonoids from the stem bark of Daphne giraldii Nitsche. Methods The flavonoids were isolated and purified by column chromatography on silica gel and Sephadex LH-20. Their structures were determined by spectroscopic methods, including IR, 1HNMR, 13 CNMR, HMBC and FAB-MS.Results Four biflavonoids daphnodorins A-D 1 (Ⅰ-Ⅳ) were isolated from the stem bark of D. giraldii. Conclusion The above four biflavonoids were isolated from the title plant only.

6. Ginsenoside Rg1 protects HL-7702 cells against oleic acid-induced injury via aldolase/AMPK/PINK1 signalling

AIM: To study the protective effects of ginsenoside Rg1 on HL-7702 cells injury induce by oleic acid (OA), and investigate its role in aldolase/AMPK/PINK1 signalling. METHODS: HL-7702 cells were cultured in vitro and divided into five groups: control group (C), oleic acid group (OA), OA+ginsenoside (Rg1) group, OA+compound C (CC) group, and OA+CC+Rg1 group. The viability of HL-7702 cells was determined by CCK8 assay and Hoechst staining. The apoptosis and mitochondrial membrane potentials of HL-7702 cells were measured using flow cytometry. ATP content in HL-7702 cells was observed. Western blot was used to detect the expression levels of Cleaved caspase-3, PINK1, MFN2, Aldolase and p-AMPK in HL-7702 cells. RESULTS:Compared with C group, the viability of cells in OA group was significantly decreased (P<0.05), the apoptotic rate and Cleaved caspase-3 expression were greatly increased (P<0.05), ATP level, mitochondrial membrane potentials (TMRE) and PINK1 expression were significantly decreased (P<0.05). Compared with OA group, the viability of cells in OA+Rg1 group was significantly increased (P<0.05), the apoptotic rate and Cleaved caspase-3 expression were greatly decreased (P<0.05), ATP level, mitochondrial membrane potentials (TMRE) and PINK1 expression were significantly increased (P<0.05). Compared with OA+Rg1 group, the viability of cells and p-AMPK expression level in OA+CC+Rg1 group was significantly decreased (P<0.05). Reducing the expression of aldolase in cells inhibited Rg1?s actions on PINK1 and p-AMPK and cell viability. CONCLUSION: Ginsenoside Rg1 can improve the injury of HL-7702 cells via regulating aldolase/AMPK/PINK1 signaling pathway.
7.Influence of different injection techniques on the quality of bolus in 99mTc-DTPA renal dynamic imaging

Objective To explore the influence of different injection techniques on the quality of bolus in 99mTc-DTPA renal dynamic imaging.Methods 395 patients accepted 99mTc-DTPA renal dynamic imaging were retrospectively analyzed.All patients were divided into three groups according to injection techniques:direct injection group (187 cases),intravenous route injection group (84 cases)and venous indwelling needle injection group (124 cases).The three groups were injected by each technique.Areas of interest (ROI) were drawn on abdominal aorta by Xeleris workstation in blood flow perfusion imaging.The time-radioactivity curves of ROI were got.The patients whose ROI curve formed a peak was successfully injected,and did not formed was unsuccessfully injected.The number of patients in three groups who were successfully or unsuccessfully injected was respectively calculated.The data of three groups was taken Chisquare test by SPSS17.0 software.Results 174 patients of the direct injection group,46 of the intravenous route injection group and 115 of the venous indwelling needle injection group were injected successfully.The successful rate respectively was 93.0%,54.8% and 92.7%.The successful rate of the direct injection group and venous indwelling needle injection group were higher than intravenous route injection group.The difference had statistical significance.The successful rate of the direct injection group and venous indwelling needle injection group hadn't statistical significance.Conclusions The successful rates of the direct injection group and venous indwelling needle injection group were similar.The venous indwelling needle injection technique can be chosen.The successful rate of the intravenous route injection group was lower than the other two groups.The intravenous route injection technique should be chosen prudently.
8.Intervention effect of erythromycin on tumor necrosis factor-α and interleukin-8 in hyperoxia-induced lung tissue of premature newborn rats

Objective To explore the effect of macrolide antibiotics(erythromycin) on tumor necrosis factor(TNF)-α and interleukin(IL)-8 in hyperoxia-induced lung tissue of premature newborn rats,and to study the intervention effect of erythromycin on hyperoxia-induced lung injury.Methods One-day old preterm Sprague Dawley rats were randomly divided into four groups by random number table method:air + sodium chloride group,air + erythromycin group,hyperoxia + sodium chloride group,hyperoxia + erythromycin group.Hyperoxia groups were continuously exposed to oxygen (oxygen > 0.85) and air group in room air.After 1,7,14 days of exposure,the preterm rats of four groups were sacrificed,whole lung of these rats were isolated,the lung histological changes were observed by hematoxylin-eosin staining,TNF-α and IL-8 in pulmonary tissue homogenate were detected by ELISA.Results The results showed that:(1) Compared with air + sodium chloride group,TNF-α and IL-8 expression in hyperoxia + sodium chloride group were significantly increased(P < 0.05) after 1,7 days of exposure [1 d:TNF-α:(16.163 ± 0.574) ng/ml vs.(21.923 ±2.066) ng/ml,IL-8:(18.214 ±3.649) ng/ml vs.(23.546 ± 5.240) ng/ml ;7 d:TNF-α:(15.940 ±0.821) ng/ml vs.(19.688 ±0.764) ng/ml,IL-8:(18.541 ± 4.114) ng/ml vs.(24.255 ±4.692) ng/ml],in particular,TNF-α expression appeared to increase earlier,their expression became significantly weak in 14 days (P < 0.05).(2) Compared with hyperoxia + sodium chloride group,TNF-α and IL-8 expression in hyperoxia +erythromycin group became significantly weak after 1,7,14 days of exposure(P <0.05) after the intervention of erythromycin [1 d:TNF-α:(21.923 ± 2.066) ng/ml vs.(18.903 ± 1.851) ng/ml,7 d:IL-8:(24.255 ±4.692) ng/ml vs.(23.508 ±3.543) ng/ml,14 d:TNF-α:(16.443 ±5.466) ng/ml vs.(14.453 ±0.963)ng/ml],but their expression became weaker in 14 days than that in 1,7 days.Conclusion The release of inflammatory mediators TNF-α and IL-8 induced by oxidation outbreak participates in the development of hyperoxia induced lung injury,erythromycin may regulate immune function,inhibits the levels of oxidant-mediated TNF-α and IL-8 induced by oxidation outbreak,and alleviate hyperoxia lung injury in premature rats.

Objective To explore the effect of entire process nursing intervention during nebulization treatment of acute wheezing episodes in school-age children.Methods One hundred and forty-one children with acute wheezing episodes were divided into the experimental group (75 patients) and the control group (66 patients) by whether willing to accept nursing intervention or not,respectively.Routine nursing such as cognition of wheezing knowledge and respiration instruction was given to both groups,while specialists accompanied the whole procedure of inhalation treatment and they gave nursing intervention such as respiration,saliva swallow,expiration through nose,grasp of inhalation tank and body position maintenance to the experimental group.The improvement of wheezing and subjective sense by the patients,transcutaneous oxygen saturation (SaO_2),and the compliance of patients were measured and evaluated.Results Improvement of wheezing and comfort feeling were found in 97.3% of the experimental group,while 80.3% of the control group.And SaO_2 value (higher than 95%) was 92.0% in the experimental group,75.8% in the control group.The compliance of the experimental group was also better than the control group.Conclusions Entire process nursing intervention during nebulization treatment can improve clinical effect and the compliance of wheezing children.
10.Combination effect of tramadol and low dose propofol on emergence agitation in children receiving sevoflurane for adenotonsillectomy procedure

Objective To investigate the combination effect of tramadol and low dose propofol on emergence agitation in children receiving sevoflurane for adenotonsillectomy procedure. Methods Ninety patients receiving sevoflurane for adenotonsillectomy procedure were randomly divided into control group (administration of 0.1 mL/kg normal saline 30 min before the end of operation), tramadol group (administration of 1 mg/kg tramadol 30 min before the end of operation) and tramadol + propofol group (administration of 1 mg/kg tramadol 30 min before the end of operation and 1 mg/kg propofol at the end of operation). Time of extubation and time stayed in postanesthetic care unit (PACU) after operation were recorded, scores of Pediatric Anesthesia Emergence Delirium ( PAED) Scale, modified Aldrete scores and pain scores were obtained immediately after entrance into PACU, and the prevalences of post-operative nausea and vomiting were observed. Results There was no significant difference in time of extubation, time stayed in PACU and modified Aldrete Scores among groups (P >0.05). There were significant differences in scores of PAED Scale immediately after entrance into PACU, with control group > tramadol group > tramadol + propofol group (P < 0.05). The pain scores of tramadol group and tramadol + propofol group were significantly lower than that of control group (P < 0.05). The prevalence of nausea and vomiting was the highest in tramadol group, and the prevalence in tramadol + propofol group was significantly lower than that in tramadol group ( P < 0.05). Conclusion The combination use of tramadol and low dose propofol can decrease the severity of emergence agitation in children receiving sevoflurane for adenotonsillectomy procedure, and reduce the prevalence of nausea and vomiting.
