1.Treatment of traumatic brain injury with endothelial progenitor cells induced from bone marrow mesenchymal stem cells in rats

Objective To observe if endothelial progenitor cells (EPCs) and bone marrow mesenchymal stem cells (BMSCs) could home to the injured region and study the effect of BMSCs-derived EPCs on traumatic brain injury (TBI) in rats.Methods BMSCs were isolated from 3-month-old SD male rats weighing 150 g,and induced to EPCs.EPCs were identified by surface markers CD133,CD34 and FLK-1.A total of 120 female adult SD rats were divided into 3 groups (n =40 each) according to the random probability table method:EPCs group,BMSCs group and 3T3 cells group.The model of moderate to severe TBI was induced by the fluid percussion device.All the cells (i.e.EPCs,BMSCs and 3T3 cells) were injected with BrdU before transplantation to the tail vein of rats.On days 2,7,14 and 28 after transplantation,rat neurological function was evaluated using the modified neurological severity score (mNSS),and injured brain tissue was harvested to detect CD34 and brain-derived neurotrophic factor (BDNF) positive cell density using the immunohistochemistry method.Sry-positive cells were evaluated using the fluorescence in situ hybridization (FISH).Results The positive rate of CD133,CD34 and Flk-1 was 52%,33% and 38%,indicating BMSCs differentiation towards an EPCs phenotype.On days 7,14 and 28 after transplantation,the mNSS in EPCs group [(10.2 ± 1.5),(8.7 ± 1.0) and (4.9 ± 1.0) points] and in BMSCs group [(10.8 ± 1.7),(10.1 ± 1.7),and (7.2 ± 1.3) points] were lower than that in 3T3 cells group [(12.4 ± 1.5),(1 1.7 ± 1.8),(10.3 ± 1.5) points] (P < 0.05).On 14 and 28 days after transplantation,BrdU-positive CD34 and BDNF in EPCs group were significantly more than those in BMSCs group (P < 0.05).Instead,there were no positive cells in 3T3 cells group.The method of Sry probe to trace the transplanted cells could detect more positive cells compared to the BrdU labeling method.Conclusion Both BMSCs and EPCs can home to the injured region,but EPCs have much better therapeutic effect.
2.Evaluation and improvement of anti-interference performance of D-3-hydroxybutyrate detection reagent

Objective To evaluate the anti-interference performance of D-3-hydroxybutyrate (D3H ) reagent kit by using the NCCLS document EP7-A2 and to improve its anti-interference performance .Methods The anti-interference evaluation test of D3H kit was performed according to NCCLS document EP7-A2 and its anti-interference performance was improved by adding vitamin C oxidase .Results 342 mol/L free bilirubin ,342 mol/L conjugated bilirubin ,1 450 FTU chyle and 5 g/L hematoglobin had no inter-ference on the high concentration D3H detection ,0 .03 g/L vitamin C interfered with the low and high concentration D3H detection . The dosage-effect experiment results showed that vitamin C produced the linear positive interference effect on D 3H detection . Adding 2 500 U/L of vitamin C oxidase could antagonize the interference of vitamin C on detection .Conclusion Vitamin C can in-terfere with the D3H detection and adding vitamin C oxidase in reagent can antagonize its interference .
3.Cognizance on the re-fracture of the operated vertebra with cemented augmentation in osteoporotic vertebral compression fractures

One option with minimally invasive surgery for treatment of painful osteoporotic vertebral compression fracture is percutaneous vertebroplasty or percutaneous kyphoplasty via an inflatable balloon tamp followed by stabilization with polymethylmethacrylate cement.The vertebral height is restored and the decreased loading ability of the fractured vertebral body is re-strengthened with pain relief.The effect of the method is widely demonstrated in those conditions.At the same time, the relative problems, such as the complications are being lucubrated and discussed.The reports on the re-fracture or height loss of the operated vertebrae with cemented augmentation are becoming more.By now, there are some unclear aspects on the problem: the definition of the re-fracture of the augmented vertebra is still unclear and not ascertained and the clinical manifestations are various.The mechanisms and correlative factors are still not clear-cut either.The rate of subsequent re-fractures after cemented augmentation from literature is fargoing, with a rate of 0% to 63%.The correlative factors on the re-fracture of the operated vertebrae chiefly include: osteoporosis, bone necrosis caused by the heat injury from cement solidification,failure to achieve endplate-to-endplate cement augmentation, intravertebral cleft or avascular bone necrosis, premature mobilization or excessive movement, overfull reduction of the vertebral height, the biological character of the spine column, and so on.Great controversy still exists on the correlation between those possible factors and the re-fractures.More attention should be paid to this problem to explore the true risk factors and mechanism, decrease the re-fracutre rate, and elevate the clinical outcome in osteoporotic compression fractures.
4.Comparative study on lumbar microdiscectomy and small-incision discectomy for the treatment of lumbar disc herniation

[Objective]To study the efficacy and related problems of lumbar microdiscectomy and small-incision discectomy in the treatment of lumbar disc herniation.[Method]A total of 82 cases were treated with lumbar microdiscectomy,70 cases were treated with small-incision discectomy.The results of two groups were compared.[Result]Compared with the small incision group,the operative time and the complication rates were significently reduced.The effective rate between the two groups had no significent difference.[Conclusion]Small-insion discectomy is as an effective operation as microdiscectomy,while lumbar microdiscectomy had the advantages of minimal invasive,more indications,less bleeding.Strict selection of indications,sufficient preoperative preparations and practiced surgical techniques are the key points of the two minimally invasive techniques to award good outcomes.
5.Anterior microsurgical decompression and fusion via cervical disc space

[Objective]To oberserve the method and efficacy of anterior microsurgical decompression and fusion via cervical disc space in the treatment of cervical spondylotic myelopathy.[Method]Forty-three patients were treated with microsurgical decompression via cervical disc space and fusion with PEEK(Polyetheretherketone)cages.Preoperative,immediate postoperative and follow-up X-ray films were utilized to oberserve the fusion rate of the segments,and to evaluate the restoration of lordosis(Cobb's angle)and intervertebral height.[Result]All patients were followed up with an average of 15.4 months.The fusion rate was 100% and the satisfactory rate was 88.4% according to the 40 score method.Immediate postoperative X-ray films showed 6.0?improvement in lordosis(P

Objective To explore the relationship between the expression of Hypoxia-induciable factor-1α (HIF-1α) and changes of hepatitis microcimulation in patients with chronic hepatitis B (CHB).Methods 120 patients with CHB were studied. The expression of HIF-1 α was observed by immunohistochemistry and the morphotogic changes were observed with transmission eletron microscope. Results Stenosis, blockage, dilataltion, capillarization of hepatic sinusoids were the main morphotogic changes of liver. The expression of HIF-1α was positive or stronger positive. There is a positive correlation between the expression of HIF-1α and microcirculation disturbance of liver.Conclusion The disturbance of liver with CHB could exacerbate the dysbolism of nutrient substance and oxygen between hepatic cells and blood circulation. Hypoxia of hepatic cells was an important reason of the liver's pathological changes.
7.Analysis of the relationship between plasma D-dimer and hepatic functional reserve status in patients with hepatitis B

ObjectiveTo investigate the relationship between the level of plasma D-dimer(D-D) and hepatic functional reserve status in patients with hepatitis B (HB).Methods77 cases with HB were divided into two groups, hepatic function compensated group and hepatic function decompensated group.Plasma D-D, hepatic functional indexes and PT of all patients were detected.ResultsThe level of D-D in the hepatic function decompensated group was higher than that in the hepatic function compensated group(299.0 ± 498.5) μg/L vs (27.9 ± 34.0) μg/L(t =3.1, P < 0.01).D-D negatively correlated with ALB and PA(r =-0.6,-0.6, all P < 0.01), and positively correlated with TBIL and PT(r = 0.4,0.6, all P < 0.01).ConclusionThe level of plasma D-D related with hepatic functional reserve status,and detecting the level of D-D could be helpful to estimate hepatic functional reserve and prognosis of patients with HB.
8.EFFECTS OF GINSENG ROOT SAPONINS ON IMMUNOSUPPRESSION IN TUMOR-BEARING MICE

Ginseng root saponins ( GRS ) was extracted from native ginseng.The dosage used was calculated according to the content of saponins.Immunosuppression in mice was induced in 15 days after implar- ting Ehrlich ascites carcinoma (EAC) subcutaneously in armpit, and in 5-11 days after implanting EAC into peritoneal cavity. GRS administered orally 50mg/kg- for 8 days did not prevent the atrophy of the thymus in tumor-bearing mice but rather aggravated it. GRS administered orally 50mg/kg for 14 days partially restored the suppressed phagocytosis of peritoneal macrophages in tumor-bearing mice. GRS administered orally 50mg/kg for 6 days somewhat restored the suppression of hemolysin formation in tumor-bearing mice 5 days after implanting EAC, But GRS given in the same route and dosage for 9 days, showed no effect on the suppression of hemolysin formation. GRS administered orally 50mg/kg for 10 days partially restored the suppression of delayed hypersensitive reaction in tumor-bearing mice.

Objective Accumulating evidence has demonstrated that the sub-anesthetic dose of ketamine exerts rapid and ro-bust antidepressant-like effects though its action mechanisms are not yet fully understood .The aim of this study was to investigate the role of gamma-aminobutyric acid ( GABA) in the antidepressant effects of ketamine . Methods Thirty-two male Wistar rats were e-qually randomized into four groups: saline, ketamine, GABA, and GABA+ketamine.All the animals were implanted with a guide cannula into the lateral ventricle and on the eighth day after operation subjected to a 15 min forced swimming test (FST) for the estab-lishment of a depression model .At 24 h after modeling , the rats of the saline and ketamine groups were treated intracerebroventricularly with 2μL isotonic saline solution, and those of the GABA and GABA +ketamine groups with 50μg (2μL) GABA, followed by intrap-eritoneal administration of 1 mL saline in the former two groups and 10 mg/kg (1 mL) ketamine in the latter two groups 10 min later.At 30 min after treatment , the open field test ( OFT) was carried out for crossing and rearing scores and a 6-min FST was performed to re-cord the immobility time in the last 5 minutes.The content of GABA in the prefrontal cortex of the rats was measured following behav -ioral tests. Results The immobility time was significantly decreased in the ketamine group ([107.5 ±21.2]sec) as compared with the saline, GABA, and GABA+ketamine groups ([167.2 ±22.1], [159.8 ±17.5], and [143.8 ±22.1]sec) (P<0.05), with no significant difference between the GABA and GABA +ketamine groups (P>0.05).The level of GABA in the prefrontal cortex was remarkably lower in the ketamine group ([12.4 ±3.4]ng/mg prot) than in the saline, GABA, and GABA+ketamine groups ([23.3 ± 6.3], [27.3 ±5.7], and [18.0 ±5.4]ng/mg prot) (P<0.05), but markedly higher in the GABA than in the GABA +ketamine group (P<0.05).OFT scores exhibited no significant difference in the lo-comotor activity among the four groups of rats ( P >0.05 ). Conclusion The antidepressant effects of ketamine are related to the decreased GABA level in the prefrontal cortex in rats receiving FST .
