1.Expression of ?-catenin during check pouch carcinogenesis in golden hamster

Objective:To investigate ?-catenin expression during check pouch carcinogenesis in golden hamster.Methods:Expression of ?-catenin was examined with immunohistochemical SABC method in 23 cases of check normal buccal epithelium of golden hamster,22 of hyperplasitic epithelium,22 of dysplastic epithelium,29 of squamous cell carcinoma.Results:Normal buccal epithelium showed strong pericellular staining in the basal,suprabasal and prickle cell layers.Positive rates(%) of ?-catenin in normal buccal epithelium,hyperplastic epithelium,dysplastic epithelium and squamous cell carcinoma were 100,100,77.3 and 41.4 respectively.Conclusion: The decreased expression of ?-catenin may be related to the carcinogenesis of golden hamster check pouch.
2.Preparation of bifunctional anti-human lingual cancer/anti-VEGF antibody

Objective:To develop bifunctional anti-human lingual cancer/anti-VEGF antibody.Methods:Antigen of lingual cancer was prepared from Tca 8113 cells and that of VEGF from endothelial cells of umbilical cord vein.The hybridoma cells of anti-human ligual cancer/anti-VEGF were prepared by the technique of secondary hybridoma.The antibody was identified by ELISA. Results:The specific conjugation rate between bifunctional antibody and lingual cancer tissue was 85.29%, and that between the antibody and vascular endothelial cells was 82.35%.Conclusion:The bifunctional antibody of anti-human lingual cancer/anti-VEGF has good targeting potential.

4.The expression of DPC4 during carcinogenesis of golden hamaster check pouch

Objective:To elucidate the role of dpc4 gene in the development of squamous cell carcinoma. Methods:DMBA at 5 g/L was locally applied in check pouch 3 times a week for 6,9 and 12 weeks respectively in three groups of golden hamasters to induce carcinoma.The animals were sacrificed after DMBA application.Normal controls were the check pouch samples of golden hamasters without any treatment and negative controls were established by local application of acetone for 12 weeks.DPC4 4 expression was examined by immunohistochemical staining.Results:DPC4 expression was observed in all(22) cases of the normal epithelium samples(100%),20 of the 21 cases of simple hyperplasia(95.2%),20 of the 27 cases of abnormal hyperplasia(74.1%) and 12 of the 25 cases of squomous cell carcinoma(48%).Conclusion:dpc4 may play an important role in the development of oral squomous cell carcinoma.

Objective To investigate the expressions and clinical significance of Ki-67,p53,and survivin in esophageal cancer and precancerosis.Methods The expressions of Ki-67,p53,and survivin proteins were detected by immunohistochemical staining in 40 normal esophageal mucosa,136 precancerosis (42 mild atypical hyperplasia,43 moderate atypical hyperplasia,and 51 severe atypical hyperplasia),and 68 esophageal cancer tissues.The correlation of three proteins expressed in esophageal carcinoma tissues was analyzed.Results The positive expression rate of Ki-67 was 0 (0/40)for normal epithelium,35.7% (15/42) for mild dysplasia,51.2% (22/43) for moderate dysplasia,74.5% (38/51) for severe dysplasia,92.6% (63/68) for squamous carcinoma,respectively.The positive expression rate of p53 protein was 0 (0/40) for normal epithelium,28.6% (12/42) for mild dysplasia,46.5% (20/43) for moderate dysplasia,52.9% (27/51) for severe dysplasia,67.6% (46/68) for squamous carcinoma,respectively.The positive expression rate of survivin protein was 0 (0/40) for normal epithelium,38.1% (16/42) for mild dysplasia,55.8% (24/43) for moderate dysplasia,64.7% (33/51) for severe dysplasia,89.7% (61/68) for squamous carcinoma,respectively.Rank correlation analysis showed that abnormal expressions of Ki-67,p53,and survivin were correlated significantly with the pathological grading of the lesions (r =0.637,0.454,0.590,P <0.01).The expressions of Ki-67,p53,and survivin were positively correlated in esophageal carcinoma (r =0.407,0.646,P < 0.01).Conclusions The abnormal expressions of Ki67,p53,and survivin were associated with the processes of the esophageal canceration,and the joint detection with three parameters has important clinical value.
8.Cloning of human bone morphogenetic protein-2 gene and construction of recombinant adenoviral vector

Objective To clone the human bone morphogenetic protein-2(hBMP2) gene cDNA and construct its replication-deficient type 5 adenoviral vector.Methods Using osteoblastic sarcoma tissue to extract cell total RNA,hBMP-2 cDNA was amplified by RT-PCR.The hBMP-2 cDNA was then inserted into cloning vector pGEM-T.The recombinant plasmid was screened and the recombinant plasmid which was confirmed by restriction enzyme,PCR and sequence analysis was gotten.To construct the recombinant adenoviral vector,the BMP-2 cDNA was subcloned into a shuttle vector and the recombinant pShuttle-BMP2 vector was obtained.It was subcloned into Adeno-X adenoviral genomic DNA and the pAd-BMP2 DNA was obtained.Results A length of 1 200 bp BMP-2 cDNA was cloned,and it was subcloned into adenoviral vector,the recombinant adenovirus was constructed successfully.After measuring the titre of virus(1?1011 nfu?L-1),the target gene was evaluated by PCR.Conclusion The hBMP-2 adenoviral is obtained.The successful construction of pAd-BMP2 may provide us useful target for the further research.

To investigate the relationship between the expression of death associated protein kinase (DAPK) gene and oral squamous cell carcinoma (OSCC). The expression of DAPK gene in OSCC was detected by semi-quantitive RT-PCR. The results showed that the expression of DAPK gene was down regulated significantly, which may relate to the tumorigenesis and development of OSCC. The detection of DAPK gene may act as an index in OSCC dignosis.
