1.FABRICATION OF TISSUE ENGINEERED CARTILAGE USING PGA AS SCAFFOLD SEEDED WITH ALLOGENIC CHONDROCYTES

To investigate the formation of tissue engineered cartilage using polyglycolic acid (PGA) as three dimensional scaffold seeded with allogenic chondrocytes. First, chondrocytes were harvested from rib cartilage of one week old infant rabbits by collagenase digestion.The isolated cells were cultured in vitro for 2~3 generations, and then the cell suspension was concentrated. Next, the suspension was seeded onto PGA scaffolds soaked with poly l lysine. Then, scaffold chondrocyte constructs were transplanted into subcutaneous tissue of adult rabbits. Finally, the chondrogenesis was studied histologically at various periods. The results showed that chondrocytes were well distributed on the PGA scaffolds in the period of culture in vitro, and could secrete matrix around one week after seeding. 4 weeks after transplantation, the immature cartilage was regenerated in recipient animals. But some inflammatory cells surrounding the new cartilage and some PGA fibers were observed. 8 weeks after transplantation, mature cartilage with abundant matrix was obtained. PGA fibers were not found. This study proves that it is possible to regenerate new cartilage in animals possessing immune function by seeding allogenic chondrocytes onto PGA scaffold. Rejectionreaction is not strong.
2.Mechanism of AMPK-enhanced chemosensitivity of breast cancer MCF-7/adr cells to adriamycin

Background and purpose: AMP-activated protein kinase (AMPK) plays an important role in the regulation of cell metabolism and energy balance and is associated with cell proliferation, survival and multiple signaling pathways. Recent reports found that AMPK is involved in tumor suppression and drug resistance. The aim of this study was to explore the effect of AMPK on the anti-tumor effect of adriamycin and underlying mechanism in breast cancer MCF-7/adr cells. Methods:The anti-proliferative effects of adriamycin was detected by methyl thiazolyl tetrazolium (MTT) assay in MCF-7/adr, MCF-7/adr-vector and MCF-7/adr-AMPKαcells. The cell morphology in each group was stained with the lfuorescent dye Hoechst 33528, and the effects on apoptosis induction were examined by lfow cytometry (FCM). The intracellular concentration of adriamycin was detected by lfuorescence assay. The resis-tance-and apoptosis-related proteins were analyzed by Western blot. Results:The growth of breast cancer MCF-7/adr cells was inhibited by adriamycin in a dose-and time-dependent manner. The IC50 values at 24 and 48 h were (36.8±2.1) and (28.8±1.3) μg/mL, respectively. AMPKαover-expression enhanced the cytotoxic effect of adriamycin in MCF-7/adr-AMPKαcells in a dose-and time-dependent manner. Its IC50 values at 24 and 48 h were (16.0±0.7) and (4.2±0.2) μg/mL, respectively. Fluorescent morphological assay showed that AMPKαoverexpression contributed to adriamycin induced apoptosis in MCF-7/adr-AMPKαcells. After treatment with 1.0 μg/mL adriamycin for 48 h, the apoptosis rates of MCF-7/adr, MCF-7/adr-vector and MCF-7/adr-AMPKα cells were (12.0±1.4)%, (12.7±1.6)% and (32.0±4.2)%, respectively, indicating that overexpression of AMPKα enhanced the adriamycin-induced apoptosis in MCF-7/adr cells. Fluorescence microplate assay showed that over expression of AMPKαsigniifcantly increased the intracellular accumulation of adriamycin, in a concentration dependent manner. Western blot analysis showed that, compared with MCF-7/adr and MCF-7/adr-vector cells, the expressions of Bax, caspase-3 and cleaved PARP proteins were increased. Meanwhile, Bcl-2 and P-gp protein expressions were decreased in MCF-7/adr-AMPKαcells. Furthermore, the release of cytochrome c from mitochondria into the cytosol was also observed in MCF-7/adr-AMPKαcells. Conclusion:AMP-Kαoverexpression can enhance the chemosensitivity of breast cancer MCF-7/adr cells to adriamycin through inhibiting the drug effux transporter and regulating the expression of apoptosis-related proteins.
3.A STUDY OF THE EFFECTS OF PROTECTANTS WITH DIFFERENT CONCENTRATION OF GLYCEROL AND SUCROSE ON CRYOPRESERVATION OF HUMAN SPERMATOZOA

The effects on frozen-thawed sperm survival rate of 16 different cryoprotective media containing different ratio of glycerol (0%, 2.5%, 5% and 7%) and sucrose (0,25,50 and 100 mmol/L) were compared. The results showed that glycerol-sucrose cryoprotective media had better effect on cryoprotection of spermatozoa than that of traditional glycerol protective medium, and appropriately increasing the concentration of sucrose and decreasing the concentration of glycerol could improve sperm motility, and especially benefit to preserve sperm linear motility at 12h postthawed. Using 5% glycerol combined with 50 mmol/L sucrose as cryoprotective medium, the sperm survival rate at 0, 6, 12h postthawed was 85.38%, 51.22%, 33.38%, respectively, the linear moving sperm survival rate was 83.74%, 33.33%, 18.38% respectively.
4.Treatment of laryngotracheal stenosis due to relapsing polychondritis

OBJECTIVE To investigate the clinical characteristics,treatment and prognosis of laryngotracheal stenosis due to relapsing polychondritis (RP).METHODS The clinical data of 6 patients with laryngotracheal stenosis due to RP were reviewed retrospectively.RESULTS From 1996 to 2006,6 patients with laryngotracheal stenosis due to RP were managed in our hospital.Respiratory tract lesions were diffuse and extensive.Of them,4 patients accepted laryngotracheal reconstruction using surgical flaps with or without pedicle,2 accepted dilation of trachea in thoracic segments.Five patients were cured without recurrence.CONCLUSION Treatment of laryngotracheal stenosis due to RP is very difficult.If the surgical opportunity and methods were selected correctly according to each patient,good results can also be obtained.
5.Reconstruction of laryngotracheal defect using the musculo-periosteum flapof the sternocleidomastoideus with clavicular periosteum

Objective:To investigate the effect of the musculo-periosteum flap of the sternocleidomastoideus with clavicular periosteum on the reconstruction of extensiveness laryngotracheal defect. Method: Retrospectively studied 49 patients, who admited in our department from 1996 to 2005 years for severe laryngotracheal defect. There were 28 males and 21 females aged from 15 to 69 years old (mean age was 47 years old). The causes of laryngotracheal defect were laryngotracheal stenosis(31 cases) and surgery for thyroid carcinoma invading trachea (18 cases). All of 49 cases were treated with the graft of musculo-periosteum flap of the sternocleidomastoideus with clavicular periosteum and placed a silicon T-tube stenting for 3 to 6 months. Result; All of 49 cases were successfully decannulated with recuperative normal airway patency and effective phonation. The follow up ranged from 2 to 10 years, and the effect of operation was steady. Conclusion:The musculo-periosteum flap of the sternocleidomastoideus with clavicular periosteum is an ideal graft for the reconstruction of cervical extensiveness tracheal defect.

Objective To observe the feasibility of using bone marrow-derived mesenchymal stem cells (MSCs) as seed cells for tissue-engineering in Otolaryngology. Method MSCs were isolated from bone marrow of human rib and purified by centrifuge and cultured in vitro. The proliferation and growth characteristics of MSCs were observed in primary and passage culture. Result Human bone marrow-derived MSCs showed active proliferation capacity in vitro in primary and passage cultures. Conclusion Human bone marrow-derived MSCs have relatively young biologic age and they can be used as the seed cells for tissue engineering.
7.Investigation about nurses'congnition of reporting adverse events and errors voluntarily in Guangxi Province

Objective In order to know nurses'congnition of reporting adverse events and errors voluntarily by questionnaire,and then analyze the related factors. Methods Investigated 275 nurses from 25 hospitals in Guangxi Province by self-desinged questionnaire to know their cognition about reporting adverse events and errors voluntarily,analyzed the datum of invetigation. Results There was different ideas about reporting adverse events and errors voluntarily in nurses with different professional title and duty.Nurses'attitude was different under the different system of reporting adverse events and errors. Conclusions The safety awarness of nurses should be strengthened.The existing reporting system must be imported. It is necessary to establish a comprehensive reporting system of care mistakes and adverse events.
8.Use of silicon T-tube in laryngotracheal reconstruction.

OBJECTIVE:
To discuss the function and operative method of silicon T tube in laryngotracheal reconstruction.
METHOD:
Two hundred and ninety-seven patients of laryngotracheal stenosis were operated with laryngotracheal reconstruction in our department. All of patients were tracheostomy dependent before reconstruction and were placed a silicon T-tube stenting for 3 to 6 months after reconstruction.
RESULT:
Two hundred and eighty-nine patients (97.3%) were successfully decannulated with good airway patency and effective phonation. They were followed up from 1 to 10 years, and no recurrence was found.
CONCLUSION
Silicon T-tube is an effective and safe stent for laryngotracheal reconstruction. Paying attention to some application details may avoid the complication and obtain a satisfactory effect.

10. Investigation on demands for antenatal care services among 2 002 pregnant women during the epidemic of coronavirus disease 2019 in Shanghai

Objective:
To identify problems and demands for antenatal care (ANC) among pregnant women in different trimesters of pregnancy in Shanghai for optimizing ANC service during the epidemic of coronavirus disease 2019 (COVID-19).
Methods:
Organized by Maternal and Child Health Care institute in the 16 districts of Shanghai, a cross sectional study was conducted among pregnant women who came to pregnancy registration in the community health centers or attended ANC in maternity hospitals from February 7 to February 12, 2020. Consented participating women completed a semi-structured online questionnaire voluntarily. Data was analyzed using frequency and scoring, chi-square test.
Results:
A total of 2 002 valid questionnaires were collected from 183 community health centers and 67 midwifery hospitals. About 94.6% of the pregnant women worried about being infected during the COVID-19 epidemic, and 14.7% demanded for psychological consultation. Appointment ANC services were requested by 87.7% of the participants for avoiding presenting themselves in people-density places. Compared with other pregnancy trimesters, pregnant women in the second trimester were more willing to reduce the frequency of ANC (48.1% VS. 39.5% VS. 35.2%,
