1.MRI Features of Dysembryoplastic Neuroepithelial Tumor

Objective To investigate MRI features of dysembryoplastic neuroepithelial tumor(DNT).Methods The MR images of ten cases with surgically and pathologically proved DNT were retrospectively reviewed.Results MR imaging showed cystic component with mural nodule in 6,mainly cystic component with septa in 3.The solid components of the tumors were long T_1 and long T_2 signal intensity,the cystic components were even longer T_1 and T_2 signal intensity which resembled the signal of cerebrospinal fluid.Mild enhancement could be found at the intra-tumoral solid nodule or septations after injection of constrast medium but the cystic component did not show any enhancement.No peri-tumoral edema and mass effect were found in most cases.Conclusion DNTs are of some special features on the MR images,the diagnosis can be exactly done before operation if MRI features in combination with clinical data of the patients.

[Abstact] Circular RNAs(circRNAs) were once mistakenly considered as the transcription splicing intermediates , affiliate by-products or splicing errors , and the understanding of circRNAs has been significantly revolutionized with the development of high-throughput sequencing technology of RNA and the improvement of the corresponding bioinformatics analysis .CircRNAs with their unique closed-loop structure and undenible important biological functions have become new favorites in both molecular biology studies and clinical trials.To our knowledge , circRNAs usually arise after transcription , stably express in a variety of biological cells and regulate or interfere genetic expression , thus affecting occurrence and progression of diseases.Recently, circRNAs′influences upon various cancers have begun to edge up and we elaborates them in detail here based on the latest researches .
3.Research of hypoxia-inducible factor in tumor angiogenesis

Hypoxia-inducible factor (HIF) is an important transcriptional factor,which plays crucial role in response to hypoxia.HIF is highly expressed in many human cancers,and it is closely related to tumor angiogenesis.HIF promotes tumor growth,invasion and metastasis via up-regulating vascular endothelial growth factor gene and starting associated angiogenesis.With the in-depth study of HIF regulation of tumor angiogenesis mechanism,HIF is expected to become an important target for anti-angiogenesis therapy.
4.Clinical and imaging evaluation in children with congenital airway abnormalities

Objective To analyse clinical and imaging characteristics of congenital airway abnormalities,and to guide the clinical diagnosis and reduce rate of missed diagnosis and misdiagnosis.Methods Clinical and imaging characteristics in children who had been diagnosed congenital airway abnormalities in Shengjing Hospital of China Medical University from 2005 to 2011 were reviewed and analysed retrospectively.Results Of 23 children,12 cases were male,11 cases were female,the age was from 1 month to 10 years,the average age was 32 months.The disease course was from 3 days to 3 months,an average was 18 days.Clinical manifestations were repeated coughing,wheezing,dyspnea and recurrent respiratory infection.Through the spiral CT and three dimensional reconstruction,enhanced CT examination,6 cases of tracheobronchial stenosis,3 cases of trachea and bronchus,1 case of dysplasia of the left pulmonary artery and vein,3 cases of pulmonary sequestration,4 cases of pulmonary cyst and 6 cases of pulmonary hypoplasia were found.Conclusion Missed diagnosis and misdiagnosis of congenital airway malformations often occurs.When the following features were observed,congenital airway malformations may be taken into account:persistent cough; repeated or persistent wheezing,treated ineffectively with anti-inflammatory agent and bronchodilators ; recurrent or persistent stridor; repeated pulmonary infections,dyspnea; repeated or persistent pneumonia in the same part and unexplainedly mediastinal shift in X ray chest radiography.Spiral CT three dimensional reconstruction and enhancement is of great value in diagnosis of airway malformation.
5.Protective effect of Akt on gastric ischemia-reperfusion injury by microinjection of oxytocin into the paraventricular nucleus in rats

Objective To observe the effect of microinjection of oxytocin(OT) into paraventricular nucleus(PVN) on gastric ischemia-reperfusion(GI-R) injury and its molecular mechanism.Methods GI-R injury was induced in rats by clamping the celiac artery for 30 min and followed by reperfusing for 1 h.A cannula was inserted into the unilateral PVN for microinjection of OT.The gastric mucosal injury index was counted grossly.The expressions of Akt and caspase-3 in rat gastric mucosa were examined by Western blot and by immunohistochemistry.Results Microinjection of OT into PVN dose-dependently allevated gastric mucosal injury subjected to GI-R.Microinjection OT into PVN significantly increased the expression of Akt protein and decreased the level of caspase-3 in gastric mucosal following GI-R.The effects of OT were prevented by pretreatment with OT receptor antagonist atosiban into the lateral cerebral ventricle.Conclusion Microinjection of OT into PVN significantly protected against GI-R injury.These effects of OT are mediated by its receptors.The mechanisms are mediated by increasing Akt expression,which in turn inhibits caspase-3 expression.
6.Optimization and application of protein C-terminal labeling by carboxypeptidase Y.

Proteolytic cleavage is one of the post-translational modifications and plays important roles in many biological processes, such as apoptosis and tumor cell metastasis. The identification of the cleavage events can improve our understanding of their biological functions in these processes. Although proteomic approaches using N-terminal labeling have resulted in the discovery of many proteolytic cleavages, this strategy has its own inherent drawbacks. Labeling of protein C-termini is an alternative approach. Here, we optimized the labeling procedure in the profiling protein C-termini by enzymatic labeling (ProC-TEL) and improved the labeling efficiency for the positive isolation of protein C-terminal peptides and mass spectrometric identification. We applied this approach to a complex protein mixture from Escherichia coli and identified many C-terminal peptides and internal cleaved peptides from more than 120 proteins. From the identified cleavages, we found several previously known internal proteolytic cleavage sites and many novel ones which may play roles in regulating normal biological processes. This work provides a potential new way, complementary to the N-terminomics, for the identification of proteolytic cleavages in complex biological systems.

7.The clinical features of Hashimoto thyroiditis with thyroid cancer

Objective To analyze the clinical features of Hashimoto thyroiditis with thyroid cancer, and provide scientific basis for clinical diagnosis and treatment. Methods The clinical data of 87 patients of Hashimoto thyroiditis with thyroid cancer and 105 patients of Hashimoto thyroiditis were retrospectively analyzed. Results The rates of Hashimoto thyroiditis with thyroid cancer in age <30, 30-39, 40-49, 50-59 and≥60 years were 1/3, 47.5%(29/61), 51.4%(38/74), 36.0%(18/50) and 1/4. The rates of Hashimoto thyroiditis with thyroid cancer in patients of age 30-39 years and 40-49 years were higher than that in patients of age ≥60 years, but there were no statistical differences (χ2=0.327 and 0.418, P>0.05). There were statistical differences in total thyroxine (TT4), thyroid stimulating hormone (TSH), thyroglobulin antibody (TgAb) and thyroid peroxidase antibody (TPOAb) levels between Hashimoto thyroiditis with thyroid cancer and Hashimoto thyroiditis, there were statistical differences (P<0.05 or <0.01). Multiariable Logistic regression analysis result showed that the decreased serum level of TT4 and the increased levels of TSH, TgAb and TPOAb were the correlated factors of Hashimoto thyroiditis with thyroid cancer (P<0.05 or <0.01). Conclusions Low level of TT4 and high levels of TSH, TgAb and TPOAb may increase the risk of Hashimoto thyroiditis with thyroid cancer. The 30-49 years male patients with Hashimoto thyroiditis should be highly suspected of the possibility of merger thyroid cancer.
8.Radiosensitizing effects of miR-101 on HeLa cancer cells and underlying mechanism

Objective To study the effects of microRNAl01 (miR-101)on radiosensitization of human uterine cervix cancer HeLa cells and underlying mechanism.Methods HeLa cells were divided into three groups including blank control,miRNA negative control and miR-101 transfection group.The cells were irradiated by 160 kVp X-ray generated from a linear accelerator at a dose rate of 1.15 Gy/min.Real-time quantitative PCR (qRT-PCR) was used to detect the expression of miR-101.The clonogenic survival assay was applied to evaluate the effect of miR-101 on radiosensitization of HeLa cells.γ-H2AX immunofluorescence and Western blot assays were performed to observe DNA double-strand breaks and the protein expressions of ATM and DNA-PKcs of HeLa cells,respectively.Results Compared with the negative control group,the expression of miR-101 was significantly increased in the HeLa cells at 48 h after transfection with miR-101 mimic,and the survival of HeLa cells over expression of miR-101 was significantly reduced(t =10.75,P < 0.05).The miR-101 had remarkable radiosensitive effect on HeLa cells(F =7.72,P <0.05) with a SERD0 of 1.29.Moreover,over-expression of miR-101 could inhibit the repair of DNA damage induced by irradiation.Compared with the control group,the protein expressions of ATM and DNA-PKcs were significantly decreased in the HeLa cells over expression of miR-101.Conclusions Over-expressions of miR-101 could inhibit cell growth and enhance radiosensitivity of HeLa cells by inhibiting the repair of radiation-induced DNA damage.
9.Analysis of peer-assisted learning on critical thinking ability of undergraduate nursing students

Objective To analyze the implementation effects of peer-assisted learning on critical thinking ability of undergraduate nursing students. Methods A total of 411 college nursing students were selected with 203 students of Class One, Class Three, and Class Five from Grade 2012 majoring in nursing were taken as the control group, 208 students of Class Two, Class Four, and Class Six as the experimental group. In the course of basic nursing practice, the control group was trained by the traditional practice, and the experimental group was trained by the way of peer mutual aid. Critical thinking disposition inventory Chinese version (CTDI-CV) was adopted to evaluate the level of critical thinking ability of the nursing students of both groups. SPSS 17.0 was used and the data of the two groups were compared by t test. Results The score of experimental group students' critical thinking ability and the score of 7 dimensions including their seeking truth, analysis ability, self-confidence, inquisitiveness, cognitive maturity, open mind and systematic ability were higher than those before the experiment (P<0.05). The total score of the experimental group students' critical thinking ability and the score of 5 dimensions including seeking truth , analysis ability, self-confidence, curiosity and cognitive maturity were higher than those of the control group (P<0.05). Conclusion Peer-assisted learning is beneficial to the improvement of critical thinking ability of nursing undergraduate students.
10.The feasibility study of radiation absorbed dose measurement using oligonucleotide dually labeled with Alexa Fluor 488 and BHQ1 probe

Objective To study the feasibility of measuring radiation absorbed dose with the fluorescent probe Alexa Fluor 488-DNA-BHQ1.Methods An oligonucleotide dually labeled at its 5'-and 3'-end with fluorescent molecular Alexa Fluor 488 and specific fluorescence inhibitors BHQ1 was prepared.The Alexa Fluor 488-DNA-BHQ1 aqueous solution was exposed with X-ray and its fluorescence intensity was measured.Results When the concentration of Alexa Fluor 488-DNA-BHQ1 was between 0.5 and 1 μmol/L,the fluorescence intensity of its aqueous solution had excellent linear dose response from 0.1 to 30 Gy (R2 =0.99) and it was stably maintained after 40-80 min of irradiation especially at 4℃.Conclusions In the dose range of 0.1-30 Gy,the Alexa Fluor 488-DNA-BHQ1 fluorescent probe can be used to measure radiation absorbed dose.
