1.Evaluation of the efficacy and safety of Qingre Zhibu Decoction combined with psychological intervention in the treatment of acute gout

Objective To Qingre ChuBi decoction combined with psychological intervention efficacy and safety in the treatment of patients with acute gout were analyzed. Methods In our hospital from March 2014 to November 2016 received 68 cases of acute gout patients as the research object, according to patients were randomly divided into control group and treatment group, 34 cases in the control group were treated with Etoricoxib Tablets and psychological intervention treatment, 34 cases of patients in the treatment group in the control group plus Chubi decoction for clearing. Results Two groups of patients before treatment VAS score were compared, no significant difference; The VAS score after treatment compared with before treatment were significantly decreased (P<0.05), comparison between groups had significant differences; compared with control group, VAS score improved more obviously (P<0.05), comparison between groups had significant differences. Conclusion Qingre ChuBi decoction combined with psychological intervention has a good effect on the treatment of acute gout, high safety, worthy of clinical application.
2.MicroRNA-1 and-16 inhibit cardiomyocyte hypertrophy by targeting cyclins/Rb pathway

AIM:MicroRNAs ( miRNAs) were recognized to play significant roles in cardiac hypertrophy .But, it remains unknown whether cyclin/Rb pathway is modulated by miRNAs during cardiac hypertrophy .This study investigates the potential roles of microRNA-1 (miR-1) and microRNA-16 (miR-16) in modulating cyclin/Rb pathway during cardiomyocyte hypertrophy .METHODS:An animal model of hypertrophy was established in a rat with abdominal aortic constriction (AAC).In addition, a cell model of hypertrophy was also achieved based on PE-promoted neonatal rat ventricular cardiomyocyte .RESULTS:miR-1 and-16 expression were markedly de-creased in hypertrophic myocardium and hypertrophic cardiomyocytes in rats .Overexpression of miR-1 and -16 suppressed rat cardiac hypertrophy and hypertrophic phenotype of cultured cardiomyocytes .Expression of cyclins D1, D2 and E1, CDK6 and phosphorylated pRb was increased in hypertrophic myocardium and hypertrophic cardiomyocytes , but could be reversed by enforced expression of miR-1 and -16.CDK6 was validated to be modulated post-transcriptionally by miR-1, and cyclins D1, D2 and E1 were further validated to be modulated post-transcriptionally by miR-16.CONCLUSION: Attenuations of miR-1 and -16 provoke cardiomyocyte hypertrophy via derepressing the cyclins D1, D2, E1 and CDK6, and activating cyclin/Rb pathway.
3.MEF2C mediates the effect of microRNA-214 on inhibiting cardiomyocyte hypertrophy

AIM:To investigate the effect of miR-214 on cardiomyocyte hypertrophy and the expression of the potential target genes . METHODS:A cell model of hypertrophy was established based on angiotensin-Ⅱ( Ang-Ⅱ)-induced neonatal mouse ventricular car-diomyocytes (NMVCs).Dual luciferase reporter assay was performed to verify the interaction between miR-214 and the 3’ UTR of MEF2C.The expression of MEF2C and hypertrophy-related genes at mRNA and protein levels was determined by RT-qPCR and Wes-tern blotting, respectively.RESULTS:The expression of ANP, ACTA1,β-MHC and miR-214 was markedly increased in Ang-Ⅱ-in-duced hypertrophic cardiomyocytes .Dual luciferase reporter assay revealed that miR-214 interacted with the 3’ UTR of MEF2C, and miR-214 was verified to inhibit MEF2C expression at the transcriptional level .The protein expression of MEF2C was markedly in-creased in the hypertrophic cardiomyocytes .Moreover, miR-214 mimic, in parallel to MEF2C siRNA, inhibited the expression of hy-pertrophy-related genes in Ang-Ⅱ-induced NMVCs.CONCLUSION:MEF2C is a target gene of miR-214, which mediates the effect of miR-214 on attenuating cardiomyocyte hypertrophy .
4.Anti-oxidative Stress Effect of Lycopene on Motor Function after Spinal Cord Injury in Rats

Objective To observe the influence of anti-oxidative stress effect of lycopene (LP) on motor function after spinal cord injury (SCI) in rats. Methods 36 healthy adult Sprague-Dawley rats were randomly divided into Groups A, B and C with 12 rats in each group.SCI model was made by Allen's mode (10 g×25 mm) on T9. Group A received no treatment, 30 mg/kg methylprednisolone sodium succinate was injected for Group B in peritoneal cavity 30 min after SCI, 20 mg/kg LP was given orally for Group C. On the 1st d, 3rd d and 7th d after operation, the motor function was assessed by slanting board test and Basso-Beattie-Bresnahan scale (BBB); superoxide dismutase (SOD) and malondialdehyde (MDA) were determined in serum. Results Compared with Group A, the scores of the slanting board test significantly increased in Group C 1 d, 3 d, and in Group B 1 d, 3 d and 7 d after operation (P<0.05); the BBB scores significantly increased in Group C 1 d, 3 d and 7 d, and in Group B 1 d, 7 d after operation (P<0.05); the activity of SOD significantly increased in Group C 1 d, 3 d and 7 d, and in Group B 3 d, 7 d after operation (P<0.05); the content of MDA significantly decreased in Group C 3 d, 7 d, and in Group B 1 d, 7 d after operation (P<0.05). Conclusion Lycopene can reduce the level of oxidative stress and promote the motor function in rats after acute SCI.

6.Construction of postoperative prognosis model for patients with colorectal cancer

Objective:To screen the factors influencing overall survival (OS) of patients undergoing radical resection for colorectal cancer (CRC) and to construct a prognostic model for OS of patients after CRC.Methods:The clinical data of 350 patients with stage Ⅰ-Ⅳ CRC who underwent radical resection in the People's Hospital of Wuhan University from March 2017 to December 2019 were collected retrospectively. Patients were divided into subgroups 0 ( n=70), 1 ( n=172), and 2 ( n=108) according to different preoperative systemic inflammation score (SIS). The relationship between different SIS, neutrophil to lymphocyte ratio (NLR), lymphocyte to monocyte ratio (LMR), systemic immune inflammation index (SII) and prognosis of CRC patients undergoing radical surgical resection were analyzed, and Cox regression models were used to perform univariate and multifactorial analyses of factors affecting patient prognosis, and column line graph models were constructed based on the results of multifactorial analyses. Results:By the deadline of follow-up, 80 of 350 CRC patients died, and the 5-year OS rate was 77.14%. The 5-year survival rates of patients in SIS group 0, group 1 and group 2 were 95.71%, 79.65% and 61.11% respectively, with a statistically significant difference ( χ2=30.19, P<0.001). Statistically significant differences in age ( χ2=19.40, P<0.001), tumor site ( χ2=8.18, P=0.017), T stage ( χ2=10.01, P=0.007), TNM stage ( χ2=14.80, P=0.001), tumor diameter ( χ2=13.91, P=0.001) and carcino-embryonic antigen (CEA) level ( χ2=10.12, P=0.006) among patients in SIS group 0, group 1 and group 2. The 5-year OS rates of patients in the low NLR and high NLR groups were 82.67% and 56.16% respectively, with a statistically significant difference ( χ2=24.96, P<0.001) ; the 5-year OS rates of patients in the low LMR and high LMR groups were 66.85% and 88.17% respectively, with a statistically significant difference ( χ2=22.45, P<0.001) ; the 5-year OS rates of patients in the low SII and high SII groups were 86.14% and 69.02% respectively, with a statistically significant difference ( χ2=14.76, P<0.001). Univariate analysis showed that age ( HR=2.58, 95% CI: 1.54-4.32, P<0.001), T stage ( HR=2.41, 95% CI: 1.24-4.68, P=0.009), N stage ( HR=3.03, 95% CI: 1.85-4.94, P<0.001), TNM stage ( HR=3.61, 95% CI: 2.15-6.04, P<0.001), nerve invasion ( HR=1.97, 95% CI: 1.27-3.08, P=0.002), vascular invasion ( HR=2.31, 95% CI: 1.49-3.59, P<0.001), preoperative SIS 1 score ( HR=5.09, 95% CI: 1.57-16.56, P=0.007), SIS 2 score ( HR=11.05, 95% CI: 3.42-35.65, P<0.001), NLR ( HR=2.97, 95% CI: 1.90-4.64, P<0.001), LMR ( HR=0.31, 95% CI: 0.19-0.52, P<0.001), and SII ( HR=2.50, 95% CI: 1.54-4.06, P<0.001) were all independent influence factors affecting the postoperative prognosis of CRC patients undergoing radical surgical resection; multivariate analysis showed that age >60 years ( HR=2.27, 95% CI: 1.31-3.91, P=0.003), TNM stage Ⅲ-Ⅳ ( HR=7.08, 95% CI: 1.89-26.59, P=0.004), and preoperative SIS 2 score ( HR=4.02, 95% CI: 1.09-14.83, P=0.037) were all independent risk factors affecting the postoperative prognosis of CRC patients undergoing radical surgical resection. The nomogram model built based on the screened variables has high prediction accuracy: the C-index of the nomogram was 0.75. Conclusion:Age>60 years old, TNM stage Ⅲ-Ⅳ, SIS 2 score are all independent risk factors for postoperative prognosis of colorectal cancer. The nomograph model constructed by this method has high prediction accuracy.
7.Blood lipid and hematoma enlargement and outcome in patients with intracerebral hemorrhage

The correlation between blood lipid and hematoma enlargement is a research hotspot at present. This article reviews the relationship between blood lipid and hematoma enlargement and outcomes in patients with intracerebral hemorrhage, as well as the correlation between statin treatment and hematoma enlargement.

9. Genetic characterization analysis of 2B genotype rubella virus strains in Liaoning province

Objective:
To analyze the genetic characterization of 2B genotype rubella virus strains first isolated in Liaoning province.
Methods:
Vero/Slam cells were used to isolate rubella viruses from throat swabs. Fragments of the E1 gene (739 nucleotides) were amplified by reverse transcription-polymerase chain reaction (RT-PCR) and the PCR products were sequenced and analyzed. The phylogenetic trees were constructed with rubella 2B genotypes reference strains.
Results:
RVi/Liaoning.CHN/13.18/1[2B] was isolateded from the case which had a history of international travel and probably imported from abroad. RVi/Liaoning.CHN/13.18/1[2B] was in the same 1ineage with RVs/Edinburgh.GBR/6.12[2B] and RVs/Kawasaki.JPN/17.11/1[2B], but far from the epidemic strains isolated in China in recent years. RVi/Liaoning.CHN/14.18/1[2B] belonged to the same lineage with rubella virus 2B genotype strains isolated in China from 2014 to 2015, and had the highest homology with RVi/Chouzhou.Anhui.CHN/17.15/2[2B] and RVi/Shaoyang.Zhejiang. CHN/13.13[2B].
Conclusions
It was the first time to isolate the 2B genotype rubella virus in Liaoning province and the strain belonged to different 1ineage.
10.Statistical analysis of adverse drug reactions of Trastuzumab for injection from 2018 to 2023 in Beijing

OBJECTIVE To provide evidence for the safe clinical application of Trastuzumab for injection. METHODS Reports of adverse drug reaction (ADR) related to Trastuzumab for injection submitted in Beijing from June 2018 to May 2023 were collected. Statistical analyses were performed using SPSS 25.0 software. The gender and age of patients with ADRs, outcome, types of cancer, and the time of onset, severity, affected organs/systems of ADRs were included for analyses. The univariate Logistic regression analysis was conducted on patient fever and ADR prognosis. RESULTS A total of 195 patients with 318 instances of ADRs were included in the study. Women (87.69%) and patients aged 60-69 (33.85%) were more likely to experience ADRs. Breast cancer dominated (86.67%) in terms of cancer types; in terms of outcomes, most patients (67.69%) showed improvement, and some patients (27.69%) achieved full recovery. Overall, 68.72% of ADRs mainly occurred on the day of medication, and 95.38% of ADRs were of “moderate” severity. The most affected organs/systems were general diseases and various reactions at the administration site (40.57%), with chills (18.87%) and fever (18.24%) being the most common. Univariate Logistic regression analysis showed that fever mostly occurred within 1 day of medication (OR=5.63, 95%CI was 2.26-14.02, P< 0.001). The time of onset of ADR greater than 1 day was a risk factor for poor ADR prognosis (OR=20.08, 95%CI was 2.45- 164.43, P=0.005), mainly manifesting as bone marrow suppression and liver function abnormalities. Neutrophilia, cardiorespiratory arrest, and mixed liver damage were new ADRs not recorded in the drug’s instructions. CONCLUSION Women and patients aged ≥60 are high-risk groups for ADRs of Trastuzumab for injection. Chills and fever remain the most common ADRs of this drug, and these symptoms mostly occur within 1 day of medication, which have better prognoses. Close attention should be paid to the patient’s temperature changes on the day of drug infusion, with timely intervention. Regular monitoring of the patient’s hematological indicators is necessary to detect any bone marrow suppression and liver function abnormalities after medication.
