Therefore,the expression level and activity of glyoxalase I are essential to pathogenesis of AD.

Objective To study the effects of transient cerebral ischemic reperfusion(IR) and flunarizine on the expression of plasminogen activator inhibitor-1(PAI-1) protein in brain tissue of gerbils.Methods A cerebral transient ischemic-reperfusion model in gerbils was established by clamping both common carotid.The gerbils were fed with flunarizine before experiment in flunarizine treatment group.The expression of PAI-1 protein in brain tissue was detected by immunohistochemistry technique at the 1,3 and 7 days after IR.Results There was slight expression of PAI-1 protein in the brain tissue of gerbils in the normal control group and sham operation group.Compared with normal control group,the expression of PAI-1 protein in neurons and gliacytes of ischemic gerbils was evidently increased at the 1,3 and 7 days in the cerebral ischemia group(all P
3.Relationship between coexpression of EGFR and ERK2 and activation of ERK2 in lung squamous cell carcinomas

AIM: To evaluate the relationship between coexpression of EGFR and ERK2 on the level of protein and mRNA and activation of ERK2 in lung squamous cell carcinomas (SCC). METHODS: Twenty cases of lung squamous cell carcinomas and their paired normal lung tissues were studied. The expression of mRNA and protein of EGFR and ERK2 was measured by reverse transcription PCR (RT-PCR) and Western blotting, respectively. The activity of ERK2 was detected by immunoprecipitation and kinase reaction. RESULTS: The results showed that the expression of EGFR mRNA and protein in paired normal lung tissues was hardly detected by RT-PCR and Western blotting, respectively, but in 20 cases of lung SCCs their expression was upregulated significantly. Compared with normal lung tissues, the expression of ERK2 mRNA and protein in lung cancer tissues was also upregulated significantly, the latter increased about 3-5-fold, and the activity of ERK2 in lung SCCs was also markedly activated. CONCLUSIONS: These data highlight that the overexpression of ERK2 and EGFR protein occurs simultaneously in lung SCCs, which mainly results from the increasing level of gene transcription, the increased activity of ERK2 in lung cancer tissues perhaps attributes to the upregulation of coexpression of EGFR and ERK2. 4.Inhibiting role of polydatin to expression of intercellular adhesion molecule-1 induced by lipopolysaccharide on vascular endothelial cells

AIM and METHODS: To investigate expression of intercellular adhesion molecule-1(ICAM-1) on human umbilical vein endothelial cells (HUVEC) induced by lipopolysaccharide (LPS) , and inhibiting role of polydatin by cellular immune fluorescent staining and laser confocal microscope scanning technology. RESULTS: Compared with basic expression of ICAM-1 on HUVEC, the ICAM-1 expression was enhanced significantly after stimulated by LPS from 8 h to 36 h, dose-dependent relation appeared between expression of ICAM-1 and LPS. ICAM-1 expression on endothelial cells treated only by polydatin had no abvious change,but inducing role of LPS to expression of ICAM-1 was inhibited significantly by polydatin pretreating endothelial cells. CONCLUSION: The expression of ICAM-1 on endothelial cells can be promoted by LPS , and polydatin can inhibit LPS-induced ICAM-1 expression

Objective To explore the relationship between the expression of CD44v6,P16 protein and the invasion and metastasis in gastric carcinoma and to investigate the correlation between the expression of CD44v6 and P16 protein.Methods Immunohistochemical methods were used to detect the expression of CD44v6 and P16 protein in 110 cases of gastric carcinoma.Results In 110 cases of gastric carcinoma,the expression of CD44v6 and P16 protein were 72.7%(80/110)and 47.3%(52/110)respectively. AIM: To study the change of intercellular adhesion molecule-1(ICAM-1) expression in intestine tissues of mice induced by LPS and regulatory effect of p38 mitogen-activated protein kinase (p38 MAPK) on ICAM-1 expression. METHODS: Protein and mRNA of ICAM-1 were measured using Western blotting and RT-PCR respectively in intestine tissue of BALB/c mice treated by lipopolysaccharide(LPS) or LPS plus SB203580, a specific inhibitor of p38 MAPK. RESULTS: Compared with control group, the expression of ICAM-1 protein and mRNA was increased significantly by LPS stimulation in dose- and time-dependent manner. ICAM-1 expression reached peak value at 12-36 h after LPS stimulation. 20.0 mg/kg of LPS could induce the maximum of ICAM-1 expression. Pretreatment of mice with SB203580 for 30 min could inhibit significantly LPS-induced expression of ICAM-1 protein and mRNA expression in mouse intestine tissues. CONCLUSIONS: These data highlight that LPS could up-regulate ICAM-1 protein and mRNA expression in intestine tissue of mice in dose- and time-dependent manner, and p38 MAPK signal pathway plays an important role in ICAM-1 expression induced by LPS. It suggests that inhibition of p38 MAPK might be a useful principle for the prevention and treatment of intestine damage of endotoxic shock.
7.Evaluation of postoperative radiotherapy for parotid gland carcinoma

Objective: To evaluate the efficacy of postoperative radiotherapy in the treatment of parotid gland carcinoma. Methods: Eighty-six postoperated patients with parotid gland carcinoma( 7 in stage Ⅰ, 28 in stage Ⅱ, 33 in stage Ⅲ and 18 in stage Ⅳ) were radiated by 60Co ?-ray or linear accelerator X-ray combined with electron beam. All patients were diagnosed by pathology and followed up for more than 5 years. Results: The five year survival rate and the local control rate were 73.3% and 87.2% respectively. Poor prognosis was observed in the cases with the neoplastic classification of undifferentiated carcinoma, sequamous cell carcinoma and malignant pleomorphic adenoma, but the better prognosis was obtained in the cases with acinic cell carcinoma and mucoepidermoid carcinoma. Poor prognosis was observed in the cases with stage Ⅲ and Ⅳ of clinical stage. Radiotherapy undertaken in 2 weeks after surgical operation gave higher 5- year survival ratio( 83.8%). The group given 51~60 Gy radiation showed 82.9% of five- year survival rate. Conclusion: The combination of surgery with radiation is effective in the treatment of parotid gland carcinoma.Radiation of 51~60 Gy 2 weeks after operation may result in better prognosis. Neoplastic type and clinical stage are important factors for prognosis.
8.Determination of polyblend miscibility by dilute-solution viscometry

BACKGROUND: Polyblends composed of thermoplastic polyurethane elastomer (TPU) and bisphenol A type of polysulfone (PSF) or polymethyl methacrylate (PMMA) can improve the properties of TPU, bisphenol A type of polysulfone or PMMA and can widen the scope of application. The property of polyblends bad or good depends on the miscibility of polyblends. The dilute-solution viscometry (DSV) is a simple and rapid way for determining the miscibility for polyblends. The study on the determination of miscibility for TPU/PSF and TPU/PMMA polyblends by the DSV has not been reported up to now. OBJECTIVE: To differentiate the miscibility of TPU/PSF polyblend and that of TPU/PMMA polyblend, and at the same time to verify the consistency of μ and α criteria in the determination of polyblend miscibility by DSV.DESIGN: Observation and contrast analysis based on the two polyblend systems.SETTING: Chemical Engineering and Pharmaceutics College, Henan University of Science and Technology.MATERIALS: TPU sample was purchased from Luoyang Jiming Chemical Industry Limited Company, China. PSF sample was purchased from the First Plastic Factory of Dalian, China. PMMA sample was synthesized by our laboratory. N, N-dimethylformamide was provided by Beijing Chemical Plant.METHODS:This study was performed at the Key Laboratory of Polymer Science and Nano-technology, Henan University of Science and Technology in May 2006. The different molar ratio of TPU / PSF and TPU / PMMA polyblends were prepared in N, N-dimethylformamide. An ubbelohde dilution viscometer (whose inner diameter was between 0.5 mm and 0.6 mm) was employed for measuring the relative viscosities of the polymer solutions in DMF at (25±0.01) ℃. The second stop-clock was used to record efflux time. From the efflux time, the relative viscosities could be obtained. Then the specific viscosities could be given by the relative viscosities value. From a series of relative viscosities, the specific viscosities and other data of two polyblends, μ values and α values of two polyblends were obtained.MAIN OUTCOME MEASURES: The efflux time of two polyblend solutions in an ubbelohde viscometer.RESULTS: The μ and α values of TPU/PSF polyblend were all above zero, which showed that TPU/PSF polyblend was miscible. But the μ and α values of TPU/PMMA polyblend were all below zero, which showed that TPU/PMMA polyblend was immiscible. CONCLUSION:The μ criterion is consistent with the α criterion in judging for polyblend miscibility. DSV method is simple, and it can be used in the determination of polyblend miscibility.
9.Comparison of mental health between aircrew and ground crew at ratio of 1:1 among 122 army airmen

BACKGROUND: The training task for aircrew of army airmen is very heavy, the evaluation of flight personnel on psychological diathesis is performed to early discover and deal with corresponding questions.OBJECTIVE: To compare the difference of mental health between aircrew and ground crew of army airmen and provide reference data for implementing corresponding norms.DESIGN: Chester sampling investigation and paired design were applied.SETTING: the 71687 Force of Chinese PLA, the 71521 Force of Chinese PLA, Department of Epidemiology, Fourth Military Medical University of Chinese PLAPARTICIPANTS: By chester sampling, 122 aircrew and 122 ground crew of one army airmen in one stationed training troop were randomized in February 2001. The pairs were made at ratio of 1:1 according to age difference less than 3 years and same sex and nationality. The investigated persons were all male, of Han and aged varied from 23 to 52 years.METHODS: Cornell health questionnaire was adopted respectively in investigation of aircrew and ground crew of army airmen. The investigated persons finished and handed in questionnaire independently on the spot in manner of secretion and centralized filling-up. Cornell health questionnaire is composed of 18 sections, including 195 questions, involving 4 aspects,named somatic symptoms, medial and family history, general health and habit and mental symptoms. The total score of Cornell health questionnaire ≥ 30 indicates somatic and mental disturbance and the score of mental section ≥ 10 indicates mental disturbance, both of which are taken as the criteria in comparison of psychological health in receptors of two groups.MAIN OUTCOME MEASURES: Every factor score and total score in somatic and mental sections in Cornell health questionnaire for aircrew and ground crew.RESULTS: The tested results of each 122 cases in either aircrew group or ground crew group entered result analysis. Evaluated results of somatic and mental symptoms in receptors of two groups: the total score was low in Cornell health questionnaire (0-14 scores), of which, the total score in aircrew group was higher than that in ground crew group (81.5% and 59.8%,P < 0.05); the score in mental section was low (0-9 scores), of which, that in aircrew group was higher remarkably than that in ground crew group (99.2% and 91.8%, P < 0.01); the score for the receptors with somatic and mental disturbance (total score≥30, score in mental section ≥ 10) in aircrew group was lower than those in ground crew group (4.1% and 12.3%, P < 0.05); the score for the receptors with mental disturbance (score in mental section≥ 10) in aircrew group was lower remarkably than ground crew group (0.8% and 8.2%, P < 0.01). For somatic section in aircrew group, except the factors of muscle, skeleton and skin, their scores were higher than those in ground crew group (P > 0.05), the scores of the rest factors were all lower than those in ground crew group, of which, the difference was significant in evaluation of factors of eye, ear, nervous system, urinary reproductive system and fatigue in comparison of two groups (P < 0.05). The score of every factor in mental section of aircrew group was lower than those in ground crew group. Except tension factor, in comparison of the rest factors in two groups, the significant difference was present (P < 0.05).CONCLUSION: Overall psychological health state in Cornell questionnaire of aircrew in army airmen was superior to ground crew. Aircrew still presents a part of psychological health questions, mainly in somatic section and inadaptability was the most positive response in mental section, suggesting that aircrew probably presents inadaptable phenomena.
10.Comparative analysis of the risk factors of ischemic stroke in the middle-aged and youth

BACKGROUND: Ischemic stroke has a younger tendency; hence to explore the risk factors of stroke in the middle-aged and youth for primary rehabilitation has extremely important clinical significance.OBJECTIVE: To investigate the risk factor-of stroke in the middle-aged and youth.DESIGN: A case-control analysis based on diagnose.SETTING: Department of neurology in a university hospital.PARTICIPANTS: Totally 100 hospitalized patients including the middle-aged and youth ( < 44 years old, n = 63 ) and the pre-elderly (45 - 49years old, n = 37) with mean age of(42. 91 ±6.8) were selected into stroke group from the Department of Neurology, Xiangya Second Hospital, Central South University between July 1997 and December 2000. Another 100 surgical patients without history of cerebrovascular diseases aged(42.70 ± 2.98) including the middle-aged and youth( n = 61 ) and the pre-elderly( n = 39) were collected randomly from the same corresponding period as control gruop.METHODS: A comparative study of 12 risk factors between two groups was studied and compared the middle-aged group and youth, the pre-elderly group with control group.MAIN OUTCOME MEASURES: Morbidity comparison of each risk factor in each group.RESULTS: Hypertension, hyperlipemia, abnormal electrocardiogram (ECG),family history of stroke, arteriosclerosis of the eyeground, diabetes, and rheumatic heart disease are risk factors of ischemic stroke in the middle-aged.CONCLUSION: It is extremely important significance to reduce blood pressure and blood-fat level in the prevention of ischemic stroke.
