The epidemic characteristics and genotype of Bacillus anthracis strains in Liaoning Province ,China was analyze in this study .Six Bacillus anthracis strains from 2001 to 2011 were studied with multiple‐locus variable‐number tandem repeat analysis (MLVA) .BioNumerics4 .0 software was used to analyze the DNA fingerprint of statistics ,and cluster analysis results were obtained .Clustering analysis found that the 6 strains could be divided into two genotypes .For anthrax outbreaks ,the ge‐netic markers of multiple‐locus variable‐number tandem repeat were highly similar .It's suggested that MLVA is quite useful for investigation of strain relatedness in regions of outbreaks .
2.SH and HN Protein Genetic Characterization Analysis of Mumps Virus Isolated in Liaoning Province from 2008 to 2014.

To analyze the genetic characterization of epidemic mumps virus strains in Liaoning Province and provide the basis for mumps control. A total of 32 mumps viruses strains were isolated during 2008-2104. The fragment of SH genes and HN genes were amplified by RT-PCR, the PCR products were sequenced and analyzed. Basing on the 316 nucleotides of SH gene, The phylogenetic analyses were processed with the data of WHO mumps reference strains downloaded from GenBank and 32 mumps viruses strains. It showed that the 31 mumps virus strains belong to F genotype except MuVi/Liaoning. CHN/16.11 which was G genotype . Comparing to the A reference strains (Jeryl-Lynn and S-79), F genotype MuV were mutated on 12 amino acids sites and 27 amino acids siteson on HN gene. F genotype MuV added one N-glycosylation site in 464th-466th amino acids. The antigenic sites on HN were mutated on 121th, 123th, 279th, 287th, 336th, 356th and 442th. Maybe, it will influence the MuV antigenic.

3.Genetic Characterization of Hemagglutinin on Measles Virus Epidemic Strain Genotype H1a in Liaoning Province (China) from 1997 to 2014.

To wished to characterize the hemagglutinin (H) gene of the measles virus epidemic strain H1a in Liaoning Province (China) from 1997-2014 to provide a basis for the control and elimination of measles. All 63 measles virus strains were the H1a genotype. Fragments of the H gene (1854 nucleotides) and nucleoprotein (N) gene (450 nucleotides) were amplified by reverse transcription-polymerase chain reaction (RT-PCR) and the PCR products sequenced and analyzed. Phylogenetic-trees were constructed with reference strains of the genotype-H measles virus downloaded from GenBank, including Chinese measles virus strains isolated in 1993-1994 and the vaccine reference strains S-191 and C-47. Sixty-three strains of the measles virus in 1997-2014 belonged to genotype H1a. The mean evolutionary rate for gene N-450 was higher than that for the H gene. All 63 strains of the measles virus were mutated from: serine (Ser S) to asparagine (Asn N) in the 240th amino acid; arginine (Arg R) to glycine (Gly G) in the 243th; and tyrosine (Tyr Y) to Asn N in the 481th amino acid. All measles virus strains in cluster 2 were mutated from proline (Pro P) to leucine (Leu L) in the 397th amino acid. The other neutralization sites showed no apparent difference when comparing the nucleotides/amino acids of the H gene of S191 vaccine strains.

4.Massive pulmonary embolism similar to acute myocardial infarction rescued by ECMO: a case report

Acute myocardial infarction (AMI) and acute massive pulmonary embolism can be characterized by no pulse electrical activity. Patients of cardiac arrest (CA) with no pulse electrical activity have a high mortality rate before the cause was corrected. Extracorporeal membrane oxygenation (ECMO), as a kind of artificial heart-lung support organ, provides treatment for CA patients. A case of massive pulmonary embolism similar to AMI was treated in the Third Central Hospital of Tianjin, who received interventional thrombolysis assisted by ECMO. Through the review of the overall development of the case, we aim to broaden the diagnosis and treatment of CA patients with no pulse electrical activity, and to improve the understanding of the complications secondary to ECMO.
5.Spatial Analysis on Measles in Liaoning Province,China,2013-2015

Objective To explore the spatial distribution of measles from 2013 through 2015 in Liaoning province,China and to provide references for measles control and prevention. Methods The GeoDa 1.4. 6 program was used to conduct exploratory spatial data analysis to identify the spatial distribution characteristics and pattern of measles in Liaoning province. Results The frequency analysis showed that the measles epidemic situation appeared to have significant positive skewing within 105 counties of Liaoning province in each year from 2013 through 2015. The global trend analysis indicated a balanced trend in 2013 and 2015,and that the high incidence measles areas were located in the eastern and northern provincial regions in 2014. The global Moran'sⅠwas 0.294 5,0.391 9,and 0.147 7,and general G values were 0.015 9,0.012 0,and 0.013 5,revealing a positive spatial autocorrelation and a high-high aggregation model for each year between 2013 and 2015. The local spatial autocorrelation analysis recognized 5 core areas and 25 hot-spot counties with a high incidence of the measles epidemic,mainly distributed in Shenyang,Fuxin,Tieling,Fushun,Benxi,Liaoyang,Panjin,and Huludao. Conclusion Measles cases were clustered geographically in Liaoning province from 2013 through 2015. Spatial epidemiology methods may offer insights for the epidemiologic study of measles.
6.Training effect of case-based learning on post competency of medical students in the teaching of medical chemistry

Objective To investigate the training effect of the case-based learning (CBL) teaching method on post competency of medical students. Methods Four classes of students majoring in nursing in China Medical University were enrolled and divided into experimental group and control group, with two classes in each group. In the teaching of medical chemistry, the CBL teaching method was adopted for the experimental group and the traditional teaching method was adopted for the control group, with the same teacher and teaching materials. Questionnaire survey and final examination were performed to assess the effect of the CBL teaching method on post competency of medical students, and SPSS 20.0 software was used for data analysis. Results The results of the questionnaire survey showed that the experimental group had better assessment indices of post competency. The results of the final examination showed that the experimental group had a significantly higher mean score of medical chemistry than the control group [(73.5 ±10.3) vs. (68.1 ±9.1)]. Conclusion The application of the CBL teaching method in medical chemistry can effectively improve the learning outcomes of medical students and train their post competency, and therefore, it holds promise for application in the teaching of basic medicine.
7.Comprehensive evaluation and analysis of laboratory resource allocation in 14 blood stations based on entropy weight -TOPSIS method

【Objective】 To investigate the resource allocation status of blood testing laboratories in 14 blood stations in Gansu Province, explore the impact of differences in basic conditions on the comprehensive testing ability of laboratories, so as to promote the homogenization and standardization of blood screening capacity in blood stations in Gansu and improve blood safety and effectivenes. 【Methods】 An evaluation index system of laboratory resource allocation was constructed and a question-naire was designed. The data of human resources, infrastructure and key equipment of 14 blood stations were collected. The entropy weight -TOPSIS method was used to evaluate and rank the resource allocation of 14 blood stations. 【Results】 In the comprehensive evaluation of blood testing laboratory resource allocation in 14 blood stations in Gansu, the top three were laboratories A, B and I, and the last three were laboratories G, M and J. On the whole, the main issue was unreasonable structure of human resources: most laboratories had unreasonable age structure; except for Laboratory A, there was no personnel with bachelor's degree or above in laboratories; most laboratories had not established a team with intermediate professional titles. In terms of infrastructure, the size of seven laboratories could not meet the needs of modern laboratory testing, and all eight blood stations had no spare nucleic acid laboratories nor a mutual spare laboratory with other blood stations As for the key equipment, 5 laboratories had no automatic blood grouping diagnostic instrument, 5 laboratories only had one set of enzyme immunoassay detection system, 3 laboratories had no spare equipment for the key equipment, which means if the equipment failure could not be repaired in time, the release of results would be affected. 【Conclusion】 There were significant differences in human resources, infrastructure and key equipment of blood testing laboratories in 14 blood stations in Gansu, which had a great impact on laboratory testing capacity and subsequent development. It is suggested that governments at all levels and health administrative departments optimize the input of laboratory resource allocation according to the blood collection volume of blood stations to gradually narrow the differences in resource distribution between different regions, improve the degree of laboratory automation and optimize the personnel structure, so as to build high-quality and efficient blood testing laboratories and ensure the safety of clinical blood use.
8.Molecular Mechanism of Erxian Decoction in Treatment of Anxiety Disorder Based on Network Pharmacology and Experimental Verification

ObjectiveTo predict the potential molecular mechanism of Erxian decoction in the treatment of anxiety disorder based on network pharmacology, and to verify the efficacy and mechanism using the animal model of maternal separation combined with restraint stress. MethodActive components and related targets of Erxian decoction were obtained by traditional Chinese medicine system pharmacology database and analysis platform (TCMSP) and SwissTargetPrediction. The targets related to anxiety disorder were screened out through GeneCards, therapeutic target database (TTD), online mendelian inheritance in man database (OMIM), and DrugBank, and the drug-disease intersection targets were obtained by taking intersections with the drug targets. The protein-protein interaction (PPI) network was constructed by the STRING database, and the core targets were screened out based on topological parameter analysis. Gene Ontology (GO) enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis were carried out for the intersection targets through the Metascape platform. Maternal separation combined with restraint stress was used to induce the mouse model of anxiety disorder. From the end of lactation on the 21st postnatal day (PD21) to the completion of restraint stress on the 97th postnatal day (PD97), the mice were fed with Erxian decoction mixed with diet. The anxiety state of mice was evaluated by open field test and elevated O-maze test. The content of plasma corticosterone (CORT) in mice was detected by enzyme-linked immunosorbent assay (ELISA). The expression levels of protein kinase B (Akt1), mammalian target of rapamycin (mTOR), brain-derived neurotrophic factor (BDNF), postsynaptic density-95 (PSD95), and synaptophysin in the hippocampus of mice were detected by Western blot and real-time quantitative polymerase chain reaction (Real-time PCR). ResultNinty-seven active components and 227 action targets of Erxian decoction were obtained. There were 3 863 targets related to anxiety disorder, with 161 drug-disease intersection targets. Among these intersection targets, core targets such as Akt1, interleukin-1β (IL-1β), interleukin-6 (IL-6), tumor necrosis factor (TNF), and mTOR were presumedly closely related to anxiety disorder. The results of KEGG pathway analysis showed that Erxian decoction mainly treated anxiety disorder through phosphatidylinositol 3-kinase (PI3K)/Akt, mitogen-activated protein kinase (MAPK), and neuroactive ligand-receptor interaction signaling pathways. The results of animal experiments showed that compared with the model group, the Erxian decoction group significantly increased the time of mice spent in the central zone and central crossing times and time spent in the opened arm and opened arm crossing times, with significantly increased expression levels of p-Akt1, p-mTOR, BDNF, PSD95, and synaptophysin (Syp). ConclusionErxian decoction has the multi-target and multi-pathway characteristics in the treatment of anxiety disorder, and its mechanism may be related to the improvement of synaptic plasticity and neuroinflammation by affecting Akt1, IL-1β, IL-6, TNF, mTOR, and other core targets and modulating PI3K/Akt, MAPK, as well as neuroactive ligand-receptor interaction signal pathways.
9.Feasibility of Erxian Decoction and Wenshen Prescription in Treatment of Depression Based on Network Pharmacology and Experimental Verification

ObjectiveTo predict the molecular mechanism of Erxian decoction and Wenshen prescription (modified Erxian decoction) in the treatment of depression based on network pharmacology and explore the feasibility of Wenshen prescription in the treatment of depression by comparing the efficacy and mechanism of the two decoctions based on a depression model induced by maternal separation combined with chronic restraint stress. MethodActive components and targets of Erxian decoction and Wenshen prescription were collected through Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP) and Bioinformatics Analysis Tool for Molecular mechanism of Traditional Chinese Medicine (BATMAN-TCM). Targets related to depression were screened out from databases such as GeneCards, Online Mendelian Inheritance in Man database (OMIM), and DrugBank. Common targets of drugs and disease were obtained and imported to Cytoscape 3.8.2 to plot the drug-active component-target-disease network. STRING platform was used to construct a protein-protein interaction (PPI) network and core targets and related core components were screened out. Gene Ontology (GO) enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) functional enrichment analysis were performed on common targets through Metascape platform. The depression model was induced in mice by maternal separation combined with chronic restraint stress. From the 21st day of maternal separation (PD21) to the 111th day of restraint stress completion (PD111), mice were fed with the diet mixed with Erxian decoction or Wenshen prescription for intervention. The depressive state of mice was evaluated according to the sucrose preference test, tail suspension test, open field test, and elevated O-maze test. The expression of ionized calcium-binding adapter molecule 1 (Iba1) in the microglia was observed by immunohistochemistry (IHC). Western blot and Real-time fluorescence-based quantitative polymerase chain reaction (Real-time PCR) were used to detect the expression levels of protein kinase B1(Akt1), brain-derived neurotrophic factor (BDNF), postsynaptic density-95 (PSD95), and synaptophysin (Syn). ResultA total of 126 and 118 targets of Erxian decoction and Wenshen prescription in the treatment of depression were screened out, with only eight more targets of Erxian decoction than Wenshen prescription. The two decoctions shared the same core targets, mainly including Akt1, interleukin-6 (IL-6), interleukin-1β (IL-1β), and tumor necrosis factor-α (TNF-α). KEGG pathway enrichment analysis predicted that Erxian decoction and Wenshen prescription mainly treated depression through the phosphatidylinositol-3 kinase (PI3K)/Akt signaling pathway, mitogen-activated protein kinase (MAPK) signaling pathway, and neuroactive ligand-receptor interaction pathway. Animal experiments showed that compared with the results in the model group, Erxian decoction and Wenshen prescription could up-regulate the sucrose preference index, prolong the time spent in the central zone, increase the number of crossings, prolong the time spent in opened arm, increase the number of crossings in the opened arm, elevate the expression levels of p-Akt1, BDNF, PSD95, and Syn (P<0.05, P<0.01), shorten the immobility time of tail suspension, and reduce the expression level of Iba-1 in the hippocampal microglia (P<0.05, P<0.01). No significant difference between the two decoctions was found. ConclusionUnder the pathogenesis and syndrome law of depression dominated by kidney yang deficiency, Wenshen prescription modified from Erxian decoction is feasible in the treatment of depression. The mechanism may be attributed to the fact that both decoctions can improve neuroinflammation and synaptic plasticity in the hippocampus by affecting Akt1, IL-1β, IL-6, TNF-α, and other core targets and regulating the PI3K/Akt, MAPK, and neuroactive ligand-receptor interaction signaling pathways.
10.Effect of Xiaoyaosan on JNK Pathway in LPS-induced Depressive-like Behavior in Mice

ObjectiveTo explore the mechanism of Xiaoyaosan in alleviating lipopolysaccharide (LPS)-induced depressive-like behavior in mice based on the c-Jun N-terminal kinase (JNK) pathway. MethodAfter adaptive feeding, C57BL/6J mice were randomly divided into normal group, model group, minocycline group (intrabitoneal injection, 50 mg·kg-1), fluoxetine group (intragastric administration, 2.6 mg·kg-1), and low-, medium-, and high-dose Xiaoyaosan groups (intragastric administration,6.012 5, 12.025, and 24.050 g·kg-1). After 14 days of administration, the model group and each administration group were intraperitoneally injected with 2 mg·kg-1 LPS, and the normal group was intraperitoneally injected with equal volume of normal saline. Depressive-like behavior in mice was assessed using the open field test and the elevated zero maze test. High-performance liquid chromatography (HPLC) was used to measure the levels of norepinephrine (NE) and epinephrine (E) in the mouse hippocampus. Enzyme-linked immunosorbent assay (ELISA) was performed to determine serum interleukin-1β (IL-1β) levels. Immunohistochemistry was used to measure the protein expression levels of ionized calcium-binding adapter molecule-1 (Iba-1), c-Fos, and c-Jun. Real-time polymerase chain reaction (Real-time PCR) was used to measure mRNA expression levels of IL-1β, c-Jun, c-Fos, and JNK3 in the mouse hippocampus. Protein expression levels of JNK and phosphorylated (p)-JNK in the mouse hippocampus were measured using capillary protein automated protein expression analysis system (Western). ResultCompared with the normal group, the model group exhibited significantly reduced central area residence time, crossing times, and travel distance in the open field (P<0.01), significantly increased serum IL-1β levels (P<0.01), significantly decreased NE and E levels (P<0.05), upregulated mRNA expression of IL-1β, JNK3, and c-Fos, and increased protein expression of Iba-1, c-Fos, and c-Jun (P<0.05, P<0.01). Compared with the model group, the Xiaoyaosan groups showed increased central area residence time and open arm residence time (P<0.05), increased NE and E levels (P<0.01), decreased mRNA expression of IL-1β, JNK3, c-Jun, and c-Fos, and decreased protein expression of Iba-1, c-Fos, JNK, and p-JNK (P<0.05, P<0.01). The minocycline group and the fluoxetine group showed decreased mRNA expression of JNK3, c-Jun, and c-Fos (P<0.05, P<0.01). The minocycline group showed decreased serum IL-1β and p-JNK protein expression (P<0.01). The fluoxetine group exhibited increased NE and E levels and decreased c-Fos protein expression (P<0.01). ConclusionXiaoyaosan can improve depressive-like behavior induced by LPS in mice, and its mechanism may be related to the inhibition of neuroinflammatory responses and the JNK pathway.
