1.Simultaneous Determination of the Contents of Dictamnolactone and Skimmianine in Root of Zanthoxylum armatum DC. from Gansu Province by HPLC

Objective To develop an HPLC method for the simultaneous determination of the contents of dictamnolactone and skimmianine in the root of Zanthoxylum armatum DC. from Gansu Province. Methods The column was kromasil-C18 (250 mm×4.6 mm, 5 μm);the mobile phase was acetonitrile-water (65∶35);UV detection was performed at 254 nm;the flow rate was 1.0 mL/min;detected drift tube temperature was set at 30 ℃. Results The calibration curves were linear in the range of 0.205-2.05μg for dictamnolactone (r=0.999 9) and 0.199 6-1.996μg for skimmianine (r=0.999 7). The average recovery was 99.69%(RSD=0.57%) for dictamnolactone and 99.49% (RSD=1.04%) for skimmianine. Conclusion The method is simple and accurate. It is suitable for content determination of dictamnolactone and skimmianine in the root of Zanthoxylum armatum DC..
2.Effects of liposome-mediated CD gene transfer plus radiation on human rectal cancer HR-8348 cells in nude mouse

ObjectiveTo investigate the effect of repeated transfer of liposome mediated CD gene plus radiation on the growth of human rectal cancer cells xenograft in nude mouse Methods HR 8348 cells transfered with liposome mediated CD gene repeatedly (at day 0,4,8,12 ) and treated with fractionated radiation at a dosage of 2 Gy/day to a total dose of 30 Gy. At the same time, mice were injected i.p.with 800 mg/kg/day of 5 FC for 12 days. Results Compared with controls tumors in treatment group grew with an inhibition rate of 78 7% in weight, and 81 5% in volume. ConclusionRepeated transfer of liposome mediated CD gene working together with radiation effect a significant inhibition on the growth of xenografted human rectal carcinoma HR 8348 cells in nude mouse.
3.Diagnosis and therapy of adrenal ganglioneuromas

Objective To investigate the clinical characteristics,manifestation and management of adrenal ganglioneuromas. Methods The data of 15 cases of ganglioneuromas diagnosed by pathology were reviewed retrospectively.Of the 15 cases,5 was male,aged 24 to 45 with an average of 29 years,and other 10 was female,aged 25 to 69 with an average of 31.Ten tumors were located in the right side,which was twice as many as that of the left side.There were 8 patients discovered by B ultrasonograpgy during health check-up,4 patients were detected because of paroxysmal abdominal pain. Three cases mainly presented with paroxysmally hypertension,and one of them had a high level of aldosterone ( 112 pmol/L). One of them had a high level of epinephrine (210 nmol) and nor-epinephrine (496 nmol). The B-ultrasound showed low-echo in all cases.Five cases showed calcification,the border was legible.The border lines of the tumors are not clear with vana cava in two cases. On unenhanced CT scanning,all cases showed low density areas.Eleven of the 15 cases underwent the enhanced CT,but 10 cases showed no enhanced areas on CT scanning.One case underwent MRI and showed low signals in T1 WI and inhomogeneous high signals in T2WI. Results Twelve cases underwent adrenal glands tumorectomy with abdominal NO.11 intercostals incision while other 3 underwent adrenal glands tumorectomy through posterior abdominal laparoscope.The tumors were completely removed in 14 cases,partly removed in 1 case.All patients recovered soon without complications such as bleeding or adrenal crisis.All cases were followed-up 13 to 74 months.The blood pressure of 3 cases with hypertension decreased to normal.The tumors recurred in 1 case,but no metastasis 55 months later. Conclusions It is difficult to differentiate adrenal medullary tumor from ganglioneuromas.The imaging data of iconography before surgery can give helpful information in the diagnosis of ganglioneuroma.Complete surgical excision is the only method for curing and the outcome is satisfactory.
4.Effect of patient's preoperative visit to operating room on the preoperative apprehension

Objective To evaluate the effect of the patient's preoperative visit to the operating room on the patient's apprehension before operation.Methods One hundred and Sixty patients aged 16-64 yr with fear visual analog scale (FVAS) score≥4 scheduled for elective surgery were randomly divided into 2 groups(n=80 each):control group and study group.In control group the anesthesiologists visited their patients the day before operation ns usual;while in study group the anesthesiologists brought their patients to the operating room,showed them the environment and anesthesia equipment and assured of the safety of the operation and anesthesia.The degree of fear was scored according to FVAS (O=no fear,1-3 mild,4-6 moderate,7-10 severe) and was evaluated on the 1st morning after admission(T1),the night before operation(T2)and before induction of anesthesia(T3).MAP and HR at T1-3 and blood glucose level at T1,3 were mensured and recorded.Results The patient's preoperative apprehension was significantly allayed by patient's preoperative visit to the operating room.The MAP,HR,incidence of moderate and severe fear at T2,3 and blood glucose level at T3 were significantly lower in study group than in control group.Conclusion Patient's preoperative visit to the operating room can allay precperative apprehension.
5.Alteration in Potassium Channel in Ventricular Myocytes in Diabetic Rats

6.Effects of Blood Sample Preserved by Different Methods and Times on Blood Lactate

This study was to examine the effects of blood sample preserved by different methods and times on blood lactate. The subjects were 25 male middle school students. Earlobe blood was collected and the blood lactate was determined respectively at once, after preserved by freezing 1 week, 2 weeks, 3 weeks, 4weeks or cold storage 1 day and 5 days. The results showed that all lactate values measured of 6 preserved samples were not significantly different but relevant with that of fresh sample.
7.Norepinephrine induced expression of interleukin-6 in human macrophages and mechanisms

OBJECTIVE To investigate the effect of norepinephrine (NE) on expression of interleu?kin-6 (IL-6) in human macrophages and explore its pro-inflammatory and pro-atherosclerotic mecha?nisms. METHODS Murine U937 macrophages were cultured and stimulated with 0.01-10μmol·L-1 of NE for 6, 9, 12, 24 and 48 h. The IL-6 mRNA level of 24 h was analyzed by RT-PCR, and IL-6 protein expression in the supernatant at 0, 6, 9, 12, 24 and 48 h was detected by ELISA. After 24 h, intracellular reactive oxygen species (ROS) generation was observed by DCF fluorescence. The cells were pretreated with antioxidant N-acetylcysteine (NAC), complexⅡinhibitor thenoyltrifluoroacetone (TIFA) and NADPH oxidase inhibitor diphenyleneiodonium (DPI) for 1 h, and stimulated with different concentrations of NE for 24 h, before the level of IL-6 protein was detected by ELISA. RESULTS The expression of IL-6 mRNA and protein increased with the concentration NE 0.01-10 μmol · L-1 and incubation time. IL-6 mRNA and protein levels in macrophages were 2.62 and 4.47-fold those in cell control group when treated with NE 1.0μmol·L-1 for 24 h. Meanwhile, as the concentration of NE increased, the generation of ROS was 1.87, 2.56, 2.91 and 5.36-fold that of cell control group (P<0.01). NAC 10 mmol · L-1 and DPI 10 μmol · L-1 significantly antagonized the effect of NE on IL-6 expression, but TIFA had no effect. CONCLUSION NE upregulates IL-6 expression, which may contribute to the formation and develop?ment of atherosclerosis via ROS mediated by NADPH oxidase in macrophages.
8.Effect of Three Kinds of Polysaccharides from Herbal Medicines on Immunomodulation in Mice

Objective To explore the features of immune responsed activated by polysaccharides from herbal medicines and provide basis for immunomodulation of polysaccharides. Methods The mice were immuned by polysaccharides of Angelica, Atractylodes and Giant typhonium rhizome respectively, then the corresponeding antibodies and cross-reactive antibodies in serum were detected by ELISA. Results Three kinds of polysaccharides from herbal medicines could all activated the mice to produce not only the corresponding antibodies, but also the cross-reactive antibodies, the ranges of non-specific antibodies activated by distinct polysaccharide were different. Conclusions Polysaccharides from herbal medicines may be a kind of non-specific and broad-spectrum immunomodulators.

10.Therapeutics, genomics and epigenomics of retinoblastoma

Retinoblastoma is the most common pediatric retinal tumor initiated by biallelic inactivation of the retinoblastoma gene ( RB1 ) , affecting roughly 1 in 15000 children with 1100 new cases each year.In China, RB remains a potentially devastating disease while lack of early diagnosis and aggressive treatment strategies.Moreover, RB patients individually show a variety of drug response to regular systemic chemotherapy since intensive chemotherapy and long-term follow-up are not as readily available.The majority of patients with advanced diseases require enucleation which affects the future quality of children life.Recent studies showed that alterations in genomics and epigenomics of RB play very important roles in RB progress and therapy.We thereby review current understanding changes of genomics and epigenomics in RB as potential prognostic and therapeutic targets.
