Objective To study the effects of hypoxia-inducible factor-1α (HIF-1α) on radiationinduced autophagic cell death in breast cancer cells. Methods MCF-7 cells were divided into four groups:control (normoxia,21% Oxygen),irradiation (8 Gy X-rays),hypoxia (Cobalt chloride,CoCl2)and irradiation with hypoxia (CoCl2).150 μmol/L CoCl2 was utilized to induce hypoxic conditions.Western blot was applied to detect the expression of HIF-1α and MAPLC3.MDC and Hoechst staining were used to detect autophagy and apoptosis.Radiosensitivity was detected by cloning formation. The short hairpin interfering RNA (shRNA) retroviral transduction particles targeting HIF-1α was transfected into MCF-7 cells to establish HIF-1 α knockdown cells, then the radiosensibility, autophagy and apoptosis were detected. Results Compared with control group and irradiation group, the protein level of HIF-1 increased obviously in the normaxia, irradiation, hypoxia and irradiation with hypoxia groups, and the values were 0,0,1.00,1.89,respectively.The expression levels of MAPLC3 were markedly up-regulated in irradiation, hypoxia and irradiation with hypoxia groups as compared with control, and the ratios of LC3Ⅱ/LC3Ⅰ were 1.15,1.73,2.38 and 3.60,respectively.The radiosensitivity of MCF-7 cells decreased in the following order:normoxia with 3MA > normoxia > hypoxia with 3MA > hypoxia.HIF-1α knockdown cell (pSUPER-HIF-1α Ri) and vector control were constructed. After treatment with CoCl2,survival fraction of MCF-7-pSUPER was significantly higher than that of control (t =3.080,6.946,6.658,6.380,P <0.05),and radiosensitivity was down-regulated after irradiation, but there was no significant difference between normoxia and hypoxia in survival fraction of MCF-7-pSUPER-HIF-1α Ri.After treatment of irradiation or hypoxia, the autophagic fractions in MCF-7-pSUPER-HIF-1α Ri significantly decreased, reduced by 21.1%,25.5%,15.5%,respectively(t =4.635,4.738,6.354,P <0.05) as compared with MCF-7-pSUPER,but there was no change in apoptosis. Conclusions HIF-1α may increase radiationinduced autophagy and decrease radiosensitivity, but have no influence on apoptosis.

Objective To observe the influence of ultra-shortwave (USW) irradiation on infarct volume and Ca2+-ATPase (SPCA) secretion after brain ischemia and reperfusion. Methods Eighty Sprague-Dawley rats were randomly divided into a sham operation group (n=8),a model group (n=36) and a USW group (n=36).The animal model of middle cerebral artery ischemia and reperfusion (MCAO/R) was established using the suture method in the rats of the model and USW groups, while the sham operation group was given the same operation but without inserting the thread plug. One day,3 days and 7 days after the intervention,12 rats were sacrificed and the infarct volumes and SPCA1 protein expression were measured using 2,3,5-triphenyltetrazolium chloride staining and western blotting. Results No white infarcted tissue was found in the sham operation group. In the model and USW groups the volume of infarcted tissue decreased with time.Significantly less infarcted volume was observed in the USW group compared to the model group at each time point. The SPCA1 levels in the brain tissue were lower than in the sham operation group after one and 3 days of USW treatment, but they were significantly lower in the model group as well.As time went by,the average SPCA1 level increased significantly in the model and USW groups.A slightly higher SPCA1 level was observed in the USW group compared to the model group after one day of treatment, but with no significance. However, significant differences were found between them after 3 and 7 days of intervention. Conclusion Ultra-shortwave irradiation can protect against MCAO/R injury by decreasing the infarcted volume, which may be related to down-regulation of SPCA1,minimizing nerve cell apoptosis and promoting neural functional recovery,at least in rats.

Objective To evaluate the effect of antiplatelet drugs and ischemic-events-free duration by thromboelastography (TEG) in patients undergoing peripheral endovascular treatment. Methods From Mar 2015 to Oct 2016,38 patients undergoing initial successful peripheral stenting and then coming back for recurrent ischemic events.TEG was used to evaluate the platelet inhibition rate induced by arachidonic acid (AA%) approach, the platelet inhibition rate induced by adenosine diphosphate (ADP%) approach, and the ADP-induced platelet-fibrin clot strength (MAADP).Clinical feature differences between patients with MAADP > 47 mm and MAADP ≤ 47 mm were compared. Results AA% less than 50% was found in 9 patients(23.9%),and ADP% less than 30% in 14 patients(36.8%).5 patients met the two conditions.Patients with MAADp > 47 mm had a significantly lower ADP% (x2 =10.755,P < 0.001),compared to patients with MAADP ≤47 mm.Univariate survival analysis showed ADP% < 30% and MAADP >47 mm were risk factors for getting shorter ischemic-events-free duration, compared to patients with ADP% ≥ 30 (P < 0.001),with MAADP ≤47 mm (x2 =4.408,P =0.036).Conclusions TEG may detect those patients who has a low response to antiplatelet drugs, and some TEG parameters may have a ischemic predictive value.

Gallstone is a common and frequent disease and frequent incidence, secondary infection and cancer seriously affect the health of patients. Academic organizations in different regions have issued multiple guidelines and consensus to promote the normative diagnosis and treatment of gallstones. However, in clinical practice, most symptomatic gallstones are treated, while the formation and prevention process of gallstones are ignored, making the concept of treating without a disease has not been strengthened. This article reviews the risk factors and mechanisms of gallstone formation, and points out the importance of effective prevention during stone formation. In the stage of gallstone formation, the high risk factors of stone formation can be analyzed through two aspects of injury factors and protective factors, and the high risk groups of stone formation can be screened out. According to the pathophysiological progression of gallstones, personalized prevention and follow-up strategies can be developed for the stone formation stage of gallstones.
