Toll-like receptor,TLR (TLR) can sense invading pathogens,the acute inflammatory response in the body,chronic airway inflammation and pathophysiology of allergic diseases and play an important role in the immune defense response.Among them,TLR2,TLR4 most prominent role and become a hot research topic.This paper summarizes the importance of TLR2,TLR4 in the pathogenesis of respiratory tract infections,and pointed out by interfering with TLR2,TLR4 signal transduction pathway or reduced TLR2,TLR4g role may help to weaken the disease or improve the prognosis,which provides a new way of thinking and clues for the treatment of clinical common diseases.
2.Effect of Resveratrol on the Expression of MCP-1 and BNP in Myocardial Cells of Rats with Acute Pulmonary Embolism

Objective To examine the effect of resveratrol on the expression of monocyte chemoattractant protein-1(MCP-1)and brain natriuretic peptide(BNP)in myocardial cells of rats with acute pulmonary embolism.Methods Ninety-six SD rats were randomly divided into 2 groups:thromboembolism group (group T)and resveratrol + thromboembolism group (group TR),each group having 3 subgroups(n=1 6 each)in terms of different time points(1 h,4 h,8 h).The model of acute pulmonary embolism was established by infusion of autologous thrombi via the catheter inserted from the right jugular vein to the pulmo-nary artery.In group TR,resveratrol at 10 mg/kg (dissolved in 1%DMSO solution)was injected to the rats via the tail vein.Animals were sacrificed at 1,4 and 8 h after the model establishment.The rat hearts were harvested,fixed in 10%paraformaldehyde and paraffin-embedded.The expression levels of MCP-1 and BNP were detected by immunohistochemistry (SP method).Results Both MCP-1 and BNP were expressed in myocardial cells in group T and TR and gradually increased with time.The expression levels of MCP-1 and BNP were much lower in group TR than in group T at 4 and 8 h,with difference being statistically significant(P<0.05).Conclusion The expression of MCP-1 and BNP tended to increase with time in both groups.Resveratrol may protect the heart tissues by suppressing the expression of MCP-1 in myocardial tissues of rats with a-cute pulmonary embolism.
3.Effects of crocetin on doxorubicin-induced damage in rats with myocardial mitochondria

AIM: To study the protective effects of crocetin on the doxorubicin-induced damage in rats with myocardial mitochondria. METHODS: Rats were given intraperitoneal injection of doxorubicin to induce cardiotoxicity. After continuous oral administration of crocetin, the rats were sacrificed, and myocardial mitochondria were isolated. The mitochondrial membrane potential (MMP), O~-_2. production, the activity of respiratory chain complex IV and glutathione peroxidase (GSH-PX), and DNA fragmentation were determined. The expression level of COII gene was determined through RT-PCR. RESULTS: Crocetin increased the activity of respiratory chain complex IV and GSH-PX, MMP and the expression level of COII and decreased DNA fragmentation and superoxide anion radical O~-_2. production in rats with myocardial mitochondria. CONCLUSION: Crocetin may decrease the damage in rat myocardial mitochondria induced by doxorubicin.
4.Influence of dietary intervention on treatment effect and life quality of young patients with hepatitis B

Objective To explore the effect of dietary intervention on treatment effect and life quality of young patients with hepatitis B. Methods 106 cases of hepatitis B patients were randomly divided into the intervention group and the control group, each group with 53 cases. The patients in the control group were given medication treatment and conventional diet, while the intervention group was given medication treatment plus dietary intervention. The therapeutic effect and quality of life were evaluated between the two groups 30 days after the intervention. Results 29 cases in the intervention group were cured (54.72%),while only 16 cases in the control group (30.19%) were cured. The total score of life quality in the intervention group was (69.72 ± 8.42), and (55.71 ± 8.31) in the control group. Significant difference existed in subsequent dimensionality: emotional role, bodily function,bodily pain, vitality, social function, mental health and general health between the two groups. Conclusions Clinical efficacy can be improved and quality of life can be enhanced through dietary intervention in patients with hepatitis B.
5.Research progress of multi-functional cancer killing cytokine, interleukin-24 in tumors

Interleukin-24 (IL-24) is a member of IL-10 family, which was cloned from human melanoma cells by the method of subtractive hybridization. IL-24 can inhibit the growth of tumor cells and angiogenesis and enhance radiation sensitivity and immune adjustment with non-toxic effects on normal cells.As a kind of cytokine with multiple anti-tumor functions, IL-24 will become a new weapon of gene therapy against cancer. The application and mechanism of IL-24 in the treatment of cancer will be discussed in this paper.
6.Effect of moving services on negative emotions and quality of life of patients with leukemia

Objective To investigate the effects of moving services on negative emotions and quality of life of the patients with leukemia.Methods A total of 128 patients with leukemia were chosen and divided into the control group(n =66)and the intervention group(n =62)according to admission date.The patients of the control group received conventional nursing mode,while the patients of the intervention group were given moving services including updates care philosophy,advocated behavior reflect personalized and multi -style health education.The self -rating depression scale(SDA),self -rating anxiety scale(SAS)before and after the intervention were used to assess changes in patients with negative emotion,while the quality of life scale QLQ -C30 core was used to estimate quality of life in patients before and after treatment.Results After intervention,QLQ -C30 functional scores including physical function, role function,cognitive function,emotional function and social function score were higher than the control group [(68.79 ±13.97)points vs (60.48 ±14.95)points,(53.49 ±15.82)points vs (31.03 ±20.68)points,(88.81 ± 16.46)points vs (73.33 ±16.50)points,(86.48 ±8.21 )points vs (70.30 ±16.43 )points,(31.03 ± 1.27)points vs (26.36 ±8.72)points],and the differences were statistically significant(t =3.094,3.606,3.265, 3.478,3.20,all P <0.05).QLQ -C30 symptom scores including fatigue,pain,nausea,vomiting,dyspnea,insomnia, appetite loss,constipation,diarrhea rating were lower than the control group[(41.81 ±14.25)points vs (52.85 ± 14.57)points,(16.89 ±10.14)points vs (24.43 ±19.05 )points,(17.41 ±13.56 )points vs (29.51 ± 13.64)points,(25.83 ±21.09 )points vs (36.56 ±21.50)points,(11.54 ±8.19 )points vs (25.76 ± 19.07)points,(40.93 ±18.30)points vs (55.87 ±20.05 )points,(15.02 ±10.24)points vs (21.54 ± 12.41)points,(8.66 ±2.41)points vs (15.47 ±9.16)points],and there were statistically significant differences (t =3.179,3.243,3.359,3.470,3.205,3.151,4.282,3.153,all P <0.05).After intervention,the scores of SAS and SDS of the intervention group and the control group were (34.96 ±10.24)points vs (48.24 ±11.71)points, (33.58 ±9.60)points vs (42.06 ±9.62)points,and the differences were statistically significant(t =6.320,5.806, all P <0.05 ).The passing rate of care satisfaction of the intervention group was higher than the control group (98.06% vs 73.08%),and the time of hospitalization of the intervention group was lower than the control group [(22.02 ±2.24)d vs (27.54 ±3.41)d],and the differences were statistically significant(χ2 =4.337,t =3.759,all P <0.05).Conclusion Moving services can improve the quality of life of patients,reduce the negative emotions of patients,improve the quality of nursing services.
7.Effect of interleukin-1 on contractile function of rat thoracic aorta

Objective To investigate the effect of interleukin-1 (IL-1) on contractile function of rat thoracic aorta.Methods Forty male Sprague-Dawley rats,weighing 250-300 g,were sacrificed to obtain the thoracic aortic rings.The experiment was performed in 2 parts.Part Ⅰ The thoracic aortic rings were divided into 2 segments and randomly divided into 2 groups (n =20 each):control group and IL-1 group.In IL-1 group,the thoracic aortic rings were incubated with Kreb solution containing 20 ng/ml IL-1 for 2 h,and contracted with cumulative concentrations of phenylephrine,ranging from 10-9 to 10-5mol/L.In control group,the thoracic aortic rings were incubated with Kreb solution for 2 h,and contracted with cumulative concentrations of phenylephrine,ranging from 10 9 to 10-5mol/L.Part Ⅱ The thoracic aortic rings were divided into 3 segments and randomly divided into 3 groups (n=20 each):IL-1 group,IL-1+ L-NAME (the NOS inhibitor) group and IL-1 +cyclooxygenase inhibitor indomethacin group (IL-1 +Ⅰgroup).The thoracic aortic rings were incubated with Kreb solution containing 20 ng/ml IL-1 for 1.5 h in the three groups.In addition,in IL-1 +L-NAME and IL-1 +Ⅰ groups,the thoracic aortic rings were incubated for 30 min with Kreb solution containing 100 μmol/L L-NAME and 2.5 mmol/L indomethacin,respectively.Contraction of the thoracic aorta was then induced with cumulative concentrations of phenylephrine,ranging from 10 9 to 10-5 mol/L.In group IL-1,the thoracic aortic rings were incubated with Kreb solution.The maximum contractile tension of the thoracic aortic rings was recorded at each concentration of phenylephrine,and the percentage of the maximum contractile tension at the concentration of 10-6 mol/L in group C was obtained.Results Part Ⅰ The percentage of contractile tension at phenylephrine 10-s,10-7,l0 6 and 10-5mol/L was significantly decreased in IL-1 group as compared with C group.Part Ⅱ The percentage of contractile tension at phenylephrine 10-7,10-6 and 10-5mol/L was significantly increased in IL-1+L-NAME and IL-1+I groups as compared with IL-1 group.Conclusion IL-1 can inhibit the contraction of rat thoracic aorta,and promoted production of NO and prostacyclin may be involved in the mechanism.

It's a multi-protein complex and not single protein that accomplish most of cellular activity.Therefore,to identify and analyze the composition of protein complex is necessary for studying the function of proteins.Tandem affinity purification technique(TAP) enables the effective purification of complex under physiological conditions without knowledge of the complex composition and function.Now,it has been adapted to analyze the composition of the protein complexes in yeast.Combined with mass spectrometry,TAP can identify the interacting proteins of target protein and offer great help for opening protein interacting network and function out.
10.Methicillin-resistant Staphylococcus aureus colonization in infants at neonatal intensive care unit and effect of mupirocin decolonization

Objective To evaluate the methicillin-resistant Staphylococcus aureus (MRSA) colonization in infants at neonatal intensive care unit (NICU) by active surveillance cultures (ASC) and the effects of mupirocin decolonization on MRSA infection. Methods Neonates adimitted to NICU of Ruian People's Hospital of Zhejiang Province, China between October 1, 2013 and September 30, 2014 underwent ASC within 24 hours of admission (ASC group). The samples from nasal vestibule and umbilicus were cultured for MRSA, and positive cultures were considered as MRSA colonization. These with negative cultures underwent repeated culture of MRSA by ASC at the 1st and 2nd week after NICU admission. Neonates admitted to NICU between October 1, 2014 and September 30, 2015 with MRSA colonization based on ASC were decolonized with mupirocin at both nasal vestibule and umbilicus twice daily for five consecutive days (decolonization group). Culture samples were obtained one day and one week after decolonization to repeat ASC. ASC was stopped if the subsequent culture was negative twice successively; and a second mupirocin decolonizaton was performed if the culture was positive. Chi-square test was used to compare the rates of colonization and infection between the two groups. Results (1) MRSA colonization and infection in ASC group: MRSA colonization rates within 24 hours, and in the 1st and 2nd week after NICU admission in ASC group were 2.2% (9/418), 3.7% (15/402) and 3.6% (13/361), respectively. Compared with those without MRSA colonization, neonates with MRSA colonization had a higher incidence of MRSA infection [13.5% (5/37) vs 3.7% (14/381), χ2=7.524, P=0.006]. (2) MRSA colonization and infection in decolonization group: MRSA colonization rates within 24 hours, and at the 1st and 2nd week after NICU admission in decolonization group were 2.8% (12/435), 2.9% (12/414) and 1.3% (5/373), respectively. The MRSA colonization rate at the 2nd week was significantly lower than that in ASC group (χ2=3.919, P=0.048). Twenty-nine cases had MRSA colonization, among which, 27 cases were decolonized once and two cases were decolonized twice, and all were successful. The rate of MRSA infection in decolonization group was 2.1% (9/435), which was significantly lower than in ASC group [4.5% (19/418)] (χ2=4.118, P=0.042). Conclusions Rate of MRSA colonization is high at NICU. Mupirocin can decolonize MRSA carriage and reduce MRSA infection in neonates.
