1.Meta-Analysis on Clinical Efficacy of Integrated Treatment of Traditional Chinese and Western Medicine on Immune Thrombocytopenia

Objective To evaluate the clinical efficacy of integration of traditional Chinese and western medicine (ITWM) or pure western medicine on the patients with immune thrombocytopenia (ITP). Methods Randomized controlled trials (RCTs) on ITWM treatment of ITP were searched in PubMed, EMBASE, Wanfang database, CBMDisc, CNKI and VIP. Quality of the RCTs was evaluated strictly, and statistical analysis was performed with Cochrane Collaboration’s RevMan5.1.7 software. Results Twenty-two trials involving 1527 patients were included. The results of Meta-analysis showed that the ITWM treatment of ITP significantly improved the total effective rate [RR=1.23, 95%CI (1.17, 1.29), P<0.000 01] compared with that of pure western medicine treatment. Funnel plot showed that publication bias was small. Meanwhile, peripheral platelet count and the number of megakaryocyte which can generate platelet in bone marrow increased, obvious advantage was showed in long-term effect, and adverse reactions were reduced with ITWM treatment in ITP patients. Conclusion The existed limited evidences suggest that ITWM treatment of ITP can significantly improve clinical efficacy. The quantity and quality of research literature were limited, and well-designed randomized double-blind controlled trials with large sample and multi-centers are required for a more reliable conclusion.
2.Determination of Baicalin in Yanyan Capsules by HPLC

OBJECTIVE:To establish an HPLC method for quantitative determination of baicalin in Yanyan capsules. METHODS:The determination was performed on Kromasil C18(250 mm?4.6 mm,5 ?m) with mobile phase consisted of meth-anol-water-phosphoric acid(47∶53∶0.2).The detection wavelength was set at 280 nm. RESULTS:The linear range of baicalin was 0.125~1.216 ?g(r=0.999 6) and its average recovery was 99.15%(RSD=0.62%,n=6).CONCLUSION:The method is simple and accurate,and it can be used for the quality control of Yanyan capsule.
3.Illusion of transparency between individuals of high and low self-monitoring in two different type of interactive situations

ObjectiveTo examines the difference in illusion of transparency between high and low selfmonitoring individuals in different situations in reference to the experimental paradigm ofdrink recognition by Gilovich(1998 ).MethodsHigh and low self-monitoring individuals (the subjects) were selected by Snyder's Self-Monitoring Scale.Two situations were designed to study the illusion of transparency.One was acquaintance situation,and the other was the stranger situation.Results 1.The differences among the high and low selfmonitoring individuals had influence on the illusion of transparency.In low self-monitoring individuals there was a stronger illusion of transparency being demonstrated among both acquaintance situation (4.53 ± 2.12,2.53 ±2.45; t (16) =4.41,P<0.05) and stranger situation (4.33 ±2.93,3.11 ±2.89; t (17) =2.11,P=0.05),while in high self-monitoring individuals,there were no illusion of transparency in either situations ( t (12) =0.38,P > 0.05 ; t (22) =1.09,P> 0.05 ).2.The level of illusion of transparency in low self-monitoring group among acquaintance situation (2.00 ± 2.17) was higher than high self-monitoring group(0.23 ± 1.87 ),t (28) =2.40,P <0.05 ),while there was no difference between two groups in stranger situation (1.22 ±2.46,0.74 ±3.25,t (39) =0.52,P >0.05).ConclusinThe differences among the high and low self-monitoring individuals have inlluence on the illusion of transparency.The interactive situations have an influence on the level of illusion of transparency.
4.Performance evaluation and results analysis of chemiluminescence method for detecting plasma BNP

Objective To analyze and evaluate the precision ,accuracy and linearity of the chemiluminescence method for detec‐ting plasma BNP .Methods According to the experimental schemes of CLSI EP15‐A2 ,EP6‐A files and other relevant documents , the precision ,accuracy and linearity of the Siemens Centaur XP chemiluminescence instrument for detecting BNP ,and the detection results were compared with the performance declared by manufacturer or the quality target formulated by laboratory .Results The imprecision of BNP detected by the chemiluminescence method was less than 1/3 TEa regulated by the Clinical Laboratory Center of Ministry of Health (allowable total error);the variation coefficient index (CVR) of internal quality control data was less than ± 2;the relative bias of the results of external quality control blind samples with the target values were less than Tea regulated by the Ministry of Health ;the linear evaluation results showed that BNP was once linearity in the range of 5 .3‐4696 .7 pg/mL .Conclusion The precision ,accuracy and linearity of the Siemens Centaur XP chemiluminescence instrument for detecting BNP can accord withthe quality objectives requirements and meet the clinical needs .
5.Effects of glucagon-like peptide-Ⅱ on residual small bowel absorption of glucose in a rat model of short bowel syndrome

Objective: To study the effect of GLP-Ⅱ on residual bowel absorption of glucose in a rat model of short bowel syndrome. Methods: Rats undertaken 75% small bowel resection were randomly assigned to SB group and SB/GLP-Ⅱ group. Another group of weight-matched normal rats(Normal) received normal diet ad libitum was used as control group for SB group. All rats were allowed normal chow on the 1st postoperative day(POD). On the POD6,the intestinal glucose absorption data per unit length as well as per unit weight of ileum were measured by in vivo circulatory perfusion experiment. Results: Length of perfused ileum in 3 groups of rats were not significantly different. Wet weight to length ratio of perfused ileum in SB group was significantly higher than that of normal group,but significantly lower than that of SB/GLP-Ⅱ group. 45min glucose absorption of per cm ileum in SB/GLP-Ⅱ group was significantly higher than that of SB group(P0.05). Conclusions: In a rat model of SBS, GLP-Ⅱ can enhance the intestinal glucose absorption measured as per unit length of resudial ileum and GLP-Ⅱ manifests a trend to increase the glucose absorption measured as per unit wet weight of resudial ileum.
6.Expression of Cripto-1 in basal-like breast carcinoma and its clinical significance

Objective To detect the protein expression of Cripto-1 and CD34 in basal-like breast carcinoma and study the functions of Cripto-1 in basal-like breast cancer angiogenesis,invasion and metastasis.Methods Immunohistochemistry (IHC) was used to detect the expression of Cripto-1 and CD34 in 60 cases of basal-like breast cancer,40 cases of non basal-like breast cancer and 20 cases of normal breast tissues.The clinical relationship between Cripto-1 and basal-like breast cancer were analyzed as well as its connection with CD34 markers MVD.Results The expression of Cripto-1 protein in basal-like breast cancer (42/60,70.00 %) was significantly higher than that of nonbasal-like breast cancer (19/40,47.50 %) and normal breast tissue (4/20,20.00 %) (both P < 0.05).The expression of Cripto-1 protein was positively correlated with the tumor size,clinical stage,lymph nodes metastasis and MVD value in patients of basal-like breast cancer (all P < 0.05),but there was no corelation with the age of the patients (P > 0.05).Conclusions The up-regulation of Cripto-1 expression may be associated with angiogenesis and development in basal-like breast cancer.The effect of Cripto-1 promoting angiogenesis may be an important factor in the acceleration of the local invasion and distant metastasis in basal-like breast cancer.
7.EFFECT OF TOTAL FLAVONOIDS OF CHRYSANTHEMUM INDICUM ON THE APOPTOSIS OF ADJUVANT ARTHRITIS RAT SYNOVIOCYTES AND THE EXPRESSION OF CASPASE-3

Objective To study the effect of the extract of total flavonoids of Chrysanthemum indicum(TFC) on adjuvant arthritis(AA) synoviocytes.Methods Totally 0.1ml of the complete Freund's adjuvant was subcutaneously injected into the right hind feet pads of 20 SD rats.24 days after immunity synoviocytes in the knee joint were treated with TFC.Cell morphology was examined with electron microscopy.Protein level of caspase-3 cleaved fragments was analyzed by Western blotting.The annexin V stain assay was applied to explore the effect of caspase-3 inhibitor on the amelioration in synovial cells apoptosis of AA rats.Results Typical morphology and biochemical feature of apoptosis in synovial cells of AA rats were observed with TFC.The protein level of caspase-3 cleaved fragments increased obviously and was related with the concentration of TFC in synovial cells of AA rats.The apoptotic cells positively stained with annexin V were markedly reduced by caspase-3 inhibitor.Conclusion TFC can induce apoptosis in AA rats synoviocytes,which may achieve therapeutical effects in AA.The activation of caspase-3 may be one of the main causations.
8.Determination of Ferric Oxide in Calamine Powder by FAAS

Objective:To determine the content of ferric oxide in calamine powder. Methods:Flame atomic absorption spectrom-etry ( FAAS) was used to determine the content of ferric oxide. The determination results were compared with those of the titration method. Results:There was a good linear relationship between the absorbance and the concentration of iron within the range of 0. 5-4μg·ml-1 , the relative standard deviation of the repeatability test was 1. 2%, the average recoveries were 100. 4% and 99. 4%, the limit of quantitation and detection was 0. 081 μg·ml-1 and 0. 024 μg·ml-1 , respectively. Conclusion:The method of FAAS is ac-curate and quick with good specificity and high sensitivity, which can be used for the determination of ferric oxide in calamine powder.
9.Expression of S100A7 in basal-like breast carcinoma and its clinical significance

Objective To detect the protein expressions of S100A7 and CD34 in basal-like breast carcinoma and analyze the clinical relationship between S100A7 and basal-like breast cancer and the connection with CD34 markers microvessel denisity (MVD).To study the function of S100A7 in basal-like breast cancer angiogenesis,invasion and metastasis.Methods Immunohistochemistry (IHC) was used to detect the expressions of S100A7 and CD34 in 60 cases of basal-like breast cancer,40 cases of non basal-like breast cancer and 20 cases of normal breast tissues.Results The expressions of S100A7 in basal-like breast cancer,non basallike breast cancer and normal breast tissues were 83.33%,62.50% and 35.00% respectively.There were significant differences among them (x2 =5.556,P < 0.051; x2 =17.107,P < 0.05; x2 =4.051,P < 0.05).There was positive correlation between S100A7 and lymph node metastasis,TNM staging and MVD of basal-like breast cancer patients.But there was no significant correlation between S100A7 and patient's age and tumor sizes.Conclusion The up-regulation of S100A7 expressions may be associated with the occurrence and development,invasion and metastasis,angiogenesis of basal-like breast cancer.The effects of S100A7 promoting angiogenesis may be an important factor in the acceleration of the local invasion and distant metastasis in basallike breast cancer.
10.Construction of tissue-engineered skin by culture of human amniotic epithelial cells on human deepidermized dermis for the repair of skin defects in nude mice: an experimental study

Objective To evaluate the feasibility of repair of full-thickness skin defects in nude mice with tissue-engineered skin which was constructed by culture of human amniotic epithelial cells (hAECs) and fibroblasts on human de-epidermized dermis (DED).Methods Healthy human amniotic tissues were treated with trypsin at a low concentration in multi-steps to prepare hAECs,and a two-step collagenase digestion was used to treat healthy children's prepuce tissues to prepare fibroblast suspensions.When fibroblasts were cultured in vitro up to passage 3-5 and hAECs up to passage 2,they were seeded on the reticular dermal surface and basement membrane surface of the DED respectively to construct the tissueengineered skin.A total of 20 heahhy male nude mice aged 3-4 weeks were enrolled into this experiment,and full-thickness skin defects were made on the middle of the back of mice.Then,these mice were randomly divided into 2 groups by using a lottery method,and reconstructed full-thickness tissueengineered skin grafts and vaseline oil gauze were used to cover the wounds in the tissue-engineered skin group and control group respectively.The whole body and transplantation sites of the nude mice were observed on day 7,14,21 and 28 after transplantation,the wound healing time and rate were compared between the above two groups,and skin tissues at the transplantation site were harvested at 4 weeks after transplantation and subjected to histological examination.Results HAECs had stem-cell characteristics and expressed octamer-binding protein-4 (OCT-4) and embryonic marker stage-specific embryonic antigen4 (SSEA-4).After 2-week organ culture,the in vitro reconstructed tissue-engineered skin showed 4-9 continuous layers of stratified epithelium,and the histological structure of the epidermis was similar to that of the normal human skin.Compared with the control group,the tissue-engineered skin group showed significantly higher wound healing rates on day 7,14 and 21 after transplantation (57.49% ± 6.11% vs.22.93% ± 4.26%,92.80% ± 3.10% vs.54.57% ± 7.94%,98.83% ± 0.25% vs.91.16% ± 4.79%,respectively;n =10,t =27.36,32.23,11.80,respectively,all P < 0.001),shorter wound healing time [(21.51 ± 1.51) d vs.(28.80 ± 1.14) d,n =10,t =42.23,P < 0.001],with the color of skin grafts closer to that of autologous skin on day 28 after transplantation.Histological examination revealed distinct stratification of the epithelium,obvious keratinization and favorable growth of cells in the dermis in the tissue-engineered skin group,but thin epithelium with some defects,indistinct stratification of the dermis,and inflammatory cell infiltration in the control group.Condusion Tissue-engineered skin constructed by the culture of hAECs and fibroblasts on human DED can survive in nude mice after transplantation,resulting in a more favorable healing of wounds,and is expected to serve as a kind of ideal tissue-engineered skin.
