Hypertrophic cardiomyopathy is an autosomal-dominant disease.Disease-causing mutations have been found in genes encoding structural components of the thick and thin filament systems of cardiac myocyte;it has therefore been named as a disease of sarcomere.Many approaches have been used to characterize the pathogenesis of the desease.Transgenic animal models have been created to gain further insight into the pathogenesis of this disease.Most of these models has been made in mice;however,recently a transgenic rabbit model has been created.In addition,there are several natural-occurring forms of HCM in animals.The discovery of responsible genes and the elucidation of the molecular mechanisms of pathogenesis through the use of animal models promise improved and early diagnosis and the potential for mechanism-based therapeutics.

Objective To study the double restriction fluorescent labeling (DRFL) method for fluorescent labeling of trace DNA samples and its effect in enhancing the pathogen detection sensitivity of microarray assays. Method SARS-CoV RNA samples were reversely transcribed and then further amplified with the restriction display (RD)-PCR and fluorescently labeled by conventional restriction labeling directly with Cy-universal primer and the novel double labeling with Cy-universal primer and CydNTP. The labeled samples were applied to the microarray with the viral probes, processed and analyzed. Results Compared with the conventional method, DRFL labeling resulted in 3. 5835 times higher fluorescent intensity of all the SARS probes on average, even though increased fluorescent intensities for different probes varied considerably. Conclusion Signal to noise ratio can be enhanced by the DRFL method which improves the sensitivity of microarray technology in trace pathogen detections.

Objective To investigate the calbindin D\|28K (CB) neurons that receive the visceral nociceptive information in the interstitial nucleus of the spinal trigeminal tract (INV) directly project to the nucleus of the solitary tract (NTS). Methods Triple\|labeled methods of fluorogold (FG) retrograde tracing combined with Fos and CB proteins immunofluorescence histochemistry after formalin stimulating upper alimentary tract. Results Most of FG\|retrograde labeled neurons distributed in the paratrigeminal nucleus (PaV) and the dorsal paramarginal nucleus (PaMd) of INV ipsilateral to the FG injection. About 71\^2% of FG\|retrograde labeled neurons contained CB and 31\^5% of FG/CB double\|labeled neurons exhibited positive Fos\|immunoreaction in INV.Conclusion The results suggested that a part of neurons containing CB in INV receive the visceral nociceptive information and directly project to NTS. CB neurons might play an important role in transmitting visceral nociceptive information from INV to NTS. [

The new course standard of nursing specialty in military hygiene was established against the main problems in military hygiene teaching and considering the peacetime and wartime need of nursing post.In this paper,guiding ideology,content choosing and the request of operational skill of this standard were discussed in detail.

Objective Based on the age of big data, the treatment mechanisms of Qishen-Yiqi(QSYQ) for myocardial infarction was focused. Methods All the data stemmed from Gene Expression Omnibus (GEO). For ensuring the efficacy genetic molecular, differentially expressed genes were discobered by Qlucore Omics Explorer (QOE). And then gene set enrichment analysis was performed by Database for Annotation, Visualization and Integrated Discovery online Gene Annotation system(DAVID) for showing the genes functions during bioprocess. In the end, the predicted genes and proteins interactions networks were demonstrated by Gene/protein interaction database (STRING). Results The main biological process involved up regulating the expression of some specil genes such as NFIL3, ARNTL, DBP, FGD4 and nuclear receptor genes like NR1D1, NR1D2 while using QSYQ treatment. In such case, BHLHE40/41 was regulated. Conclusion Traditional Chinese medicine, QSYQ, could influence the expression of genes of cytothesis, inflammatory and enzymatic activity as its effect of the molecular mechanism, in order to treat and cure the myocarditis infarction.

Objective To develop a better probe preparation method of gene chips for the further gene chips fabrication and its application in the gene expression. Method Using human leukemia K562 cells as material,applying a new kind of technology for gene differential display restriction display(RD PCR),we isolated a lot of cDNA fragments as probes of gene chips. Results This method has overcome the shortcoming of more pseudopositives in DD PCR,and was easy to be manipulated.Compared with the chip fabricated with full cDNA probes,the probe length is comparatively homogeneous,and the hybridization dynamics is apt to be controlled.Conclusion\ RD PCR technology could provide a new strategy for the probe preparation of gene chips.\;[
