1.Relationship between choroidal telangiectasia and subfoveal choroidal thickness in patients with central serous chorioretinopathy

Objective To investigate the relationship between choroidal telangiectasia and subfoveal choroidal thickness in the patients with central serous chorioretinopathy.Methods Eighty-four patients (84 eyes) with central serous chorioretinopathy in our hospital from February 2015 to April 2016 were selected,based on the fluorescein angiography the patients were divided into 3 groups:Mild group (leakage at 9-10 minutes after injection,26 cases),moderate group (leakage at 5-8 minutes after injection,37 cases),severe group (leakage at 5 minutes after injection,21 cases).The choroidal capillary dilatation was assessed by OCTA,and subfoveal choroidal thickness in three groups and patients with different foveal choroidal expansion degree were measured by EDI-OCT,and correlation analysis was performed.Results The proportion of severe dilation of choriocapillae in mild group,moderate group,severe group were 7.69%,13.52% and 23.81%,respectively.The subfoveal choroidal thickness was sequentially increased in mild group,moderate group,severe group (P < 0.05).The subfoveal choroidal thickness in patients with mild,moderate and severe dilatation of choroidal capillaries were (306.59 ± 74.18) μm,(367.21 ± 85.04) μm and (416.27 ± 104.56) μm,respectively (P < 0.05).The degree of choroidal telangiectasia in patients with central serous chorioretinopathy had a positive correlation with the thickness of subfoveal choroid (r =0.812,P =0.037).Conclusion The degree of choroidal telangiectasia and subchondral choroidal thickness in patients with central serous chorioretinopathy are significantly increased with the severity of the disease,and there is a positive correlation between them.
2.Establishment of plantlet regeneration system for Aconitum carmichaeli

Object To establish an effective plantlet regeneration system of Aconitum carmichaeli Debx. for the purpose to obtain a large number of high quality seedling in a short time. Methods Leaves in vitro were tried as the explants and cultivated in different media with the various portion of hormones. Results The medium of MS+BA 1.0 mg/L+KT 0.5 mg/L+NAA 0.2 mg/L was the most suitable one for the induction of shoots; the medium of MS+BA 1.5 mg/L+NAA 0.1 mg/L was beneficial to the propagation of shoots; the medium of MS+BA 0.5 mg/L+NAA 0.1 mg/L was good for the elongation of shoots; and the medium of 1/2 MS without any hormone was suitable for the induction of roots. Conclusion Rapid propagation of A. carmichaeli could be achieved by tissue culture and this will lead to the possibility for its seedling in the industrial production.
3.Effects of gefitinib combined with DC-CIK on advanced lung adenocarcinoma

Objective To investigate the short-term efficacy and safety of gefitinib combined with dendritic cells-cytokine induced killer (DC-CIK) cells in the treatment of advanced lung adenocarcinoma.Methods Fifty patients with lung adenocarcinoma who in previous had underwent radiotherapy and first-line chemotherapy failure received DC-CIK in combination with gefitinib treatment,blood count changes,imaging data,carcinoembryonic antigen and changes in the quality of life before and after treatment were compared and evaluated.Results DC-CIK could improve effectively and relieve bone marrow suppression response after chemotherapy and significantly increase the WBC content of blood (P < 0.01).After treatment,the tumor carcinoembryonic antigen (CEA) was significantly lower in patients (P < 0.01).Fifty cases of patients in complete remission (CR) were 0 cases,partial remission (PR) were 24 cases,stable disease (SD) were 17 cases and progression (PD) were 9 cases,and the response rates (RR) were 48%;The quality of life of patients was significantly improved (P < 0.05).Common adverse reactions were rash,diarrhea,chill and fever,but mild symptoms could be relieved after symptomatic treatment.Conclusions Gefitinib with autologous DC-CIK cell infusion second-line treatment of advanced lung cancer have a certain short-term efficacy,without significant adverse reactions.Gefitinib with autologous DC-CIK cell therapy can mitigate the response of bone marrow suppression in patients with advanced lung cancer and improve the quality of life of patients.Long-term effect remains to be investigated.
4.Effects of electroacupuncture on the pain threshold and the NMDA R1 mRNA in DRG on neuropathic pain rats.

To observe the effect of multiple electroacupuncture (EA) on the pain threshold and the regulation of N-methyl-D-aspartate (NMDA) receptor in dorsal root ganglia (DRG) of neuropathic pain rats. Rats were prepared with a unilateral chronic constriction injury (CCI) to the sciatic nerve. EA was done in acupoints "Huan Tiao" and "Yang Ling Quan" for 30 min every day and the thermal thresholds were detected after EA at 3, 5, 7, 10, 14 days after operation. On day 14 after nerve injury, the in situ hybridization method was used to investigate the change of NMDA R1 mRNA in L4-L5 DRG. The thermal threshold reduced significantly from day 3 after operation in CCI rats. After multiple EA treatment, the ipsilateral thermal hyperalgesia relieved gradually and the thermal threshold had no difference with control side after day 5 (P > 0.05). From Day 7 after operation, the thermal threshold at each time point were significantly different compared with CCI group respectively (P > 0.05). Moreover the EA had accumulative effect. On Day 14 after operation, the NMDAR1 mRNA positive neurons and the mean optic density in ipsilateral L4-5 DRG were less than that of control side (P < 0.05), mainly in medium and small neurons. After EA treatment, the NMDAR1 mRNA positive neurons in ipsilateral DRG had no considerable difference comparing with those of control side, significantly increased comparing with CCI group (P < 0.05). It's concluded that the NMDA receptors in DRG relate closely with the generation and development of neuropathic pain. The multiple EA treatment can attenuate the thermal hyperlagesia of neuropathic pain rats and regulate the NMDA receptor.

5.Impact of different adjuvants on immunogenicity of the HBV particle vaccine containing the S + preS1 fusion antigen in Balb/C mice.

We previous reported the development of novel hepatitis B virus(HBV) vaccine containing the surface antigen(S) plus PreS1 fusion derived from Chinese hamster ovary (CHO) cells system. In this study, we analyzed the impact of different adjuvants on immunogenicity of the HBV particle vaccine in Balb/C mice, including alum alone, CpG oligodeoxynucleotides (CpG-ODN) alone and CpG-ODN in combination with alum adjuvant. We first detected the antigen specific humoral response in mice, including total IgG antibody and IgG subtyping. Then, we characterized the specific cell-mediated immune (CMI) response by detection of gamma-interferon secreting splenocytes after stimulating with S or PreS1 peptide pool. Our results showed that: CpG-ODN adjuvanted vaccine could rapidly induce higher level of anti-PreS 1 and anti-S antibodies, and a higher ratio of IgG2a/IgG1 antibody than that of alum adjuvanted vaccine. At the same time, CpG-ODN adjuvanted vaccine induced robust antigen-specific cellular immune responses in mice, which was superior to that of alum adjuvanted vaccine and CpG-ODN in combination with alum adjuvanted vaccine; however, the vaccine candidate with CpG-ODN in combination with alum adjuvant induced highest anti-S antibody and mixed IgG subclasses in mice after twice immunization. There exists dominant HBV CMI epitopes in the N-terminal of S antigen. These results provided important evidence that CpG-ODN adjuvanted HBSS1 particles vaccine may serve as a novel candidate in the development of new preventive and therapeutic agents against hepatitis B infection.
