1. Determination of imatinib in rat plasma by HPLC and study of its pharmacokinetics

OBJECTIVE: To develop a high performance liquid chromatography method for the determination of imatinib in rat plasma and study its pharmacokinetics. METHODS: Plasma was deposited by perchloric acid. The analytical column was ZORBAX SB-C18 (4.6 mm × 150 mm, 5 μm). The mobile phase was acetonitrile-water-0.1% trifluoroacetic acid (19:61:20) and the flow rate was 1.0 mL · min-1. The LV detection wavelength was 282 nm. Six male SD rats were given a single dose of 50 mg · kg-1 imatinib by garage. Blood samples were collected from tail vein at different time points. The concentrations of imatinib in plasma were determined by the established HPLC method. The pharmacokinetic parameters were analyzed by DAS program. RESULTS: Excellent liner relationship was obtained in the range of 0.10-20.00 mg · L-1 (r=0.9998), and the lower limit of qauntification was 0.05 mg ·L-1. The recoveries were (98.86±2.77)%, (100.35±2.31)% and (100.14±1.66)% respectively at three concentrations (0.25, 5.00, 15.00 mg · L-1), the intra-day RSDs were 2.64%, 2.50% and 1.62% with REs of -1.60%, -1.00% and 0.10%, and the inter-day RSDs were 3.51%, 2.77% and 1.34% with REs of -0.80%, 0.72% and 0.21%, respectively. After oral administration in rats, the pharmacokinetic profile of imatinib was fitted with a two-compartment model, the half-lives of α phase and β phase were (2.40±0.84) h and (7.82±0.87) h, respectively. CONCLUSION: The method is accurate, simple, rapid and can be used to determine imatinib concentration in rat plasma and study its pharmacokinetics.

Objective:
To investigate the effect of Shugan Jiayu capsule on depression in patients with schizophrenia.
Methods:
From January 28, 2016 to January 24, 2017, 82 schizophrenic patients in Wenling Psychiatric Rehabilitation Hospital were selected in the study.All the patients had depressive symptoms and were divided into two groups by random number table method, with 41 cases in each group.Both two groups were treated with routine therapy.The control group was treated with quetiapine, and the observation group was given Shugan Jieyu capsule.The improvement of the disease and the occurrence of adverse events in the two groups were observed.

Objective To evaluate the feasibility of prospective electrocardiogram(ECG)-gating computed tomography coronary angiography(CTCA).Methods Sixty patients with suspected or known coronary artery disease underwent 64-slice CTCA using prospective ECG-gating.Multi-planar reconstruction (MPR),curved-planar reconstruction(CPR),maximum intensity projection(MIP)and volume rendering (VR)were used to demonstrate the coronary arteries.The image quality and radiation dose was evaluated.Results The mean effective radiation dose was(2.7±0.2)mSv.93.3%(720/772)segments of all coronary arteries were of diagnostic image quality,44.2%(341/772)Was classified as excellent and 49.1%(379/772)was good.Non-diagnostic coronary segments were found in 6.7%(52/772)of all coronary arteries.Therewere 5(8.3%)cases with severe coronary stenosis(>75%)or occlusion,17(28.4%)cases with moderate stenosis(50%-75%),18(30.0%)cases with mild stenosis(<50%)or irregular lumen,20(33.3%)cases with normal coronary artery.Conclusion With a low radiation dose,prospective electrocardiogram(ECG)-gated coronary 64-MSCT angiography has a good potential for the detection of coronary stenosis,especially for excluding coronary artery disease.

OBJECTIVE To assess the prevalence and analyze the influence factors of laryngopharyngeal reflux disease(LPRD) in the Fuzhou region, in order to provide a theoretical basis for the development of prevention and control measures for LPRD. METHODS A questionnaire survey in residents in Fuzhou by a random cluster sampling was carried out. Individual information, reflux symptom index(RSI) of Belafsky and risk factors were included. Patients more than 13 scores of RSI were defined as LPRD. Data were statistically analyzed. RESULTS A total of 4100 residents were investigated, 4063 of them were available. The prevalence of LPRD was 5.00%. Often eating too much, often drinking strong tea, menolipsis, rhinitis, tonsillitis were closely related to LPRD. CONCLUSION The prevalence of LPRD in Fuzhou region were closely related to many factors.

8.Prognostic significance of pelvic lymphadenectomy in patients with bladder cancer

Objective:To evaluate the prognostic significance of standard pelvic lymphadenectomy on the disease-free survival (DFS) rate of bladder cancer patients undergoing radical cystectomy (RC) and to discuss the influencing factors of lymph node positivity and the relationship between positive lymph nodes and lymphadenectasis. Methods:This prospective analysis includes 120 cases of bladder cancer treated with pelvic lymphadenectomy and RC in Tianjin Medical University Cancer Institute and Hospital between 2008 and 2013. The cases were divided into two groups, namely, the standard pelvic lymphadenectomy group (Group A) and the nonstandard pelvic lymphadenectomy group (Group B). The relationships among positive lymph nodes, lymphadenectasis, tumor stage, and patho-logical grade were retrospectively analyzed. Results:The 1-, 3-, and 5-year overall survival rates of 120 patients were 84%, 69.9%, and 57.9%, respectively. Group A was significantly correlated with a better 3-year overall survival rate than Group B, i.e., 78.4%vs. 46.2%(P<0.05). Lymphadenectasis influenced the DFS rate of bladder cancer patients after RC with pelvic lymphadenectomy, i.e., 50.0%vs. 86.4%(χ2=9.303, P<0.05). Meanwhile, lymphadenectasis was positively correlated with lymph node positivity (P<0.001). Tumor stage, histological subtype (urothelial carcinoma and non-urothelial carcinoma), and age were the prognostic factors for bladder cancer (P<0.05). Conclusion:Intraoperative lymphadenectasis is the influencing factor of lymph node positivity. This study determined that standard pel-vic lymphadenectomy and lymphadenectasis may influence the DFS rate after RC and are the independent risk factors for the prognosis of bladder cancer. Creating evidence-based guidelines of standardized lymphadenectomy for further improvement of the surgical quali-ty and survival of bladder cancer patients is essential.

Objective:To evaluate the promoting surgery incision healing effect of recombinant human acidic fibroblast growth fac-tor ( rh-aFGF) in the treatment of children with open fractures. Methods:A multicenter, randomized and controlled clinical trial was conducted to study the efficacy of rh-aFGF. Totally 120 cases of injured children (age<14y) were randomly divided into two groups, the treatment group (n=60) was given rh-aFGF washing during the operation and spraying after the operation, and the control group ( n=60) was treated with normal saline. Both groups were given traction, screw or Kirschner wire fixation. The healing time, healing status and delayed healing were observed and compared in the two groups. Results:Stage I healing rate, the complete healing time and delayed healing rate in the treatment group was 86. 6%, (20. 3 ± 5. 6)d and 3. 3%, respectively, while that in the control group was 56.7%, (23.4 ±6.2)d and 15.0%, respectively. The differences between the two groups were significant (P<0.05 or 0.01). Conclusion:Rh-aFGF can effectively promote wound healing and shorten the healing time. For all these positive aspects,rh-aFGF de-serves wider clinical application in postoperative rehabilitation after open fractures.
