1.Color Doppler flow imaging in acute cholecystitis to predict technical difficulties during laparoscopic cholecystectomy

Objective To evaluate the value of preoperative color Doppler flow imaging findings for predicting possible difficuulties encountered during laparoscopic cholecystectomy (LC).Methods Eighty-six Datients with acute cholecystitis underwent color Doppler flow imaging examination were divided into operation difficult group(67 cases)and operation easy group(19 cases)according to the diffculty score.The parameters were measured pre-operation including the volume of gallbladder,the thickness of the gallbladder wall,the condition of arterial flow in the gallbladder wall,the conditions of gallbladder cavity and gallbladder fossa and the intra-and extra-hepatic bile duet.The relationships among imaging results,operation difficulties and operation findings were investigated.Results Gallbladder volume,gallbladder wall thickness.the presence rates of plentiful arterial flow in the gallbladder wall,adhesion of gallbladder and stone incarceration In operation difficult group were significantly different from those in operation easy group [(52.6±14.6)mm~3 vs(32.6±10.4)mm~3,(9.7±4.1)mm vs(3.8±0.9)mm,89.5%(17/19)vs 17.9%(12/67),78.9%(15/19)vs 11.9%(8/67),10.5%(2,19)vs 0(0/67)](P<0.05 or<0.01).The accurate rate was 94.2%(81/86)in predicting.Conclusion Preoperative color Doppler flow imaging is helpful in predicting difficulties of LC.
2.Color Doppler ultrasonography in monitoring thrombosis of lower limb deep vein after malignantthoracic tumor surgery

Sixty patients underwent surgery for malignant thoracic tumor were evenly randomized into control group (group A) and treatment group (group B).0.3 ml low-molecular-weight heparin was injected per 12 h for 3 d in group B from 24 h after surgery.Bilateral venous sonography of lower extremities were performed before and d1,d3 after operation.Blood coagulation marks PLT,PT,APTT,Fibrinogen (FIB)and D-dimer were determined at the same time.Diagnosis of calf vein thrombosis after surgery was confirmed by color Doppler sonography in 7 patients:1 in group B (3%) and 6 in group A (20%) (P < 0.05 ).Thrombosis at calf intermuscular vein was found in 6 patients and thrombosis at posterior tibial vein in 1 patient.FIB and D-dimer had no significant difference on the dl ( P > 0.05 ) between two groups,but significantly lower on the d3 after surgery in group B than that in group A ( P < 0.05 ).Low-molecularweight heparin reduced blood hypercoagulation state and thrombosis.Color Doppler sonography can be a valuable method for detecting asymptomatic deep vein thrombosis in the early stage after surgery and monitoring the efficacy of thrombosis prevention.
3.Tet: novel anti-tumor drug target based on DNA demethylation

Tet (ten-eleven translocation) proteins belong to α-ketoglutaric acid (α-KG or 2-OG) and Fe2+ dependent dioxygenases. Tets are found to be involved in the unique mammalian DNA active demethylation process by specifically oxidizing the methyl group of 5-methylcytosine (5mC) in mammalian genome, and play critical roles in gene regulation in early embryonic development and stem cell differentiation via regulating the dynamic balance distribution of 5mC. Abnormal expression and function of Tets are closely associated with various hematological malignances, including myelodysplastic syndrome, chronic myelomonocytic leukemia, and acute lymphoblastic leukemia, as well as solid tumors. Hence, Tets and Tets-mediated DNA demethylation are novel anti-tumor drug targets. Investigation of biological function and catalytic mechanism of Tets is helpful for further understanding mechanisms of tumor incidence and development relevant to DNA demethylation pathway and can provide reference for developing new anti-tumor targeted drugs.
4.Expression and clinical significance of miR-34b in children with acute leukemia

Objective To research the expression,methylated regulation and clinical significances of miR-34b in chidren with acute leukemia(AL).Methods The methylation status of miR-34b promoter CpG islands were detected with methylation-specific polymerase chain reaction (MSP) in patients with AL.Then the expression of miR-34b was compared,which was detected by Taqman real-time fluorescence quantitative polymerase chain reaction (qRT-PCR),between the AL patients and normal group,in order to analyze their relationship with the clinical indicators.Resuits In 8 leukemia cell lines (U937,HL-60,MV4-11,M2R,K562,Raji,CCRF,DAMI) showed methylation,the positive rate of the methyl was 100%.Thirty-one acute lymphocytic leukemia(ALL) pediatric patients were newly diagnosed,and 24 cases showed methylation,the methylation positive rate 77.42% (24/31).Nineteen acute myeloid leukemia(AML) patients were newly diagnosed,and 8 cases showed methylation,the methylation positive rate was 42.11% (8/19 cases).There was no methylation in the 23 cases of normal children.The relative expression levels of miR-34b in the normal group,the group of leukemia cell lines,the group of ALL pediatric patients with newly diagnosed,the AML group and the group with mixed lineage leukemia gene rearrangement MLL+ were 5.22 ± 1.15,0.03 ± 0.03,1.65 ± 0.69,0.18 ± 0.06,0.64 ± 0.34,respectively.The findings indicated that there were significant differences in the relative expression levels of miR-34b between the normal group and the group of leukemia cell lines,the ALL group,the AML group,and the MLL+ group.The relative expression and methylated level of miR-34b had no statistically difference in gender,age at diagnosis,WBC count,chromosome,fusion gene,MLL gene rearrangement,and the minimal residual disease(LDH) levels in newly diagnosed AML patients(all P > 0.05).And the relative expression and methylated level of miR-34b had no statistically difference in gender,age at diagnosis,WBC count,immunophenotype,chromosome,fusion gene,MLL gene rearrangement,TEL/AML1 gene,risk stratification,the minimal residual disease (MRD) in thirty-three days and the LDH levels in newly diagnosed ALL patients(all P > 0.05).But as for the response to prednisone experiment,there was a significant difference between the sensitive group and the non sensitive group(P < 0.05).Conclusions The expression level of miR-34b in AL was significantly lower and it was regulated by methylation mechanism,which implies that miR-34b may play a role of a tumor suppressor gene in the pathogenesis of leukemia.MiR-34b may affect the early treatment response of ALL patients,and it may be an indicator of risk stratification and poor prognosis in pediatric ALL.
5.A comparative study on the quality of Radix Angelicae dahuricae Dispensing Granules produced by pharmaceutical factories

AIM:To compare the quality of Radix Angelicae dahuricae Dispensing Granules produced by different pharmaceutical factories.METHODS:The main coumarin compounds were identified by TLC and HPLC respectively;imperatorin was determined by HPLC.RESULTS:Imperatorin and other main coumarin compounds of Radix Angelicae dahuricae Dispensing Granules in the market could not be determined because of low content.CONCLUSION:Radix Angelicae dahuricae Dispensing Granules produced by different pharmaceutical factories in the market have distinct differences in quality,and were of poor quality in the mass.
6.The diagnostic value of different anthropometric indexes in the metabolic syndrome of Xinjiang Uygur population by ROC curve

Data were gathered from epidemiological survey of Xinjiang Hetian area in 2008 and 2 228 subjects were included.Waist-height ratio (WHtR),body mass index (BMI),Waist-hip ratio (WHpR) were calculated.Kappa test was applied to determine the concordance among different methods.The results showed that according to International Diabetes Federation 2005 diagnostic criteria for metabolic syndrome,it was better to apply WHtR in predicting metabolic syndrome by the receiver operating characteristic (ROC) curve than BMI,abdominal circumference,and WHpR.When the cutoff of WHtR was 0.53,the sensitivity and specificity in diagnosing MS were 91.2% and 71.8 %,with its area under ROC curve 0.878.Kappa test showed an intensive concordance between WHtR and waist circumference (WC).The specificity and sensitivity of diagnosing MS would be significantly raised by using both WHtR and WC.
7.Nursing care of difficulty in decannulation of PICC in 1 case of very low birth weight infant

Objective To summarize the nursing experience of 1 case of very low birth weight infant decannulation difficult in PICC. Methods The nursing key points included: full assessment analysis decannulation difficult reason, consult the PICC catheter outpatient health nurses, give magnesium sulfate hydropathic compress, mucopolysaccharide polysulfate cream local besmear outside, sanyrene outside, at the same time give low-molecular-weight heparin calcium injection subcutaneous injection such as anticoagulant active treatment and nursing. Results After 7 days ,the infant left axillary mass dispel, PICC pull out smoothly. Conclusions Decannulation difficult of very low birth weight infant requires full evaluation, multidisciplinary cooperation and specialist consultation, can give targeted personalized nursing safety smooth tube drawing, is worthy of reference for clinic.
8.Correlation of serum ADAMTS-4 and aβ2-GPl levels with white matter lesions in elderly patients with acute cerebral infarction

Objective To explore the correlation of serum levels of ADAMTS-4 and aβ2-GP1 anti-body with WML in elderly patients with ACI.Methods A retrospective study was conducted on 200 elderly ACI patients admitted in Department of Neurology of from January 2022 to January 2024.Based on Fazekas scoring,they were categorized into the control group(n=32),mild group(n=96),moderate group(n=49),and severe group(n=23).Their neurological function and ser-um levels of ADAMTS-4 and aβ2-GP1 were compared among the different groups.Results The NIHSS score and serum levels of ADAMTS-4 and aβ2-GP1 were significantly higher in the severe group than the moderate group,mild group and control group in order,and statistical difference were observed among them(P<0.05).Serum ADAMTS-4 and aβ2-GP1 levels were significantly positively correlated with the severity of WML(P<0.05).Serum ADAMTS-4 and anti-β2-GP1 were influencing factors for the occurrence of WML in elderly ACI patients(OR=2.149,95%CI:1.375-3.360,P=0.001;OR=1.702,95%CI:1.185-2.446,P=0.004).Conclusion The levels of serum ADAMTS-4 and aβ2-GP1 are closely related to the severity of WML and nerve damage in elderly ACI patients.
9.Effect of DNA methylation on the proliferation of leukemia cells regulated by miR-34b.

OBJECTIVETo study the expression level and CpG island methylation status of miR-34b in leukemia cell lines and to research the effect of DNA methylation on the proliferation of leukemia cells regulated by miR-34b.
METHODTaqman real-time fluorescence quantitative polymerase chain reaction (qRT-PCR) was carried out to detect the relative expression of miR-34b in control group (bone marrow cells of 20 children without blood disease) and 8 leukemia cell lines (U937, HL-60, MV4-11, M2R, K562, Raji, CCRF, DAMI). Methylation-specific polymerase chain reaction (MSP) was carried out to detect the methylation differences of miR-34b in control group (Bone marrow cells of 23 children without blood disease), 8 leukemia cell lines. HL-60 and K562 were treated with methyltransferase inhibitor (5-aza-2-dC) for further detection of its methylation status and expression of miR-34b. Hsa-miR-34b mimics was transfected into K562 cell by liposome, the transfection efficiency was detected by flow cytometry. The cell proliferation of hsa-miR-34b transfected group in each stage was measured with CCK-8 assay, and then compared with non-transfected group and negative control group.
RESULTThe relative expression level of miR-34b in the group of children without blood disease and the group of leukemia cell lines were 5.22 ± 1.15, 0.03 ± 0.03. The results showed that, the group of leukemia cell lines was significantly different from the control group (t = 4.538, P < 0.01) . Eight leukemia cell lines showed methylation, the positive rate of the methyl was 100%. There was no methylation in the 23 cases of control group. After leukemia cell lines HL-60 and K562 were treated with 5-aza-2-dC, the methylated bands became obviously weakened, and the relative expression levels of miR-34b substantially increased 49.5 times and 18.8 times respectively. After hsa-miR-34b mimics was transfected into K562 cell by liposome, its transfection efficiency detected by flow cytometry was 61% and the cell proliferation was measured with CCK-8 assay from which it was found that the cell proliferation was significantly suppressed compared with the control group at 48 h (t = 9.303, P < 0.01), 72 h (t = 65.617, P < 0.01), 96 h (t = 36.878, P < 0.01) and 120 h (t = 18.748, P < 0.01) in hsa-miR-34b transfected group, with the inhibition rate of 12.2% (48 h), 45.7% (72 h), 32.5% (96 h) and 22.9% (120 h).
CONCLUSIONThe hypermethylation of promoter leads to decrease in the expression levels of miR-34b in leukemia cell lines, which attenuate mechanism of proliferative inhibition may be one of the reasons of occurrence or development of childhood leukemia.

10.Interpretation on Dietary Guidelines for Chinese School aged Children(2022)

Abstract
School aged children grow and develop rapidly, and adequate nutrition is the material basis for their normal intellectual and physical development. It is necessary to formulate the dietary guidelines for school aged children on the basis of the dietary guidelines for the general population, and revise in time according to the changes in the nutritional health status of school aged children. The revision of the dietary guidelines for school aged children is based on the content of Dietary Guidelines for Chinese School aged Children (2016). In addition, according to the revision principles and procedures of the Dietary Guidelines for Chinese Residents, as well as comprehensive consideration of the current prominent dietary and nutritional problems of school aged children in China, Dietary Guidelines for Chinese School aged Children (2022) is finally issued after evidence based researches, discussion and consultation. Dietary Guidelines for Chinese School aged Children (2022) highlights the improvement of nutritional literacy, the cultivation of healthy eating behaviors, the building of healthy food environment, as well as updating the recommendation concerning diet and active physical activity. Dietary pagodas for school aged children of different three ages including 6-10 years, 11-13 years and 14-17 years are added to the guideline. The Guideline provides practical references for parents, teachers, school health workers, medical staff in child health services and workers in centers for disease control and prevention.
