1.A study on platelet indices and platelet membrane glycoprotein in children with primary immune thrombocytope-nia

ObjectiveTo investigate the change of the platelet function in children with primary immune thrombocy-topenia (ITP) before and after treatment.MethodsThe platelet indices, immature platelet fraction (IPF%), immature platelet counts (IPC), granule membrane glycoproteins of platelet (CD62p, PAC-1, CD42b) were tested by automatic blood cell analyzer and lfow cytometry (FCM) in 18 children with ITP before treatment and after complete response and in 17 children undergoing elective surgery (control gourp).ResultsCompared with control group, MPV, PDW, P-LCR and IPF% were signiifcantly higher and PLT, PCT, IPC were signiifcantly lower in children with ITP. The expressions of three platelet membrane glycoproteins (PAC-1, CD62p, CD42b) were signiifcantly lower in children with ITP. After treatment, children with ITP achieved completely response. Compared with before treatment, MPV, PDW, P-LCR and IPF% were signiifcantly lower, and PLT, PCT, IPC, and the expressions of three platelet membrane glycoproteins (PAC-1, CD62p and CD42b) were significantly higher. ConclusionsThe platelet function is depressed and in status of low activity in children with ITP. The relative platelet indices provide new references for the diagnosis and the judgment of therapeutic effect in ITP patients.

Objective To research the effects of metformin on proliferation and apoptosis in human breast cancer MCF-7 cells and investigate the role of Foxp3 in the effect.Methods The inhibit rate of MCF-7 cells was measured by MTT assay after 48 h metformin treatment(5 mmol/L、10 mmol/L、20 mmol/L).Metformin-induced cell apoptosis was measured by Hoechst 33258 stai-ning and flow cytometry.The expression levels of Foxp3 and caspase-3 mRNA were detected by real-time PCR.Expression of Foxp3 protein was detected by Western blot.Results Metformin markedly inhibited proliferation of MCF-7(P<0.05)compared with control group.And the early apoptotic rates increased to 3.76%,8.96%,and 18.67% with 5 mmol/L、10 mmol/L、20 mmol/L metformin treatment,respectively.Metformin increased caspase-3 mRNA expression levels,and decreased the expression levels of Foxp3 mRNA and protein expression levels(P<0.05,vs.control).Conclusion Metformin could inhibit the proliferation and in-duce apoptosis of MCF-7,and its mechanism maybe related to down-regulated expression of Foxp3.
4.Study on formation of biofilm in bronchi of old patients during application of mechanic air

Objective To investigate the status of biological membrane(biofilm) formation in the bronchi of old patients treated with mechanical air,and clarify its correlations and risk factors.Methods The retaining time in 25 bronchi from 22 cases of old patients and the results of bacterial culture were analyzed.Results Bacteria culture-positive samples are 22 cases isolated from 25 endotrached tube,accounting for 88%.Negative samples are 3 cases,accounting for 12%.Conclusion The density of bacteria is corresponding to the time of tubing in the bronchi.With the extension of tubing time,the quantity of bacteria is greatly increased.

Raman spectroscopy has generated many branches during the development for more than 90 years. Surface enhanced Raman spectroscopy (SERS) improves SNR by using the interaction between tested materials and the surface of rough metal, as to quickly get higher sensitivity and precision spectroscopy without sample pretreatment. This article describes the characteristic and classification of SERS, and updates the theory and clinical application of SERS. It also summarizes the present status and progress of SERS in various disciplines and illustrates the necessity and urgency of its research, which provides rationale for the application for SERS in microbiology.

7.The effect of warm ischemia duration on renal function early after laparoscopic partial nephrectomy

Objective To assess the effect of variable durations of warm ischemia on renal function early after laparoscopic partial nephrectomy ( LPN ) and make the definite safety duration of renal warm ischemia.Methods The clinical data of 76 patients treated with LPN from October 2012 to June 2014 were retrospectively analyzed.The patients were divided into 3 groups based on warm ischemic time,namely group A (28 cases) with warm ischemia time less than 20 min,group B (34 cases) with warm ischemia time more than 20 min and less than 30 min, group C ( 14 cases ) with warm ischemia time more than 30 min.LPN was performed with renal artery clamping alone in all the patients.Preoperative and postoperative renal scintigraphic scan was performed to access glomerular filtration rate ( GFR) in all patients.The GFR values were compared among before, 1 week, 1 month and 3 months after operation.The factors predicting the early renal injury were assessed by multivariate regression analysis.Results The renal GFR of the kidney underwent LPN decreased 19.43(17.70,22.06) ml/min at 1 week,17.04(13.94,20.70) ml/min at 1 month,13.82(10.72,18.73) ml/min at 3 months after the surgery in group C,respectively.In group A, the renal GFR of the operated-side decreased 12.07(10.91,13.42) ml/min,10.04(9.16,11.75) ml/min, 8.44(7.07,9.72) ml/min,respectively.In group B, the renal GFR of the operated-side decreased 13.64 (12.48,16.72) ml/min,10.29(9.17,14.27)ml/min,9.63(7.85,12.59) ml/min,respectively.The GFR decreased greater in group C than that in group A and B(P<0.05).The total renal GFR decreased (10.70 ± 4.93)ml/min at three months in group C,compared with (5.64 ±4.12)ml/min in group A and (6.37 ± 4.32)ml/min in group B,respectively.The decreased value in group C was greater than that in group A and B(P<0.05).However,the differences of the total renal GFR among the 3 groups were not significant at 1 week and 1 month(P>0.05).The multivariate regression analysis revealed that warm ischemia duration was the independent risk factor of the early renal injury.Conclusions Warm ischemia duration is the major factor regarding the early renal recovery after LPN.Warm ischemia time more than 30 min may not only greatly affect the renal function but also the renal function recovery rate.
8.Provision of medical-care service and referral for children suffering from hand-foot-mouth diseases in rural areas

A survey with self-administered questionnaire conducted in 357 families with children suffering from with hand-foot-mouth disease showed that 74. 5 percent of patients' home was at a distance of less than one kilometer to medical-care institutions, and 41.7 % of patients went to village clinics or community health-care service stations as their first visit. Diagnosis could be established for 31. 5% of them in their first visit, and 78. 4% of them underwent referral taking public vehicles.
9.Relationships between seven single nucleotide polymorphisms and female breast cancer among Hui and Han nationalities

Objective To investigate the relationships between seven single nucleotide pdymorphisms(SNPs), including 6788895(SIAH2 gene), rs10941679(5p12), rs889312(MAP3K1), rs13387042(2q35), rs6504950(17q23), rs17530068(6q14) and rs2284378(20q11), and female breast cancer of Hui and Han nationalities.Methods A hospital-based case-control using age and nationallity as frequency matching conditions was conducted in Affiliated General Hospital of Ningxia Medical University.Results ①A total of 225 breast cancer patients and 225 controls were enrolled in this study.There was no significant differentce between case group and control group in age(48.27±9.27 vs 48.48±9.36, t=0.238,P=0.812) or nationallity (181 Han people and 44 Hui people vs 184 Han people and 41 Hui people, x2=0.131, P=0.718).②The subjects of study conformed to Hardy-Weinberg equilibrium principles.③The genotype and allele distribution of the above seven SNPs were not significantly different between cases group and control group of Hui and Han nationalities.Conclusion The seven SNPs mentioned above may not be related to breast cancer among people Hui and Han nationalities.
10.Expression and significance of telomerase in development process of scar formation

ObjectiveTo study the expression of telomerase of fibroblasts in the formation and development of scar,and to investgate the relationship between the telomerase activity and the formation and development of scar.MethodsExpression of telomerase was detected by immunohistochemical techniques in 18 specimens from granulation tissue,17 keloid,16 specimens from hypertrophic scar,28 specimens from mature scar,32 specimens from normal skin,and SPSS16.0 statistics software was used to analyze the relationship between telomerase and scar.Results It was shown that the positive expression rate of telomerase in the granulation tissue group was 94.4 %,that in the keloid group was 58.8 %,that in the hypertrophic scar group was 18.8 %, and that in the mature scar and in the normal skin was zero. comparisons between groups,in addition to the normal scar group and normal skin group,other groups were statistically significant differences.ConclnsionsScar formation is a process with multi-factor participation,while telomerase activation is an important factor. Detecting telomerase activity in the development process of scar formation may determine the prognosis.Through the control of telomerase activity in the pathogenesis of scar may become a new approach of scar treatment.
