1.Study on diagnosis of prostate cancer by PAP combined with PSA and the effect of estrogen

Objective To study the serum levels of PSA, PAP joint detection in diagnosis of prostate cancer and the effect of estrogen on it.Methods60 patients with prostate cancer were selected in Jinyun County People's Hospital department of urology patients, giving diethylstilbestrol 3mg, oral once a day, and give Aspirin Enteric-coated Sustained Release Tablets 100mg oral, once daily, for 2 consecutive months.At the same time, we selected 60 cases of patients with benign prostatic hyperplasia, normal control group of 60 healthy subjects were the same period.Using radioimmunoassay, were observed inprostatic hyperplasia group, normal control group, the prostate cancer group before treatment, the prostate cancer group after treatment, serum PSA, PAP levels, as well as the group before and after treatment in patients with prostate cancer IPSS score index.ResultsThe prostate cancer group, serum PSA, PAP levels were significantly higher than that of benign prostatic hyperplasia group and normal control group, the difference was statistically significant (P<0.05);no significant difference was found between the prostatic hyperplasia group and normal control group, PSA in prostate cancer positive detection rate and the detection sensitivity is 78.33% the specificity was 70%, the index of PAP in prostate cancer, and the positive detection rate and the detection sensitivity was 53.33%, specificity index reached 93.33%;PSA for prostate cancer detection positive rate and sensitivity was higher than that of PAP, but the specificity is less than PAP, the difference is statistically significant (P<0.05) after treatment;the prostate cancer group, serum PSA, PAP levels were improved, compared with that before treatment, serum PSA, PAP levels decreased significantly, the difference was statistically significant (P<0.05);and after the treatment, the prostate cancer group of prostate symptoms have improved, compared with before treatment, IPSS score decreased significantly, the difference was statistically significant (P<0.05).ConclusionThe combined detection of serum PSA, PAP in the diagnosis of prostate cancer can improve the sensitivity and specificity, and can be used for early diagnosis of prostate cancer;estrogen can lower their level, improve the symptoms of prostate cancer, inhibit the progress of the disease.
2.Preparation of chitosan scaffold tube

BACKGROUND: Chitosan is commonly prepared using emulsion freeze-drying methods, which is complexly operated. OBJECTIVE: To introduce a simple methods for the preparation of chitosan and analyze its biological characteristic. DESIGN, TIME AND SETTING: Controlled observation experiments were carried out in the Central Laboratory, the Second Affiliated Hospital of Chinese Medical University (Shenyang, Liaoning, China) from April to December in 2007. MATERIALS: Ten Wistar rats of 8 weeks old and SPF grade were adopted. Chitosan powder in separated packs, with 99% purity and 99% deacetylation degree were the product of Shandong Jinan Ocean Bio-Product Co., Ltd. (China). METHODS: Acetic acid solution was added into chitosan to prepare 8% chitosan acetic acid hydrosol, which was then stayed for one day. Flowing the immersion into the hydrosol and immediate dislodgement, plastic tube was soaked in 1 mol/L sodium hydroxide for 3 minutes. Hydrosol out of the tube etiolated and coagulated into jells, the tube was removed when the jells were semi-dried. Subsequent to complete drying, chitosan tubes were prepared as an internal diameter of 5 mm, and were cut into many segmentations (length 2 mm). The prepared tubes were subcutaneously implanted into rat muscular tissues at right McBurney point, while rat left muscular tissues were only incised and sutured, taking as controls. MAIN OUTCOME MEASURES: Gross observation of chitosan tube, degradation percentage of chitosan tubes in rat muscular tissue, and tissue inflammation change around chitosan tube detected by hematoxylin-eosin stain. RESULTS: The surface of chitosan tube was smooth and the tenacity was good. Intramuscular implantation test showed that 50% and 90% chitosan tubes were absorbed by tissues in 4 weeks and 8 weeks postoperatively. Inflammatory reaction surrounding tubes was decreased gradually. CONCLUSION: It gives a new kind of way to prepare chitosan tube, with few materials, low cost, simple operation and short time. The prepared chitosan tubes approve the good histocompatibility and biological degradation.
3.Content Measurement of Bulb Total Alkaloid and Saponin in Zhejiang Fritillariae and Thuubergii

[Objective] To mensurate and compare with the mensuration of alkaloid and saponin in the flower,pedicel and bulb of fritillariae thuubergii,make full use of all parts of fritillariae thuubergi.i[Method]To mensurate the mensuration of alkaloid and saponin in the three parts of fritillariae with the twophase titration method and gravimetric method.[Result]The mensuration of alkaloid in the flowers of fritillariae thuubergii is 0.182%,while it is 0.047% and 0.341% in the pedicel and bulb separately,and the mensuration of saponin in the flowers of fritillariae thuubergii is 5.112%,while it is 8.840% and 1.724% in the pedicel and bulb separately.[Conclution] The difference of major component in the fritillariae thuubergii offers the foundation to make full use of the flowers,pedicel and bulb.

Diabetic cognitive dysfunction is one of the important complications of diabetes mellitus .Insulin plays an important protective role in cognitive function through MAPK and PI 3-K/Akt signaling pathway .Insulin resistance may give rise to excessive tau protein phosphorylation , formation of neurofibrillary tangles inhibits insulin degrading enzyme , reduces the degradation of amyloid β;enhances oxidative stress , further damages the integrity of the neu-ronal structure and function and eventually leads to cognitive dysfunction .
5.Expression and significance of c-myc and CD24 in colorectal cancer

Objective To determine the expression of c-myc and CD24 in colorectal carcinoma,colo-rectal polyp and normal mucosa,and to explore the role and correlation of them in the carcinogenesis of colorec-tal carcinoma. Methods The expression of c-myc and CD24 in colorectal carcinoma(n = 60),colorectal ade-nomatous polyp(n = 45),colorectal hyperplastic polyp(n = 15)and the adjacent non-cancerous tissue(n =30)was observed by immunohistochemical assay. Conclusion The expression of c-myc and CD24 are high in colorectal cancer,having a significant correlation with some of the clinicaopathological features. c-myc is likely to act as a downstream target gene of CD24 signaling pathway,whose expression is probably regulated by CD24 in colorectal carcinoma tissue.
6.Comparative Evaluation of Gene-set Analysis Methods

Objective To compare the efficiency of χ~2-Fisher's exact test which is one of the competitive null hypothesis approaches with SAM-GS which belongs toself-contained null hypothesis approaches. Methods The two methods were used to analyze a simulation experiment which contained five different scenarios. The results were compared with the simulated initialization,and assessing indexes were calculated to compare the efficiency. Results Under the same conditions,SAM-GS always have higher power than that of χ~2-fisher's exact test. However, the final inference is equivalent, namely if the difference between the two groups are smaller than 0.30,the two methods can not be better to identify differences between them. By contrary, when the differences between the two phenotypes are larger than 0.30, the two ways can both identify differences. Conclusion SAM-GS tends to have slightly higher power thanχ~2-Fisher' s exact test. The two methods can be used for screening enrichment gene sets of gene expression profile.χ~2-Fisher's exact test has the important advantage of being able to analyze multi-class phenotype.
7.Preoperative symposium about a program for treatment of scoliosis

[Objective]To determine a surgical plan for the treatment of scoliosis. [Methods]A consecutive series of scoliosis patients were admitted from Oct. 1,2007 to Feb. 29,2008. The preliminary operation programs (including diagnosis,operative approach,upper instrumented vertebra,lowest instrumented vertebra,whether to need selective fusion of main thoracic curve and multiple operations) were formulated by the attending physicians.Preliminary operation programs were decided on by a symposium. A comparison was made between the two kinds of programs.[Results]Data from 143 consecutive scoliosis patients were reviewed. The average age at the time of operation was 16.1 years (2-49 years) and the average curve was 56.9? (range,15?~115? ). There were 6 spine surgeons involved in the study. The accepted programs formulated by the symposium occurred in 46 (32.2%) cases. One of these cases was changed in surgical approach and need selective fusion of main thoracic curve,19 in upper instrumented vertebra and 29 in lower instrumented vertebra. There were no changes in diagnosis and need multistage operations. Operation programs were difficult to formulated in patients aged less than 20,with Cobb's angle≤70?and thoracic curve patterns. [Conclusion]Preoperative symposium for treatment of idiopathic scoliosis influenced the preliminary operation programs in 32.2%. Sympoium appears to have a significant role in drawing up a more perfect operation program.

9.Advance in the influence of melatonin to pregnancy related rhythm

Melatonin holds a wide range of biological effects, its multi-channel and multi-target impacts on reproductive sys-tem are the basis of pregnancy related rhythm, including conception, the duration of pregnancy and delivery.This paper describes the synthesis of melatonin, the characteristics of melatonin receptors distribution and melatonin′s biological property.It also illuminates the influence of melatonin rhythm on the above-mentioned courses, hoping to provide theory basis for clinical suggestions to modulate rhythm around pregnancy.
10.Stem cell-induced proliferation of myocardial cells and exercise-induced regeneration of myocardial cells

BACKGROUND:Stem cel transplantation has incomparable superiorities in the treatment of cardiovascular diseases. But stem cel s have a very low efficiency to differentiate into myocardial cel s spontaneously, and there are many factors influencing stem cel differentiation. OBJECTIVE:To clarify the pros and cons of different sources of stem cel s in the treatment of myocardial infarction, to investigate the methods for improving the differentiation efficiency of myocardial cel s, optimal differentiation conditions and mechanism underlying exercise-induced stem cel mobilization and endogenous myocardial cel regeneration. METHODS:A computer-based search of CNKI and PubMed databases was performed by the first author for articles related to stem cel therapy for myocardial infarction and stem cel differentiation into myocardial cel s, exercise effect on stem cel proliferation and myocardial cel regeneration published from 1985 to 2015. The key words were“stem cel s, myocardial infarction, myocardial regeneration, cardiac cel , exercise”in Chinese and English, respectively. Final y, 54 articles were included in result analysis. RESULTS AND CONCLUSION:In recent years, a variety of chemical inducers and biological components have been used in the myocardial differentiation of stem cel s. Simulation of myocardial microenvironment and vascular cel growth factors are the main methods of inducing myocardial differentiation. Aerobic exercise-induced stem cel mobilization can induce ischemic cardiac angiogenesis and upregulate a variety of vascular endothelial growth factors so as to promote myocardial proliferation and repair. However, in-depth exploration is stil needed in the harvesting of stem cel s, transplant rejection, and regulatory mechanisms underlying the directed differentiation of stem cel s into cardiomyocytes.
