BACKGROUND: Chitosan is commonly prepared using emulsion freeze-drying methods, which is complexly operated. OBJECTIVE: To introduce a simple methods for the preparation of chitosan and analyze its biological characteristic. DESIGN, TIME AND SETTING: Controlled observation experiments were carried out in the Central Laboratory, the Second Affiliated Hospital of Chinese Medical University (Shenyang, Liaoning, China) from April to December in 2007. MATERIALS: Ten Wistar rats of 8 weeks old and SPF grade were adopted. Chitosan powder in separated packs, with 99% purity and 99% deacetylation degree were the product of Shandong Jinan Ocean Bio-Product Co., Ltd. (China). METHODS: Acetic acid solution was added into chitosan to prepare 8% chitosan acetic acid hydrosol, which was then stayed for one day. Flowing the immersion into the hydrosol and immediate dislodgement, plastic tube was soaked in 1 mol/L sodium hydroxide for 3 minutes. Hydrosol out of the tube etiolated and coagulated into jells, the tube was removed when the jells were semi-dried. Subsequent to complete drying, chitosan tubes were prepared as an internal diameter of 5 mm, and were cut into many segmentations (length 2 mm). The prepared tubes were subcutaneously implanted into rat muscular tissues at right McBurney point, while rat left muscular tissues were only incised and sutured, taking as controls. MAIN OUTCOME MEASURES: Gross observation of chitosan tube, degradation percentage of chitosan tubes in rat muscular tissue, and tissue inflammation change around chitosan tube detected by hematoxylin-eosin stain. RESULTS: The surface of chitosan tube was smooth and the tenacity was good. Intramuscular implantation test showed that 50% and 90% chitosan tubes were absorbed by tissues in 4 weeks and 8 weeks postoperatively. Inflammatory reaction surrounding tubes was decreased gradually. CONCLUSION: It gives a new kind of way to prepare chitosan tube, with few materials, low cost, simple operation and short time. The prepared chitosan tubes approve the good histocompatibility and biological degradation.

[Objective] To mensurate and compare with the mensuration of alkaloid and saponin in the flower,pedicel and bulb of fritillariae thuubergii,make full use of all parts of fritillariae thuubergi.i[Method]To mensurate the mensuration of alkaloid and saponin in the three parts of fritillariae with the twophase titration method and gravimetric method.[Result]The mensuration of alkaloid in the flowers of fritillariae thuubergii is 0.182%,while it is 0.047% and 0.341% in the pedicel and bulb separately,and the mensuration of saponin in the flowers of fritillariae thuubergii is 5.112%,while it is 8.840% and 1.724% in the pedicel and bulb separately.[Conclution] The difference of major component in the fritillariae thuubergii offers the foundation to make full use of the flowers,pedicel and bulb.

Objective To study the serum levels of PSA, PAP joint detection in diagnosis of prostate cancer and the effect of estrogen on it.Methods60 patients with prostate cancer were selected in Jinyun County People's Hospital department of urology patients, giving diethylstilbestrol 3mg, oral once a day, and give Aspirin Enteric-coated Sustained Release Tablets 100mg oral, once daily, for 2 consecutive months.At the same time, we selected 60 cases of patients with benign prostatic hyperplasia, normal control group of 60 healthy subjects were the same period.Using radioimmunoassay, were observed inprostatic hyperplasia group, normal control group, the prostate cancer group before treatment, the prostate cancer group after treatment, serum PSA, PAP levels, as well as the group before and after treatment in patients with prostate cancer IPSS score index.ResultsThe prostate cancer group, serum PSA, PAP levels were significantly higher than that of benign prostatic hyperplasia group and normal control group, the difference was statistically significant (P<0.05);no significant difference was found between the prostatic hyperplasia group and normal control group, PSA in prostate cancer positive detection rate and the detection sensitivity is 78.33% the specificity was 70%, the index of PAP in prostate cancer, and the positive detection rate and the detection sensitivity was 53.33%, specificity index reached 93.33%;PSA for prostate cancer detection positive rate and sensitivity was higher than that of PAP, but the specificity is less than PAP, the difference is statistically significant (P<0.05) after treatment;the prostate cancer group, serum PSA, PAP levels were improved, compared with that before treatment, serum PSA, PAP levels decreased significantly, the difference was statistically significant (P<0.05);and after the treatment, the prostate cancer group of prostate symptoms have improved, compared with before treatment, IPSS score decreased significantly, the difference was statistically significant (P<0.05).ConclusionThe combined detection of serum PSA, PAP in the diagnosis of prostate cancer can improve the sensitivity and specificity, and can be used for early diagnosis of prostate cancer;estrogen can lower their level, improve the symptoms of prostate cancer, inhibit the progress of the disease.

Objective To compare the efficiency of χ~2-Fisher's exact test which is one of the competitive null hypothesis approaches with SAM-GS which belongs toself-contained null hypothesis approaches. Methods The two methods were used to analyze a simulation experiment which contained five different scenarios. The results were compared with the simulated initialization,and assessing indexes were calculated to compare the efficiency. Results Under the same conditions,SAM-GS always have higher power than that of χ~2-fisher's exact test. However, the final inference is equivalent, namely if the difference between the two groups are smaller than 0.30,the two methods can not be better to identify differences between them. By contrary, when the differences between the two phenotypes are larger than 0.30, the two ways can both identify differences. Conclusion SAM-GS tends to have slightly higher power thanχ~2-Fisher' s exact test. The two methods can be used for screening enrichment gene sets of gene expression profile.χ~2-Fisher's exact test has the important advantage of being able to analyze multi-class phenotype.

Objective To explore the protection of the facial-acoustic nerve during microsurgery for acoustic neuroma via the suboccipital retrosigmoid approach. Methods The clinical data of 25 cases with acoustic neurinoma treated by the suboccipital retrosigmoid approach were retrospectively analyzed. Results Total removal was achieved in 22 patients and subtotal in 3 patients. 20 with Cochlear nerve anatomic preservation at 2 weeks after surgery and 3 patients with effective hearing and 13 patients lost effective hearing but retained measurable hearing and 9 patients completely lost hearing( the New Hannover Classification V grade). The facial nerve was preserved anatomically in 18,H-B Grade I ～ IV in 15,Grade V ～ VI in 10. There were not cerebrospinal fluid leakage, intracranial infection and death in all cases. Conclusion When operating,the relation between the tumor and the important structure surrounding could should be understood fully. Then removed the tumor piece by piece with microsurgical techniques. That the effective way to keep the facial nerve and its function being protected,and could improve the prognosis of the operation.

Objective To study the reasonable treatment to the patients with raptured intracranial aneurysms of hunt-hess Ⅲ-IV grade in the cerebral vasospasm stage. Methods The clinical data of 80 patients with ruptured intracranial aneurysms of hunt-hess Ⅲ-IV grade were analyzed retrospectively. All of the patients were clipped closed by microsurgery (36) or embolized intervention(44) in 3～18 days after disease. According to the time of surgical intervention after disease,they were divided into 42 patients with early(3～10 days) surgical group and 38 patients with advanced(11～18 days) surgical group. The different operative methods and different treatment periods were compared in order to evaluate the effect to the prognosis, the DIND incidence and the ruptured again aneurysms during operation incidence. Results There was no statistical significance in the effect about early or advanced surgical intervention to the prognosis, the DIND incidence and the ruptured again aneurysms during operation incidence (P > 0.05). There was no statistical significance in the effect about advanced interventional embolization or clipping closed by microsurgery to the prognosis,the DIND incidence and the ruptured again aneurysms during operation incidence(P > 0. 05). There was statistical significance in the effect about early interventional embolization or clipping closed by microsurgery to the prognosis,the DIND incidence and the ruptured again aneurysms during operation incidence(P< 0.05). The interventional embolization had better prognosis,more lower DIND incidence and more lower ruptured a-gain aneurysms during operation incidence than the clipping closed by microsurgery. Conclusion The patients with ruptured intracranial aneurysms of hunt-hess Ⅲ-IV grade in the cerebral vasospasm stage should be earlier intervened surgically,then it seems that interventional embolization is more reasonable which is worth referencing in clinic. The good operation habits during operation and the effectively strengthening management after operation had positive significance to ensure operative effect and to improve prognosis.

Objective To study the possible reasons and the processing strategy of brain injury operation complicated with intracranial hemorrhage.Methods The clinical data about 22 patients who were digged skull operation because of brain injury and complicated with intracranial hemorrhage in operation in neurosurgery from 2007 June to 2008 February were retrospectively analyzed,and different types intracranial hemorrhage were classified.Results In 22 patients,12 patients were complicated with epidural hematoma,in which the patients (9/12,75.0%) complicated with opposite hematoma were common;5 patients were complicated with intracerebral hematoma,in which the patients (3/5,60%) complicated with ipsilateral hematoma were common;4 patients were complicated with acute subdural hematoma,and all the patients complicated with opposite hematoma;1 patient was complicated with sub-flap hematoma.Conclusions In the patients who are complicated with intracranial hemorrhage because of digging skull operation to brain injury,epidural hematoma are most,then intracerebral hematoma,subdural hematoma,sub-flap hematoma.The important measures of preventing secondary intracranial hemorrhage are definiting force mechanism and distinguishing fracture line before operation,gradiently decompressing,completely removing crushing brain tissue,protecting reflux veins,rigorously stopping bleeding during operation.

Objective To explore the expression of interferon(IFN)-γ,interleukin(IL)-4 and IL-17 in the patients with hepatitis B virus(HBV)infection,which can reflect the function of helDer T lymphocyte(Th)1,Th2 and Th17 cells.Methods There were 34 chronic hepatitis B(CHB)Datients,31 liver cirrhosis(LC)patients,26 primary hepatocellular carcinoma(PHC)patients,22 chronic severe hepatitis patients and 15 healthy controls involved in our research.The levels of IFN-γ,IL-4 and IL-17 in serum of the patients with chronic HBV infection and healthy controls were determined by enzyme linked immunosorbent assays(ELISA). And the dala were analysed via t-test and Pearson correlation analysis.Results IFN-γ/IL-4 ratio of the CHB group(1.08±0.66,P＜0.01)was lower than that of healthy controls(2.60±0.60),and the ratio in the chronic severe hepatitis B group(4.81±0.87,P＜0.01)was higher than that in healthy control group.IFN-γIL-17 ratio in the CHB(1.13±0.85,P＜0.01),the LC(1.69±0.92,P=0.010)and the PHC group(1.76±0.84,P=0.011)were all lower than that in healthy conlrols group(2.66±0.70),and the ratio in the chronic severe hepatitis B group(3.68±0.42)was higher than that in healthy controls group (P=0.004).IL-4/IL-17 ratio in the LC(0.72±0.38,P=0.026)and the PHC group(0.63±0.19,P＜0.01)were both lower than that in heahhy controls(1.04±0.23).Conclusiolls The dominant expression of IL-4,IL-7 and IFN-γ is respectively found in the CHB patients,LC and PHC patients,and chronic severe hepatilis B patients.

AIM To investigate the curative effects of Kuntai Capsules (Coptidis Rhizoma,Scutellariae Radix,Asini Corii Colla,etc.) on rat model for primary ovarian insufficiency (POI) caused by 4-vinylcyclohexene diepoxide (VCD).METHODS Seventy-five weaned 21-day old rats were divided equally into five groups randomly:control group,model group,low-,medium-,and high-dosage Kuntain group.Except the control group was given sesame oil rats,other four groups were given intraperitoneal injection of VCD solution.According to vaginal smear of rats,eight rats in each group were selected,the levels of FSH (follicle-stimulating hormone),LH (leuteinizing hormone),E2 (estradiol) and AMH (anti-Mullerian hormone) were measured,and morphological changes of ovary and uterus were observed.RESULTS Compared to the control group,FSH level in rats' serum of the model group significantly increased while AMH and E2 levels significantly decreased,and LH level rose;Kuntai Capsules significantly decreased FSH level while increased AMH level;The wet weight of rats' ovary in the model group was lighter than that in the control group.The ovary weight of Kuntai Capsules low-dosage group was increased;the number of follicles at all levels and corpora lutea of rats in the model group were decreased.After the treatment with Kuntai Capsules,the number of follicles at all levels and corpora lutea were increased;light microscopy manifested decreased uterine glands of the tube wall,reduced glandular cavity.After the treatment with Kuntai Capsules,the number of uterine glands increased and the gland cavity expanded.CONCLUSION Kuntai Capsules may slowdown the functional decline rate of ovarian.

Objective To investigate the effectiveness of arthroscopic reduction and percutaneous fixation (ARPF) for the treatment of tibial plateau fractures. Methods Seventeen cases of tibial plateau fractures from April 1999 to June 2002 were reviewed. The fractured tibial plateau was reconstructed by “drag-and-draw” reduction and iliac crest bone graft under arthroscope. And percutaneous cancellous bone screw fixation was conducted. Results Clinical bone union was achieved in all the patients at the third postoperative month. No infection or severe osteoarthritis occurred. Conclusions ARPF for tibial plateau fractures has advantages of reduction under direct view, reliable fixation and facilitation for early activities.
