OBJECTIVE: To introduce the basic functions, working conditions and requirements of bioreactot, additional, to discuss the research process of bioreactor.METHODS: PubMed database (http://www.ncbi.nlm.nih.) and CNKI database (www.cnki.nat/index.htm) were retrieved by the first author with search terms of "bioartificial liver, bioreactor, membrane material" both in English and Chinese. The time was limited between January 1994 and August 2009. Inclusion criteria: ①literatures closely linked to the article; ②papers published in more authoritative journals in recent years. ③the contents of old or duplicate documents were excluded.RESULTS: Totally 150 documents were seized by primary screen, 88 irrelative papers and 32 duplicate documents were excluded, finally, 30 literature entered further analysis. Bioartificial liver system has become an effective in vitro supportive treatment for hepatic failure patients. Bioreactor is an important ingredient of bioartificial liver, which provided a platform for the hepatocytes growth metabolism, substances exchange and immune isolation. The main cellulose semipermeable bioreactor comprises fiat membrane bioreactor and hollow fiber bioreactor, and the main types of semipermeable membranes contain mixed cellulose ester membrane, cellulose acetate membrane, cuprophan membrane, PVDF membrane, as well as poly(ethylene terephthalate) membrane.CONCLUSION: As a dynamic system, the optimal of control system in bioreactor is conductive to regulating mass transfer and establishing bionic physical or chemical gradients, which can implement the construction of hepatocyte with smallest unit. The selection of membrane is the key link in bioreactor construction of bioartificial liver system

Objective To explore effect of small dose of diuretics combined with Dahuangzhechong pill on testosterone, Cystatin C(CysC), leptin and β2 microglobulin(β2-MG) in serum in patients with chronic renal failure.Methods 69 cases of kidney disease patients were collected and divided into two groups.Testosterone, Cystatin C, leptin and β2 microglobulin in serum were compared before and after the treatment.The control group was given hydrochlorothiazide tablets, treatment group was given Dahuangzhechong pill based on the control group.All patients were treated for 3 months. Results After treatment, levels of testosterone elevated (P <0.05), CysC, leptin and β2 microglobulin decreased (P <0.05) in the two groups compared with before treatment;Compared with the control group , level of testosterone was higher (P<0.05), levels of CysC ,leptin and β2-MG were lower (P<0.05).Conclusion Low dose diuretics combined with Dahuang Zhechong pill has good effect in treatment of chronic renal failure curative.The mechanism may be related to elevate serum testosterone levels and reduce serum CysC , leptin and β2-MG level in patients.

Aim To investigate the anti-angiogenic effect of 12a, the mastoparan-like peptide from wasp ( Vespa magnifica ) venom. Methods The tube for-mation and proliferation of human umbilical vein endo-thelial cells ( HUVEC ) and chicken chorioallantoic membrane ( CAM) model were used to observe the an-ti-angiogenic effect of 12a in vitro and in vivo, respec-tively. Results In the CAM model, 0. 1 μg or 1 μg of 12 a could markedly inhibit angiogenesis induced by 0. 2 μg rh-bFGF with heavy loss of color, decreasing density and obscure frame of the vessels. The inhibi-tion rates of angiogenesis were 60 . 2 % for 0 . 1 μg and 90. 3 % for 1 μg, respectively. Accordingly, in HU-VEC culture experiment, the proliferation and angio-genesis of HUVEC treated by 1 mg·L-1 and 10 mg· L-1 of 12 a were decreased by 55. 4 %, 39. 3 % and 51. 6 %, 26. 7 %, respectively. Conclusion 12a has a significant anti-angiogenic effect in a concentra-tion-dependent manner.

To study the toxicity of monoamine in high concentrations to neurons. Neurons were treated in vitro with different doses of dopamine, norepinephrine and serotonin for various lengths of time. The toxicity of these reagents on neurons was observed. Apoptosis of neurons was analyzed by agar electrophoresis, in situ DNA end labeling technique and flow cytometry. The results showed that a high concentration of monoamine could induce apoptosis of the culturing neurons. At 24h after treatment with dopamine or norepinephrine (80, 160 and 320?mol/L) as well as with 320?mol/L serotonin, apoptotic rates of neurons were significantly higher than that of the controls, respectively ( P

Objective To establish the PDGF-BB-induced proliferation of rat pulmonary artery smooth muscle cells(PASMCs), and investigate the effects of the AMPK agonist AICAR on the cell cycle of PASMCs, in order to search new drugs for prevention of pulmo-nary vascular remodeling. Methods 20ng/ml PDGF-BB was used to induced the proliferation of PASMCs, and the effect of 0. 5mmol/L AICAR on the proliferation of PASMCs was observed. Western blot was used to detect the total and phosphorylated AMPK. The prolifer-ation of PASMCs was determined by CCK-8. The mRNA expression of cyclinD1, cyclinE and CDK2/4/6 were detected by flow cytometry analysis cell cycle,quantitative real-time PCR. Results Western blot Results indicated AICAR could promote the activation of AMPK. CCK-8 test Results showed that AICAR blocked the proliferation of PASMCs induced by PDGF-BB. Flow cytometry analysis indicated that AICAR arrested the cell cycle in G0/G1 to S phase. RT-PCR Results demonstrated that AICAR inhibited the mRNA expression of cy-clinD1, cyclinE and CDK2/4/6. Conclusion The AMPK agonist AICAR can block the proliferation of PASMCs induced through arrest cell cycle in G0/G1-S phase by regulation the mRNA expression of cyclin D1, cyclinE, CDK2/4/6, and AICAR has a potential applica-tion in preventing pulmonary vascular remodeling.

Aim To investigate the gene sequences and pro-angiogenic activities of thymosin-β4 from skin of Nanorana yunnanensis. Methods Two cDNA se-quences of thymosin-β4 from Nanorana yunnanensis designated as pTβ4-1 and pTβ4-2 were obtained by PCR. The tube formation and proliferation of human umbilical vein endothelial cells ( HUVEC ) and chick-en chorioallantoic membrane ( CAM) model were used to observe the pro-angiogenic effects of synthetic pep-tides according to the reduced amino acid sequences of pTβ4-1 and pTβ4-2 in vitro and in vivo, respectively. Results The cDNA sequences of Tβ4-1 and pTβ4-2 were composed of 662 bp and 673 bp, respectively, and their deduced amino acid sequences contained 44 residues which shared highly similarity with those from human , mouse , chicken , claw frog , zebra fish , turbot and Amolops loloensis. Furthermore, the mature amino acid sequences of pTβ4-1 and pTβ4-2 had highly con-served structural regions when compared with other thy-mosin-β4 previously reported. Synthetic peptides could significantly promote proliferation and tube formation of HUVEC and angiogenesis of CAM. Conclusion There are two thymosin-β4 peptides in skins of Nanorana yunnanensis which have pro-angiogenic ac-tivities.

Clinical teaching includes performing ward inspection and clinical practice, moreover, the teaching should be based on how to solve clinical problems and focused on pediatrics characteristics. The practice and experience shows that clinical study in department of pediatrics wiu improve the students' comprehensive ability and help them lay a solid foundation for further work.

Objective To investigate the expression of RGC32 gene in pulmonary adenocarcinoma tissue and to explore the influence on proliferation and apoptosis of A549 cells.Methods Real-time PCR was applied to detect the expression of RGC32 gene in 36 cases of pulmonary adenocarcinoma and pericancerous tissues.RNA interference was used to inhibit the expression of RGC32 gene.After RNA interference,the expression of RGC32 gene was detected by real-time PCR,the apoptosis of the transfected cells was detected by flow cytometry and the inhibition ratio of cell proliferation was detected by methyl thiazolyl tetrazolium (MTT).Results The expression of RGC32 gene was upgraded in pulmonary adenocarcinomas tissues(1:2.2736,t=-29.185,P=0.01).After RNA interference,the expression of RGC32 gene transfected A549 cells was down-regulated significantly[(2.47±0.17)％ vs(4.65±0.26)％,t=-202.868,P=0.000].Comparing to the control cells,the apoptosis of experimental group cells increased significantly (2.9 ％ vs 45.4 ％,t=-37.915,P=0.01),and the inhibition ratio of cell proliferation increased significantly.Conclusion The expression of RGC32 gene shows an obvious upgraded in pulmonary adenocarcinoma.The low expression of RGC32 gene can induce apoptosis and inhibit proliferation of A549 cells.

Background Retinal vascular recanalization is key to the treatment of retinal vascular occlusive disease.Studies confirmed that urokinase by intravitreal injection inhibits the expression of occludin protein at tight junction complexes among retinal capillary endothelial cells.Objective This study was to observe the effects of urokinase via eye local injection on the outward permeability of blood-retinal barrier by detecting the concentration of intravitreal Evans blue (EB).Methods Sixty healthy Sprague-Dawley (SD) rats were randomly assigned to four groups,and the right eyes of the rats were used as experimental eyes.Urokinase of 4 μl (350 U) and the equal volume of PBS (0.01 mol/L) was intravitreally injected separately in the intravitreal urokinase group and the intravitreal PBS group,and 10 μl urokinase (1000U) and the equal volume of PBS was injected via retrobulbar tissue respectively as the retrobulbar urokinase group and the retrobulbar PBS group.Twenty-four hours after injection of drugs,0.5％ EB 4 μl was intravitreally injected.Four hours later,the rats were sacrificed and the right eyeballs were excised for the extraction and drying.EB was extracted from dried vitreous by formamide.Then,the concentration of EB in formamide was determined by a formamide extraction-ultraviolet spectrophotometry method to calculate the concentration of EB in vitreous.The use and care of experimental animals followed the Regulations for the Administration of Affair Concerning Experimental Animals by State Science and Technology Commission (2011 version).Results The rat vitreous body showed the light blue color in intravitreal urokinase group and the retinal vessels were visible under the microscope,and that in the retrobulbar urokinase group presented blue color.However,in the intravitreal and retrobulbar PBS group,rat vitreous exhibited the deeper blue color and retinas were invisible.Absorbance of EB in formamide was 0.181 ±0.008,0.450±0.017,0.330±0.009 and 0.436±0.012 in the intravitreal urokinase group,intravitreal PBS group,retrobulbar urokinase group and retrobulbar PBS group,respectively.The intravitreal EB concentrations in the intravitreal urokinase group were (0.266±0.014)g/L,which was lower than (0.667±0.026) g/L,(0.496±0.015) g/L and (0.657±0.017) g/L of the intravitreal PBS group,retrobulbar urokinase group and retrobulbar PBS group,showing significant different among the four groups (F =100.406,P＜0.01),and the intravitreal urokinase group showed the lowest value in comparison with other three groups (all at P＜0.01).Conclusions Local application of urokinase around eye can augment the outward permeability of blood-retinal barrier in rats.Intravitreal assay of EB after intravitreal injection is a feasible approach to the determination of outward permeability of blood-retinal barrier.

Objective To evaluate the value of early preventive tracheal intubation in the early first‐aid of serious severe trau‐ma and improve the early first‐aid ability of serious severe trauma .Methods Using retrospective study ,among 258 patients of acute airway obstruction caused by severe trauma ,those who have reached the standard of obstruction and respiratory failure according to observation were as the contrast group (n=84) ,and those who had the tendency of airway obstruction instant treatment of preven‐tive trachea intubation were as early preventive tracheal intubation group (n=174) .The average in hospital days ,the occurrence rate of multiple organ failure and death rate were contrasted .Results In the early preventive tracheal intubation group ,in hospital days were 17 .2 on average ,the occurrence rate of multiple organ failure was 44 .59% and the death rate was 10 .67% .In the con‐trast group ,it was 27 .9 d ,67 .51% and 33 .75% ,the two groups had shown significant difference(P<0 .05) .Conclusion Early tra‐chea intubation have a good trachea management of serious severe traumas ,improve inspiration ,shorten the hospitalization days ,and reduce the occurrence rate of complication .It is important to improving rescuing rate .
