1.Cloning Expression and Functional Verification of recF and recO Gene from Deinococcus radiodurans R1 in Escherichia coli

Objective To construct expressing recombinant of recF and recO from Deinococcus radiodurans R1and express the target protein in E.coli BL21(DE)3.Methods recF and recO gene were amplified by PCR from genomic DNA of Deinococcus radiodurans R1 and inserted into expression plasmid vector pET30b(+) to construct pET30b(+)-recF and pET30b(+)-recO.The recombinant plasmids were transformed into E.coli BL21(DE)3 and the recombinant proteins were expressed by the isopropyl-?-D-thiogalactopyranoside(IPTG) and were analyzed with SDS-PAGE.The changes in the survival rate of bacteria in each group before-and after UV-irradiation were calculated.Results The recombinant plasmid pET30b(+)-recO and pET30b(+)-recF was obtained and the recombinant protein could be highly expressed in E.coli BL21(DE)3.Conclusion This study has provided a foundation for further studies and applications of the recombinant RecF and RecO,and initial detection shows that recO and recF gene can increase the resistant ability of E.coli.
2.Reactive Oxygen Species are Involved in Nitric Oxide-InducedApoptosis of Neurons

With redox-sensitive fluorescene probes DCFH-DA and DHR123, the formation of cytosolic and intramitochondrial reactive oxygen species (ROS) inside immature rat cerebellar granule cells during the apoptosis induced by nitric oxide donor S-nitroso-N-acetyl-pennicillamine (SNAP) was monitored by laser confocal scanning microscopy. The cytosolic and intramitochondrial ROS increase significantly after 0.5 mmol/L SNAP treatment for 1 h. Pre-treatment with the nitric oxide scavenger hemoglobin can effectively inhibit the formation of cytosolic and intrarnitochondrial ROS and protect neurons from apoptosis. Adding glutathione can also protect neurons from apoptosis, and the cytotoxity of nitric oxide increases significantly while the synthesis of glutathione is inhibited. The results indicated that ROS might be involved in NO-induced apoptosis in neural cells and glutathione might be the endogenesis antioxidant to protect neurons from oxidative injury.
3.A preliminary study on relationship between single nucleotide polymorphism ofmiR-196a2 and risk of leukemia

Background and purpose:miR-196a2 functions as an oncogene during tumor initiation and pro-gression. The up-regulation promotes tumor cell proliferation, invasion and metastasis. Therefore, it is promising to be an important tumor biomarker. The aim of this study was to investigate whether rs11614913, a gene polymorphic site ofmiR-196a2, is associated with the risk of leukemia.Methods:A case-control analysis was employed. Bone marrow or periph-eral blood was collected from 210 leukemia patients diagnosed from Jan. 2009 to Jul. 2015 in Yantaishan Hospital (case group) as well as 250 healthy people who were physically examined during the same period (control group). Polymerase chain reaction-restriction fragment length polymorphism (PCR-PFLP) was used to detect the genotype of rs11614913. Application test was used to compare the difference in the frequency of each genotype between case group and control group. The odds ratio (OR) of SNP allelic genes was calculated using logistic regression analysis and 95%CI represented the risk of leukemia for each genotype.Results:The distribution differences in the frequency of T/T, C/C, C/T genotype of miR-196a2 rs11614913 between case group and control group were statistically significant (P<0.05). The risk of leukemia for individuals who carried mutant homozygous C/C was 2.661-fold higher than those carried wild-type homozygous T/T, and the difference was statistically significant (P<0.05).Conclusion:ThemiR-196a2 gene polymorphic site rs11614913 was associated with the risk of leukemia. Mutant homozygous C/C or C allelic gene carrying was probably a risk factor for leukemia.
4.Efficacy Observation of Interleukin-2 Combined with Cisplatin in the Treatment of Malignant Pleural Effusion

Objective: To investigate the effect and safety of interleukin-2 combined with cisplatin in the treatment of malignant pleural effusion.Methods: Totally 86 patients with malignant pleural effusion were randomly divided into the observation group (44 cases) and the control group (42 cases).The control group was treated with cisplatin, and the observation group was treated with interleukin-2 additionally.The treatment course was 3 weeks.The ascites relief, quality of life, survival status and tumor markers before and after the treatment, and the adverse drug reactions were compared between the groups.Results: The total ascites remission rate and the effective rate in the observation group were significantly higher than those in the control group (P<0.05).Compared with those before the treatment, CEA, CA19-9 and CA450 decreased significantly after the treatment in both groups (P<0.05), those in the observation group were significantly lower than those in the control group (P<0.05).After the treatment, the physiological function, physiological occupation, emotional function, body pain and the other life quality scores in the observation group were significantly higher than those in the control group (P<0.05).The 24-month survival rate in the observation group was significantly higher than that in the control group (P<0.05).There were no significant differences in adverse drug reactions between the groups (P>0.05).Conclusion: Interleukin-2 combined with cisplatin can effectively relieve ascites and improve quality of life, which can also prolong the survival time of patients.
5.Symptomatic characteristics and factors related to severity of irritable bowel syndrome

Objective To analyze symptomatic characteristics and the factors related to severity of irritable bowel syndrome (IBS).Methods Total 516 IBS patients met Rome Ⅲ criteria were enrolled in the study from June 2009 to February 2016.A questionnaire survey was conducted in face-to-face manner,including intestinal and extra-intestinal symptoms,sleeping and psychological status.Severity of IBS was determined by symptom scores.Results Among 516 patients,there were 135 cases with mild symptoms (26.2%),267 with moderate symptoms (51.7%) and 114 with severe symptoms (22.1%).Periumbilical pain/discomfort associated with defecation was a common symptom location (34.6%,178/516),however,53.3% (275/516) patients also reported having abdominal pain/discomfort during non-defecation period.In IBS patients,51.2% (264/516) were coexisted with gastroesophageal reflux disease (GERD),59.1% (305/516) with functional dyspepsia (FD),and 46.9% (242/516) with depression.The prevalence of ordinary abdominal pain/discomfort [57.0% (217/381) vs.43.0% (58/135),x2 =7.841],sensation of incomplete defecation[74.5% (284/381)vs.61.5% (83/135),x2 =8.277] and coexisted GERD[54.1% (206/381) vs.43.0%(58/135),x2 =4.920],FD[64.0%(244/381)vs.45.2% (61/135),x2 =14.664],headache[50.7% (193/381)vs.38.5% (52/135),x2 =5.889] and depression[52.2% (199/ 381)vs.31.9% (43/135),x2 =16.623] in patients with moderate to severe symptoms were significantly higher than that in patients with mild symptoms (all P < 0.05).The predictors to moderate-severe IBS symptoms were coexisted FD (OR =1.798,95% CI:1.162-2.783,P =0.010) and depression (OR =1.696,95% CI:1.043-2.759,P =0.014).Conclusion The symptoms of IBS patients are various,and patients coexisted with FD and depression are more likely to have more severe symptoms.
6.Clinical characteristics analyses of multiple myeloma with peripheral neuropathy

Objective To investigate the clinical manifestations and nerve electrophysiological abnormalities of the patients with multiple myeloma peripheral neuropathy (MMPN). Methods The clinical data of 35 patients with MMPN diagnosed in Shanxi Provincial People's Hospital from March 2010 to June 2018 were retrospectively analyzed. All patients received nerve electrophysiology examination, including nerve conductive velocity (NCV) and skin sympathetic response (SSR). Results The most common symptoms of peripheral nerve involvement for MMPN patients were sensory abnormalities (25 cases, 71%), including root pains (3 cases, 9%), pain ablation of the upper or lower limbs (18 cases, 51%), dyskinesia (15 cases, 43%), disappeared or reduced knee or ankle reflex (13 cases, 37%) and automatic sympathetic disorders (20 cases, 57%). Nerve electrophysiological tests showed 29 cases (83%) were involved in abnormal sensory nerve conduction velocity (SCV) or motor nerve conduction velocity (MCV), including 13 cases of upper limb abnormality in NCV, 5 cases of prolonged incubation in upper limb, and 10 cases of decreased amplitude;8 cases of lower limb abnormality in NCV, 6 cases of prolonged incubation in lower limb, and 4 cases of decreased amplitude. SSR test showed abnormalities were found in 17 patients (49%), including 8 cases of upper limb abnormality in SRR, 2 cases of prolonged incubation in upper limb, and 7 cases of decreased amplitude, 1 case of disappeared waveform; 6 cases of lower limb abnormality in SRR, 1 case of prolonged incubation in lower limb, and 4 cases of decreased amplitude, 2 cases of disappeared waveform; 3 cases of abnormal upper and lower limbs. Conclusions The most common peripheral nerve damages of MM are "sock and gloveˉlike" sensory ablation, accompanied with the involvement of automatic nerve damage. NCV is the major method to diagnose MMPN, and SSR plays an important role in the detection of sympathetic nerve damage of MMPN.
7. Value of serum HBV RNA in HBeAg-negative patients with chronic hepatitis B

Objective:
To analyze serum HBV-RNA levels in patients with chronic hepatitis B whose serum HBV-DNA has dropped to undetected levels after treatment with entecavir, and to explore the correlation between HBV-RNA level and liver biochemical parameters, which lay the research foundation for the clinical significance of new serological marker HBV-RNA.
Methods:
HBeAg negatively detected 107 cases with chronic hepatitis B whose serum HBV-DNA test results were lower than detection level for six consecutive months after receiving standard nucleoside therapy for more than 12 months were included. HBV-RNA level was detected by Perkin-Elmer reagent. HBV-DNA level was detected by Roche Cobas. Hitachi automatic biochemical analyzer was used to detect ALT and AST. Architect chemiluminescence analyzer was used to detect HBsAg, HBeAg, anti-HBe and anti-HBc. RStudio software was performed to analyze the correlation between HBV-RNA level and liver biochemical parameters. Logistic regression was used to analyze the independent factors influencing HBV-RNA level.
Results:
The positive detection rate of serum HBV-RNA in patients with chronic hepatitis B whose serum HBV-DNA had dropped to undetected levels after ETV treatment was 22.43%. HBsAg, ALT and AST levels in HBV-RNA positive group were slightly higher than HBV-RNA negative group, while anti-HBc levels were slightly higher in HBV-RNA negative group. There was no difference in the level of anti-HBe between the HBV-RNA negative and the positive group. Objective:
To study the distribution of sub-health and occupational stress as well as their correlation among middle school teachers in Tianjin, then provide evidences for prevention and control of the status of sub-health.
Methods:
A total of 3 522 middle school teachers from six districts of Tianjin were recruited with stratified cluster sampling strategy for the investigation of Sub-Health Measurement Scale version 1.0 (SHMS V1.0) and Occupational Stress Inventory-Revised Edition (OSI-R) .
Results:

9.Construction of training indexes of post competency of dental hygienist

Objective:To construct training indexes of post competency of dental hygienist, and to provide objective basis for the establishment of the index system of the transition from dental nurse to dental hygienist in the future.Methods:The relevant literature of post competency of dental hygienist was searched from the databases such as PubMed, Medline, Web of Science, China National Knowledge Internet, Wanfang database. The search time was from the establishment of the database to March 2021. The expert letter inquiry questionnaire was designed through preliminary consultation.Delphi expert consultation was conducted for 20 dental experts from Beijing city, Chongqing city, Jiangsu province, Sichuan province, Jilin province from May to November 2021, and model indexes and weight assignment of post competency of dental hygienist were determined.Results:Three rounds of Delphi expert consultation were conducted, the effective recoveries of the questionnaires were 100%, the expert authority coefficients were 0.81, 0.81, 0.83, respectively, the coefficient of variation of expert consultation was 0.000-0.386, 0.000-0.300 and 0.000-0.250, respectively, the coordination degree of expert opinions in the third round of consultation was 0.679, 0.428 and 0.389 (all P<0.01). The formed training indexes of post competency of dental hygienist included 4 first-level indexes, 20 second-level indexes and 60 third-level indexes. Conclusions:The training index system of post competency of dental hygienist in this study is scientific, reliable, and practical, to provide reference for training and assessment of post competency of dental hygienist and objective basis for establishing the indexes system of dental nurse to dental hygienist in the future.
10.Effectiveness of different specific exercise therapies in treatment of adolescent idiopathic scoliosis:a network meta-analysis

OBJECTIVE:At present,there are a variety of treatment methods for scoliosis using specific exercise therapy,but there is a lack of comparison of efficacy between different specific exercise therapy.This article compared the effectiveness of different specific exercise therapies to treat adolescent idiopathic scoliosis through a network meta-analysis. METHODS:Domestic and foreign electronic databases of relevant studies were searched for randomized controlled trials of specific exercise therapy for adolescent idiopathic scoliosis.Search time was from January 2000 to July 2023.The literature was screened by two reviewers using RevMan 5.4 and Stata 16.0 software to extract data and assess the bias risk of of inclusion studies. RESULTS:(1)This article includes 20 randomized controlled trials with 1 377 patients.Of them,12 studies involved Schroth therapy;2 studies involved BSPTS therapy,and 6 studies involved SEAS therapy.(2)The network meta-analysis indicated that in terms of improving Cobb angle and reducing trunk rotation angle in scoliosis patients,the BSPTS therapy group and Schroth therapy group were better than the conventional control group[WMD=-4.60,95%CI(-8.37,-0.82),P<0.05;WMD=-3.37,95%CI(-4.98,-1.75),P<0.05;WMD=-3.20,95%CI(-5.50,-0.90),P<0.05;WMD=-2.13,95%CI(-3.16,-1.09),P<0.05].The Schroth therapy group performed better than the conventional control group effective in improving the International Society for Scoliosis Research-22 Questionnaire quality of life score[WMD=1.41,95%CI(0.07,2.75),P<0.05]. CONCLUSION:Given the current evidence,BSPTS therapy group and Schroth therapy group were better than the conventional control group in improving Cobb angle and reducing trunk rotation angle.In the comparison of different specific exercise therapies,BSPTS therapy can be preferred to improve Cobb angle and reduce trunk rotation angle in adolescent idiopathic scoliosis patients.In addition,Schroth therapy may be the best treatment to improve the quality of life of adolescent idiopathic scoliosis patients.Limited by the quantity and quality of the included studies,the above conclusions should be interpreted with caution and need more high-quality studies to further validation.
