1.The relationship among resilience, general self-efficacy and coping styles in patients with tibia and fibula fracture

Objective To explore the relationship among resilience, general self-efficacy and coping styles of inpatients with tibia and fibula fracture. Methods Totally 104 inpatients with tibia and fibula fracture who met the conditions were investigated with General Self-Efficacy Scale, Connor-Davidson Resilience Scale and Medical Coping Modes Questionnaire. Results The total score of resilience with tibia and fibula fracture inpatient was 52.36 ± 9.56, which had a positive relationship with general self-efficacy and self-improvement coping style. Educational level, general self-efficacy, facing coping and yielding coping styles were influencing factors of resilience in patients with tibia and fibula fracture. Conclusions The level of resilience in patients with tibia and fibula fracture needs to be improved. Health care workers should pay attention to improve patient′s general self-efficacy, focus on patient′s coping styles and strengthen psychological health education of lower degree patients.

DEC1 is a basic helix-loop-helix(bHLH)transcription factor involved in the regulation of cell prolif-eration;cell differentiation;lymphocytes maturation;circadian rhythms;immune response and response to hypoxia.Recent studies have revealed that the expression of DEC1 is abnormally high in various tumor tissues and cells.In addition;the expression of DEC1 in the tumor tissue is related to the malignancy of various cancer types.This paper;therefore;focuses on the relationship between DEC1 and proliferation;apoptosis;cellular senes-cence;invasion and metastasis of tumor cells;in order to elaborate the role of DEC1 in the pathogenesis and pro-gression of tumor.
3.Effect of Livin RNA interference on radiosensitivity of colorectal cancer HT-29 cell xenograft in nude mice

Objective To explore the effects of silencing Livin gene by RNA interference mediated by lentiviral vector on colorectal cancer HT-29 cell xenograft growth and sensitivity to radiotherapy in nude mice.Methods BALB/c nude mice models were established by subcutaneously inoculating differently treated HT-29 cells into nude mice and the tumor growth situation of tumors was observed by measuring the volume of tumors and the weight of the nude mice at different time points after cell seeding.Livin expression was detected by RT-PCR and immunohistochemistry,respetively.Apoptosis rate was detected by TUNEL.Normal saline,lentivirus carring unrelated sequences,lentivirus caning Livin shRNA were injected intratumorally.All the nude mice were given 10 Gy of 6 MV X-ray irradiation.The changes of mice weight and the tumor volume were measured at different time points and the weight and tumor growth curves were drawn.Results The inhibition rate of tumor volume was(50.04 ± 0.07)% and the tumor weight of the RNA interfering group was significantly less than that in experimental group compared to the blank and negative groups(F=4.85,P<0.05),and the inhibition rate of tumor weight was(50.27 ±0.17)%.Relative Livin mRNA expression level in the RNA interfering experimental group was(17.75 ±0.08)%,and was significantly lower than that of the blank group(67.60 ± 0.05)% and the negative group(68.54 ± 0.03)%(F=89.97,P<0.01).Livin protein expression level in the RNA inferring group was also significantly lower[(36.00 ± 3.40)% versus(85.00 ± 3.15)%,(80.33 ± 3.08)%,F=107.32,P<0.01].The apoptosis rate in the RNA interfering experimental group was significantly higher than that in the blank and the negative groups[(23.67 ± 2.25)% versus(5.00 ± 1.50)%,(8.33 ± 1.82)%,F=56.94,P<0.01].Combined with radiotherapy,the tumor volume at different groups had significant difference(F=10.70,P<0.01),and RNA interfering group was significantly less than negative group and blank group(F=7.01-9.32,P<0.01).Conclusions Silencing of Livin gene expression by lentiviral vector-mediated RNA interference could inhibit the growth of colorectal HT-29 cell xenograft and increase the sensitivity of the transplanted tumors to radiotherapy.
4.Human cytomegalovirus infection and its significance in peripheral blood leukocytes of patients with systemic lupus erythematosus

Objective To detect the prevalence of human cytomegalovirus (HCMV) in peripheral blood of patients with systemic lupus erthematosus (SLE) and explore its role in the pathogenesis of SLE.Methods HCMV DNA was isolated from the peripheral blood leucocytes (PBLs) of 60 patients with SLE and 111 healthy controls.Nested polymerase chain reaction (nPCR) technology was used to investigate the gene of HCMV glycoprotein gB (UL55) in these specimens.HCMV infections in the PBLs of SLE patients were confirmed by HCMV-UL55 detection.Two-sample t test,nonparametric test,Chi-square test and Fisher probabilities were used to analyze.Results Agarose gel electrophoresis and sequencing analysis showed that established nPCR could specifically detect HCMV-UL55 gene,the HCMV infection rate was significantly higher in patients with SLE than in the healthy controls (P<0.01).Positive rates of HCMV infection in SLE group and controls were 41.7% (25/60) and 1.8% (2/111),respectively.Compared to the SLE patients with HCMV-negative PBLs,the positive rate of Rib-P [26%(9/35) vs 56%(14/25),x2=5.659,P=0.017],the positive rate of direct Coomb's test [37%(13/35) vs 72%(18/25),x2=7.096,P=0.008] and the level of antiβ2GP1 [21.3 (9.9,51.8) U/ml vs 13.6 (5.9,23.1) U/ml,U=2.017,P=0.044] were significantly higher than those in the SLE patients with HCMV-positive PBLs.Compared to the SLE patients with HCMV-negative PBLs,the number of red blood cells [(3.65±0.10)×1012/L vs (3.17±0.17)×1012/L,t=2.574,P=0.013] and lymphocytes [(1.37±0.14)×1012/L vs (0.90±0.13)×1012/L,t=2.456,P=0.017] in peripheral blood and the hemoglobin levels [(110±19) g/L vs (98±5)g/L,t=2.034,P=0.048] of the SLE patients with HCMV-positive decreased significantly.At the same time,the positive rate of hematuria [40%(14/35) vs 72%(18/25),x2=6.000,P=0.014] and 24 h proteinuria [0.80 (0.53,2.37)g vs 0.48 (0.13,1.21)g,U=2.140,P=0.032],which indicated kidney damage were also significantly increased in SLE patients with HCMV-positive PBLs.Conclusion The infection of HCMV in peripheral blood cells may take part in the development of SLE.
5.Effect of Small Interfering RNA Silencing the Fatty Acid Synthase Gene on Lipid Metabolism in Human Hepatic Cell Line HepG2

Objective: To investigate the effect of the gene interfering technology on fatty acid synthase (FAS) gene silencing for lipid contents in human hepatic cell line HepG2 and to study the lipid metabolism related gene expression in HepG2 cells. Methods: A total of 3 pairs of small interfering RNA (siRNA) targeting different sequences of FAS mRNA were synthesized as FAS-siRNA-1, FAS-siRNA-2 and FAS-siRNA-3, meanwhile, 2 controls were established as Blank control group, in which HepG2 cells were not treated, and Negative control group, in which HepG2 cells were transfected by non-effective siRNA. The mRNA, and protein expression levels of FAS in HepG2 cells were examined by real-time lfuorescence quantitative RCR and Western blot analysis to screen the most effective pair of siRNA for FAS gene silencing; and that speciifc siRNA was transtected to HepG2 cells for 48 hours to detect the intra-/extra-cellular TG, TC levels and the mRNA expression related to lipid metabolism in HepG2 cells. Results: The screening experiment indicated that FAS-siRNA-3 was most effective for FAS gene silencing. Compared with Blank control group, the mRNA and protein expressions in FAS-siRNA-3 transfected HepG2 cells (Transfected group)decreased to (52.33 ± 3.07) % and (51.57 ± 3.14) % respectively. Compared with Blank control group, Transfected group had the reduced intra-/extra-cellular TG levels and reduced extracellular TC level; while increased mRNA expression of hepatic lipase,P<0.0001 and decreased mRNA expression of TG transfer protein in HepG2 microsome,P<0.05. Conclusion: FAS gene silencing could signiifcantly decrease the intra-/extra- cellular TG level and extracellular TC level in HepG2 cells, those ifndings need to be conifrmed by furtherin vivo andin vitro studies.
6.Use of Fibroscan for Evaluating Efficacy of Combined Therapy with FuFang BieJia RuanGan Tablet and Antiviral Drugs in Patients with Chronic Hepatitis B Virus-related Cirrhosis

Background:Fibroscan is the noninvasive method widely used to evaluate quantitatively the liver fibrosis and monitor the long-term efficacy of anti-fibrosis therapy. Aims:To study the use of Fibroscan for evaluating the efficacy of combined therapy with FuFang BieJia RuanGan tablet and antiviral drugs in patients with hepatitis B virus( HBV)-related cirrhosis. Methods:A total of 90 patients with HBV-related cirrhosis from March 2013 to September 2014 at Shanghai Ren Ji Hospital were recruited,and divided into treatment group and control group. Patients in treatment group received FuFang BieJia RuanGan tablet,and patients in control group received conventional liver-protective drugs,all the patients took nucleoside antiviral drugs at the same time. The treatment courses in both groups were 6 months. Liver stiffness measurement( LSM)was detected by Fibroscan before and after treatment. Biochemical parameters,width of portal vein and clinical symptoms were recorded. Results:After treatment,LSM was significantly decreased in both groups( P <0.05). Liver function,width of portal vein and Child-Pugh score were improved in both groups(P <0. 05),and no significant differences were found between the two groups(P>0. 05). LSM was closely associated with Child-Pugh score both before and after treatment(r=0. 484,P<0. 01;r=0. 523,P<0. 01). Patients with Child-Pugh A had lower LSM than those with Child-Pugh B or Child-Pugh C(P<0. 01). Conclusions:FuFang BieJia RuanGan tablet combined with oral antiviral drugs can remarkably improve the liver function of cirrhotic patients and prevent progression of cirrhosis. Dynamic detection of LSM can be used for monitoring drug efficacy and disease progression in patients with cirrhosis.
7.Effect of bifunctional IL2-GMCSF in promoting dendritic cell activation in vitro in simulated tumor-induced immune suppression.

OBJECTIVETo test the effect of bifunctional molecule IL2-GMCSF in promoting the activation of dendritic cells (DCs) cultured in tumor conditioned medium.
METHODSWe prepared a tumor conditioned medium using mouse melanoma cell line B16F10 supplemented with IL2-GMCSF, GM-CSF, IL-2, or the combination of the latter two. After culturing mouse DC cell line DC2.4 in the conditioned medium for 24 h, the DCs were examined for phagocytosis, proliferation, maturation phenotype, cytokine secretion, and signal pathway activation.
RESULTSDC2.4 cells displayed characteristics of immature DCs. After cell culture in the conditioned medium, the cells showed enhanced phagocytosis but significantly suppressed cell proliferation activity. Culture in the conditioned medium also promoted DC cell maturation and secretion of macrophage-derived chemokine (MDC), but inhibited IL-12 secretion. Supplementation of the conditioned medium with IL2-GMCSF promoted phagocytosis, proliferation, maturation, and cytokine (including both IL-12 and MDC) secretion of DC2.4 cells. Compared with GM-CSF, IL2-GMCSF induced a higher level of NF-κB signal pathway activation but suppressed STAT3 activation.
CONCLUSIONCompared with GM-CSF, IL2-GMCSF can better promote DC activation in the context of tumor-induced immune suppression, and thus shows potentials in anti-tumor therapy.

8.Pain management effect of quality control circle mode on patients with lower limb fractures

Objective To compare the pain management effects between painless wards with quality control circle mode and conventional pain management mode.Methods A retrospective case control study was conducted on the clinical data of 233 patients with lower limb fracture admitted from August 2015 to August 2016.There were 124 males and 109 females,aged 18-74 years [(48.3 ±3.3)years].The patients were divided into observation group (n =117) and control group (n =116) according to the pain management mode.The observation group followed the standard continuous quality improvement program and combined with professional team and patients Wechat group to implement pain management,and further measures were taken in accordance with the feedbacks.The control group adopted routine painless ward nursing model for perioperative analgesia nursing intervention.The pain score VAS,the start time of functional exercise,the compliance of rehabilitation activities,the length of hospital stay,and the healing time of fracture were compared between the two groups.Results There was no significant difference in VAS scores between the two groups at 12 hours before operation and 6 hours after operation (P > 0.05).The observation group had lower VAS scores at 12 hours (3.2± 1.4),24 hours (2.8 ±0.9),48 hours (1.6 ± 0.7),and 72 hours (1.5 ± 0.8) after operation than the control group (P <0.05).The observation group started functional exercises earlier [(18.9 ± 0.4) hours after operation]than the control group earlier [(48.1 ± 1.7) hours after operation] (P < 0.01).The observation group had a rehabilitation compliance rate of 62.6%,higher than that of the control group (17.6%) (P <0.05).The hospital stay [(12.18 ± 0.14) days] and fracture healing time [(97.86 ± 0.83) days] of the observation group were shorter than those of the control group (P < 0.05).Conclusion The pain management model of standardized continuous quality improvement can significantly relieve pain in patients with lower limb fracture,shorten hospitalization time,bring forward the start time of functional exercise,improve the compliance of rehabilitation activities,and promote fracture healing.
9.Design and application of energy monitoring system for laboratory animal facilities

Objective Experimental animal facilities account for a significant proportion of the energy consumption by scientific research institutions;however,the energy consumption characteristics of these facilities differ from those of ordinary buildings,and thus require specialized monitoring and management.Methods A set of energy consumption monitoring systems was designed for experimental animal facilities and deployed in the specific pathogen-free-level experimental animal facility of Huazhong University of Science and Technology.Results The system achieved real-time collection and recording of the facility's electricity consumption data,and proposed energy-saving measures for three application scenarios.Conclusions This energy consumption monitoring system designed for experimental animal facilities is reliable,efficient,and user-friendly,and has the potential to guide and promote energy management programs at experimental animal facilities.
10.Correlation between birth weight and blood pressure of child and adolescent: A Meta-analysis

Objective To systematically review the correlation between birth weight and blood pressure of child and adolescent, and provide a theoretical basis for the etiologic research and the prevention for hypertension of child and adolescent.Methods Comprehensive electronic searches of Chinese Web of Knowledge (CNKI), Wang Fang, Wei Pu (VIP), PubMed and Web of Science were conducted to identify any study reporting the correlation of birth weight and blood pressure of child and adolescent.SPSS 13.0 software was used to convert the data.Meta-analysis was conducted with STATA 12.0 software.Results Thirty four researches were included in this study.The results of meta-analysis showed that low birth weight children or adolescents had a higher risk of hypertension with a significant difference compared to the group which birth weight greater than 2 500 g (OR =1.20;95% CI:1.09-1.33;P < 0.01).Its systolic blood pressure and diastolic blood pressure was on the high side.In addition, it showed that no significant differences were found between hypertension and high birth weight (OR =1.03;95% CI:0.89-1.18;P > 0.05).But its systolic blood pressure and diastolic blood pressure was on the high side too.Conclusions Abnormal birth weight is probably a risk factor of child and adolescent high blood pressure.Low birth weight has greater influence on blood pressure of children and adolescents.It can be detected early to prevent abnormal birth weight and child hypertension.
