1.Study on inhibitory effects and mechanism of amlodipine on human breast carcinoma cell line MCF-7

Aim To observe the effects of amlodipine on cell cycle,cell cycle-related genes and cyclin expression of human breast carcinoma MCF-7 cells.And to probe effects of amlodipine on cell cycle and its mechanism of human breast carcinoma MCF-7 cells.Methods In vitro growth inhibitory effects of amlodip-ine on human breast carcinoma MCF-7 cells were determined by MTT assay,cell cycle distribution was detected by flow cytometry,the Mrna expression levels of cell cycle-related genes cyclinD1 and p21 were treated by RT-PCR,and the protein expression of cell cycle protein cyclinD1 and p21 were assessed by Western blot.Results With dose and time dependently,amlodipine could inhibit the proliferation of human breast carcinoma MCF-7 cells in vitro.The IC_(50) was 14.439 μmol·L~(-1).When breast cancer cells MCF-7 were treated with 7.22 μmol·L~(-1)(1/2 IC_(50)),14.439 μmol·L~(-1)(IC_(50))and 28.88 μmol·L~(-1)(2IC_(50))amlodipine for 48 h,the ratios in the G_0/G_1 phase were significantly increased as compared with control group(P<0.05).Amlodipine inhibited the expression of Mrna and protein of cyclinD1,while increased the expression of Mrna and protein of p21.Conclusions Amlodipine exhibits obvious anti-tumor activities on human breast carcinoma MCF-7 cells and arrest cell cycle in G_1 phase.The mechanism of G_1 phase arresting may be related to modulating the Mrna and protein expression of cell cycle-related gene cyclinD1 and p21.
2.An experimental study of spongy and putty-like demineralized bone matrix (DBM) in the repair of segmental defects

0.05), whereas the value of new bone grafted with pDBM was significantly lower than that of the normal group (P0.05); but the CUS of the pDBM grafted group was significantly lower than that of normal radius (P0.05). Histological analysis exhibited that most of the DBM was absorbed and substituted by matured new cortical bone in the treated defects of both groups 6 weeks postoperatively, whereas in the untreated group, the defects were only filled with fibrous connective tissue in their mid-portion. Conclusion The sDBM and pDBM are both effective in repairing segmental bone defects. The properties of new bone induced by grafts with sDBM are superior to that of pDBM in biomechanics. These materials can be used in clinical practice as bone graft extenders or enhancers.
3.Effect of boningmycin on expression of capsid protein L1 and serum immune factors in cervical HPV infection patients

Objective To investigate the effect of boningmycin on expression of capsid protein L1 and serum immune factors in cervical HPV infection patients.Methods 60 patients with cervical HPV infection were divided into experimental group and control group, 30 cases in each group.The control group were given recombinant human interferon α2b suppository, 100 000 IU, once every other day, and the experiment group were given boningmycin 300 mg intramuscular injection,once every other day.Treatment of 10 times for a course,2 consecutive courses After treatment, the HPV capsid protein L1, serum TNF-α, IL-2, IFN-αand HPV were compared.Results Compared with control group after treatment, the percentage of HPV capsid protein L1 increased ( P <0.05 ) , serum TNF-α, IFN-αlevels increased ( P <0.05 ) , IL-2 decreased ( P <0.05 ) , and HPV positive rate decreased( P <0.05 ).Conclusion Boningmycin could reduce the percentage of capsid protein L1 in patients with cervical human papilloma virus infection, elevate serum TNF-αand IFN-αlevels, reduce the level of IL-2 and HPV positive rate.
4.Clinical analysis of 19 AIDS patients coinfected with cryptococcus meningitis

Objective To observe the clinical features of AIDS coinfected with cryptococcus meningitis. Methods The clinical data of 19 AIDS patients coinfected with cryptococcus meningitis were retrospectively analyzed.Results Among 19 patients, the main clinical manifestation including headache (100.0%), nausea/vomiting (94.7%), fever (78.9%) and neck stiffness (84.2%).Seven cases revealed increased CSF protein, 10 cases showed depressed CSF glucose levels and 11 patients revealed elevated CSF pressures.The mean CD4+cell count was (58.9 ±27.8)/mm3.Imaging examination showed the neurological complications were hydrocephalus, cerebral infarction, cerebral atrophy and cerebral hernia.The usage rate of amphotericin B, amphotericin B liposome, fluconazole and fluorine cytosine was 31.6%, 21.1%, 47.4% and 47.4%, respectively.Only 2 cases received antiretroviral therapy.The misdiagnosis rate was 31.6%, and mortality rate was 21.1%.Conclusions AIDS coinfected with cryptococcus meningitis onsets hidden, with the clinical manifestation is atypical, and the misdiagnosis rate is higher.Early diagnosis and early usage of appropriate antifungal therapy/antiretroviral therapy can help to prevent the development of it.
5.To confirm the structure validity of the job satisfaction questionnaire of the rear service personnel

Objective To confirm the structure validity of questionnaire through a probe into latent factorial structure of the rear service personnel′s job satisfaction from polyclinics hospitals .Methods The questionnaire′s construct validity was estimated by ex‐ploratory factor analysis (EFA) and confirmatory factor analysis(CFA) .Results Exploratory factor analysis made the question‐naire including six factors which belonged to job satisfaction and occupation loyalty .Every factor had reasonable explanation and six factors remained 70 .306% information .Exploratory factor analysis of job satisfaction showed five factors ,explaining 70 .514% in‐formation .The confirmatory factor analysis model 1 and model 2 were conducted by LISREL8 .70 software .The results showed that X2 freedom degree was less than 5 .In addition to adjust the goodness of fit index was less than 0 .9 ,all other fit indices were greater than 0 .9 .At the same time ,SRMR and RMSEA were less than 0 .08 .Conclusion The results of exploratory factor analysis(EFA) and confirmatory factor analysis(CFA) both reveal that the questionnaire′s construct validity was good .
6.Theoretical research and progress of basal body and bio-coatings in biomaterial field

BACKGROUND: In the research of biomaterial, especially bio-coatings field, how to find a good biological and mechanical properties basal body, and to prepare coatings with uniform and compact surface and strong bonding force, is the key factor in clinical Application of artificial prosthesis. OBJECTIVE: To review the progress of bio-coatings.METHODS: Databases of CNKI, VIP, and Science Direct were retrieved with key words of "Biocoatings, HAp, Titanium, Carbon/carbon composites" to search papers published from January 1998 to October 2009. PApers underlying bio-coatings were selected. Totally 216 literatures were initial searched, according to inclusive criteria, 31 literature were summarized. RESULTS AND CONCLUSION: By contrasting the basal bodies made of titanium or carbon/carbon and different coating preparation, we found that the basal body composed of carbon/carbon composites exhibited good biocompatibility, which had similar strength to natural bone. Using hydrothermal method, the composites coating prepared by taking carbon/carbon as basal body and hydroxyApatite, chitosan, or polymethyl methacrylate as coatings provided a new idea for increasing the combing strength between coatings and basal body.
7.Immunomodulatory effects of probiotics on rat models with ovalbumin induced food allergy

Objective To investigate the effects of probiotics on the occurrence of ovalbumin (OVA) induced food allergy and the regulatory effects on immune function of rat models. Methods Thirty female Brown-Norway rats aged 3 weeks were randomly divided into blank control group,food allergy group and probiotics intervention group(n=10).The levels of serum OVA-IgE and intestinal sIgA were measured by ELISA method.Splenic lymphocytes were isolated and cultured in vitro,and the Treg lymphocyte subgroups in the spleen were analyzed by flow eytometry.The levels of IFN-γ,IL-4 and IL-10 in the supernatant of cultured splenic lymphocytes were measured by ELISA method. Results The serum OVA-IgE level in food allergy group was significantly higher than that in blank control group(P<0.05),while that in probiotics intervention group was significantly lower than that in food allergy group(P<0.05).Compared with food allergy group and blank control group,the level of intestinal sIgA in probiotics intervention group was significantly higher(P<0.05).The percentage of CD4~+CD25~+T lymphoeytes in food allergy group was significantly lower than that in blank contml group(P
8.Protective role of heme oxygenase in myocardial dysfunction postresuscitation

Objective To study the cardioprotection effects of heme oxygenase-1(HO-1)on cardiopulmonary resuscitation(CPR).Method Male Sprague-Dawley rats were asphyxiated for 9 minutes and resuscitated.Rats wefe randomly divided into 4 groups,namely,sham asphysiation group,CPR group,Hemin group and Heroin +ZnPP group(zinc protoporyphyrin IX).Resuscitated groups were further divided into two subgroups according to various intervals:6 hours and 24 hours after resuscitation.Hemodynamic was observed.Serum creatine phosphokinase-MB(CPK-MB)and lactate dehydrogenase were determined.HO-1 in heart tissue homogenates was assayed.Ultrastructure of rats hearts was examined.Statical evaluation was performed with analysis of variance.Results The mean blood pressure(MBP)in resuscitated groups was significantly reduced after resuscitation,hadn't any difference between supgroups.The scores of dp/dt 40 and-dp/dt were significantly decreased in CPR group and Hemin+ZnPP group after resuscitation(all P<0.05),but dP/dt40 in Heroin group did nol differ significantly after resuscitation.and-dp/dt decreased only 0.5 hours and one hour after resuscitation and returned to baseline values two hours after resuscitation.The scores of dp/dt 40and-dp/dt in Heroin group at different intervals after resuscitation were significantly higher than those in CPR group and Hemin+ZnPP group(all P<0.05).Serum CPK-MB and LDH in CPR group and Hemin+ZnPP group at 6 hours and 24 hours after resuscitation were significantly higher than those in Hemin group(all P<0.05).The cardiac tissue ultrastructure of rats in Hemin group was more intact than that of CPB group and Hemin+ZnPP goup.HO-1 levels in heart tissue homogenates of Hemin group at 6 hours and 24 hours after resuscitation were significantly higher than those in CPR group and Heroin+ZnPP group(all P<0.05).Conclusions HO-1 expression induced by Heroin can effectively improve post-resuscitation myocardial dysfunction,alleviate cardiac injury,keep the ultrassructure integrity of cardiac myocytes.It may be a new approach to treat myocardial dysfunction after resuscitation.
9.Biological characteristics of umbilical cord mesenchymal stem cells in different conditioned media

BACKGROUND:The growth of mesenchymal stem cells in vitro in different conditioned media is different evidently. So it is necessary to choose a more suitable medium. OBJECTIVE:To contrast and observe the proliferation of human umbilical cord mesenchymal stem cells in three kinds of media and to check the immunophenotype and differentiation ability of mesenchymal stem cells. METHODS:Human umbilical cord mesenchymal stem cells were col ected by explant method in sterile conditions. After subculturing by T75 incubation bottles, the third generation of mesenchymal stem cells were cultured in Dulbecco’s modified Eagle’s medium/Ham’s nutrient mixture F-12 medium containing 5%fetal bovine serum, Dulbecco’s modified Eagle’s medium/Ham’s nutrient mixture F-12 containing 10%fetal bovine serum and Mesen PRO RSTM medium. After 1, 3, 5, 7 days of culture, the cells were counted to draw a growth curve. Immunophenotype of the third generation of umbilical cord mesenchymal stem cells were determined by flow cytometry and the ability of osteogenic and adipogenic differentiation was also detected. RESULTS AND CONCLUSION:The third generation of cells cultured highly expressed CD44, CD73, CD90, CD105, but did not express CD29, CD31, CD34, HLA-DR. The oil red O staining showed a lot of little red lipid drops after adipogenic induction;alizarin red staining showed osteoblast-like cells after osteogenic induction, indicating umbilical cord mesenchymal stem cells in vitro have the potential of multi-directional differentiation. After observation and counting, the colony and shape of cells cultured in Dulbecco’s modified Eagle’s medium/Ham’s nutrient mixture F-12 containing 10%fetal bovine serum were superior to those cultured in the other two media. Therefore, it is concluded that Dulbecco’s modified Eagle’s medium/Ham’s nutrient mixture F-12 containing 10%fetal bovine serum is preferred for cellsubculture.
10.The value of MRI in diagnosing and classifying of plasma cell mastitis

Objective To evaluate the value of the magnetic resonance imaging (MRI)in the diagnosis and classification of plas-ma-cell mastitis (PCM).Methods The breast MRIs of eighteen pathologically confirmed PCM patients were retrospectively ob-served and analyzed.The manifestations of lesions,including shape,size,margin,and characteristic enhancement on T1 WI,T2 WI with fat suppression and contrast enhanced T1 WI were observed and documented respectively.Results PCM were divided into three types according to MRIs:the inflammation type 4 cases,the abscess type 9 cases,and the mixed type 5 cases.In the inflammation type,3 cases of lesions were wide extending while the other 1 limited in nipple and areola.All 4 cases of lesions were hypointensity on T1 WI and hyperintensity on T2 WI with fat suppression.And in the contrast enhanced scanning,2 cases showed patchy hyperin-tensity and 2 cases showed branch-like enhancement.In abscess type,6 cases of lesions were wide and 3 cases limited in one quad-rant.Of all 9 cases,there were 3 cases with single abscess and 6 with multiple abscess;and all cases showed hypointensity on T1 WI and hyperintensity on T2 WI with fat suppression.In the enhanced scanning,there were 4 cases with honeycomb-like hyperintensity and 5 cases with annular and patchy enhancement.In mixed type,inflammation was accompanied with abscess and fistula.All cases were hypointensity on T1 WI and hyperintensity on T2 WI with fat suppression.In the enhanced scanning,3 cases showed honeycomb-like enhancement,1 case showed multiple annular enhancement and 1 case showed annular and patchy enhancement.Regions of in-terest were selected and time-signal intensity curves were obtianed in every enhancement cases,all of which were influent.Of all ca-ses,5 showed high-protein deposition in the expansion ducts,exhibiting hyperintensity on T1 WI and hypointensity on T2 WI with fat suppression.Conclusion The abscess type is the most common in PCM and the second is the mixed type.Annular and honeycomb-like enhancement are characteristic appearance of these two types,which are helpful in the diagnosis and classification of PCM.
