Acta Nutrimenta Sinica 1956;0(04):-
Objective:To investigate the effects of genistein (Gen) on apoptosis in human osteoblast. Methods:Human periosteum was maintained in DMEM medium containing 10% newborn calf serum through four passages to isolate and proliferate osteoblast. Then, cells were maintained in phenol red-free DMEM supplemented with 5% charcoal-stripped FCS for 4 d before addition of treatments. Flow cytometer and DNA fragment were used to explore apoptosis in osteoblast, and immunohistochemistry was used to study protein expression of Bax, Bcl-2 and caspase-3. Results:Estrogen depletion could induce apoptosis in human osteoblast. With the similarity of 10-8 mol/L E2, at the conentrations of 10-6 mol/L and 10-5 mol/L, Gen could inhibit apoptosis induced by estrogen depletion. Compared with vehicle control, Gen significantly decreased protein expression of Bax and caspase-3 and increased protein expression of Bcl-2. Conclusion:Genistein could inhibit apoptosis induced by estrogen depletion in human osteoblast.

Journal of Huazhong University of Science and Technology (Medical Sciences) 2007;27(6):660-3
To explore the relation of angiotensin-converting enzyme (ACE) and angiotensin II type 1 receptor (AT1R) gene polymorphism with coronary heart disease (CHD) and the severity of coronary artery stenosis, 130 CHD patients who underwent coronary angiography were examined for the number of affected coronary vessels (> or = 75% stenosis) and coronary Jeopardy score. The insertion/deletion of ACE gene polymorphism and AT1R gene polymorphism (an A-->C transversion at nucleotide position 1166) were detected by using polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP) in CHD patients and 90 healthy serving as controls. The results showed that DD genotype and of ACE were more frequent in CHD patients than that in control group (38.5% vs 14.4%, P<0.001). The frequency of the AT1R A/C genotypes did not differ between the patients and the controls (10% vs 13.1%, P>0.05). The relative risk associated with the ACE-DD was increased by AT1R-AC genotype. Neither the number of affected coronary vessels nor the coronary score differed among the ACE I/D genotypes (P>0.05). But the number of affected coronary vessels and the coronary score were significantly greater in the patients with the AT1R-AC genotype than in those with the AA genotype (P<0.05). In conclusion, DD genotype may be risk factor for CHD and MI in Chinese people, and is not responsible for the development of the coronary artery stenosis. The AT1R-C allele may increase the relative risk associated with the ACE-DD genotype, and may be involved in the development of the stenosis of coronary artery.

China Occupational Medicine 2022;49(2):180-185
Objective To compare and analyze the occupational exposure limit(OEL)of hexavalent chromium compounds in
China and foreign countries. Methods The OEL and background information of hexavalent chromium compounds released by
nine official and unofficial organizations in seven countries/regions were collected and sorted. The classification, limit levels,
formulation principles and compound characteristics labeling of OEL were compared. Results The OEL values published by
nine organizations ranged from 0.000 02 to 0.050 00 mg/m3
. Among them, the limits of seven organizations(including China)
did not clearly distinguish the soluble types of hexavalent chromium compounds;the limits of eight organizations(including
China)were calculated in terms of chromium;seven organizations(including China)have only developed OEL for long-term
exposure. The OEL of hexavalent chromium in China was 0.050 00 mg/m3 which is relatively loose. Its carcinogenicity label was
consistent with international standards, its sensitization label was consistent with some countries, and its percutaneous
absorption label had not been included in the characteristic labeling of limit values. Conclusion It is suggested that the OEL of
hexavalent chromium in China should be revised timely, and the systemic adverse effects caused by percutaneous absorption of
hexavalent chromium compounds should be focused on. At the same time,it is recommended to systematically adjust the
management concepts and principles of all carcinogens.

Military Medical Sciences 2017;41(6):498-501
Objective To investigate the mechanism by which hydrogen(H2) helps prevent acute lung injury induced by hyperoxia (HALI) in rats. Methods Thirty male Sprague-Dawley rats were randomly divided into three groups: control group, HALI group and H2 group, with 10 rats in each group. The control group was exposed to air at atmospheric pressure.Rats in HALI and H2 groups were exposed continuously to pure oxygen (100%O2) for 60 hours and during this period, 10 ml/kg of normal saline or H2-saturated normal saline was given every 12 hours by intraperitoneal injection to the HALI and H2 groups, respectively. After treatment, the arterial partial pressure of oxygen was examined and histopathological examination was conducted in each group.Then,RT-qPCR and Western blotting were performed to measure the transcriptional level and protein expression of heme oxygenase 1 (human heme oxygenase 1, HO-1) in rat lung tissue. Results Compared with the HALI group, H2 group showed significantly decreased severity of lung injury and a marked increase in the arterial oxygen saturation.Besides, H2 treatment induced up-regulation of HO-1 mRNA and protein levels. Conclusion The findings suggest that HO-1 may play an important role in the protection against HALI by H2.
