Purpose:To study the expression and clinical significance of PTEN in non-small cell lung cancer. Methods:Immunohistochemical staining was used to determine the expression of PTEN in 10 cases of normal lung issues and 56 cases of non-small cell lung cancer. Results:The positive incidences of PTEN expression in NSCLC was 59% while positive expression was found in all normal lung issues. The expression of PTEN was correlated to degree of differention of cancer, lymph node metastasis and clinical stages. The PTEN negative group was seen to have the worst survival rate among all groups. Conclusions:The results suggest that PTEN gene plays an important role in carcinogenesis and progress of NSCLC. Expression of PTEN may be valuable in prognosis assessment of NSCLC.

OBJECTIVE: To observe the effects of Astragalus polysaccharides (APS) on the proliferation and differentiation of 3T3-L1 preadipocytes and to elucidate its possible mechanism. METHODS: The proliferation of 3T3-L1 preadipocytes was detected by XTT method. Lipid droplets accumulated in cytoplasm of the differentiated preadipocytes were observed by using red O staining and quantified by colorimetry. The expressions of peroxisome proliferation activated receptor gamma (PPAR gamma) and CAAT/enhancer binding protein (C/EBP alpha) mRNAs and proteins were detected by real-time polymerase chain reaction (RT-PCR) and Western blotting respectively. RESULTS: APS at different concentrations (0.025-0.8 g/L) affected 3T3-L1 preadipocyte proliferation and differentiation dose-dependently. 3T3-L1 preadipocytes treated with 0.4 g/L APS had lots of lipid droplets in the cytoplasma, which were similar to cells treated with rosiglitazone (ROS). APS significantly increased the mRNA and protein expressions of PPAR gamma and C/EBP alpha (P<0.05, P<0.01, compared with the normal control group) in the course of 3T3-LI preadipocyte differentiation. CONCLUSION: APS can promote the proliferation of 3T3-L1 preadipocytes, enhance the accumulation of lipid drops, and increase the terminal differentiation of preadipocytes, which may be associated with its effects in increasing the expressions of PPAR gamma and C/EBP alpha mRNAs and proteins. The study suggests that APS has potential in the treatment of metabolic syndrome.

Objective To study diffusion characteristic, diffusion-weighted imaging (DWI) technique and its value in lumbar osteolytic metastases. Methods Twenty patients with lumbar osteolytic metastases and 20 controls were performed with sagittal SE T_1WI, FSE T_2WI, fat saturation FSE T_2WI(FS-FSE T_2WI)and single-shot spin-echo echoplanar DWI, respectively. Contrast noise ratios (CNRs) of the metastases on various series were compared. On DWI, signal attenuation ratios (SARs) and apparent diffusion coefficients (ADCs) of lumbar osteolytic metastases and normal vertebrae were analyzed. Results (1) Routine MRI appearance: There were 23 lesions in the 20 patients. The signal intensity of all lesions showed hypointense on SE T_1WI, hyperintense on FS-FSE T_2WI and DWI, and mixed signal intensity (5 cases), isointense (12 cases) or slight hyperintense (6 cases) on FSE T_2WI. (2) CNR: The CNRs of patient group on FSE T_2WI(5.70?3.82)were lower than those on SE T_1WI(25.62?11.73), FS-FSE T_2WI(23.37?7.48)and DWI (b=600 s/mm2)(24.69?9.87)(U values were 5.393, 6.359, and 5.547, respectively,P

Objective To explore the features of leukoencephalopathy with cerebral calcifications and cysts (LCC) in clinic, radiology and pathology in order to improve skills in diagnosis. Methods Two female patients had CT and/or MRI scan, and case 2 had contrast enhancement MRI scan additionally. Both cases had blood biochemistry examinations including calcium, phosphorus and alkaline phosphatase et al. Case 2 had lumbar puncture and cerebrospinal fluid test. The major cystic lesion was surgically removed in both patients and offered a histopathological examination. Results CT scan reveaed diffuse calcifications in the bilateral basal ganglia, white matter of frontal lobe and/or dentate nuclei of cerebellum in both cases, and major cystic lesion in right frontal lobe (case 1) and the left parietal lobe (case 2). The rim of enhancement was observed in cystic lesion on MRI. Histopatbological examination revealed angiomatous rearrangements of the microvessels with fibroid, hyaline degeneration and haemosiderin deposits, brain tissue associated with areas of demyelinization, some Rosenthal fibers, gliosis, calcium deposits and hemorrhage, fibrinoid necrosis occurs in partial vessels associated with thrombogenesis and stenosis as changes in arteriolitis. Blood biochemistry examination showed normal. Cerebrospinal fluid test in case 2 showed increased intracranial pressure(350 mm H_2O,I mm H_2O =0. 0098 kPa). Conclusions The onset of LCC varies and occurs from early infancy to adult. The asymmetrical calcification is characteristic in LCC. Hemorrhage could be involved in the pathogenesis of cystic formation. LCC is characterized by a cerebral obliterative microangiopathy, both demyelinization and the edematous changes could probably result in white matter abnormalities on neuroimaging.
