Objective To detect the expressions of Numb and α-catenin in the gastric cancer and matched normal gastric mucosa, and to analyze their relationship with clinicopathological factors of gastric cancer and explore their roles in gastric carcinogenesis and progression. Methods Immunohistochemical method was used to detect the expressions of Numb and α-catenin in 109 cases of tumor specimens and 97 cases of matched normal gastric mucosa. The correlations between Numb,α-catenin and clinicopathological factors were analyzed. Results The positive rates of Numb and α-catenin in gastric cancer were significantly lower than those in normal gastric mucosa (60.6% ∶ 100.0%,x2 =48.361,P =0.000;76.1% ∶ 100.0%,x2 =26.480,P=0.000).The expression of Numb protein was correlated with Lauren type (x2 =17.018,P =0.000) and tubular adenocarcinoma differentiation (x2 =17.586,P =0.000).The expression of α-catenin protein was correlated with Borrmann type (x2 =6.700,P =0.035),Lauren type (x2 =11.098,P =0.001),tubular adenocarcinoma differentiation (x2 =8.203,P =0.017) and lymph node metastasis (x2 =6.402,P =0.011).The expressions of Numb and α-catenin were statistically correlated in 109 cases of gastric cancer (rk =0.184,P =0.028).Conclusion Compared with normal gastric mucosa, both Numb and α-catenin expressions are down-regulated and the two expressions are correlated in gastric cancer.Numb and α-catenin may be involved in the regulation of histological differentiation of gastric cancer by certain passages,Numb protein may be adjusted by α-catenin pathway which is involved in Wnt-β-catenin pathway, and thus plays an important role in promoting cell proliferation, invasion and metastasis in human gastric cancer.

Objective To analyze the clinicopathological characteristics and the related factors influencing the prognosis of pancreatic neuroendocrine carcinoma. Methods The clinicpathological and follow-up data of 21 patients with pancreatic neuroendocrine carcinoma admitted between April 2000 and August 2013 in Zhongshan Hospital of Fudan University were analyzed retrospectively. The influence facts on patients' prognosis were analyzed statistically. Results Univariate analysis showed that age, stage, tumor size, degree of tumor differentiation and lymph node metastasis were factors separately influenced patients' survival.Multivariate analysis (Cox regression) revealed that age (P =0.019) was the only independent factor affecting the prognosis. Conclusions Patients with pancreatic neuroendocrine carcinoma were mostly non-functional and presented no specific clinical features.Tumors tended to metastasize and the prognosis was poor.Age was the independent factor affecting the prognosis.

Human astrovirus (HastV) is recognized as one of the leading causes of acute viral diarrhea in infants. The HastV non-structural protein, nsPla, and C-terminal protein, nsPla/4, contain various conserved functional domains, and may play an important role in virus replication, transcription and the virus-host interactions of HastV. This study used an E. coli system to investigate the expression of nsPla and nsPla/4 proteins. Firstly, the nsPla and nsPla/4 genes of HAstV-1 were cloned into the prokaryotic expression vector,PGEX-4T-1, to build the PGEX-4T-1a and PGEX-4T-la/4 fusion protein plasmids. Then, the recombinant plasmids were transformed into Escherichia coli BL21 (DE3) and induced with isopropyl-β-D-thiogalactopyranoside (IPTG). The optimal expression conditions of the two fusion proteins were identified and then analyzed by polyacrylamide gel electrophoresis (SDS-PAGE) and western blotting, respectively. The results showed that the pGEX-4T-la fusion protein was maximally expressed at 30 °C after 12 hours of induction with 1.0 mM IPTG. The pGEX-4T-la/4 fusion protein was maximally expressed at 20 °C after 8 hours of induction with 0.5 mM IPTG. Western blot analysis showed that the two fusion proteins specificity reacted with the anti-nsPla and anti-GST monoclonal antibodies, respectively. This study successfully obtained the HAstV non-structural protein, nsP1a, and its C-terminal protein nsP1a/4 protein using an E. coli system. This novel study lays the foundation for future research into the pathogenic mechanisms of human astrovirus and the functions of its non-structural protein.

Objective To study the clinical effects and side effects of recombinant human vascular endostatin combined with pemetrexed and cisplatin in the treatment of advanced lung adenocarcinoma. Methods Forty-seven patients with advanced lung adenocarcinoma were recruited from Jan 2011 to Jan 2013,and they were randomly divided into experimental group and control group. The experimental group (n =24) was added with recombinant human vascular endostatin based on pemetrexed and cisplatin, whereas the control group(n =23) was administered with pemetrexed and cisplatin only. The objective response rate (ORR),disease control rate (DCR),progressive disease (PD) rate,progression-free survival (PFS),overall survival (OS) and the side effects of 2 groups were evaluated. Results In the experimental group,ORR,DCR,PD rate,PFS and OS were 41.7 % (10/24),79.2 % (19/24),20.8 % (5/24),8.0 months and 12.5 months respectively, while those of control group were 34.7 % (8/23),47.8 % (11/23),52.2 % (12/23),6.2 months and 10.0 months.DCR,PD rate and PFS of experimental group had significant differences compared with control group (P < 0.05).OS of experimental group had no significant difference compared with control group (P > 0.05).The side effects of 2 groups were mainly hematologic toxicities, digestive reactions and fatigue, and the incidence rates were not significantly different between 2 groups (P > 0.05).Conclusions Recombinant human vascular endostatin combined with pemetrexed and cisplatin in treatment of patients with advanced lung adenocarcinoma improves the DCR,decreases the PD rate, prolongs the PFS.There is an increasing trend in the OS of experimental group, and with tolerable side effects.

Journal of Biomedical Engineering 2010;27(3):490-494
In order to deliver power to the capsule endoscope, whose position and orientation are always changing when traveling along the alimentary tract, a wireless power transmission system based on electromagnetic coupling was proposed. The system is composed of Helmholtz transmitting coil and three-dimensional receiving coil. Helmholtz coil outside the body generates a uniform magnetic field covering the whole alimentary tract; three-dimensional coil inside retrieves stable power regardless of its position and orientation. The transmitter and receiver were designed and implemented, and the experiments validated the feasibility of the system. The results show that at least 320 mW of usable power can be transmitted to capsule endoscope when its position and orientation are changing at random and the transmitting power is 25W.
