1.VALUE OF SERUM ANTI HP-IGG DETECTION IN DIAGNOSIS OF PEPTIC ULCER

Objective To provide the convenient and efficient screening program for the early detection and diagnosis of peptic ulcer by evaluatingthe clinical value of serum anti HP -IgG detection in the diagnosis of peptic ul-cer in health examination .Methods 116 participants underwent serum anti HP -IgG detection and gastroscopy ex-amination, taking the pathological biopsy results of gastroscopy examination as the gold standard .Their consistency and the diagnostic value of serum anti HP -IgG detection were compared by the Kappa consistency test .Question-naires were filled by participants after the examinations to evaluate the comfort level and conveniences during the ex -aminations as well as their overall satisfaction , and to evaluate the value of HP -IgG detection .Results 87 partici-pants were positive in serum anti HP -IgG detection , accounting for 75%, while 29 participants were negative , ac-counting for 25%.According to the results of gastroscopy examination , 29 participants were diagnosed with duodenal ulcer (25%), and 25 participants with gastric ulcer (21.55%), and 34 participants with hybrid ulcer (29.31%), and 28 participants without peptic ulcer (24.14%).Based on the results of serum anti HP -IgG detection and gas-troscopy examination, Kappa=0.744 (p=0.073) was found by the Kappa consistency test , and the consistency was good.After the calculation , the sensitivity ( the true positive rate ) was 94.25%, and the specificity ( the true negative rate) was 79.31%.The anxiety, comfort level, conveniences and patients'satisfaction of anti HP-IgG group were bet-ter than those of control group (p<0.05).Conclusion Serum anti HP-IgG detection is worthy of the clinical promo-tion as a screening program for the early detection and early diagnosis of peptic ulcer , because it boasts of high sensitivi-ty, a good consistency in gastroscopy examination ,less anxiety , convenience and satisfaction for patients .
2.Telemetric Localization of Detecting Capsule in Gastrointestinal Tract

Objective To develop a method for localizing a detecting capsule in the gastrointestinal tract.Methods A three-axis magnetic sensor was sealed in the detecting capsule.Three flat coils were fixed on patients' back.The coils were excited one by one to generate electromagnetic fields.The position and the orientation of the capsule could be determined by telemetering the signals from the magnetic sensor incorporated in the capsule.Results The telemetric localization system worked successfully.A localization model describing the relationship between the magnetic field strength and the position and orientation of the capsule,was correctly established basing on the principle of magnetic dipoles,and neural network algorithm was employed to solve a non-linear equation set.Conclusion The results showed that the localization method is feasible and the precision is higher than existing methods.The localization device can perform consecutive tracking and can be made into a portable type.After putting into practice,the device can also be used in food selection for astronauts by monitoring their gastrointestinal parameters in space.
3.Proteomics analysis of Sut melanocytes in response to xCT deficiency

Objective To investigate the mechanism of Sut melanocytes growth inhibition in response to xCT -deficiency. Methods TTotal proteins were extracted from xCT-deficient Sut melanocytes and wild melanocytes, respectively, and were separated by 2-dimensional electrophoresis. Altered expression profile of proteins of Sut melanocytes was analyzed by PDQuest software and compared with that of wild melanocytes.Proteins with significant change were chosen to be identified by mass spectrometry and database query.Results Twenty proteins in Sut melanocytes altered significantly compared with wild melanocytes. Ten of the proteins were up-regulated, while the other tens were down-regulated. Four proteins from both up-regulated and down-regulated were identified respectively: up-regulated proteins were Tubulin alpha-1b, S100-A6,Nucleoside, S-formylglutathione, and down-regulated proteins were Calumenin,NDRG1 ,DPYSL2, 14kDa unknown protein. Conclusion The identification of the xCT-deficiency related proteins may provide supporting evidence for the mechanism research of Sut melanocytes' growth inhibition caused by xCT-deficiency.
4.Detection of hepatitis B virus YMDD variants using a matrix-assisted laser desorption time of flight mass spectrometric analysis

Objective To investigate the value of detecting hepatitis B virus (HBV) YMDD variants by matrix-assisted laser de-sorption time of flight mass spectrometry (MALDI-TOF MS). Methods The assay is based on polymerase chain reaction (PCR) amplifica-tion and mass measurement of oligonucleotides containing sites of mutation of the YMDD motif. Result The MALDI-TOF MS-based genoty-ping assay was sufficiently sensitive to detect as few as 100 copies of HBV genome per milliliter of serum, and this method had superior spe-cificity for determining mixtures of wild-type and variant viruses. When sera of 40 patients were analyzed, the MALDI-TOF MS-based assay correctly identified known viral variants and additional viral quasi-species not detected by previous methods, as well as their'relative abun-dance. Conclusion The sensitivity, specificity and amenability to high-throughput analysis make MALDI-TOF MS-based assay suitable for mass screening of HBV infected patients who are receiving lamivudine.
5.STUDY ON PHYSIOLOGICAL AND BIOCHEMICAL PROPERTIES OFα-ACETOLACTATE DECAROBOXYLASES FROM DIFFERENT MICROBES

The enzyme activity of α-Acetolactate Decaroboxylases (ALDC)from different microbes was studied, the results demonstrated that it was quite different among them. There were diversities of their enzyme reaction velocities. It was clear that the enzyme activity was affected by the pH of the enzyme reaction system, for example, the optimum pH of ALDC from Lactococcus lactis was 6.6, while for Aerobacter Aerogenes it was 5.8. Addition leucine,valine and isoleucine into enzyme reaction system obviously affected the enzyme activity of ALDC from different microbes.
6.Comparison of different analyzing methods for detection of free amino acids in plasma

Objective To evaluate the accuracy of different methods in detecting free amino acids in plasma.Method 40 blood samples from healthy volunteers were analyzed by an automatic amino acid analyzer ( Li+system) and the results compared with previous reports using other analyzers.Results The results ob-tained by this analyzer for major amino acids [asparagine (Asn), glutamic acid (Glu), glutamine (Gln), valine (Val), tyrosine (Tyr), phenylalanine (Phe), ornithine (Orn), arginine (Arg)] were similar with those previously reported using other amino acid analyzers ( all P>0.05 ) and liquid chromatography tandem-mass spectrometry (LC-MS/MS) (all P>0.05).Except for Glu and Tyr, the results for major amino acids showed large deviation compared to the results from high performance liquid chromatography ( HPLC) ( all P<0.05).Conclusion The amino acid analyzer (Li+) and LC-MS/MS (iTRAQ kit) could accurately detect free amino acids in plasma.
7.Establishing Physiological Body Fluid Method to Evaluate the Normal Value of Plasmic Free Amino Acid in Normal Healthy Volunteers

Objective To analyze the plasma amino acid levels and evaluate the free amino acids value of normal healthy volun-teers in plasma by establishing physiological fluid method.Methods ①Establishment of methodology:including repeatabili-ty of blood sample,recovery rate,linearity,and stability;②The free amino acid values in plasma in 40 normal healthy volun-teers were analyzed.Results With the standard solution of different concentrations (2,4 and 8 nmol)analyzed and linear re-gression of amino acid calculated,found that R2 of all the amino acids were within the range from 1 to 0.9 9 9.The repeatabili-ty with in batch and between batches of different amino acid in blood samples were respectively 0.30~2.90 (rsd)and 0.40~5.16(rsd);recovery rates were 93.1%~107.4%.Then evaluated the normal values of plasma amino acid in 40 healthy volunteers (male vs female:1 vs 1 ),and compared the results with that measured by other instruments.Conclusion With lithium citrate buffer system to analyze the free amino acid in blood samples,it displayed good reproducibility,recovery rate, linearity and stability.Comparison of different instruments measuring the free amino acids in plasma of normal healthy vol-unteers showed good agreement,confirming that the method of physiological fluid analysis is reliable.
8.Study on the relationship between the neovascularization of carotid atherosclerotic plaques,plasma myeioperoxidase(MPO)concentration and cerebral infarction

Objective To study the relationship between the neovascula rization of carotid atherosclerotic plaques with real-time contrast-enhanced ultrasonography(CEUS),plasma myeloperoxidase (MPO)concentration and cerebral infarction.Methods Seventy-three cases with eighty-nine carotid atherosclerotic plaques were examined with real-time CEUS.According to the imaging features,the seventythree cases were divided into three groups:soft plaques group,hard plaques group and mixed plaques group.The differences of the incidence rate of cerebral infarction events,plasma MPO concentration and contrast media persistent enhancement time with real-time CEUS in plaques among the three groups were analyzed.The cerebral infarction events were diagnosed by CT or MRI routinely.Results The incidence rate of cerebral infarction events,plasma MPO concentration and contrast media persistent enhancement time of soft plaques group were significantly higher than that in hard plaques group and mixed plaques group.The risk of cerebral infarction was higher in the patients with elevated level of MPO.It showed positive correlation between the contrast media persistent enhancement time and plasma MPO concentration,and the correlation coefficient was 0.838. Conclusions The type of the plaque is correlated with the neovascularization of plaques and plasma MPO concentration.The study of relationships among them can help to evaluate the plaques,stability thoroughly and guide the clinician to prevent the incidence of cerebral infarction events.
9.The effect of operative modality on the prognosis of gastric carcinoma patients suffering from different types of intra-abdominal metastasis

Objective To study the effect of operative modalities on the prognosis of gastric carcinoma patients suffering from various types of intra-abdominal metastasis. Methods In this study, 150 cases of gastric cancer with abdominal metastasis screened from data base, were divided by the location into peritoneal dissemination group (n=106), hepatic metastasis group (n=19) and both peritoneal dissemination and hepatic metastasis group (n=25). The surgical interventions include total resection, palliative resection and no resection. The clinicopathological parameters and prognosis were analyzed. Results The total tumor resection rate in peritoneal dissemination group (48.1%) and hepatic metastasis group (63.2%) was significantly higher than that in both peritoneal and hepatic metastasis group (20.0%), with the mean survival time (months) being 31.1, 12.8, 9.7 respectively (P<0.05). In peritoneal group, the mean survival (months) for tumor total resection, palliative and no resection subgroup was 46.3,1.7,4.8 respectively (P<0.05), whereas in hepatic metastasis group, the mean survival (months) was 17.2,4.0,5.4 respectively (P<0.05), in both peritoneal and hepatic metastasis group, the mean survival (months) was 11.2,8.9,5.0 respectively (P>0.05). Lymph nodes resection significantly prolonged survival in peritoneal dissemination group. Conclusions The prognosis of gastric cancer with peritoneal dissemination is better than with hepatic metastasis. Tumor total resection and lymph node dissection significantly improve the survival of patients suffering from peritoneal or hepatic dissemination.
10.Value of contrast-enhanced ultrasonography on grading extracranial carotid stenosis

Objective To assess value of contrast enhanced ultrasonography(CEUS)in diagnosis of extracranial carotid stenosis.Methods Baseline ultrasonography and CEUS were performed in 108 patients with extracranial carotid stenosis.The location and degree of stenosis were judged and compared with the results of digital subtraction angiography(DSA).Results Of 108 patients,DSA diagnosed 81 vessels in 62 cases of mildintermediate stenosis which consisted of 33 common carotid arteries(CCAs),43 internal carotid arteries(ICAs)and 5 external carotid arteries(ECAS),51 vessels in 39 cases Of severe stenosis(46 ICAs and 5 CCAs)and 7vessels in 7 cases of complete occlusion(7 ICAs).The surface of intima and the interface of media-adventitia were depicted clearly by CEUS.The consistent rates in diagnosis on mild-intermediate stenosis,severe stenosis and complete obstruction by baseline ultrasonography and CEUS were 76.5%and 9 1.4%(P<0.05).60.8%and94.1%(P<0.001),28.6%and 100%(P<0.05)respectively when judged by DSA as the golden standard.CEUS and DSA were correlated significantly on calculating the stenosis rate(r=0.975,P<0.001).Conclusions CEUS can assess the degree of extracranial carotid stenosis accurately and non-invasively which is superior to baseline ultrasonography and correlated significantly with DSA.
