1.Variation of blood biochemical indices in liver cirrhosis rats after adipose-derived mesenchymal stem cell transplantation

BACKGROUND:Stem cel transplantation is a promising treatment of advanced liver disease, and adipose-derived mesenchymal stem cel s have become another kind of popular cel s fol owing bone marrow mesenchymal stem cel s.
OBJECTIVE:To investigate the influence of adipose-derived mesenchymal stem cel transplantation on blood biochemical indices of liver cirrhosis rats.
METHODS:Sixty rats were equal y randomized into normal control, model and cel transplantation groups. Model rats of liver cirrhosis were made in the latter two groups through intragastric administration of carbon tetrachloride. One week after successful modeling, rats were given intraperitoneal injection of adipose-derived mesenchymal stem cel suspension in the cel transplantation group, and given normal saline in the other two groups.
RESULTS AND CONCLUSION:Compared with the normal control group, the model group showed a significant increase in the levels of alanine aminotransferase, aspartate aminotransferase, total bilirubin, total protein in liver tissues, serum level of malondialdehyde, 15-minute indocyanine green retention rate and degree of hepatic fibrosis, and a significant decrease in serum albumin level, serum albumin/globulin, levels of glutathione peroxidase and cyclic guanosine monophosphate in liver tissues. On the contrary, these indicators were al improved in the cel transplantation group compared with the model group.
Moreover, CM-Dil-positive cel s were visible in the liver tissue of rats undergoing adipose-derived mesenchymal stem cel transplantation. Al these findings indicate that adipose-derived mesenchymal stem cel transplantation can reduce liver cirrhosis in rats by acting on blood biochemistry levels.

3.Human umbilical cord blood mononuclear cell transplantation for treatment of radioactive premature ovarian failure in nude mice

BACKGROUND:Stem cel transplantation, in recent years, has become a preferred treatment for premature ovarian failure. Umbilical cord blood mononuclear cel s containing a large number of mesenchymal stem cel s, immature stem/progenitor cel s, and endothelial progenitor cel s can be used as an important source of stem cel transplantation.
OBJECTIVE:To study the therapeutic efficacy of human umbilical cord blood mononuclear cel transplantation on radioactive premature ovarian failure in nude mice.
METHODS:120 female BALB/C nude mice were randomly divided into four groups:blank control group without any intervention;model group, intravenous transplantation group, and in situ transplantation group exposed to 60Coγrays, 0.5 Gry per day, for 30 days. After 30 days, premature ovarian failure models were made in the latter three groups. Then, nude mice in the latter three groups were given bilateral ovary injection of 10 μL DMEM, tail vein injection of 10 μL human umbilical cord blood mononuclear cel s (1×1013/L), and bilateral ovary injection of 10 μL human umbilical cord blood mononuclear cel s (1×1013/L), respectively. Thirty days after cel transplantation, serum levels of estradiol, fol icle hormone, luteinizing hormone, inhibin B and vascular endothelial growth factor were detected, cel apoptosis in the ovary tissue and cel survival were observed pathological y.
RESULTS AND CONCLUSION:After modeling, the serum levels of estradiol, inhibin B and vascular endothelial growth factor were significantly reduced (P<0.01), while fol icle hormone and luteinizing hormone levels increased (P<0.01). After transplantation, these indexes were al improved in the in situ transplantation group (P<0.01), and reduced fol icle hormone and luteinizing hormone levels were visible in the intravenous transplantation group (P<0.05, P<0.01). Compared with the model group, in situ transplantation and intravenous injection of human umbilical cord blood mononuclear cel s could effectively reduce cel apoptosis in the ovary tissue (P<0.05), and transplanted cel s were able to survive in the ovary of nude mice. Al these findings show human umbilical cord blood mononuclear cel transplantation do have curative effects on premature ovarian failure in nude mice through the inhibition of apoptosis and the regulation of hormone secretion.
4.Experimental study on the toxicity from PMNLs during ischemia reperfusion cerebral injury

Objective To study the change of activity and toxicity of superoxide (O - 2 ) produced in polymorphonuclear leukocytes(PMNLs) during ischemia reperfusion cerebral injury.Methods The rats were administrated by both PMA (an activator of single transduction of O - 2 produced in alkaline phosphatase(ALPase) positive granules of PMNLs) and the inhibitor BCA respectively; the model of middle cerebral artery (MCA) occlusion was made by suture cleat mothod,the activity changes of both myeloperoxidease (MPO) and O - 2 were measured at 6, 12, 24, 48, 72 and 168h reperfusion following ischemia 1h, and the pathological ultrastructural changes were observed. Results The MPO activity of both PAM group and BCA group reached the peaks at 24h after reperfusion; however,there were no remarkable differences in MPO activity between these two groups in the same time point. The O - 2 activity in the PAM group was significantly higher than those in the BCA group. The O - 2 activity reached the peak at 72h of ischemia reperfusion. In the same experimental time point, the pathological changes of the ultrastructure in ischemic reperfusive injury brain of the PAM group were much more serious than the those of the BCA group, which showed obviously the neurons edema, the abnormal structures of nerve felt and synapse in the ischemia reperfusion injured brain.Conclusion The increase of brain O - 2 activity from PMNLs during cerebral ischemia reperfusion injury was direct ratio to the degree of cerebral injury. BCA might depress the activity and the toxicity of the O - 2.
5.Clinical therapeutic effect of three-dimensional conformal radiation therapy combined with chemotherapy of paclitaxel on brain metastases from breast cancer in the elderly

Objective To evaluate the efficacy and adverse reaction of three-dimensional conformal radiation therapy (3D-CRT) combined with chemotherapy of paclitaxel in treatment of brain metastases from breast cancer in the elderly.Methods The 50 patients were randomly divided into observation group (n=26,radiation combined with chemotherapy) and control group (n= 24,simple radiation).In the early stage,both groups received common two-dimesional conformal radiation therapy.The total dose (DT) of whole brain irradiation was 30-40 Gy.In the later stage,the reduced field for the local lesion of brain metastases would be altered to 3D-CRT for the post period with 2 Gy 5 times a week.DT was added from 10-24 Gy up to total DT of 50-64 Gy.The patients were given paclitaxel 65-85 mg/m2 by intravenous drip at 1st and 8th day with synchronization of 2-4 weeks,having paclitaxel chemotherapy of 2-4 circle,28 days a circle.After 2 month treatment,the efficacy and adverse effects of the two groups were observed.follow up for 2 years,the long-term efficacy and survival rate were evaluated.Results The effective rate was 76.9% in observation group and 45.8% in control group,respectively (x2 =5.120,P<0.05) and the KPS score was 80.8% and 54.2%,respectively.The quality of life was improved in observation group versus control group (x2 =4.059,P<0.05).Compared with control group,hypoleukemia was significant in observation group (P<0.05).The complications such as nausea and vomiting,hepatic dysfunction were more in observation group than in control group,but there was no statistical significance between two groups.There was statistic ally significant difference in 2-year survival rate between two groups (x2= 4.7260,P<0.05).Conclusions The 3D-CRT combined with paclitaxel chemotherapy is a prefered choice for locally advanced brain metastases from breast cancer.More side effects and adverse reaction are observed in observation group.However,all the patients could tolerate them.It is worthy of popularization and application.
6.Genetic variants in the promoter of cyclooxygenase 2 interacting with Hp infection and the risk of esophageal cancer

Objective To evaluate the association of COX2 genetic variants with the risk of esophageal cancer and the interaction of COX2 genetic variants with Hp infection. Methods A total of 119 patients with esophageal cancer and 238 frequency-matched controls were genotyped by polymerase chain reaction-restriction fragment length polymorphism method. Odds ratios (OR) and 95% confidence intervals ( CI) were estimated by logistic regression. Results Case-control analysis showed an increased risk of developing esophageal cancer for 1195 GA(OR =2.69,95% CI= 1. 46-5. 14) and 1195AA ( OR = 2. 30,95% CI = 1.23-4. 89) genotype carriers,respectively, compared with non 1195 GG carriers. When stratified by Hp status, the significantly increased risk of esophageal cancer was found among Hp carrier with OR (95%CI) =2.74 (1.35-5.96) ,but not among Hp non-carriers. Conclusion Genetic polymorphism in COX2 promoter region may play an important role in esophageal cancer by Hp infection.
7.Effect of Jinkoubao on rabbit oral ulcer model

Objective To explore the therapeutic effect of Jinkoubao on oral ulcer in rabbits and its action mechanism.Methods Among 60 SPF NewZealand rabbits,6 cases were randomly selected for the ulcer model identification,and the rest was randomly and equally divided into 3 groups:the control group (NC group),normal saline drug film group (NS group) and Jinkoubao film group (JK group).The rabbit model of oral ulcer was constructed by applying 40 % glacial acetic acid to burn rabbit oral mucosa for duplicating the oral ulcer model.of the change situation of oral ulcer was observed on the same day for constructing model,on 3,5,7 d after medication.The EGF level in oral mucosal tissue was detected by using RT-PCR and the local histopatho1ogical changes in oral ulcer was observed by using HE staining method.Results Compared with the NS group,the ulcer area on 3,5,7 d after medication in the JK group was significantly deceased (P<0.01).Compared with the NS group,the EGF level in oral buccal mucosal tissue in the NS group and JK group was markedly increased (P<0.01).But the EGF level increase in the JK group was faster than that in the NS group,the difference was statistically significant(P<0.01).The HE staining section of rabbit oral ulcer on 3,5,7 d after medication showed that the inflammatory cells decrease in the JK group was more obvious than that in the NS group,fibroblasts proliferation was obvious and epithelial hyperplasia was good.Conclusion Jinkoubao could greatly improve the symptoms of oral ulcer in rabbits and promotes its healing,which is possible to strengthen the repair capacity of oral ulcer by regulating the EGF level.
8.Observation of the clinical curative effect of flupentixol/melitracen combined with mosapride citrate in the treatment of functional dyspepsia

Objective To investigate the clinical curative effect of flupentixol/melitracen (deanxit) combined with mosapride citrate in the treatment of functional dyspepsia.Methods The clinical data of 240 patients with functional dyspepsia were collected,According to different treatment method,they were divided into two groups,120 patients of the control group were treated with mosapride citrate combined with omeprazole,120 patients of the observation group were treated with deanxit combined with mosapride citrate.One course of treatment was 4 weeks.After one course of treatment,the clinical curative effects and adverse reactions of the two groups were evaluated.Results After treatment,the abdominal distension,abdominal pain,early satiety,belching and other symptoms scores of the two groups were significantly lower compared with before treatment,the differences were statistically significant (all P < 0.05).After treatment,the abdominal distension,abdominal pain,early satiety,belching and other symptoms scores of the observation group were significantly lower than those in the control group,the differences were statistically significant(all P < 0.05).The clinical total effective rate of the observation group was 90.8%,which was significantly higher than 67.5 % of the control group,the difference was statistically significant (x2 =8.432,P < 0.05).Before and after treatment,the liver and kidney function and blood in the urine routine examination in the two groups had no significant changes.The incidence rate of adverse reactions of the observation group was 11.67%,which of the control group was 15.83 %,there was no statistically significant difference (x2 =0.911,P > 0.05).Conclusion Deanxit combined with mosapride citrate in the treatment of functional dyspepsia is one of the important method,and its clinical effect is satisfactory,relatively safe,it is worthy of popularization and application.
9.Effect of PD-WEBB training on balance impairment and falls in people with Parkinson's disease.

OBJECTIVE:
To determine the effect of Parkinson's disease-weight bearing exercise for better balance (PD-WEBB) exercise on balance impairment and falls in people with Parkinson's disease (PD).
METHODS:
A single-blind, randomized controlled clinical trial was conducted. The falls efficacy scale score, unified Parkinson disease rating scale (UPDRS) score and Mini-BESTest score were measured and compared between a PD-WEBB group and a control group.
RESULTS:
The falls efficacy scale score, UPDRS-2 score, UPDRS-3 score and Mini-BESTest score were improved in the PD-WEBB group compared with the control group (P<0.05), with no significant change in UPDRS-1 score between the two groups.
CONCLUSION
PD-WEBB training can significantly improve the balance impairment and quality of life to prevent falls. PD-WEBB training is suitable for PD patients in China, and is a reasonable, effective and sustainable training of family and community assessment model.

10.Role of astrocytic NLRP3 in lateral hypothalamus in anxiety-like behaviors after hemorrhagic shock and resuscitation in mice

Objective:To evaluate the role of astrocytic NOD-like receptor protein 3 (NLRP3) in the lateral hypothalamus (LHA) in anxiety-like behaviors after hemorrhagic shock and resuscitation in mice.Methods:Forty-eight clean-grade male C57BL/6 mice, aged 10 weeks, weighing 25-30 g, were divided into 4 groups ( n=12 each) using a random number table method: sham operation group (group C), hemorrhagic shock and resuscitation group (group H), hemorrhagic shock and resuscitation + adeno-associated virus group (group HI), and hemorrhagic shock and resuscitation + control virus group (group HIV). The model of hemorrhagic shock and resuscitation was developed by bleeding and re-transfusion through the femoral vein in H, HI and HIV groups. At 21 days before developing the model, AAV-GfaABC1D-EGFP-Cre was injected into bilateral LHA in group HI, and AAV-GfaABC1D-EGFP was administered as a control in group HIV. Anxiety-like behaviors were evaluated by EPM-maze and bead-burying tests at 14 days after resuscitation. Mice were immediately sacrificed at the end of behavioral tests, and LHA-containing brain tissues were obtained for determination of co-localization of NLRP3 with glial fibrillary acidic protein (GFAP), the fluorescence intensity of Wisteria floribunda agglutinin was measured using immunofluorescent staining to reflect the expression of extracellular matrix in the LHA, and the percentage of cleaved caspase-1/GFAP and IL-18/GFAP positive cells in total cells was calculated. Results:Compared with group C, the number of buried beads and percentage of time of staying at the open arm were significantly decreased, the expression of extracellular matrix in the LHA was down-regulated, and the percentage of cleaved caspase-1/GFAP and IL-18/GFAP positive cells was increased in H, HI and HIV groups, and the co-localization coefficient of NLRP3 and GFAP was significantly decreased in group HI ( P<0.01). Compared with group H, the number of buried particles and percentage of time of staying at the open arm were significantly decreased, the expression of extracellular matrix in the LHA was up-regulated, the co-location coefficient of NLRP3 and GFAP was decreased, the percentage of cleaved caspase-1/GFAP and IL-18/GFAP positive cells was decreased ( P<0.01), and no significant change was found in the parameters mentioned above in group HIV ( P>0.05). Conclusions:Anxiety-like behaviors after hemorrhagic shock and resuscitation is associated with astrocytic NLRP3-induced pyroptosis in the LHA and reduction of extracellular matrix in mice.
