1.Effective components against HIV-1 replicative enzymes isolated from plants.

Plant active components characterized of many different structures and activities on multiple targets, have made them to be the important sources of inhibitors on HIV-1. For finding leading compounds with new structure against HIV-1, three key HIV-1 replicative enzymes (reverse transcriptase, protease and integrase) were used as screening models. The in vitro activities of 45 plant derived components isolated from Schisandraceae, Rutaceae and Ranunculaceae were reported. Within twelve triterpene components isolated, eight compounds were found to inhibit HIV-1 protease, in these eight active compounds, kadsuranic acid A (7) and nigranoic acid (8), inhibited both HIV-1 protease and integrase; Among fifteen lignans, meso-dihydroguaiaretic acid (15) and kadsurarin (16) were active on HIV-1 reverse transcriptase, and 4, 4-di(4-hydroxy-3-methoxyphenly)-2, 3-dimethylbutanol (13) active on HIV-1 integrase. All of the six alkaloids, seven flavones, and five others compounds were not active or only with low activities against HIV-1 replicative enzymes. Further studies of the triterpene components showing strong inhibitory activities on HIV-1 were warranted.
2.HTLV-1 bZIP Factor (HBZ): Roles in HTLV-1 Oncogenesis.

Human T-cell leukemia virus type 1 (HTLV-1) is a retrovirus demonstrated to be associated with human disease. Infection by the HTLV-1 can cause T-cell leukemia (ATL) in adults. HTLV-1 bZIP factor (HBZ) is a viral protein encoded by the minus strand of the HTLV-1 provirus. Among the regulatory and accessory genes of HTLV-1, HBZ is the only gene that remains intact and which is expressed consistently in all patients with ATL. Moreover, HBZ has a critical role in the leukemogenesis of ATL. Here, we review the function of HBZ in the oncogenesis of HTLV-1 and its molecular mechanism of action.

This paper describes the anti-inflammatory, antipyretic and analgesic effects of Sudoxicam ( Sud) synthesized by Shenyang Pharmaceutic College.Sud is found to possess significant anti-inflammatory, antipyretic and analgesic quality in such animal models as carrayeenan-induced paw oedema ( ED50=5.26mg/kg ) , tylolinduced ear oedema in mice, adjuvant-induced arthritis in rats, yeast-induced fever in rats, acetic acid-induced arithing ( ED50= 5.0mg/kg ) and hot plate-induced pain reaction in mice.
4.STUDIES ON THE TOXICITY OF SUDOXICAM

The acute iv and ig LD50 of sudoxicam synthesized by Shanyan Ph-armaceutic College was calculated to be 135?10. 7 and 320 ? 96 mg/kg in mice. In polyarthritic rats, Sud at 2 and 4 mg/kg/d for 28 days did not induce changes in serum AKP, SGPT, BUN the morphal-ogic characteristics of the gastrointestinal tract, liver and kidney. In 3 dogs orally given Sud 16 mg/kg/d for 5 consecutive days, 2/3 died at days 10 to 13. In other 3 dogs given Sud 16 mg/kg/d every other day, plasma concentrations of Sud were determined by ultra violet spectrophotometry. It was found that on the days 10 to 20 , the concentration time curves had attained a intoxicant steady state with plasma sudoxicam concentrations as high as 44. 9 ? 73. 9?g/ml ,and all of them died after 17~25 days. Their bleeding time and coagu- lation time were prolonged to 2 or 3 times of the control and serum BUN was increased. In one dog autopsy revealed the interstitial inflammation of the kidney and small local necrosis of the live, but not in other dogs.Gastric ulceration in rats can be produced by the oral administration of verious doses of Sud. The UD50 of Sud was 15.47mg/kg. It was found that the gastric ulceration in rats is related to the plasma levels
5.Synergistic inhibitory effect of static magnetic field and antitumor drugs on Hepa1-6 cells.

Chemotherapy as a routine method for clinical treatment of cancer has disadvantages such as significant toxicity and strong resistance. In order to improve the efficacy of the drugs and reduce the by-effects, we tried to combine static magnetic field (SMF) with cisplatin or adriamycin. The growth of Hepa1-6 cells treated with the static magnetic field (SMF) combined with cisplatin or adriamycin was significantly inhibited, as detected with MTT (3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide) test. Combined treatment group cells underwent significant morphological changes as observed by HE (Hematoxylin and eosin) staining under optical microscope. Cell cycle analysis indicated that SMF increased the ratio of cells arrested in G2/M phase caused by cisplatin, and when treated with SMF combined with adriamycin, cells were almost arrested in G1 and G2/M phase. SCGE test showed that SMF can enhance the ability of cisplatin or adriamycin to promote cell DNA damage. Atomic force microscope observation found that the combination of antitumor drugs and magnetic field treatment induced larger and deeper holes on the cell membrane, and surface structure damage is serious. The combination of antitumor drugs and magnetic field technology effectively inhibits the growth of tumor cells, and reduces drug doses. The results implicate this method as potential cancer therapy.

6.Study on perioperative nursing quality evaluation index system of benign prostatic hyperplasia

Objective To establish the benign prostatic hyperplasia evaluation index system for nursing quality. Methods Indicators framework for perioperative nursing quality evaluation index system was built up by the theoretical research, clinical research, expert meetings approach firstly, then the Delphi method was used to screen and verify the indicators. Results Two rounds of consultation questionnaires were collected with rate of 86.0%, 95.3% respectively, the first round of consultation had the retention of 13 indicators, removed 4 indicators of those. After the second round, the average score of important target was no less than 8.0, and more than half of indicators had a full mark, coefficient of variation targets no higher than 0.20 were up to 100%. Coefficient of concordance of two rounds were 0.241 and 0.433 respectively. ConclusionsAfter two rounds of consultation, there were 13 quality indicators left and experts had well authoritativeness and representative.
7.Experimental Detection Study on Cerebral Hemorrhage in Rabbits Based on Magnetic Induction Phase Shift Spectroscopy Under the Feature Band.

This study was aimed to improve the sensitivity of magnetic induction phase shift detection system for cerebral hemorrhage. In the study, a cerebral hemorrhage model with 13 rabbits was established by injection of autologous blood and the cerebral hemorrhage was detected by utilizing magnetic induction phase shift spectroscopy (MIPSS) detection method under the feature band. Sixty five groups of phase shift spectroscopy data were obtained. According to the characteristics of cerebral hemorrhage phase shift spectroscopy under the feature hand, an effective method, B-F distribution, to diagnose the severity of cerebral hemorrhage was designed. The results showed that using MIPSS detection method under feature band, the phase shift obviously growed with increase of injection volume of autologous blood, and the phase shift induced by a 3-mL injection reached -7.750 3 degrees ± 1.420 4 degrees. B-F distribution could effectively diagnose the severity of cerebral hemorrhage. It can be concluded that the sensitivity of the cerebral hemorrhage magnetic induction detection system is improved by one order of magnitude with the MIPSS detection method under the feature band.

8.Effect of inhibition of TGF-β1 receptors on lymphocytes during infection of Echi-nococcus granulosus

Objective:To investigate the effects of TGF-β1 on T lymphocytes of BALB/c mice infected with Echinococcus granulosus( E.granulosus ) in vitro.Methods: The inhibitor group:the spleen cells of BALB/c mouse were co-cultured with E.granulosus and SB525334.The control group:the spleen cells of BALB/c mouse were co-cultured with E.granulosus and PBS.The blank group:the spleen cells of BALB/c mouse were co-cultured with RPMI-1640 medium and SB525334.The lymphocytes were collected at 48 h post-infection.The T lymphocyte subsets, the number of CD4+CD25+T cells, the number of NK cells, and the expression of NKG2D receptor were detected by flow cytometry.The NK cell activity was determined with the lactate dehydrogenase leakage assay(LDH).Results:The inhibition the TGF-β1 receptors resulted in the increase of in the number of CD4+T cells,the decrease in the number of CD8+T cells,the increase of in the ratio CD4+/CD8+T cells,the decrease of in the number of CD4+CD25+T cells,the increase in the expression of the NKG2D receptors,the increase in the lysis rate of Yac-1 cells by NK cells,and a positive cor-relation between the expression of activity receptor NKG2D and killing activity of NK, which were mediated by E.granulosus.Conclusion: The inhibition of TGF-β1 receptors can enhance the immune response of T lymphocytes against E.granulosus infection in vitro.
9.Reflectance confocal microscopy features of irritant cutaneous reactions to sodium lauryl sulphate in healthy adults

Objective To investigate reflectance confocal microscopy (RCM) features of irritant cutaneous reactions to sodium lauryl sulphate (SLS) in healthy persons aged from 18 to 60 years,to analyze effects of age and gender on cutaneous reactions,and to estimate the value of RCM in objective evaluation of cutaneous reactions.Methods An occlusive patch test was performed on the back of 120 healthy testees with 0.1% and 0.5% SLS solution (0.1% and 0.5% SLS groups) and distilled water (negative control group) for 48 hours.At different time points after the patch removal,clinical evaluation and RCM were performed.Results RCM imaging in the 0.1% and 0.5% SLS groups showed parakeratosis,indistinct structure of the stratum corneum,spongiosis and infiltration of inflammatory cells in the epidermis,and telangiectasia in the papillary dermis.The incidence of RCM features reached the peak until 24 hours after the removal of 0.1% and 0.5% SLS patches,and the incidence of telangiectasia in the dermis was up to 66.7% and 95.0% in the 0.1% and 0.5% SLS groups respectively.At 24 hours after the removal of 0.5% SLS patch,the incidence of spongiosis was significantly lower in the males than in the females (68.9% [42/61] vs.84.7% [50/59],x2 =4.24,P < 0.05).However,the incidence of spongiosis was significantly higher in testees aged 18-40 years than in those aged 41-60 years at 24 hours after the removal of 0.1% SLS patch (53.3%[32/60] vs.35.0%[21/60],x2 =4.09,P < 0.05).For the other RCM features,there were no significant differences in their incidence between different genders or age groups after the removal of 0.1% and 0.5% SLS patches (all P > 0.05).Clinical evaluation showed that after the removal of 0.1% and 0.5% SLS patches,no significant difference in the incidence of irritant cutaneous reactions was observed between the males and the females or between the testees aged 18-40 years and those aged 41-60 years (all P > 0.05).There were good correlations between the clinical evaluation results and RCM features.At 24 hours after the removal of 0.1% SLS patch,the correlation coefficient between spongiosis and clinical evaluation results was up to 0.77,so was that between telangiectasia in the dermis and clinical evaluation results (both P < 0.001).However,at 0.5 hour after the removal of SLS patches,clinical evaluation showed that the positive reaction rates were 2.5% (3/120) and 12.5% (15/120) in the 0.1% and 0.5% SLS groups respectively.In the meantime,there were 17.5 % (21 / 120) and 51.7% (62/120) of testees manifesting more than 2 RCM features in the 0.1% and 0.5% SLS groups respectively,which were more similar to the clinical evaluation results at 24 hours after the removal of SLS patches (34.2% [41/120] and 85.0% [102/120] in the 0.1% and 0.5% SLS groups respectively) compared with the clinical evaluation results at 0.5 hour after the removal of SLS patches.Conclusions Neither gender nor age affects irritant cutaneous reactions to 0.1% and 0.5% SLS.Compared with clinical evaluation,RCM can evaluate cutaneous reactions more objectively and accurately in the early stage of irritant reactions.
10.Experimental study of the effect of GOLPH3 on proliferation, apoptosis and radiosensitivity of OE33 esophageal cancer cell line

Objective To evaluate the effect of GOLPH3 on the proliferation, apoptosis and radiosensitivity of OE33 esophageal cancer cell line. Methods The expression levels of GOLPH3 mRNA and protein in the esophageal cancer cells and normal esophageal epithelial cells were detected by qRT-PCR and Western blot, respectively. The OE33 esophageal cancer cells were transfected with GOLPH3 siRNA and subject to irradiation treatment simultaneously. The cell proliferation was detected by MTT assay. The cell apoptosis was detected by flow cytometry. The radiosensitivity was assessed by cell cloning test. The expression levels of cleaved Caspase-3, Bax and cleaved Caspase-9 protein levels were quantitatively measured by Western blot. Results The expression levels of GOLPH3 mRNA and protein in the esophageal cancer cells were significantly higher than those in the normal esophageal epithelial cells ( both P<0.05) . GOLPH3 siRNA could obviously down-regulate the expression levels of GOLPH3 mRNA and protein in the OE33 esophageal cancer cells. The proliferation activity of esophageal cancer cells was decreased, whereas the apoptosis rate was increased and the expression levels of cleaved Caspase-3, Bax and cleaved Caspase-9 were up-regulated after down-regulating the expression of GOLPH3 or irradiation treatment ( all P<0.05) . After down-regulating the expression of GOLPH3 in the esophageal cancer cells treated with irradiation, the cell proliferation activity was more significantly decreased, whereas the apoptosis rate was elevated and the expression levels of cleaved Caspase-3, Bax and cleaved Caspase-9 were more evidently up-regulated ( all P<0.05) . In the irradiated OE33 esophageal cancer cells after down-regulating the expression of GOLPH3, the radiosensitization ratio of the cells was 1.673. Conclusions GOLPH3 is highly expressed in the esophageal cancer cells. Down-regulating the expression of GOLPH3 can increase the radiosensitivity, induce the apoptosis and inhibit the proliferation of OE33 esophageal cancer cells.
