Chinese Journal of Tissue Engineering Research 2017;21(12):1819-1825
BACKGROUND: Tissue-engineered materials containing osteoblasts have been widely used in the treatment of orthopedic diseases. How to improve hypoxic tolerance and the biological behavior of osteoblasts under hypoxic conditions is an important goal of the research on tissue engineering.OBJECTIVE: To explore the effect of M2 macrophage supernatant and Eucommia flavonoids on the biological behaviorof osteoblasts under hypoxia to pave ways for tissue engineering. METHODS:After MC3T3E1 cells were resuscitated, and M2 macrophages were isolated, induced and cultured, all cells were divided into control, M2 macrophage, Eucommia flavonoids and combination groups. The control group received no intervention. Cells in the M2 macrophage, Eucommia flavonoids and combination groups were exposed to M2 macrophages or/and 100 mg/L Eucommia flavonoids, respectively. RESULTS AND CONCLUSION:The cell viability in the M2 macrophage, Eucommia flavonoids and combination groups was significantly higher than that in the control group. The percentage of cells in the S and G2/M phases in the combination group was significantly higher than that in the control group. The relative mRNA expression levels of Runx2, collagen type Ⅰ and alkaline phosphatase, alkaline phosphatase activity and relative concentration of calcium ions in the combination group were significantly higher than those in the other three groups. To conclude, M2 macrophage supernatant combined with Eucommia flavonoids can promote osteoblast proliferation and differentiation under hypoxic conditions, maintain normal mineralization and osteogenesis of osteoblasts, and improve the tolerance ability of osteoblasts to hypoxia.
2.Advances in pulmonary fibrosis caused by thoracic radiotherapy combined with EGFR-TKIs
Practical Oncology Journal 2015;(2):162-166
Thoracic radiotherapy is an important means of local treatment for non -small cell lung cancer (NSCLC).Epidermal growth factor receptor tyrosine kinase inhibitors (EGFR-TKIs)have the effect of systemic therapy.Studies have shown that NSCLC patients with EGFR exons 19,21 mutation have a synergistic effect in the combination therapy .Radiotherapy activates EGFR signaling pathway ,inducing cell proliferation and DNA damage repair, leading to radiation resistance .Therefore,EGFR-TKIs have the effect in increasing radiosensitivity .Lung injury is one of the most common side effects when the two therapies combined .Studies suggest that radiotherapy combined with EGFR-TKIs may have conflicting functions in the development of pulmonary fibrosis ,the discrep-ancy between these studies may depend on the differences in the experimental systems ,the differences in pulmo-nary fibrosis models,as well as the differences between different species and individuals .Therefore,a more com-plete understanding of the etiology for pulmonary fibrosis is necessary to the development of improved treatments .

Objective To establish a method for TLC identification of Guangdongzizhu in Kanggongyan tablets. Method Developer was chloroform-methyl alcoho-Formic acid (20∶4∶0.8), spraying with the test solution of chloride ferric. Result The same color spots in the TLC graphs of sample existed at the corresponding position compared with the control. Conclusion The method is proved to be sensitive and precise, and can be used for the quality control of Kanggongyan tablets.

Objective To explore the effects of taurine-zinc(TZC) on blood lead content and blood-brain barrier(BBB) permeability of growing rats exposed to lead. Methods Male SD rats aged 3 weeks were randomly divided into four groups(8 in each group):control group, lead exposure group, dimercaptosuccinic acid(DMSA) group and TZC group. The exposure group treated with 0.12% lead acetate(0.2 ml/100 g body weight) by intraperitoneal injection,DMSA group and TZC group were treated with the same dose of lead acetate but were respectively supplemented with 5 mg/ml DMSA(2 ml/100 g body weight) by ig and 0.46 g/L TZC solution through drinking.Three weeks later, blood lead content was measured by atomic absorption spectroscopy(AAS) and changes of BBB permeability were observed using lanthanum nitrate tracer method. Results Compared with the lead exposure group, the blood lead level in TZC group and DMSA group was significantly reduced(P

Objective To study the quality assurance(QA) and quality control(QC) of PACS image display device in the department of radiology equipped with PACS and radiology information system (RIS). Methods Routine maintenance and periodic calibration were performed using photometer and automatic calibration software according to American Association of Physicists in Medicine Task Group 18 (AAPM TG18) test patterns and evaluating criterion. Digital image and communication in medicine (DICOM) gray scale standard display function (GSDF) standard calibration ,maximum luminance and minimum luminance, luminance uniformity, display resolution and geometry distortion adjustment were performed quarterly on BARCO CRT displays. Results All the results were well conformed to the criteria recommended by the AAPM TG18. About 95 percents of radiological images were interpreted on the PACS diagnostic image display. Conclusion The QC program of PACS image display device is an essential to ensure high-quality in digital medical environment.

In recent years, the rapid development of stem cell transforming technology and regeneration pose great re?search value. With maturation of transformation induction, other cell lineages were shown to be able to transform into neu?rons. In addition, astrocytes are widely distributed in the gray and white matter of the central nervous system, whose exces?sive proliferation contribute to glial fibrillary scar formation, which is the key obstacle in recovery of neurological function. Therefore, the study of transforming astrocytes into neurons draw attention from many scientists. Astrocytes transformation not only prevent the formation of glial scar, but also generate new neurons. This article summarized relevant studies that re?port function of astrocytes and its transformation into neurons.

Radiation therapy is one of the main therapeutic means of thoracic malignant tumors .When re-ceived certain radiation ,lung may cause the common complication of chest tumor radiotherapy -Radiation -in-duced Lung Injury ( RILI) ,which restricts the radiation dose for the tumor region and may affect patient outcomes . RILI is closely associated with multiple cytokines ,such as interleukin family, tumor necrosis factor, transforming growth factor, etc.Currently, research on RILI control methods has also made some breakthrough ,this article elab-orates on this individually .

Objective To present the experience of repairing advanced scrotal hypospadias with the combined use of tubed transverse preputial island flap (Duckett) and inversed scrotal raphe flap urethroplasty. Methods 19 cases were operated on with repairing distal urethral tube by Duckett preputial island flap and scrotal raphe flap urethroplasty with suturing together the adjoining ends of the two tubes on an oblique plane. The age of the patients ranged from 2 to 12 years. The length of urethroplasty was 5~9 cm. Results All the cases were followed up for 6 months to 4 years. 17 cases recovered fully on one stage operation. 2 were complicated by urethral fistula. Conclusions The results are satisfactory. The use of a casing tube is reasonable and nontraumatic.

Objective To study the relation of type Ⅱ topoisomerase gene mutation and mechanism of Ureaplasma urealyticum resistant to quinolones. Methods 13 isolates of Uu resistant to 6 quinolones were selected from 184 clinical isolates by using broth dilution method, and their gyrA, gyrB, parC, parE were amplified by PCR.After sequencing, results were compared with the nucleotide sequence of susceptible reference strain. ResultsMICs of resistant Uu isolates were 4 to 32 fold higher than susceptible reference strain counter parts; sequence comparison revealed a C to A change at 87nt of gyrA QRDR led to the substitution of aspartic acid by glutamic acid and a C to T change at 50nt of parC QRDR led to the substitution of serine by leucine, with no amino acid change observed in GyrB and ParE. Conclusion These results suggest that a C to A change at 87nt of gyrA QRDR and a C to T change at 50nt of parC QRDR are associated with quinolone resistance of Uu.
