Objective To investigate the expression level of miR-21, miR-146a and miR-224 and their clinical significances in cervical cancer .Methods miR-21, miR-146a and miR-224 were extracted from the normal cervical tissues and cervical cancer tissues , and their expression levels were quantified by real-time quan-titative polymerase chain reaction analysis .t test was used to analyze the expression difference of the two tissues and examine the associations between miR expression and the clinicopathological characters .The overall survival was analyzed by log-rank test, and survival curves were plotted according to Kaplan-Meier.Results miR-21 ex-pression levels of cervical cancer tissues and normal cervical tissues were 11.02 ±4.83 and 10.36 ±4.79, and miR-146a expression levels were 24.68 ±7.92 and 22.43 ±7.25, and there was no significant difference for both miR-21 and miR-146a expression between groups (P>0.05).miR-224 expression levels were 5.78 ±1.03 and 3.35 ±0.93 in cervical cancer tissues and normal cervical tissues , and the difference had statistical significance (P<0.001).For cervical cancer tissues, the miR-224 levels were significantly higher in tumors less differentia-ted, in advanced FIGO stage and with lymph nodes metastasis (P<0.05).miR-224 levels showed obvious corre-lation with survival time and the median survival time was shorter in patients with higher expression level of miR -224(P <0.05).COX multivariate analysis showed miR-224 expression level was an independent factor in patients'survival and prognosis ( P<0.05 ) .Conclusion miR-224 is independent predictor for aggressive pro-gression and poor prognosis in cervical cancer , and it might serve as a biomarker for predicting clinical outcome of cervical cancer patients .

Objective To study the effect of ganoderma lucidum polysaccharides on T cell subsets and AQP1, AQP3 expression of bladder cancer T24 cell line bearing mice.Methods 60 BALB/C nude mice were selection as experimental animals, bladder tumor bearing animal models were made by bladder cancer T24 cells subcutaneous injection, and were randomly divided into control group, cisplatin group, ganoderma lucidum polysaccharides and cisplatin group, cisplatin group given 25 mg/kgcisplatin intraperitoneal injection, cisplatin combined ganoderma lucidum polysaccharide group given 200 mg/kg ganoderma lucidum polysaccharide lavage, 25 mg/kg cisplatin intraperitoneal injection, the control group given quite a volume normal saline lavage.Then tumor volume and weight, peripheral blood T cell subsets and AQP1, AQP3, Caspase-3, Bax mRNA content in tumor tissue were determined.Results Cisplatin group, cisplatin combined ganoderma lucidum polysaccharide tumor volume and mass were significantly lower than the control group (P<0.05), cisplatin and ganoderma lucidum polysaccharide tumor volume and mass were significantly lower than that cisplatin group ( P<0.05 ); Cisplatin group, cisplatin combined ganoderma lucidum polysaccharides group CD4 + T cells, CD8 +T cells , CD4 +/CD8 + were significantly higher than that of control group (P<0.05), cisplatin combined ganoderma lucidum polysaccharide group CD4 +T cells, CD4 +/CD8 +were significantly higher than that of cisplatin group (P<0.05); Cisplatin group, cisplatin combined ganoderma lucidum polysaccharide group mices tumor tissues Bax, Caspase 3 mRNA content were significantly lower than the control group (P<0.05), cisplatin combined ganoderma lucidum polysaccharide group Bax,caspase 3 mRNA content were significantly lower than that of cisplatin group (P<0.05); Cisplatin group, cisplatin combined ganoderma lucidum polysaccharide group mice tumor tissue AQP1, AQP3 mRNA content were significantly lower than the control group (P<0.05); Cisplatin combined ganoderma lucidum polysaccharide group AQP1, AQP3 mRNA content were significantly lower than cisplatin group (P<0.05 ). Conclusion ganoderma lucidum polysaccharide can inhibit tumor growth and enhance cellular immune function of bladder cancer T24 cell line bearing mice, and adjust the expression of pro-apoptotic genes, AQP1 and AQP3.

BACKGROUND: Hydroxyapatite is a kind of ideal orthopedic material, but its Iow strength and brittleness need to improve. The research suggested that the improvement of the toughness and strength of bioceramics which composite hydroxyapatite (HAP) and zirconia (3% mol yttria-stabilized cubic zirconia) and fabricated ceramic scalpel possesses of the wound recover faster with smaller side. So it is significant that research on the incision concrescence of nano-sized hydroxyapatite-zirconia bioceramical scalpel.OBJECTIVE: To observe the effects of incision using nano-sized HAP-TZP bioceramics as scalpel.DESIGN: A single sample study.SETTING: Second Department of Orthopaedics, the Second Affiliated Hospital of Harbin Medical University and Harbin Institute of Technology.MATERIALS: The experiment was carried out in the Animal Laboratory of the Second Affiliated Hospital of Harbin Medical University (Provincial Laboratory) from March to May 2006. A total of 54 SD rats, of 4 months old, weighing 120-180 g, of both genders, were selected in this study. The experiment materials included nano-sized hydroxyapatite Quartz Clock Company), ammonia (Suihua Chemical Reagent Company), and anhydrous ethanol (Tianjin Chemical Reagent Company).METHODS: The nano-sized Hydroxyapatite/zirconia composite powder was synthesis by rubbing according to 40% hydroxyapatite + 60% zirconia powders (mass ratio) were mixed and milled. The knife-edge biomaterials, which fabricated by hot pressed sintering, was grinding and used in incision experiment after autoclaving. The back of the rats was shaved and removing a patch of skin under ether anesthesia made a length of 2 cm open excision wound and stitched up the tear by 1# string. The each 18 rats were sacrificed on 3, 7 and 14 days post-wounding and used for analyses respectively. A full-thickness specimen containing the wound was dissected out from each of the surgical sites. The hematoxylin-eosin (HE) staining was applied in the pathological observation.MAIN OUTCOME MEASURES: The tissue sections of back and the results of pathological observation. RESULTS: All 54 rats were involved in the results analysis without any loss. There was long gap with minimal cellular reaction in the scalpel wounds that has not healed on day 3. There was inflamed cells infiltration at the wounded margin edge. By week 1, the tiny detritus which some of them appear the repaired response of epidermis rend were still obviously found in the area of the scalpel wounds where the detritus was found to be significantly less, the wounds changed smooth and started to heal. The optical microscopy observed indicated that there was incomplete epithelialization and hyalinization of dermal collagen at the wounded area. There were obvious inflamed cells to infiltrate in the margin subcutaneous tissue of the wounds where the newborn loose collagenous fibers filled in scalpel wounds was the tissue repairing response phase. There was a great deal of the infiltrated white blood cells in the subcutaneous tissue. The tiny detritus of epidermis by the optical microscopy observed rend could not be found in the area of the scalpel wounds which were healed by week 2. The newborn capillary nets appeared in the newborn repaired collagen fibers tissue. The inflamed cells to be infiltrated were reduced. The compact and strong collagenous fibers filled in scalpel wounds were the normal wound repairing and tissue. The white blood cells infiltrated obvious reduction in the subcutaneous tissue. The normal blood cells in the newborn capillaries could be seen. CONCLUSION: The experiments prove that a HAP-ZTP bioceramic scalpel, which possesses excellent bioactive, is nontoxic, nonallergenic and noncarcinogenic for incisions.

Objective To investigate whether HER-2/neu oneogene amplification exists in 08- teosarcomas. Methods Fluorescence in situ hybridization(FISH)and DNA probes for HER-2/neu and centromere 17 were used to examine the HER-2/neu gene amplification in samples taken from 23 patients with osteosarcoma. Results None of the 23 osteosarcoma samples showed the HER-2/neu gene amplification.Condusion Since HER-2/neu gene amplification is rare in osteosarcomas, HER-2/neu target medicine should not be applied to patients with osteosarcoma

Objective To evaluate the effects of fructose-1,6-diphosphate (FDP) on the preservation and high-energy phosphate metabolism of hypothermic SD rat hearts. Methods Sixteen SD rats were randomly divided into 2 groups. Their hearts were harvested and preserved in UW solution at 4℃ for 12 hours. In the study group (n=8), FDP (5mmol/L) was added to the UW solution, whereas in the control group, FDP was not added. Langendorff perfusion was established with the isolated hearts, and cardiac functions were examined, as well as the activities of lactate dehydrogenase (LDH) and creatine kinase (CK) in the perfusion fluid collected from coronary. Then the high-energy phosphate concentration, ATP, ADP, and AMP contents in the myocardium were determined with HPLC. Method:Based on three diagnostic criteria and clinic experience,the questions were collected after item revision and item analysis of three samples. A total of 425 undergraduates responded to the formal questionnaire. The data of formal questionnaire were used to evaluate the reliability and quality.Result:The whole questionnaire was compose of 160 items including five repeated items for reliability of the respondents. Objective To investigate the status of vitamin(Vit)B12 deficiency in elderly inpatients in department of neurology.Methods 827 patients admitted to the department of neurology of Shanghai Punan hospital from March 2007 to July 2008,aged 60 and older,with no severe hepatosis and renal disfunction,and without using VitB12 preparation for 3 months,were included in the study.The levels of serum VitB12,folate,homocysteine(Hcy) were tested to screened patients with VitB12 deficiency.The results of symptoms,positive signs of neurological system and the laboratory examination were analysed and compared between patients with or without VitB12 deficiency.Results There were 827 patients,with the mean age of(77.1?7.5)years old,fulfilled the enrolling criteria.VitB12 deficiency was found in 163 patients(19.7%).The prevalence was higher in female(21.9%)than male(17.4%),which had a tendency of increased with age.In the patients with VitB12 deficiency,the rate of complicating gastrointestinal diseases was higher.Compared with patients with normal serum VitB12 level,there were more cases suffered from unstable gait(27.0%),hypopallesthesia(34.4%),and more cases combined with some chronic diseases such as cerebral infarction(62.6%),hypertension(68.7%),coronary heart disease(39.9%).77% cases had neuroelectricphysiological abnormalities,but only 9.82% cases had megaloblastic anemia.Conclusions VitB12 deficiency is common in elderly inpatients in neurology department,whose clinical manifestations are various and atypical,and the neurological symptoms are more common than megaloblastic anemia.

Objective To detect the change of the plasma hydrogen sulfide in patients with chronic heart failure (CHF) and explore its clinical significance.Methods A total of 118 patients with CHF was enrolled in this study.According to the NYHA standard,they were classified into grade Ⅱ (34 cases),grade Ⅲ (54 cases),grade Ⅳ (30 cases).The patients were divided into normal treatment group and sodium nitroprusside treatment group with 59 cases each.Another 30 healthy person were selected as control group.The plasma hydrogen sulfide level was detected by spectrophotometer.Results The plasma hydrogen sulfide level in CHF patients and healthy person was (32.45 ± 3.86),(54.26 ± 5.63) μ mol/L,and there was significant difference (P ＜ 0.05).The plasma hydrogen sulfide level in CHF patients of grade Ⅱ,Ⅲ,Ⅳwas (43.26 ±4.73),(31.87 ±4.12),(23.66 ±3.54) μmol/L,and there was significant difference (P＜0.05).The plasma hydrogen sulfide level before treatment in normal treatment group and sodium nitroprusside treatment group was(31.66 ± 3.75),(33.04 ± 4.02) μ mol/L,and there was no significant difference between two groups (P ＞ 0.05).The plasma hydrogen sulfide level after treatment in normal treatment group and sodium nitroprusside treatment group was (36.83 ± 4.32),(44.54 ± 4.68) μ tmol/L,and there was significant difference (P ＜0.05).The plasma hydrogen sulfide level after treatment in two groups was increased compared with that before treatment,and there was significant difference (P ＜ 0.05).The total effective rate in normal treatment group and sodium nitroprusside treatment group was 42.4％ (25/59) and 67.8％(40/59),and there was significant difference between two groups (P ＜ 0.05).Conclusions The abnormal plasma hydrogen sulfide level may be involved in the occurrence and development process of CHF,which could be used as a factor to monitor the disease.The upregulation of plasma hydrogen sulfide level can significantly improve the prognosis of CHF patients.
