2.Expression Difference and Its Mechanisms of miR-497 and miR-34a in Platinum-sensitive and Platinum-re-sistant Epithelial Ovarian Carcinoma Patients
China Pharmacy 2017;28(17):2308-2313
OBJECTIVE:To investigate the expression difference and its mechanism of miR-497 and miR-34a in platinum-sen-sitive and platinum-resistant epithelial ovarian carcinoma(EOC)patients. METHODS:A total of 72 EOC patients underwent ovari-an cancer staging surgery or cytoreductive surgery were selected from department of gynaecology and obstetriscs of our hospital dur-ing Jan. 2008-Jan. 2012. They received standardized platinum chemotherapy after surgery and were followed up (during Jul.2008-Jul.2016). According to the sensitivity to platinum, those patients were divided into platinum-sensitive group (42 cases) and platinum-resistant group (30 cases) . Real-time fluorescent quantitative PCR was adopted to detect the expression of miR-497 and miR-34a in tumor tissue, and the relationship of it with total survival period was investigated. The levels of DNA methylation of miR-497 and miR-34a promoter region were determined by nest type land type methylation specific PCR. Western blot assay was used to detect the H3K9 dimethylation(H3K9me2)levels. The H3K9me2 levels of miR-497 and miR-34a promoter region were de-termined by chromatin immunoprecipitation method. RESULTS:The expression levels of miR-497 and miR-34a in platinum-sensi-tive group were significantly higher than platinum-resistant group, with statistical significance (P<0.05). Total survival period of 72 patients was(45.7±17.5)months,which was positively correlated with the expression levels of miR-497 and miR-34a(r2 were 0.2714,0.3782,P<0.01). The DNA methylation of miR-497 and miR-34a promoter region in platinum-resistant group was signif-icantly higher than platinum-sensitive group, and H3K9me2 level of promoter region was significantly higher than platinum-sensi-tive group, with statistical significance (P<0.05 or P<0.01). H3K9me2 levels of platinum-resistant group were slightly higher than that of platinum-sensitive group, but there was no statistical significance (P>0.05). CONCLUSIONS:The expression of miR-497 and miR-34a in tumor tissue of EOC patients are related to the sensitivity of platinum chemotherapy and the survival time of patients. DNA methylation and histone methylation of promoter region may be one of the mechanisms of their expression changes.

Objective To explore the health inspector effect on the score of hygienic degree of hostels. Methods Using the scale for hygienic degree of hostels, 38 health inspectors from 9 sections in the Bureau for Health Inspection and Supervision of Haidian District in Beijing, scored 621 hostels in the district. The multilevel statistics model has been used to analyze the health inspector effect, of which the index is intra-class correlation (ICC), in the double level model structured by hostels as the level 1 unit and inspectors as the level 2 unit, and in the triple level model structured by hostels as the level 1 unit, inspectors as the level 2 unit and the sections as the level 3 unit. Results In the double level model, ICC of the level 2 unit is 0.142 8. In the triple level model, ICC of the level 2 unit is 0.0000 and ICC of the level 3 unit is 0.154 1. Conclusion There is inspector effect influencing the score of hygienic degree. And the effect seems to be more similar in one section comparing with the other ones.

Objective To explore the basic health state of the hotels in Haidian District. Methods 736 hotels were chosen to investigate on the hygiene state, in Haidian District, Beijing, during Feb.-Jul., 2006, based on the tables for the hygienic quantified and classified management of hotel health supervision. Results The hygienic quantified and classified standard score of 736 hotels was 53.7?22.6, and the standard score of the common hotels, one or two stars hotels and three or more stars hotel were 50.9?21.5, 58.9?24.7, 76.4?17.1. 50 households (6.8%) within them were A degree, 144 households (19.6%) were B degreee, 121 households(16.4%) were C degree and 421 households(57.2%) were D degree. Conclusion The general hygienic state of the hotels should be paid more attention to. The hygienic state changes with the level of the hotels.

Cancer Research and Clinic 2012;(11):721-724
Objective To prepare and identify the monoclonal antibody (mAb) specific to epithelial cell adhesion molecule (EpCAM) and explore the function of the mAb.Methods The EpCAM antigen expressed by the prokaryotic expression systems was used to immunize the BALB/c mice, and then the splenic cells from the mice were fused with the Sp2/0 cells to produce hybridomas secreting specific mAb.The positive clones were screened by the ELISA.The western blot analysis was used to identify the reactivity of the mAb to the antigen. Then the immunohistochemistry staining was used to detect EpCAM expression in the 3 primary colorectal carcinoma tissues. Results Three mAb specific to EpCAM were obtained by ELISA tests.Western blot results indicated that these three kinds of antibodies could react to the EpCAM antigen, but no response to the GST tag.Immunohistochemical staining results identified that these mAb could give positive signals to the primary colorectal carcinoma tissues from patients. Conclusion Three mAb specific to EpCAM are obtained and identified, which contributes to the diagnosis and therapy of the carcinoma in the future.

Journal of Environment and Health 2007;0(11):-
Objective To explore the effects of taurine-zinc(TZC) on blood lead content and blood-brain barrier(BBB) permeability of growing rats exposed to lead. Methods Male SD rats aged 3 weeks were randomly divided into four groups(8 in each group):control group, lead exposure group, dimercaptosuccinic acid(DMSA) group and TZC group. The exposure group treated with 0.12% lead acetate(0.2 ml/100 g body weight) by intraperitoneal injection,DMSA group and TZC group were treated with the same dose of lead acetate but were respectively supplemented with 5 mg/ml DMSA(2 ml/100 g body weight) by ig and 0.46 g/L TZC solution through drinking.Three weeks later, blood lead content was measured by atomic absorption spectroscopy(AAS) and changes of BBB permeability were observed using lanthanum nitrate tracer method. Results Compared with the lead exposure group, the blood lead level in TZC group and DMSA group was significantly reduced(P

OBJECTIVE:To study the anti-inflammatory,antalgic effects and irritation of Fuyan suppository. METHODS:Mouse auricle swelling test(Fuyan suppository 4.5,3.0 and 1.5 g/kg for administration)and mouse vola pedis swelling test(5 min and 0.5,1,4,6 h)were employed to study the anti-inflammatory effect of Fuyan suppository. Mouse writhing response test(Fuy-an suppository 4.5,3.0 and 1.5 g/kg) was used to study the antalgic effect. Rabbit vagina mucosa irritation test was adopted to study the vaginal irritation(Fuyan suppository 1.0 g/rabbit,once daily for one week). RESULTS:Fuyan suppository at a dose of 4.5,3.0 or 1.5 g/kg could inhibit auricle swelling of mice and reduce the times of mice’s writhing response. 3.0 g/kg Fuyan suppos-itory could,at 0.5-6 h from administration, inhibit vola pedis swelling of mice and be nonirritating to the rabbit’s vagina. CON-CLUSIONS:Fuyan suppository has better anti-inflammatory and antalgic effects and is nonirritating to the rabbit’s vagina. Theoreti-cally,vaginal administration of Fuyan suppository can be considered as safe.

Injection Stauntoniae (IS ) was applied ou a part of the saphenous nerve of rat 4mm in length. With 50% IS, amplitudes of A??, A? and C components of the compound action polential were reduced and their latencics were delayed. In particular, the amplitudcs of A? and C components reduced to 47.0% ? 23.o% and 44.0?20.0% (MEAN?SD, n = 8, P
