1.Early Face Processing of Internet Addiction Patients by Face-specific N170

Objective To study early face processing of internet addiction patients by using ERP method. Methods Ten healthy young man 18~21 years and 10 internet addiction patients 17~22 years served as subjects in experiment. Two runs of 300 stimuli (duration: 50 ms) of 3 facial and 3 non-facial pictures were randomly presented with equal probability (ISI: from 1 000 ms to 1 500 ms randomly), and the subjects were asked to react to facial stimuli and non-facial stimuli by pressing the left button and right button respectively as quickly as possible. Thirty two channel electroencephalogram (EEG) was recorded by Neuroscan Nuamps System. Results 1) Specific-face component Nd170 (face N170 minus object N170) was found in both groups, which was distributed at the temporal-occipital region in control groups but at occipital region in internet addiction group; 2) Compared with Nd170 in internet addiction group, Nd170 was significantly lower at occipital negion and delayed in control group. Conclusion The early face processing mechanism of internet addiction patients may be different from normal people.
2.Effect of the emodin thermosensitive hydrogel on coilagenase in gingival crevicular fluid of patients with chronic periodontitis

Objective To observe the effect of the emodin thermosensitive hydrogel on collagenase in gingival crevicular fluid of patients with chronic periodontitis and to evaluate the clinical efficacy.Methods Forty patients with chronic periodontitis were selected.For each patient after scaring and root planning,the one teeth on one side in a person was assigned at the test group and treated with the emodin thermosensitive hydrogel in the pocket once a week for four weeks.The teeth on the other side were assigned as the control group,just with the primary periodontal treatment.The PLI、SBI、PD、AL、MD and content of collagenase in gingival crevicular fluid were examined at baseline,before administration and after administration four weeks.The COL-II level in GCF was detected using ELISA method.Results There were no significant differences in PLI、SBI、PD、AL、MD and content of collagenase in gingival crevicular fluid before the treatment(P > 0.05).While there are some evidences that periodontal indexes dropped down and the type Ⅱ collagenase level in gingival crevicular fluid also declined after using the emodin thermosensitive hydrogel than the control group(t =3.46,4.02,4.18,3.03,2.79,4.29,all P < 0.05).Conclusion Ultrasonic scaling using the emodin thermosensitive hydrogelas was more effective in reducing the type Ⅱ collagenase level in gingival crevicular fluid.And improving clinical parameters associated with periodontal health in patients with chronic periodontitis.

5.Effects of PI3K/Akt signaling pathway on the proliferation and cell cycles of MC3T3-E1 cells

Objective Recent studies show that the PI3K/Akt signaling pathway is a key regulatory signal in both bone for-mation and bone remodeling .The experiment aimed to investigate the effects of inhibiting the PI 3K/Akt signaling pathway on the prolif-eration and cell cycles of MC3T3-E1 cells. Methods We treated MC3T3 -E1 cells with PF-04691502 the inhibitor of the PI3K/Akt signaling pathway .Then we determined the proliferative activity of the cells by MTS assay , detected the cell cycles by flow cytometry , and measured the expression of cell cycle-related proteins by Western blot . Results After inhibition of the PI3K/Akt signaling path-way with PF-04691502 at 30, 150 and 750 nmol/L, the proliferation of MC3T3-E1 cells was reduced in a dose-dependent manner to 0.647 ±0.041, 0.423 ±0.011 and 0.159 ±0.004, respectively, as compared with 1 ±0.056 in the control group (P<0.01).Sim-ultaneously , the percentage of the cells in the sub-G1 phase was significantly higher in the 1 μmol/L group than in the control ([1.45 ±0.43]%vs [0.27 ±0.21]%, P<0.01) and the expression of cyclin D1 was decreased. Conclusion Inhibition of the PI3K/Akt signaling pathway can block MC 3T3-E1 cells in the sub-G1 phase and reduce the proliferation of the cells .

Objective To study the relationship between lymph node micrometastasis and clinicopathological parameters. Methods Immunohistochemical method was adopted to detect CK19,CK20 and CD44v6 in 509 lymph nodes from 53 cases of gastric cancer, in which, conventional pathological diagnosis showed no lymph node metastasis. Results Micrometastasis was found in 47 lymph nodes (9.2%) of 21cases ( 39.6 %). 7.Effects of small RNA interference targeting mammalian target of rapamycin on paraquat-induced pulmonary fibrosis in rats

Objective To investigate the effects of small RNA interference targeting mammalian target of rapamycin (mTOR) expression on paraquat-induced pulmonary fibrosis in rats.Methods Human embryonic kidney cells HEK-293 were culturedin vitro. The mTOR small interfering RNA (mTOR-siRNA) expression plasmid transfection lentivirus was constructed, and non-specific sequence plasmid with no homology to mTOR gene was set as the control. Seventy-two healthy male Sprague-Dawley (SD) rats were randomly divided into normal saline (NS) control group, paraquatmodel group, mTOR unrelated sequence group, and mTOR-siRNA group, with 18 rats in each group. Paraquat poisoning animal model was reproduced by intraperitoneally injecting 20% paraquat solution 15 mg/kg, while the NS control group was intraperitoneally injected the same volumes of NS. Rats in the mTOR unrelated sequence group and mTOR-siRNA group were injected 1×109 TU/mL lentivirus solution 50μL into the airway, respectively, while in the NS control group and paraquat model group were injected the same volumes of NS. At 7, 14 and 28 days after treatment, 6 rats in each group were sacrificed respectively for lung tissue, the pathological changes and fibrosis of lung tissues were observed under light microscope. The levels of hydroxyproline (HYP) in lung tissues were determined by alkaline hydrolysis. The mRNA and protein expressions of mTOR in lung tissues were determined by reverse transcription-polymerase chain reaction (RT-PCR) and Western Blot.Results Under light microscope, there was no obvious pathological changes in the lung tissues in the NS control group, while in the paraquat model group and mTOR unrelated sequence group, lung tissue in rats were damaged, there were a lot of inflammatory cell infiltration, a large number of matrix collagen and fibrous tissues hyperplasia, and gradually increased with time, and it was consistent with paraquat-induced lung tissue fibrosis process. The pathological and fibrotic changes in lung tissue of mTOR-siRNA group were obviously reduced after silencing mTOR gene. The levels of HYP and the expression levels of mTOR mRNA and mTOR protein of lung tissues in the paraquat model group and mTOR unrelated sequence group were continuously increased in time-dependent manner, and they were significantly higher than those in the NS control group at all of the time points, but no significant difference was found between mTOR unrelated sequence group and paraquat model group. In mTOR-siRNA group, silencing mTOR gene could inhibit paraquat poisoning induced HYP increase in lung tissue, and the expressions increase in mTOR mRNAand mTOR protein, the values were close to the levels of NS control group, and the significant difference was found as compared with paraquat model group at 7 days or 14 days, and the change was maintained to 28 days [7 days: HYP (μg/mg) was 1.13±0.06 vs. 1.25±0.07; 14 days: HYP (μg/mg) was 1.19±0.09 vs. 1.29±0.12, mTOR mRNA (2-ΔΔCt) was 0.99±0.11 vs. 1.94±0.12, mTOR protein (gray value) was 0.39±0.08 vs. 0.75±0.09; 28 days: HYP (μg/mg) was 1.28±0.06 vs. 1.40±0.05, mTOR mRNA (2-ΔΔCt) was 1.15±0.13 vs. 2.85±0.15, mTOR protein (gray value) was 0.45±0.10 vs. 0.86±0.12, allP < 0.05].Conclusion Lentivirus-mediated mTOR-siRNA could effectively inhibit the expressions of mTOR in lung tissues of paraquat-poisoned rats, and reduce the damage and fibrosis of lung tissues caused by paraquat.
8.The postoperative complications and nursing experience of hand assisted laparoscopic J-type ileal pouch-anal anastomosis in familial adenomatous polyposis

Objective To investigate the postoperative complication and nursing experience of hand-assisted laparoscopic J-type ileal pouch-anal anastomosis (IPAA) in familial adenomatous polyposis (FAP). Methods A total of 36 patients with FAP who had undergone hand-assisted laparoscopic J-type ileal pouch- anal anastomosis (IPAA) were analyzed retrospectively. Results The postoperative complication included ileal pouch bleeding in 2 cases, urinary retention in 3 female patients, external hemorrhoids swelling in 3 cases, and anastomotic fistula in 2 cases. Six cases had ileum pouch inflammation during the postoperative follow- up. All the complications were relieved after conservative medical therapy. Conclusions IPAA operation puts forward high technical requirements with more postoperative complications. Therefore, it is important to strengthen nursing of the patients, reduce postoperative complication and increase quality of life of FAP.
9.Analysis of the Clinicl Characters and the Incidence Features of Neonatal Congenital Hypothy Roidism

Objective To explore the incidence features of neonatal congenital hypothy roidism in Lianyungang area.Methods The heel blood samples dried on filter paper was collected from newborns after 72 hours.Serum thyroid stimulating hormone(TSH)level was detected by Dissociation enhancement lanthanide fluoroimmuno assay(DELFIA).The cases diagnosed were analyszd for the clinicl characters and the incidence features.Result One hundred and fifty-eighet children with CH were diagnosed and the patients lived in 63 towns(streets)(66.3%).The distribution of CH was diffuse in Lianyungang.There was significant difference in incidence of CH between 2007 to June of 2008 and before 2006.Eighty-two patients were examined by thyroid gland color ultrasound,among which there were sixty-eight patients whoes thyroid glands were normal(82.9%)and fourteen patients were unusal(absent,replacement,hypogenesis)(17.1%).A couple of twins(male)and two embryoes from the same parents were screened for CH.Conclusion There are iodine lack(MUI300?g/L)individual among people.The iodine nutrition condition of pregnant women is one of important factor to influence the TSH level of newborns.Neonatal screeing is the only valid means to find patients with CH.
10.Effects of prostaglandin E1 on the levels of myocardium NF-κB and plasma TNF-α in patients underwent heart valve replacement

Objective To investigate the effects of prostaglandin E1 (PGE1) on the levels of myocardium nuclear factor‐κB (NF‐κB) and plasma tumor necrosis factor‐α (TNF‐α) in patients underwent heart valve replacement .Methods Forty ASA Ⅱ orⅢ degree patients (NYHA Ⅱ or Ⅲ degree) ,aged 32 to 67 years with body mass index of 17 to 28 kg/m2 ,underwent heart valve replacement ,were randomly divided into 2 groups (n=20):control group (group C) and PGE1 group (group P) .After induction of anesthesia ,PGE1 20 ng · kg -1 · min-1 was infused intravenously in group P ,while the equivalent volume of normal saline was giv‐en in group C ,and the infusion was completed at the end of operation .Arterial blood samples were collected at the time of before cardiopulmonary bypass (T0 ,baseline) ,30 min after the begin of cardiopulmonary bypass (T1 ) ,the end of cardiopulmonary bypass (T2 ) ,and 12 h(T3 ) ,24 h(T4 ) after the end of cardiopulmonary bypass .Myocardial specimens were obtained from right auricular appendage at T0 and T2 for microscopic examination and determination the activity of NF‐κB .Results The plasma mass concentra‐tions of TNF‐αwas significantly lower (P<0 .05) at T1 to T4 ,while the myocardial histopathological damage was lighter and the activity of NF‐κb significantly attenuated (P<0 .05) at T2 in group P than that in group C .Conclusion PGE1 can attenuate myo‐cardial injury in patients underwent heart valve replacement ,the mechanism is related to inhibit the activity of myocardium NF‐κB and decrease the plasma mass concentrations of TNF‐α.
