1.Effect of the emodin thermosensitive hydrogel on coilagenase in gingival crevicular fluid of patients with chronic periodontitis

Objective To observe the effect of the emodin thermosensitive hydrogel on collagenase in gingival crevicular fluid of patients with chronic periodontitis and to evaluate the clinical efficacy.Methods Forty patients with chronic periodontitis were selected.For each patient after scaring and root planning,the one teeth on one side in a person was assigned at the test group and treated with the emodin thermosensitive hydrogel in the pocket once a week for four weeks.The teeth on the other side were assigned as the control group,just with the primary periodontal treatment.The PLI、SBI、PD、AL、MD and content of collagenase in gingival crevicular fluid were examined at baseline,before administration and after administration four weeks.The COL-II level in GCF was detected using ELISA method.Results There were no significant differences in PLI、SBI、PD、AL、MD and content of collagenase in gingival crevicular fluid before the treatment(P > 0.05).While there are some evidences that periodontal indexes dropped down and the type Ⅱ collagenase level in gingival crevicular fluid also declined after using the emodin thermosensitive hydrogel than the control group(t =3.46,4.02,4.18,3.03,2.79,4.29,all P < 0.05).Conclusion Ultrasonic scaling using the emodin thermosensitive hydrogelas was more effective in reducing the type Ⅱ collagenase level in gingival crevicular fluid.And improving clinical parameters associated with periodontal health in patients with chronic periodontitis.
2.Early Face Processing of Internet Addiction Patients by Face-specific N170

Objective To study early face processing of internet addiction patients by using ERP method. Methods Ten healthy young man 18~21 years and 10 internet addiction patients 17~22 years served as subjects in experiment. Two runs of 300 stimuli (duration: 50 ms) of 3 facial and 3 non-facial pictures were randomly presented with equal probability (ISI: from 1 000 ms to 1 500 ms randomly), and the subjects were asked to react to facial stimuli and non-facial stimuli by pressing the left button and right button respectively as quickly as possible. Thirty two channel electroencephalogram (EEG) was recorded by Neuroscan Nuamps System. Results 1) Specific-face component Nd170 (face N170 minus object N170) was found in both groups, which was distributed at the temporal-occipital region in control groups but at occipital region in internet addiction group; 2) Compared with Nd170 in internet addiction group, Nd170 was significantly lower at occipital negion and delayed in control group. Conclusion The early face processing mechanism of internet addiction patients may be different from normal people.

4.Effects of PI3K/Akt signaling pathway on the proliferation and cell cycles of MC3T3-E1 cells

Objective Recent studies show that the PI3K/Akt signaling pathway is a key regulatory signal in both bone for-mation and bone remodeling .The experiment aimed to investigate the effects of inhibiting the PI 3K/Akt signaling pathway on the prolif-eration and cell cycles of MC3T3-E1 cells. Methods We treated MC3T3 -E1 cells with PF-04691502 the inhibitor of the PI3K/Akt signaling pathway .Then we determined the proliferative activity of the cells by MTS assay , detected the cell cycles by flow cytometry , and measured the expression of cell cycle-related proteins by Western blot . Results After inhibition of the PI3K/Akt signaling path-way with PF-04691502 at 30, 150 and 750 nmol/L, the proliferation of MC3T3-E1 cells was reduced in a dose-dependent manner to 0.647 ±0.041, 0.423 ±0.011 and 0.159 ±0.004, respectively, as compared with 1 ±0.056 in the control group (P<0.01).Sim-ultaneously , the percentage of the cells in the sub-G1 phase was significantly higher in the 1 μmol/L group than in the control ([1.45 ±0.43]%vs [0.27 ±0.21]%, P<0.01) and the expression of cyclin D1 was decreased. Conclusion Inhibition of the PI3K/Akt signaling pathway can block MC 3T3-E1 cells in the sub-G1 phase and reduce the proliferation of the cells .

Objective To study the relationship between lymph node micrometastasis and clinicopathological parameters. Methods Immunohistochemical method was adopted to detect CK19,CK20 and CD44v6 in 509 lymph nodes from 53 cases of gastric cancer, in which, conventional pathological diagnosis showed no lymph node metastasis. Results Micrometastasis was found in 47 lymph nodes (9.2%) of 21cases ( 39.6 %). Objective To discuss the effectiveness of standardized management of hypertension and its significance of economics.Methods Taking prospective,randomized and controlled trial design to screen hypertension in four communities of Changping Town.The patients in the four communities were randomly divided into an intervention group and a control group.The intervention group was taken the standardized management and the control group was taken conventional management.Results The treatment rate and control rate of the intervention group was higher than the control group.Necessary inputs of per capita administration costs for every 1% increase of treatment or control rate in the intervention group were less than the control groups by 1.08 and 1.36 Yuan respectively.The per capita administrative costs of intervention group were higher than control group,while the per capita comprehensive costs of intervention group were lower than control group.Compared with the control group,every 1 Yuan of per capita administrative costs increases,the comprehensive costs would be reduced 7.58 Yuan in the intervention group.Conclusion Standardized management of hypertension in community can significantly improve the prevention and treatment effects.Moreover,it has good cost-effective value and saving social resources.
8.Analysis of the Clinicl Characters and the Incidence Features of Neonatal Congenital Hypothy Roidism

Objective To explore the incidence features of neonatal congenital hypothy roidism in Lianyungang area.Methods The heel blood samples dried on filter paper was collected from newborns after 72 hours.Serum thyroid stimulating hormone(TSH)level was detected by Dissociation enhancement lanthanide fluoroimmuno assay(DELFIA).The cases diagnosed were analyszd for the clinicl characters and the incidence features.Result One hundred and fifty-eighet children with CH were diagnosed and the patients lived in 63 towns(streets)(66.3%).The distribution of CH was diffuse in Lianyungang.There was significant difference in incidence of CH between 2007 to June of 2008 and before 2006.Eighty-two patients were examined by thyroid gland color ultrasound,among which there were sixty-eight patients whoes thyroid glands were normal(82.9%)and fourteen patients were unusal(absent,replacement,hypogenesis)(17.1%).A couple of twins(male)and two embryoes from the same parents were screened for CH.Conclusion There are iodine lack(MUI300?g/L)individual among people.The iodine nutrition condition of pregnant women is one of important factor to influence the TSH level of newborns.Neonatal screeing is the only valid means to find patients with CH.
9.Modified technique of whole retinal capillary network mounting for cells and capillaries counting

Background Previous retinal vascular mounting only obtained part of retinal vessels for the study of retinal diseases,and thus it is difficult to comprehensively assess these diseases.So optimizing the trypsin digestion method to show the complete retinal capillary network is very important for the study of retinal diseases.Objective This study was to modify the preparing way of trypsin digested whole retinal capillary network and offer a basis for a quantitative analysis of cells and capillaries.Methods Thirty Wistar rats were divided into model group (20 rats) and control group (10 rats).Streptozotocinum(STZ) of 1.25% dissolved in 0.05 mmol/L sodium citratehydrochloric acid buffer was intraperitoneally injected to establish diabetes models in the model group,and the equal volume of solvent was used in the same way in the control group.Eight months after injection,100 ml PBS was injected via ventriculus sinister and released via cut right atrium,and then 100 ml 4% paraformaldehyde was injected into the ventriculus sinister.The rat retinas containing part of the optic nerve were entirely isolated,and digested by trypsin,and vitreous,inner limiting membrane and neural retinal tissue were removed.The whole retinal capillaries network was mounted on the slide.The ghost pericytes and acellular capillaries in the middle retinal area were identified and counted under the optical microscope.Results The retinal mount exhibited that the retinal vessels were stained by hematoxylin and periodic acid schiff.The vessels network presented with the entire type in shape with the radical central retinal arteries and veins and their branches.The capillary showed the shallow-and deep-layer networks between the small arteries and veins.Pericytes distributed and protruded vessel wall and formed the ghost cells without nuclei.The diameter of acellular capillaries was 20% or more than that of near capillaries,and no cellular nuclei or ghost cell was found through the vessel.Conclusions The experimental technique for setting-up of cleaned vasculature and mounting vessels on glass microscopic slide provides intact vessels,which is helpful for the evaluation of retinal vascular morphology and quantitative analysis.
10.Clinical study and significance of the test of heart rate turbulence in patients with diabetic cardiomyopathy

Objective To study the changes and clinical significance of heart rate turbulence(HRT)in patients with diabetic cardiomyopathy.Methods 24h Holter ECG recordings were examined in 33 patients with diabetic cardiomyopathy(DC)and 30 patients in the control group.Turbulence onset(TO)and turbulence slope (TS)were calculated from Holter recordings.The mean value of TO,TS between the two groups were compared,the relativity among TO,TS,SDNN and LEVDD were analyzed as well.Results TO was significantly higher in the DC group compared with the control group[(-0.38 ± 0.12)% vs(-1.88 ± 0.13)%,t =7.38,P =0.01)].TS in the DC group was significantly lower than that in the control group[(2.02 ± 0.82)ms/R-R vs (9.72 ± 1.18)ms/R-R,t =18.69,P =0.02)].TO was significantly related to SDNN(r =-0.35,P =0.02)and LEVDD(r =0.68,P =0.01),TS was also significantly related to SDNN(r =0.45,P =0.01)and LEVDD (r =-0.39,P =0.03).Conclusion The manifest of heart rate turbulence is significantly impaired in patients with diabetic cardiomyopathy.
