1.Correlation between glucose variability and coronary stenosis in type 2 diabetes mellitus combined with coronary heart disease patients

Objective To investigate the correlation between glucose variability and coronary stenosis in type 2 diabetes mellitus (T2DM) combined with coronary heart disease (CHD) patients.Methods Eighty patients with T2DM who had undergone coronary angiography were selected.According to the result of coronary angiography,the patients were divided into 2 groups:48 T2DM combined with CHD patients as CHD group,and 32 T2DM combined without CHD patients as control group.All patients underwent continuous glucose monitoring (CGM) for 72 h,and the mean blood glucose (MBG),standard deviation of blood glucose (SDBG),daily mean amplitude of glycemic excursion (MAGE),absolute means of daily differences (MODD),largest amplitude of blood glucose excursion (LAGE) and mean postprandial glucose excursion (MPPGE) were computed.The risk factor of coronary stenosis (using Gensini score) was analyzed.Results The LAGE,SDBG,MAGE,MPPGE,MODD in CHD group were significantly higher than those in control group:(11.4 ± 3.6) mmol/L vs.(8.2 ± 1.9) mmol/L,(3.4 ± 1.4) mmol/L vs.(1.9 ± 0.6) mmol/L,(7.1 ± 2.7) mmol/L vs.(4.8 ± 1.4) mmol/L,(6.6 ± 1.8) mmol/L vs.(4.6 ± 1.5) mmol/L,(3.3 ± 1.2) mmol/L vs.(2.5 ± 0.9) mmol/L,and there were statistical differences (P < 0.05).There was no statistical difference in MBG between CHD group and control group:(10.2 ± 2.8) mmol/L vs.(9.3 ± 2.2) mmol/L,P > 0.05.The Pearson correlation analysis result showed that the MPPGE,MODD and LAGE had positive correlation with Gensini score (r =0.509,0.357 and 0.338;P < 0.05);and the LVEF had negative correlation with Gensini score (r =-0.372,P < 0.05).Conclusions Glucose variability in patients with T2DM combined with CHD is markedly enhanced.MPPGE,MODD and IAGE are positively correlated with the severity of coronary stenosis.

3.Effect of ginsenoside Rg3 on Cx26 gene expression and gap junctional intercellular communication in human breast cancer cell line MCF-7

Objective To study the function of ginsenoside Rg3 on proliferation in human breast cancer cell line MCF-7 and effect of ginsenoside Rg3 on Cx26 gene expression and gap junctional intercellular communication (GJIC) in MCF-7, cultured in vitro. MethodsHuman breast cancer cells MCF-7 was exposed to ginsenoside Rg3 at differential concentrations for 24 h, respectively. The cell proliferation inhibition was measured by 3-[4,5-dimethylthiazo-2-yl]-2,5 diphenyl tetrazolium bromide (MTT) assay. The expression of Cx26 mRNA was measured by RT-PCR in experimental groups and control goup. The GJIC function of MCF-7 cell was examined with scrape-loading dye transfer assay.ResultsHuman breast cancer cell line MCF-7 was exposed to ginsenoside Rg3 at a concentration of 10, 20, 40, 80, 160 μg/ml, respectively.The inhibition ratio was 3.1%, 5.2 %, 16.0 %, 26.3 %, 29.1% respectively after 24 h. Compared with control group, the concentration of 40 μg/ml above could significantly inhibit MCF-7 cell proliferation (P <0.05), so the experimental groups were exposed to ginsenoside Rg3 at a concentration of 40, 80, 160 μg/ml,respectively. The expression of Cx26 mRNA in every experimental group compared with control group was enhanced when MCF-7 cell was exposed to ginsenoside Rg3 at a higher concentration. It was observed that Lucifer yellow fluorescent staining was limited to a single cell in control group through fluorescent microscope,but Lucifer yellow fluorescent transfered through gap junction cells to neighboring cells, then came into being flake fluorescent staining in experiment groups. ConclusionGinsenoside Rg3 can enhance the expression of Cx26 mRNA in MCF-7 cell and restore the gap junctional intercellular communication, which may be one of important mechanisms of ginsenoside Rg3 in antitumor.
4.Effects of Group Counseling on Depression and Anxiety of Patients with Schizophrenia in Rehabilitation

Objective: To study the effects of group counseling on depression and anxiety of patients with schizophrenia in their rehabilitation.Methods: Eighty patients with schizophrenia in rehabilitation stage were randomly divided into research group and control group. In research group, the patients received group counseling in addition to conventional medications. Before and after the counseling, their anxiety and depression were assessed by self-rating scale SAS and SDS. 5.Effect of viral core protein mutant on HBV encapsidation

Objective To observe a recombinant mutant of HBV core protein for dominant negative gene therapy against HBV encapsidation in vitro. Methods C gene and S gene of HBV were acquired through PCR and subcloned into pGEM T to construct pGEM T C and pGEM T S respectively. After digestion and ligation of these two plasmids, pGEM T CS was constructed. The cloned gene was inserted into pcDNA3.1 + to construct pcDNA3.1 + CS, which was identified by DNA sequencing. The recombinant plasmids were transformed into HepG2 cells, and screened with G418. The resistant HepG2 cell clones were chosen to test the expression of core surface protein by RT PCR, and the expressing HepG2 clones were cultured with 10% HBV DNA positive human serum for 72 hours. The intracellular HBV particles were extracted and the DNA was subjected to dot hybridization. Results The analysis showed that the HepG2 cells expressing mutant C protein had capabilities to resist HBV invasion in varied degrees. The mutant C protein had a dominant negative role in the encapsidation of HBV compared with the naive part of core protein. Conclusions The production of recombinant mutant core protein has a potential value for gene therapy against HBV infection.
6.Study on the effect of bacillus licheniformis and live combined bifidobacterium and lactobacillus on chemotherapy- induced diarrhea

Objective To study the effect of bacillus licheniformis and live combined bifidobacterium and lactobacillus on chemotherapy-induced diarrhea (CID). Methods Seventy-two gastric cancer patients with CID were collected and the clinical data were analyzed retrospectively. The patients were divided into 4 groups according to different treatment methods with 18 cases each. The patients in group A were treated with montmorillonite powder, the patients in group B were treated with montmorillonite powder and bacillus licheniformis, the patients in group C were treated with montmorillonite powder and live combined bifidobacterium and lactobacillus, and the patients in group D were treated with montmorillonite powder, bacillus licheniformis capsule and live combined bifidobacterium and lactobacillus. The patients with severe diarrhea in 4 groups were treated with rehydration, maintaining water and electrolyte balance and nutritional support. The Karnofsky performance scale score (KPS score), diarrhea grading before and after treatment and treatment effect were recorded. Results The KPS score after treatment in group A, group B, group C and group D were significantly higher than that before treatment: (70.6 ± 10.6) scores vs. (62.2 ± 12.2) scores, (76.1 ± 7.8) scores vs. (61.7 ± 9.9) scores, (77.2 ± 7.5) scores vs. (61.1 ± 10.8) scores, (83.9 ± 5.0) scores vs. (63.9 ± 10.9) scores. Moreover, The KPS score in group B, group C and group D were significantly higher than that in group A, the KPS score in group D was significantly higher than that in group B and group C, and there were statistical differences (P<0.05). The diarrhea grading after treatment in 4 groups were improved compared with that before treatment. Furthermore, the diarrhea grading in group B, group C and group D were significantly better than that in group B and group C, and there were statistical differences (P<0.05). In group A, there were 2 cases with excellent effect, 10 cases with effect and 6 cases with no effect;in group B, there were 8 cases with excellent effect, 8 cases with effect and 2 cases with no effect;in group C, there were 7 cases with excellent effect, 9 cases with effect and 2 cases with no effect; in group D, there were 9 cases with excellent effect, 9 cases with effect and 0 case with no effect. The treatment effect in group D was significantly better than that in the other 3 groups, and there were statistical differences (Hc = 10.81, P<0.05). Conclusions Bacillus licheniformis and live combined bifidobacterium and lactobacillus is more effective in the treatment of gastric cancer patients with CID.
7.Hydroxyapatite ocular prosthesis implants wrapped by human amniotic membrane

Objective To observe the clinical effects of human amniotic membrane as an alternative substrate for wrapping hydroxyapatite ocular prosthesis implants.Methods After enucleation, hydroxyapatite orbital implants wrapped by human amniotic membrane were implanted with four rectus sutured to the implants .Among them, 4 cases were given primary implantation and 3 cases secondary implantation.Results After 8 to 39 week follow-up, it was found that in 6 cases, the eyelids were plump and prosthesis moved well, but one implant was removed because of failure of scleral patch graft for implant exposure.Conclusion The clinical results were satisfactory for ocular prosthesis implantation of hydroxyapatite sphere wrapped by human amniotic membrane, which may be considered as an alternative substrate for ocular prosthesis implants wrapper.
8.Effects of sufentanil pretreatment on the TRPV1 mRNA expression in the hypothalamus and gastric mucosa in the rats with acute gastric mucosa lesion induced by water immersion and restraint stress (WIRS)

Objective To observe the protective effect of sufentanil pretreatment on the rats with acute gastric mucosa lesion (AGML) induced by water immersion and restrain stress (WIRS) and its effect on TRPV1 mRNA expression in the hypothalamus and gastric mucosa. Methods Thirty male Wistar rats were randomly designed into 3 groups, including the normal control group (Group NC, n = 10), the group treated with WRIS for 6 h (Group WIRS, n = 10) and the group pretreated with sufentanil (Group SF, n = 10). The model of AGML was established by the classic WIRS method , and observed for the general extent of gastric mucosal injury at WIRS for 6 hr, and calculated gastric mucosal injury ulcer index (UI) and the PH value of gastric juice; The quantification of TRPV1 mRNA expression in hypothalamus and gastric mucosa was performed using quantitative real-time PCR; In addition, the activity of super oxide dismutase (SOD) and the level of malondialdehyde (MDA) in serum were detected. Results Compared with group NC, gastric mucosal in Group WIRS was injured more seriously , and the UI and the activity of MDA were also obviously increased , but the change of SOD activity was not apparent; The TRPV1 expression in gastric mucosal decreased apparently. Sufentanil pretreatment could effectively relieve gastric mucosal injury induced by WIRS , and make the UI and the activity of MDA decreased , and up-regulate TRPV1 mRNA expression in the hypothalamus and gastric mucosa. Conclusions Sufentanil pretreatment can effectively relieve AGML induced by WIRS , which may be related to the control of oxidative stress response , the reduced gastric acid secretion , and the upregulation of the TRPV1 mRNA expression in the central and periphera nerve.
9.Effect of restraint stress at different periods on SOD and MDA of normal rat serums and mucosal tissues

Objective To investigate the effects of restraint stress (RS) in different periods on SOD and MDA of normal rat serums and mucosal tissues. Methods 32 adult male Wistar rats were randomly divided into normal control group (NC group, n=8), and restraint stress groups of 3 days, 5 days and 7 days (RS3, RS5 and RS7 groups, n=8 at each group). The comparisons were done between all the groups in terms of the general extent of gastric mucosal injury, gastric mucosal injury ulcer index (UI), water content, the activity of super oxide dismutase (SOD), and the level of malondialdehyde (MDA) in serum and mucosal. Results Compared with NC group, gastric mucosal injuries in RS group were more severe, the UI and water content was proportionally related with the restraint periods. Meanwhile, the serum and mucosal SOD in the RS groups were all increased, and with prolonged restraints, the SOD were decreased, but the serum and mucosal MDA were increased (P<0.01) and SOD/MDA in the mucosa lowered as well (P<0.01). Conclusion Restraint stress may cause mucosal injury proportional to the stress time and related with lowered SOD/MDA in mucosal tissues.
10.Effect of APETx2 on the expression of ASIC3 in dorsal root ganglia in rat AGML model

Objective To study the effect of APETx2 on the expression of ASIC3 APETx2 in a rat model of acute gastric mucosal lesion(AGML). Methods Twenty-four Wistar rats were randomly assigned to three groups in equal number : normal control group, water immersion restraint stress (WIRS) group, APETx2 treatment group. AGML was induced by WIRS for 6 hours, and APETx2 (25 μg/kg) was injected intraperitoneally before the onset of stress. Intragastric pH and gastric histopathological changes were measured and the expression of ASIC3 mRNA in DRG neurons projecting to rat stomach was examined by real-time PCR. Immunohistochemistry was performed to detect the localization of ASIC3. Results Compared with the normal control group, the WIRS group showed obvious gastric injury with lower values of intragastric pH and extensive expression of ASIC3 in the DRG neurons (P < 0.05). The treatment with APETx2 before the onset of WIRS significantly alleviated the gastric mucosal injury, decreased gastric acidity and reduced ASIC3 expression in DRG neurons (P < 0.05). Conclusions ASIC3 expression in DRG neurons projecting to rat stomach is strongly associated with gastric mucosal lesion and acidosis in the WIRS model. APETx2 can improve gastric acidosis and prevent the occurrence of these lesions.
