1.Value of ERG in the diagnosis of vascular tumor

Purpose To study the expression of ERG in various vascular tumors, and to explore the value of ERG in the diagnosis of vascular tumor. Methods Immunohistochemical EnVision method was used to detect the expression of ERG and CD31 in 30 cases of vascular tumor and 20 cases of non-vascular tumor. Results All vascular tumors diffusely expressed ERG and CD31. Expression of ERG was unrelated to tumor histologic type and benign or malignant. Expression of ERG was consistent with CD31. Of 20 non-vascular tumors, all except for one cases of epithelial sarcoma did not express ERG. The sensitivity and specificity of ERG in the diagnosis of vascular tumor were 100% and 96. 7%. Conclusion ERG is a high sensitive and specific marker for vascular-derived tumor. The combinated detection of ERG and CD31 may useful for the diagnosis and differential diagnosis of vascular tumor.
2.Advance on insulin-like growth factor binding protein in human malignant tumors

Insulin-like growth factors, their receptors and insulin-like growth factor binding protein constitutes insulin-like growth factor system. Studies in vivo and vitro suggest that they can regulate cell growth and proliferation. Insulin-like growth factors have a potent mitogenic effects which dependent on interaction between insulin-like growth factors and their receptors. IGFBP family contains six high-affinity members with variable functions and mechanisms of actions. In addition to functioning as simple carrier proteins, IGFBP can inhibit or promote IGFs by IGF-dependent or IGF-independent mechanism. Recent findings show that the IGFBP is correlated with the initiation and progression of tumor growth.

With the quick development revelant techniques, proteomics is a very active subject in modern biological fields. The key techniques of proteomics are two dimensional electrophoresis, mass spectrometry and bioinformatics. From the new point of view, proteomics may play a vital role in the early detection of neoplasm by looking for tumor specific protein and elucidating the relationship between the change of expression and different phases of tumorigenesis. The article briefly review the relevant technique, strategy and application for the early diagnosis of in digestive system tumors.
4.The diagnostic value of soluble scavenger receptor for the patients of malignant tumor associated with fever in early infection

Objective To discuss the diagnostic value of soluble scavenger receptor(sCD163)for the patients of malignant tumor associated with fever in early infection .Methods Collect 102 cases of patients confirmed with malignant tumor in this hospi-tal since January to December 2014 ,all the patients were examined with blood culture .102 patients were divided into two groups ac-cording to the results of blood culture :infection group (60 cases) and uninfected group (42 cases) .ELISA ,electrochemical lumines-cence and immune transmission turbidimetric methods were used to detect the levels of sCD163 ,PCT and CRP in serum ,to compare the differences in the above indicators between two groups .The receiver-operating characteristic curve (ROC curve) was applied to evaluate the application value of sCD163 in diagnosing of malignant tumor associated with fever in early infection .Results The lev-el of sCD163 ,PCT and CRP had statistically difference in two groups(P< 0 .05) ;the critical values of sCD163 、PCT and CRP were 110 .80 ng/mL ,0 .45 ng/mL ,15 .60 mg/L respectively which can suggest the patients with malignant tumor complicated with fever in early infection .The area under the curve were 0 .894 ,0 .835 ,0 .743 respectively ,among that sCD163 area was the largest ;The corresponding sensitivity were 88 .9% ,77 .9% and 88 .0% ;specificity were 77 .0% ,74 .0% and 50 .0% respectively .Conclusion Compared with PCT ,CRP ,sCD163 has more value for diagnosing the patients of malignant tumor associated with fever in early in-fection .
5.Improvement in Quality Control Methods for Belladonna Oral Solution

Objective:To improve the identification and determination methods in the original quality standard for belladonna oral solution.Methods:Belladonna oral solution was identified by a specific chromatogram of HPLC,and scopoletin and hyoscyamine sulfate in belladonna oral solution were detected by dual wavelength spectrophotometry.The detection of fingerprints was performed on a Waters SunFire C18 (250 mm×4.6mm,5 μm) column.The mobile phase was methanol-0.05% phosphoric acid solution at a flow rate of 1.0 ml·min-1.The detection wavelength was 344 nm and the column temperature was 30℃.The detection of scopoletin and hyoscyamine sulfate was performed on the same C18 column.The mobile phase was 10 mmol·L-1 heptanesulfonate sodiumat (pH value was 3.3 adjusted by glacial acetic acid)-absolute ethanol-acetonitrile (68.75∶6.25∶25) at a flow rate of 1.0 ml·min-1.The detection wavelengths were 344 nm and 210 nm,and the column temperature was 30℃.Results:The specific chromatogram of belladonna oral solution was accordance with that of belladonna tincture raw material.The retention time and relative peak area of each characteristic peak and reference peak all met requirements of Chinese Pharmacopoeia.Scopoletin and hyoscyamine sulfate were completely separated from the other compositions under the above mentioned conditions.The calibration curves were linear within the range of 5.168-103.360 μg·ml-1 (r=1.000 0) for scopoletin and 50.560-758.400 μg·ml-1 (r=0.999 9) for hyoscyamine sulfate.The average recovery was 101.79% (RSD=1.05%,n=6) and 100.92% (RSD=0.97%,n=6),respectively.Conclusion:After the quality control method improvement,the identification shows high specificity and the quality of belladonna oral solution can be better controlled by the two selected index components.The method is easy and accurate,which can provide a reliable way to control the quality.
6.Expression of podoplanin-positive stromal cells in esophageal squamous cell carcinoma and its clinical significance

Purpose To study the correlation between the expression of podoplanin-positive stromal cells and clinicopathologic features in squamous cell carcinoma ( SCC) of esophagus, and to explore the role of podoplanin-stromal cells in the progression of SCC of e-sophagus. Methods Immunohistochemical EnVision method was used to detect the expression of podoplanin in the stromal cells of e-sophageal SCC. The correlation between the expression of podoplanin-positive stromal cells and clinicopathologic features in SCC of e-sophagus was analyzed. Results Of 69 cases of SCC, podoplanin-positive stromal cells were seen in 43 and the positive rate was 62. 3%. Lymphatic vessel density in SCCs with podoplanin-positive stromal cell was 45. 72 ± 14. 36, which was higher than that with-out podoplanin-positive stromal cell (21. 23 ± 10. 49) (P<0. 01). Expression of podoplanin in stromal cells in SCCs with lymphatic vessel invasion was higher than those without lymphatic vessel invasion (P<0. 05). There was no correlation between podoplanin-posi-tive stromal cells and age, sex, tumor size, the depth of invasion, differentiation, lymph node metastasis, and clinical stage ( P>0. 05). Conclusion Podoplanin-positive stromal cells may promote the lymphangiogenesis and lymph vessel invasion of esophageal SCC.
7.Clinical Observation of Lumbar Intervertebral Disc Protrusion Patients Treated with Acupotomy Therapy Combined with Acupuncture Based on Distal Points Selection

Objective To evaluate the clinical effect of acupotomy therapy combined with acupuncture based on distal points selection for lumbar intervertebral disc protrusion ( LIDP) . Methods Sixty-eight LIDP patients were randomly divided into treatment group (N=33) and control group(N=35). The treatment group were given acupotomy therapy on the proximal positive response points in the lumbosacral region for relaxing the tendons,and acupuncture therapy based on distal points selection of Shugu, the Shu-stream point of the foot Taiyang bladder meridian, Zulinqi and Weizhong, the shu-stream point and the blood xi-cleft point of the foot Shaoyang gallbladder meridian respectively, twice a week, 2 weeks constituting one course of treatment. The control group was given conventional electroacupuncture therapy on Huatuo clip points, Dachangshu point, bilateral Shenshu points, Yinmen point, bilateral Zibian points and bilateral Yanglingquan points, and together with TDP radiation therapy, 5 times a week, 2 weeks constituting one course of treatment. The two groups received follow-up for one month. The low back pain scores of Japanese orthopaedic society ( JOA) and visual analog scores ( VAS) of pain were used for the evaluation of therapeutic effect after the treatment. Results ( 1) After treatment, the JOA scores were increased and VAS scores were decreased in both groups compared with those before treatment ( P< 0.05) , and the increase of JOA scores and the decrease of VAS in the treatment group were superior to those in the control group ( P< 0.05) . There was statistically significant difference of VAS scores between the two groups ( P < 0.05) during the follow-up period. ( 2) The total effective rate was 87.9%in the treatment group, and was 77.1%in the control group, the difference being significant (P<0.05) . Conclusion The acupotomy therapy combined with acupuncture based on distal points selection is effective and safe for lumbar intervertebral disc protrusion, which has the advantages of reliable long-term effect and low recurrence rate, showing good prospects for extensive clinical application.
8.The analyse of examining the bone marrow involvement in ninety-three patients of non-Hodgkin′s lymphoma

Objective To explore the value of combined detection of bone marrow smear(BMS) and flow cytometry(FCM) to examine bone marrow involvement(BMI)of B cell non‐Hodgkin′s lymphoma(B‐NHL) .Methods Patients with B‐NHL were detec‐ted by BMS and FCM simultaneously .Smears were stained with Wright‐Giemsa′s staining to differentially count neoplastic cells and the processed sample with fluorescent staining for the FCM .Results 93 cases of patients with B‐NHL were detected .The detection rate of BMS and FCM were 11 .8% and 29 .0% .There was statistical difference of detection rate between the two method(P<0 .05) .In 11 cases patients of BMI with the method of BMS ,while using the method of FCM 10 cases were positive .Conclusion Both BMS and FCM are useful to diagnose BMI of B‐NHL ,each of them has individual characteristics .Combined detection of these two methods can increase the diagnostic of BMI .

Objective It was designed to observe the effects of ? amyloid protein on the functions of neurotransmitter receptors and to disclose the possible pathophysiological mechanism of Alzheimer disease(AD) mediated by ? amyloid protein and neurotransmitter receptors. Methods Messenger RNA was taken from brains of aged Wistar rats with Promega kits and microinjected into Xenopus oocytes for receptor expression. The currents of expressed receptors and the effects of A? 1 40 on them were detected with the double electrode voltage clamps technique. Results ACh, glutamate, dopamine and GABA receptors were successively expressed in Xenopus oocytes. The currents of these expressed receptors detected with voltage clamps were carried by chloride ions with their equibrillium potentials close to 22 mV. Currents of expressed ACh, glutamate, dopamine receptors in the aged rats decreased more significantly than those in adult young rats. A? had a reverse effect on the functions of ACh and glutamic acid receptors. A? 1 40 enhanced significantly the currents of expressed glutamate receptors when it inhibited markedly the currents of expressed ACh receptors.
