1.Gastrointestinal stromal tumors:imaging features with intraoperative and clinicopathological correlation

Objective To analyze imaging of gastrointestinal stromal tumors(GIST),and to compare their imaging features with operational and pathological findings.Methods Clinical,imaging,and pathological data of 20 patients with GIST were collected.Results Imaging findings were endophytic or exophytic tumors with heterogeneous density or signal intensity,corresponding to hemorrhage,necrosis,and cystic changes.Imaging was correct for the location of the lesion in 11 of 16 primary GIST and 4 cases of relapsed tumors.Preoperational CT did not detect mesenteric,peritoneal,and omental metastasis in 5 cases. Hepatic metastases detected at CT (3 cases )were identified by operational findings.Conclusions GIST has some imaging features.CT is a useful tool in detecting and characterizating of lesions rather than detecting mesenteric,peritoneal,and omental metastasis.
2.Study on the Optimum Culture for Growth of Bacillus Subtilis B115 andthe Antibacterial Effect of B115 on Aeromonas

The essential medium of B115 composed of 1% tryptone, 0.25% yeast extract and 0.5% sodium chloride was determined by using an orthogonal design. The orthogonal design was also employed in testing the optimum additions. It was composed of 0.1%(NH_(4))_(2)SO_(4),1.4%K_(2)HPO_(4), 0.6% KH_(2)PO_(4) and 0.1% (Na_(3)C_(6)H_(5)O_(7)). The yield of B115 cultured in optimum medium was compared with the one in essential medium. Statistic analysis showed that the growth of B115 was most significantly improved by adding K~(+)、NH~+_(4) and (Na_(3)C_(6)H_(5)O_(7)) to essential medium. The antibacterial effect of Bacillus subtilis strain B115 on pathogenic Aeromonas was studied. The results showed different antibacterial effects of B115 on different aeromonads. There were obvious antibacterial effects on BSK-10 and CL990920, while no effect on the growth of TL970424.
3.Research on radix notoginseng for improving CKD rat renal fibrosis

Objective To investigate the effects of notoginseng and guiding medicinals mediated notoginseng for improving the renal inter stitial fibrosis in rats with chronic kidney-disease(CKD)by regulating TGF-β signaling pathway.Methods A total of 100 male SD rats were randomly divided into five groups:normal group(NOR,n =20),model group(CKD,n =20),radix notogin-seng group(RN,n =20),radix notoginseng plus platycodi group (RNP) and radix notoginseng plus cinnamon group (RNC,n =20).Except for the NOR group,the CKD rat model in other groups was established by adenine gavage.After modeling,the NOR group and CKD group were given the same volume of normal saline by gavage,while the group RN,RNP and RNC were given corresponding drugs by gavage,for 4 weeks.After 4 weeks,the rats in each group were sacrificed for collecting serum and detecting the renal function(serum Scr,BUN),the renal tissues were taken for conducting HE and Masson staining.Then the renal tissue pathological damage severity was observed.The expressions of FN and LN in kidney tissue were detected by immunohistochemistry and the expressions of TGF-[β,α-SMA were detected by Western blot method.Results Compared with the NOR group,the model group exhibited the renal dysfunction(P<0.01),renal interstitial severe fibrosis manifestation and increased collagen deposition(P<0.05),and the expression of kidney tissues α-SMA(P<0.01),TGF-β(P<0.01),FN and LN were significantly increased.Compared with the model group,the renal function in various treatment groups was improved,Scr(P<0.01)and BUN(P<0.01)were significantly decreased,the renal interstitial fibrosis degree was reduced,collagen desposition was decreased(P<0.05),renal tissue α-SMA(P< 0.05),TGF-β(P<0.05),FN and LN expression were reduced to some extent,in which the effect of RNC group was stronger than that of the RN group and RNP group.Conclusion Notoginseng and guiding medicinals mediated notoginseng can retard the progression of renal interstitial fibrosis caused by adenine in CKD rat in varying degrees,its mechanism maybe reduce the expression of TGF-β protein.
4.Endoscopic mucosal resection for lesions at gastroesophageal junction

Objective To evaluate the long-term efficacy and safety of endoscopic mucosal resection (EMR) for lesions at gastroesophageal junction (GEJ). Methods A total of 51 lesions located at GEJ from 51 patients, with an average size of 10. 9±4. 0 mm (3-28 mm), were treated with EMR between November 2005 and March 2009, among which 39 were diagnosed as low grade dysplasia and 12 as high grade dysplasia pathologically after EMR. The histopathologic results were compared between biopsies and EMR samples. All patients were followed up endoscopically. Results Complete resection was achieved in 44 ( 86. 3% ). The histopathologic diagnosis of 25 cases (49. 9% ) was inconsistent between biopsy and EMR samples, with 24 exhibited higher grade dysplasia after EMR. Main complication during EMR was bleeding in 17 patients ( 33. 3% ), all of which were successfully managed. No perforation or postoperative stenesis occurred. Endoscopic follow-up was carried out in 8 patients for more than 3 years, in 17 for 2-3 years, in 12 for 1-2 years and in 14 for less than 1 year. No death occurred during follow-up. Conclusion EMR can be adquately adopted as an effective treatment for pre-cuncerous lesions at GEJ, which is important in blocking malignant progression of dysplastic lesions.
5.Application research and design strategy on smart responsive mesoporous silica anti-tumor nanodelivery systems

Malignant tumors are major diseases that endanger human health. Due to their complex and variable microenvironment, most anti-tumor drugs cannot precisely reach the focal tissue and be released in a controlled manner. Intelligent responsive nano carriers have become a hot spot in the field of anti-tumor drug delivery systems. As an excellent nano material, mesoporous silica has the advantages of non-toxic, stable, adjustable pore volume and pore diameter, and easy functional modification on the surface. By virtue of its perceptive response to the tumor microenvironment or physiological changes, it can achieve the targeted drug release or controlled drug release of the drug delivery system in the tissue, making it an ideal carrier for intelligent response drug delivery system. In this paper, we review the design strategies and current research status of smart responsive anti-tumor drug delivery systems based on mesoporous silica, in order to provide a reference for the development of anti-tumor drug nanoformulations.
6.Progress of cell-free fetal RNA in maternal plasma in prediction of preeclampsia

Due to the advantages of being unaffected by fetal gender, ease of detection, and good stability, circulating cell-free fetal RNA (cffRNA) is a potential biomarker in obstetric practice. Current evidence has shown that placenta is the main source of circulating cffRNA. In view of the abnormal expression levels in women with preeclampsia and intrauterine growth restriction, circulating cffRNA is proposed as a potential tool to predict or diagnose these diseases. A summary of the molecular characteristics and the applications in preeclampsia of circulating cffRNA is reviewed, in order to evaluate the hypothesis for the prediction of preeclampsia by cffRNA.
7.Uncultured chorionic villus samples for fetal chromosome karyotype analysis: a retrospective analysis of 438 cases

Objective:To understand the possible detected mosaicism chromosome karyotyping using uncultured chorionic villus samples.Methods:This study retrospectively analyzed the clinical data of singleton pregnant women who underwent fetal chromosome karyotyping of uncultured chorionic villus samples at the Prenatal Diagnosis Center of Tianjin Central Hospital of Gynecology and Obstetrics from January 2016 to January 2019. Prenatal diagnosis indicators, fetal karyotypes, the incidence of chromosomal mosaicism and subsequent diagnosis, and perinatal outcomes were analyzed. Amniocentesis was performed when chromosomal mosaicism was identified. Descriptive statistical analysis was used for data analysis.Results:(1) A total of 438 pregnant women with available follow-up data were enrolled. Increased nuchal translucency (56.6%, 248/438) was the major indication for prenatal diagnosis. The karyotype analysis indicated that 79.5% (348/438) were normal, and 2.7% (12/428) were mosaicism. (2) Of the 438 cases, 336 cases (76.7%) were delivered at term, of which 327 cases were uncomplicated. There was one case of premature rupture of membranes within one week after amniocentesis and eight cases of abortion/fetal death between one week after the amniocentesis and 28 weeks of gestation. Of these nine cases, four had chromosomal abnormalities, and five had normal karyotypes. Termination of pregnancy was selected in 65 cases (14.8%) and 28 cases (6.4%) delivered before term. (3) Among the 12 (2.7%) cases of chromosomal mosaicism verified by fetal karyotyping through amniocentesis, four were confined placental mosaicism; six were abnormal chromosomal karyotypes in chorionic villous and amniotic fluid; one was true fetal mosaicism; one was a false positive. Among the 12 cases, three continued to term, one was preterm delivered, and eight selected labor induction, including three cases each of trisomy-21 and ultrasonographic structure abnormalities, and one case each of fetal growth restriction and labor induction based on patient preference.Conclusions:Karyotype analysis of uncultured chorionic villus samples may detect a certain proportion of mosaicism. Therefore, combining fluorescence in situ hybridization to achieve an accurate diagnosis and a detailed and systematic ultrasonic scan are recommended.

10.Protection of hypothermic preserved isolated rat hearts by resveratrol and its underlying mechanism.

OBJECTIVETo investigate whether resveratrol (RES) plays a protective role in hypothermic preserved isolated rat hearts and whether it is mediated by regulation of silent information regulator protein-1 (Sirt-1) expression.
METHODSThe Langendorff model of isolated rat heart was used. After stored in different Celsior solution at 4 degrees C for 9 h, SD rat hearts were randomly divided into 7 groups: blank control group;9 h group (soley hypothermic preservation for 9 h); RES group (3, 10, 30 micromol/L RES treatment plus hypothermic preservation for 9 h ), niacinamide (NAM) group (40 micromol/L NAM added in Celsior solution plus hypothermic preservation for 9 h), RES + NAM group (30 micromol/L RES and 40 micromol/L NAM were added in Celsior solution plus hypothermic preservation for 9 h). The morphological changes of cardiomyocytes were detected by the HE staining with the light microscope. The mRNA and protein expression levels of Sirt-1 were detected by Real-Time PCR and Western blot respectively.
RESULTS(1) Compared with the blank control group, myocardiocytes were injured remarkably in the 9 h group and the Sirt-1 mRNA and protein expression levels were decreased significantly (P < 0.01); (2) Compared with the 9 h group, rat myocardial injury was alleviated gradually in 3, 10, 30 micromol/L RES group and the Sirt-1 mRNA and protein expression levels were increased in a dose-dependent manner (P < 0.05); (3) The above protective effects of RES were attenuated by Sirt-1 inhibitor NAM.
CONCLUSIONRES can protect myocardiocytes from injury caused by long range hypothermic preservation and this protective effect maybe mediated by upregulation of Sirt-1 expression.
