2.The role of dendritic cells mediated antitumor immunity in esophageal carcinoma

Purpose:To study the role of DCs in tumor infiltrating T lymphocytes activation of esophageal carcinomas.Methods:46 specimens of patients with esophageal carcinoma were used to investigate the DCs (CD1a), the subsets of tumor infiltrating T lymphoctyes (CD3,CD4,CD8, CD25) and costimulatory B7 using monoclonal antibodies by immunohistochemistry staining LSAB method and reverse transcription polymerase chain reaction (RT PCR). Results:There was significant correlation between CD1a(+) DCs and CD3(+),CD4(+) T lymphocytes, but no marked correlation between CD1a(+) DCs and CD8(+), CD25(+) T lymphocytes. No specific B7 mRNA expression was found in all specimens. Conclusions:DCs may not activate T lymphocytes efficiently. B7 gene was expressed at low level or deficiency in esophageal carcinoma may be one of the important reasons of failure to activate lymphocytes and induce the immunosuppression.
3.Study on the in vitro Transdermal Absorption Properties of Galangin

OBJECTIVE:To investigate the in vitro transdermal absorption properties of galangin and effects of different pene-tration enhancers on its transdermal behaviors,and provide reference for developing skin preparations using galangin as APIs in the treatment of vitiligo. METHODS:HPLC was used to determine the galangin content. Using cumulative permeation rate (Q) and the transdermal rate(J)of galangin as indexes,the effect of absorption of receiving solution [20%,40% polyethylene glycol 400 (PEG400)solution and 30% ethanol solution] and rotating rate(200,300,400 r/min)on galangin in complete skin of mice were investigated,as well as the azone(1%,3%,5%)and propylene glycol(10%,20%,40%)alone or combination on its penetra-tion promotion. And the transdermal properties of galangin in complete skin,exfoliating skin,dermis skin of rats and mice were de-tected. RESULTS:The best permeability of complete skin of mice showed in 40% PEG400 solution at rotating speed of 300 r/min with 5% azone alone,J was 3.2570 μg/(cm2·h). Js of complete skin,exfoliating skin,dermis skin of mice were 2.7199,34.016, 33.874 μg/(cm2·h),respectively;and those of rats were 0.4996,9.5124,17.406 μg/(cm2·h). CONCLUSIONS:Galangin can penetrate the complete skin of mice and rats,however,the penetration quantity is far lower than exfoliating skin and dermis skin.
4.Study on the correlation between serum lipid level and anti-Müllerian hormone with embryo quality in vitro fertilization cycle

Objective To study the relationship between serum lipid level with embryo quality in vitro fertilization cycle.Methods A total of 603 pairs of infertile couples who were treated with in vitro fertilization and embryo transfer technique (IVF-ET) in our hospital from October 2015 to June 2016 were collected.All subjects underwent long-term ovarian hyperstimulation,blood lipid and the embryo quality were measured and evaluated.Results Triglyceride (TG),total cholesterol (TC) and body mass index(BMI) were significantly negatively correlated with fertilization rate,2PN fertilization rate,cleavage rate and embryo rate (P<0.05).Density lipoprotein (LDL) and non-high density lipoprotein (NHDL) and lipoprotein (a) [Lp(a)] were negatively correlated with fertilization rate,2PN fertilization rate,high density lipoprotein.The cholesterol (HDL) were positively correlated with fertilization rate,2PN fertilization rate,cleavage rate and embryo rate(P>0.05).Conclusion There is a linear correlation between blood lipid level and AMI and embryo quality.Serum lipids and AMI can be used to evaluate the quality of embryo and provide suggestion for the quality control of IVF-ET.
5.Application of functional neck dissection for lower collar-shaped incision at hair margin behind ear in the treat-ment of differentiated thyroid cancer

Objective To explore role of functional neck dissection for lower collar-shaped incision at hair margin behind ear in the treatment of differentiated thyroid cancer. Methods The clinical data of 11 patients with thyroid papillary carcinoma who underwent func-tional neck dissection for lower collar-shaped incision at hair margin behind ear were retrospectively analyzed. Results The incisions of all patients were unseen in daily life and patients were satisfied with the postoperative appearances. No case of shoulder syndrome,postoperative local recurrence and distant metastasis was observed. Conclusion Functional neck dissection for lower collar-shaped incision at hair margin behind ear is effective in the treatment of differentiated thyroid cancer. Meanwhile it can obviously improve patients’ postoperative appearance and life quality,therefore it is worth clinical application.
6.Three-dimensional perfusion imaging in acute cerebral infarction

Objective To assess the value of whole cerebral perfusion weighted map (PWM) and whole cerebral perfusion blood volume (PBV) integrating in scan protocol about CT perfusion (CTP) combined with CT angiography (CTA) in acute cerebral infarction. Methods Twenty-three patients with acute cerebral infarction proved clinically underwent CTP examination combined with CTA. The color-coded images of PWM and PBV were attained using workstation, and the raw data of contract CTA images and subtractive images between contract CTA and non-contract CTA were processed. The diagnostic sensitivity and the value of CTP and PWM, PBV integrating CTP in acute cerebral infraction were evaluated. Results Seven of 9 patients with negative results on CTP images had positive expressions on PWM and PBV images. The sensitivity of CTP was 60.87% and the sensitivity of PWM and PBV integrating CTP was 91.30%. Conclusion The scan protocol of PWM, PBV integrating CTP not only increases detection rate of acute cerebral infraction, but also has ability to predict the clinical prognosis of patients with cerebral infraction.
7.Risk factors for recurrence of ovarian endometriomas after surgical excision.

Ovarian endometrioma is a common form of endometriosis, which may cause infertility, dysmenorrhea and pelvic pain in women of reproductive age. Although surgery is the treatment of choice for endometriomas, recurrence poses a formidable frustration. This study investigated potential risk factors of endometriomas recurrence, aiming to better understand its pathogenesis. A total of 307 patients with endometriomas were followed up for an average of 28.6 months and the 1-, 2- and 3-year cumulative recurrence rate was 9.5%, 21.9%, and 29.2%, respectively. Twenty-one potential risk factors for endometriomas recurrence were evaluated using Cox's proportional hazards models. Total revised American Fertility Society (rAFS) score was significantly associated with higher recurrence (OR=1.858, 95% CI=1.122-3.075, P=0.016), as well as younger age at surgery (OR=0.953, 95% CI=0.915-0.992, P=0.020). Semiradical surgical treatment was defined as surgical removal of cyst plus hysterectomy with preservation of bilateral or unilateral ovary, and was a significant factor that was associated with lower recurrence than the conservative surgery (OR=0.318, 95% CI=0.107-0.951, P=0.040). Postoperative pregnancy was favorable factors for disease recurrence (OR=0.217, 95% CI=0.102-0.460, P=0.000). The results suggest that endometrioma recurrence is inversely associated with age at surgery and postoperative pregnancy, and may correlate with total rAFS score and conservative surgery method.
8.Combined pegylated liposomal doxorubicin and carboplatin in the treatment of recurrent epithelial ovarian cancer

Objective To evaluate the efficacy and toxicity of combined pegylated liposomaldoxorubicin (PLD) and carboplatin in the treatment of patients with recurrent epithelial ovarian cancer.Methods We retrospectively reviewed 67 patients with recurrent epithelial ovarian cancer or primaryperitoneal adenocarcinoma (8 eases) who were treated with combined PLD and earboplatin. The responserate, survival and toxicity were evaluated. The mean age for 67 patients was 52.1 (39-76) years. All ofthem received eytoreductive surgery followed by platinum-based chemotherapy either with paclitaxel orcyclophosphamide and doxorubicin after diagnosis. Combined PLD and carboplatin was used as first orsecond-line treatment or even after multiple lines of treatment after disease recurred. Patients were treatedwith PLD at 35-40 mg/m2combined with carboplatin at an area under curve ( AUC ) of 5 once every 4weeks. Results Forty-nine. Patients were evaluable for response. Twenty-three (47%) patients had acomplete response, 13 (27%) had a partial response, 3 (6%) had stable disease and 10 (20%) hadprogressive disease. The estimated median progression-free survival (PFS) was 8 months. The 1-year and 2-year survival rates were 73% and 55%, respectively. All of the 67 patients were evaluated for toxicity. Thetreatment was terminated in 2 patients due to allergic-like infusion reaction. Four patients who had acuteinfusion reaction with shortness of breath and tightness of chest did not terminate the treatment because nosuch reaction occurred when restarted the infusion. There were 2 patients with G2and 3 patients with G3hand-foot syndrome, 2 patients had G4stomatitis, and 8 patients had G3leukopeni,, No G4leukopenia orcardiotoxicity eceurred. Conclusion The combination of PLD and carboplatin is an active and wellotolerated regimen in the treatment of patients with recurrent epithelial ovarian cancer.
9.Behavioral defects induced by chronic social defeat stress are protected by momordica charantia polysaccharides via attenuation of JNK3/PI3K/AKT neuroinflammatory pathway

OBJECTIVE The aim of the present study was to evaluate the protective effects of momordica charantia polysaccharides(MCP)on depressive animal model induced by chronic social defeat stress(CSDS)and explore the underlying mechanisms.METHODS We established CSDS depressant mouse model and treated CSDS mice with MCP.Sucrose preference,forced swim test(FST)and social interaction test(SIT)were used to measure behaviors changes.We used ELISA,Q-PCR and western blot to test the levels of cytokines in the hippocampus. RESULTS The results showed that chronic administration of MCP(100,200 and 400 mg·kg-1)significantly prevented depressive-like behaviors in mice as assessed by social interaction (SIT), tail suspension (TST) and sucrose preference tests (SPT).It was showed that the elevation of proinflammatory cytokines(TNF-α,IL-6,and IL-β)concentra-tions,up-regulation of JNK3,c-Jun,and P-110β protein expressions in the hippocampus of CSDS model. Moreover,reduction activity of PI3K and phosphorylation level of protein kinase B(AKT)was also observed in the hippocampus of CSDS model.All above phenomenon were reversed after MCP intervened.Further-more,the protective effects of MCP on the CSDS mice were partly inhibited by the specific phosphati-dylinositol 3-kinase (PI3K)inhibitor, LY294002. CONCLUSION The protective effects of MCP against depressive-like effects in CSDS mice might reduce neuroinflammatory and involve in attenuation of JNK3/PI3K/AKT pathway in the hippocampus.
10.Clinical observation on improving senile postprandial hyperglycemia and insulin sensitivity with combination of acarbose and liuwei nengxiao capsule.

OBJECTIVETo observe the effect of combination therapy of acarbose and Liuwei Nengxiao capsule (LWNXC) in improving senile postprandial hyperglycemia and insulin sensitivity.
METHODSSeventy-four patients with simple postprandial hyperglycemia were divided into the control group and the treated group, 37 in each group, who were treated with acarbose alone and the combination therapy respectively for 1 month, and the changes on 2 hrs postprandial glucose (2 hPG), blood pressure (BP), blood lipid, body mass index (BMI), insulin functional index as well as the adverse reaction of acarbose in the two groups were observed.
RESULTSAfter treatment, the levels of 2 hPG, fasting blood glucose (FBG), BMI, total cholesterol and lowdensity lipoprotein-cholesterol were improved in the treated groups more significantly than those in the control group (P < 0.05 or P < 0.01). Insulin sensitive index (ISI) and insulin resistance were improved in the two group (P < 0.05 or P < 0.01), but the improvement in the treated group was more significant than that in the control group (P < 0.01). Moreover, the adverse reactions were less in the treated group than in the control group.
CONCLUSIONThe combination therapy of acarbose and LWNXC could not only improve the postprandial hyperglycemia, but also markedly increase the insulin sensitivity, and shows obvious improving effect on parameters of blood lipid, BP and BMI. The adverse reaction could be evidently reduced by combined use with LWNXC.
