1.Study on maternal serum total bile acids in intrahepatic cholestasis of pregnancy on neonatal lung injury

Objective To explore the correlation between maternal serum total bile acid (TBA) of pregnant women with intrahepatic cholestasis of pregnancy (ICP) and varying degrees of neonatal lung injury.Methods A total of 52 cases of pregnant women with ICP and their corresponding newborns were enrolled into the ICP group from March 2014 to December 2015 in the People's Hospital of Shapingba District.Other 52 cases of pregnant women received cesarean delivery whose gestational age and birth weight of newborns were close to the ICP group and their corresponding newborns were selected as the control group.The conditions of neonatal lung injury were recorded,and the correlations of maternal serum level of TBA and exposure time of high serum level of TBA to degrees of lung injury were analyzed as well.Results The incidence rate of neonatal lung injury in the ICP group (67.3 %) was higher than that in the control group (17.3 %),there was statistically significant difference (P<0.05).The degree of lung injury was positively correlated with maternal serum level of TBA and exposure time of high serum level of TBA (r=0.687,P=0.000;r=0.523,P=0.001).Conclusion The probability of neonatal lung injury of corresponding pregnant women with ICP is significantly increased,and the extent of lung injury is positively correlated with concentration of maternal serum TBA and exposure time of high serum level of TBA.
2.Technics and significance of preserving intercostobrachial nerve during axillary node clearance for breast cancer

Objective To investigate the significance of preserving the intercostobrachial nerve (ICBN) during axillary node clearance for breast cancer and share, some technical experiences of this procedure. Methods ICBN was preserved integally or partially in 78 patients of breast cancer (ICBN preserved group) and resected integrally (ICBN resected group) in 18 patients. Sensory disorders and motorial recoveries as well as tumor recurrence were compared between the two groups one week and one year after operation. Results Morbidity of sensory disorders in ICBN preserved group was less than that in ICBN resected group. Motorial recoveries were better in ICBN preserved patients without decreasing the number of axillary nodes resected and without increasing the recurrence of tumor. Conclusion ICBN should be preserved as far as possible during axillary node clearance for breast cancer.

4.TGIF antagonizes the apoptosis of gastric cancer cells induced by TGF-?

Objective To investigate the effect of TGIF on apoptosis of TGF-?-induced gastric canceer cell.Methods After TGIF was stably transfected into gastric cancer cell line BGC823,apoptosis was examined with flow cytometry,and the expressions of TGIF,caspase8 and caspase9 were analyzed with Western blot.Results The apoptosis rate of BGC823 cells with the treatment of TGF-? obviously increased,and was accompanied by activation of caspase9 but not caspase8.Moreover,compared with controls,pcDNA3.1-TGIF-transfected BGC823 cells significantly decreased the sensitivity of TGF-?-induced apoptosis.Conclusions TGF-? induced the apoptosis of gastric cancer cell BGC823 via caspase9 pathway;furthermore,TGIF could inhibit TGF-?-induced apoptosis.
5.Interleukin 8 is involved in the invasion and metastasis of CD133+hepatocellular carcinoma stem cells

BACKGROUND:Interleukin-8 is an important inflammatory chemokine that plays an important role in the regulation of tumor cel proliferation and angiogenesis.
OBJECTIVE:To investigate the effect of interleukin-8 on the invasion and metastasis of CD133+hepatocel ular carcinoma stem cel s.
METHODS:After isolation and culture of MHCC97-H cel lines, CD133+/CD133-MHCC97-H cel s were sorted using immunomagnetic beads. CD133 expression was detected using flow cytometry, and interleukin-8 level in supernatant was measured using ELISA method. Cloning efficiency, tumorigenic capacity, cel migration and invasion ability were detected through colony formation assay, tumorigenesis experiment in nude mice, and Transwel detection. Additional y, other cel s were neutralized using interleukin-8 neutralizing antibody. Measurement results were compared between cel s undergoing different treatments.
RESULTS AND CONCLUSION:The CD133 level, interleukin-8 level, cloning efficiency and cel membrane permeability of CD133+MHCC97-H cel s were significantly higher than those of CD133-MHCC97-H cel s (P<0.05). Transplantation of CD133+MHCC97-H cel s at 1×106/L and 1×107/L resulted in subcutaneous tumors in some mice, whereas no subcutaneous tumors appeared in mice undergoing transplantation of CD133-MHCC97-H cel s at the same concentrations. After interleukin-8 neutralizing antibody treatment, the CD133 level, interleukin-8 level, and cloning efficiency of CD133+/CD133-MHCC97-H cel s were significantly decreased (P<0.05), especial y in the CD133+MHCC97-H cel s (P<0.01);the migration and invasion ability and cel membrane permeability of CD133+MHCC97-H cel s were significantly reduced (P<0.05), but these changes were not obvious in CD133-MHCC97-H cel s (P>0.05). These results show that interleukin-8 could be specifical y involved in the invasion and metastasis of CD133+MHCC97-H cel s.
6.Tumor-killing effects of gastric cancer stem cells as antigens to stimulate dendritic cells combined with cytokine-induced killer cells

BACKGROUND:As the existence of tumor stem cel s, it is difficult to completely eliminate tumors in clinic.
OBJECTIVE:To explore the tumor-kil ing effect of gastric cancer stem cel s as antigen to stimulate dendritic cel s combined with cytokine-induced kil er cel s.
METHODS:Side population cel s from human gastric cancer cel lines were isolated, and tumor antigen was prepared by freeze thawing method. After coculture with dendritic cel s, dendritic cel s combined with cytokine-induced kil er cel s, gastric cancer cel antigen, and gastric cancer stem cel antigen, kil ing rates of gastric cancer cel s were detected using MTT assay. Expression rate of CD83, a mature dendritic cel surface marker, was also detected.
RESULTS AND CONCLUSION:The CD83 expression level and kil ing rate of gastric cancer cel s were both significantly lower in the gastric cancer stem cel antigen group than the other groups (both P<0.05). These results indicate that gastric cancer stem cel s as antigen to stimulate dendritic cel s combined with cytokine-induced kil er cel s can promote the proliferation of gastric cancer cel s and elevate the ability to kil ing gastric cancer cel s.
7.Molecular action mechanism of desoxyrhaponticin and serum albumin characterized by spectroscopy combined with molecular modelling.

OBJECTIVETo study the molecular action mechanism of active constituents desoxyrhaponticin (DES) and human serum albumin (HSA).
METHODUnder the simulated physiological condition, computer analog technology, fluorescent spectrometry and ultraviolet spectrum were combined to study the binding mechanism between drug and protein.
RESULTMolecular modeling was adopted to establish the binding model between DES and HSA, suggesting that the interaction force maintaining drug and protein is mainly the hydrophobic interaction with a hydrogen-bond interaction. The results from spectroscopy indicated that the interaction between DES and HSA is a dynamic binding process with a high intensity. The value of the binding distance (r) between DES and HSA was low, which demonstrate the occurrence of energy transfer. DES made an impact on HSA' structural domain microcell conformation, which resulted in hydrophobic changes in binding areas. According to the fluorescent phase diagram technical analysis, the changes in the DES-HSA reaction conformational pattern showed a "two-state" model. According to the obtained thermodynamic parameters for the DES-HSA interaction, the interactional force between DES and HSA was mainly a hydrophobic interaction. The fluorescence polarization proved that a non-covalent compound was generated during the interaction between DES and HSA.
CONCLUSIONThe spectrum experiment showed consistent results with the computer analog technology, which could provided certain reference for studies on the interaction between DES and HSA.

8.Apolipoprotein e4 allele is associated with the decrease of endothelium-dependent arterial dilation in female patients with type 2 diabetes mellitus

Objective To investigate the relationship between apolipoprotein (Apo) e4 allele and endothelium dependent arterial dilation in females with type 2 diabetes mellitus. Methods One hundred and one female type 2 diabetic patients without angiopathy, aged 40 71 years, and 95 healthy female individuals were selected. PCR/allele specific oligonucleotide probes were used to determine Apo e genotypes, and high resolution ultrasound was applied to measure brachial artery flow mediated endothelium dependent dilation and glyceryltrinitrate (GNT) mediated endothelium independent dilation. Objective:To explore the mechanisms of the inhalation of atomized 1.0% anti-IL-1βand TNF-αimmunoglobulin yolk ( IgY) on treating guinea pigs with allergic asthma induced by the inhalation of aerosolized ovalbumin ( OVA).Methods:Healthy guinea pigs were randomly divided into the normal controls ( group C ) , the allergic asthma model group ( group M )-treated by the inhalation of atomized ovalbumin ( OVA ) , the inhalation of atomized 1.0% anti-IL-1βand TNF-αimmunoglobulin yolk ( IgY ) treatment group (group Z1)-treated asthma model guinea pigs by the inhalation of atomized 1.0% anti-IL-1βand TNF-αIgY,and positive control the inhalation of atomized budesonide treatment group (group Z2)-treated asthma model guinea pigs by the inhalation of atomized budesonide.The blood was gotten by cardiac puncture and the bronchoalveolar lavage fluid ( BALF ) was collected by bron-choalveolar lavage at 2 h,4 h,8 h and 24 h after the last time atomization.The inflammatory cells in the peripheral blood ( PB) were counted by methylene blue and eosin staining.Cytokine concentrations of IL-1β,IL-4,IL-6,IL-8,IL-13,IL-16,TNF-α,TGF-β1 and IgE in the plasma and bronchoalveolar lavage fluid (BALF) were measured using an enzyme-linked immunosorbent assay (ELISA).Results:In PB,eosinophils was decreased from 2 h to 8 h in group Z1 compared to group M.In plasma,the levels of IL-1βat 4 h and 24 h,IL-16 at 2 h,4 h and 24 h,TGF-β1 from 4 h to 24 h and IgE at 24 h,as well as the levels of IL-1βand TNF-αfrom 2 h to 8 h,IL-4,IL-6,IL-8 and IL-13 from 4 h to 24 h,IL-16 at 8 h,and TGF-β1 and IgE from 4 h to 8 h,especially the level of IL-1βand TNF-αstarting at 2 h,in BALF were significantly reduced in group Z 1 compared to group M ( P<0.05 ).The levels of IL-1βand TNF-αwere positively cor-related with that of IL-4,IL-6,IL-8,IL-13,IL-16,TGF-β1 and IgE (P<0.05).Conclusion: The inhalation of aerosolized anti-IL-1βand TNF-αIgY effectively alleviates inflammatory responses in guinea pigs with allergic asthma induced by aerosolized OVA inhalation may be due to the significant decrease in the levels of various allergic inflammatory cytokines .
10.Effect of resveratrol on the oxidative stress and antioxidant levels of different skeletal muscle mitochondrial subpopulations and insulin sensitivity in rats fed with high-fat diet

ObjectiveTo observe the effect of resveratrol on the oxidative stress and antioxidant levels of different mitochondrial subpopulations in the skeletal muscle and insulin sensitivity of rats fed with high-fat diet.MethodsMale SD rats,aged 8 weeks,were divided into normal chow (NC) group,high-fat diet(HF) group,high-fat diet plus resveratrol ( HFR ) group.After intervention for 8 weeks,the impacts of resveratrol on oxidative stress levels and antioxidant enzymes activities in subsarcolemmal ( SS ) and intermyofibrillar ( IMF ) mitochondria from skeletal muscle as well as general and skeletal mascle insulin sensitivity were assessed.ResultsCompared with NC group, insulin sensitivity was significantly reduced while reactiveoxygenspecies (ROS)and malondialdehyde(MDA) levels in SS and IMF mitochondria increased in HF group ( all P<0.01 ).In addition,antioxidant enzyme activities were significantly decreased in SS mitochondrial and increased in IMF mitochondrial ( both P < 0.05 ).Compared with HF group,the insulin sensitivity in HFR group was significantly improved.Moreover,the activities of antioxidant enzymes in SS and IMF mitochondrial were increased,and the oxidative stress levels returned to normal ( P < 0.05 ).ConclusionResveratrol notably improves the oxidative stress of different skeletal muscle mitochondrial subpopulations and insulin resistance in rats fed with high-fat diet.
