1.ULTRASTRUCTURAL STUDIES ON PARIETAL CELLS OF MOUSE STOMACH USING CL~- LOCALIZATION AND POLYSACCHARIDES STAINING TECHNIQUES

Cl~- localization and periodic acid-silver methenamine techniques have been used for ultrastructural studies on parietal cells of mouse stomach. Cl~- is located only in the intracellular canaliculi (IC) and intercellular space (IS)of parietal cells. The tubulovesicular system (TS) shows various reactions in periodic acid-silver methenamine staining. The positive reaction was demonstrated in the TS which is located near the apical membranes. This indicates that acid secretion is accompanied by polysaccharides secretion in which the TS takes part, and the polysaccharides secretory activity shows its cyclicity which is characterized by the changes of amounts of secretion. The cycle of parietal cells do not synchronize.

Though implementing the quality management system of ISO9000 series standard in the hospital,it enables to make the nursing quality in a controlled state and make every service and operation of nursing staff have regulations to abide by.According to ISO9000 series standard,implementing the management of“procedureization”makes the nursing work closed to the need of patients and satisfies them by stipulating each workflow clearly.
5. Anti-thrombus activity of a novel anti-thrombus protein EPR-hirudin

OBJECTIVE: To evaluate the anti-thrombus activitity of EPR-hirudin (EH), a derivative of hirudin obtained by adding three amino acids at the nitrogen terminus. METHODS: The anti-thrombus activitity of EH was investigated by chromophore substrate method in vitro as well as by rat models of carotid arterial thrombosis and inferior vena cava thrombosis in vivo. RESULTS: The result of anti-thrombin experiment in vitro showed that EH did not produced anti-thrombin activity unless it was cleaved by blood coagulation factors. Then the anti-thrombus feature of EH was confirmed in vivo separately in the rat models of carotid arterial thrombosis and inferior vena cava thrombosis. The formation of arterial clot was inhibited and the weight of venous clot was decreased after administration of EH in a dose-independent manner. However, the blooding time, activated partial thromboplastin time (APTT) and thrombin time (TT) did not significantly increase in EH groups compared with HV and low molecular heparin. CONCLUSION: EPR-hirudin can inhibit the formation of arterial and venous clot and has low risk of blooding.
8.Advance in Rehabilitation of Upper Limb Function in Hemiplegic Patients after Stroke (review)

The incidence of upper limb dysfunction of stroke patients was high, and the recovery was difficult, which seriously impact-ed their activities of daily living and quality of life. This article reviewed the new techniques and methods for hemiplegic patients after stroke. The main techniques and methods includeed bilateral isokine-matic training, mirror neuron system, gymnastics rods training, music therapy, motor imagery, upper limb rehabilitation robot, virtual reality, constraint induced movement therapy, functional electrostimulation, electromyographic biofeedback therapy, transcranial magnetic stimulation, neurophyisological therapy and so on.
9.A viewpoint of environment concerned therapy through exploring the body constitute distribution of Shimian county

Objective To investigate the difference of TCM body constitution betweenpeople of the Shimian county and Chengdu city. Methods 200 sub-health patients in clinic of Shimian TCM Hospital were selected randomly. Another 200 sub-health patients in clinic of the Second Sichuan Province Hospital of TCM wererandomly selected. The two groups were identified with TCM body constitution based on the clinical symptoms, tongue, pulse condition, etc. Comparisons on body constitution were made between the two groups. Results In Chengdu, patients withbalance constitution and Qi deficiency constitution (respectively have the proportion of 33.0% and 20.0%)were significantly higher than thoseof Shimian County(the proportion of the two wererespectively 4.0% and 6.0%);while Shimian County have a significantly higher ratio of phlegm dampness and blood stasis constitution(43.5% and 13.0%)than those in Chengdu city(the proportion of the two wererespectively 8.0%and 4.0%), P<0.01. Conclusion Because of different regions, the population constitution ratio wasdifferent, so the disease tendency wasalso somewhat different. Therefore, physicians should give full consideration to the regional factors in diagnosing and treating patients.
10.Activity assay and preservation of S-homocysteine methyltransferase

Objective To construct a simple method for the measurement of activity of S-homocysteine methyltransferase (HMT),and explore the best processing condition for HMT and the preservation of HMT.Methods HMT was expressed in pro-karyotic system by using genetic engineering technology,then was purified by using affinity and Sephadex G1 5 chromatography. Sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE)was performed to identify the physicochemical and bio-logical properties of target protein.Based on the principle of 5 ,5′-disulfide-double (2- nitro benzoic acid)(DTNB)could react with sulfydryl compounds rapidly,the reduction of homocysteine was detect to evaluate the activity of enzyme,then the best processing conditions of HTM were determined.Activity of enzyme,preserved in preservation solution with or without glycerol and preserved under different temperatures,was detected.Activity remaining ratios were detected and compared between HMT preserved in pres-ervation solution with different protective agents of different concentration and preserved by cryodesiccation.Results The purity of recombined HMT was above 95%,with molecular weight of 36 000 and excellent catalytic activity,and the catalytic activity was 2 000 U/mg.The optimum condition for the detection of biological activity was using HEPES buffer of pH 7.4 at 37 ℃ and reac-tion for 25 min.Glycerol could significantly prolong the preserving time of HMT,and half activity of HMT could be remained for six months.The reservation rates of activity of HMT,preserved in preservation solution with mannitol and trehalose,were 104%and 100%,respectively.Conclusion HMT could be obtained through genetic engineering.A simple test method of HMT was es-tablished,and the best processing conditions and preserving methods of HMT were determined,which laid a foundation for clinical application.
